Abstract

The aim of this study is to investigate the expression of heat shock protein 70 (HSP70) on grouper infected by Nervous Necrosis Viral (NNV) with involvement of pigment-protein fraction (PPF) from Nannocloropsis oculata (N.oculata). Fish treatments were divided into three group, which are control, fish with PPF from N.oculata, and fish with PPF and NNV. After treatment for 23 d, fish were sacrificed, and their brain was harvested for protein analysis using SDS-Page. Fish blood was collected to examine the expression of HSP70 by hemagglutination and western blot analysis. The results showed that the expression of HSP70 was observed after treatment with PPF in NNV infected brain tissue and blood cell proved by SDS-Page and Western blot analysis. Thus, PPF is responsible for the expression of HSP70 in infected tissues, indicating that PPF appears to have a function as an inhibitor of proliferation of NNV.

Abstract

Myxobolus merupakan ektoparasit yang berbahaya dan dapat mengakibatkan kematian hingga 80%. Dalam praktek akuakultur, probiotik telah digunakan dalam beberapa tahun terakhir probiotik menjadi bagian integral dari praktik budaya untuk meningkatkan pertumbuhan dan ketahanan terhadap penyakit. Tujuan penelitian ini mengetahui pengaruh pemberian probiotik terhadap hematologi dan respon imun ikan koi yang terserang Myxobolus sp. yakni eritrosit dan leukosit serta ekspresi Nuclear Factor-kappa Beta  (NF-kB). Metode yang digunakan yaitu metode eksperimen. Dalam penelitian ini dibagi ke dalam 4 perlakuan yaitu, perlakuan (A) Kontrol, (B) ikan koi terinfeksi Myxobolus sp., (C) ikan koi terinfeksi Myxobolus sp. dengan pemberian probiotik dosis 0,55 ml dan (D) ikan koi terinfeksi Myxobolus sp. dengan pemberian probiotik dosis 1,1 ml. Metode pemberian perlakuan dengan menambahkan probiotik pada 30 liter air pada bak pemeliharaan. Kemudian dilakukan pengamatan eritrosit dan leukosit. Serta dilakukan Imunohistokimia menggunakan antibodi NF-kB pada jaringan insang. Berdasarkan hasil pengamatan eritrosit diperoleh nilai rata – rata pada perlakuan A sebesar 1.666.667 sel/mm3, perlakuan B sebesar 1.940.000 sel/mm3, perlakuan C sebesar 1.776.667 sel/mm3 dan perlakuan D sebesar 1.836.667 sel/mm3. Hasil pengamatan leukosit diperoleh nilai rata – rata pada perlakuan A sebesar 119.800 sel/mm3, perlakuan B sebesar 492.800 sel/mm3, perlakuan C sebesar 308.533 sel/mm3 dan perlakuan D sebesar 318.400 sel/mm3. Selanjutnya, berdasarkan hasil Imunohistokimia  didapatkan hasil pada perlakuan A nilai DAB sebesar 15,1%, perlakuan B nilai DAB sebesar 31,7%, perlakuan C nilai DAB sebesar 53,5% dan pada perlakuan D nilai DAB sebesar 47,5 %. Dari penelitian ini dapat disimpulkan bahwa pemberian probiotik berpengaruh terhadap eritrosit dan leukosit serta dapat meningkatkan ekspresi NF-KB sebagai respon imun pada ikan koi (Cyprinus carpio) yang terinfeksi Myxobolus sp. Dosis optimal pemberian probiotik yaitu pada dosis 0,55 ml. Myxobolus is a dangerous parasite that can kill up to 80% on koi carp pond.. Probiotic intake has been proven to change the composition of the microbiota, and therefore helps in the recovery of microbiota that are disrupted (by antibiotics or other risk factors) into favorable or normal compositions. The purpose of this study was to determine the effect of probiotic administration on hematology and the immune response of koi fish attacked by Myxobolus sp. namely erythrocytes and leukocytes and expression of Nuclear Factor-kappa Beta (NF-kB). The method used was an experimental by providing probiotics to fish that promoted Myxobolus sp. In this study it was divided into 4 treatments namely, (A) Control, (B) koi carp infected by Myxobolus sp., (C) koi carp infected by Myxobolus sp. with probiotic doses of 0.55 ml and (D) koi fish infected by Myxobolus sp. by administering a probiotic dose of 1.1 ml. The treatment methods were the fish immersed into 30 L of water that added by Probiotic. The hematological observation of koi fish,  erythrocytes and leukocytes, was observed and immunohistochemistry using NF-KB antibodies in the gill tissue which is the target of the entry of Myxobolus sp. on Koi fish. Based on the results of the treatment of erythrocytes obtained the value of A handling is 1,666,667 cells / mm3, handling B is 1,940,000 cells / mm3, handling C is 1,776,667 cells / mm3 and handling D is 1,836,667 cells / mm3. The results of leukocyte observations obtained an average value on treatment A of 119,800 cells / mm3, treatment B was 492,800 cells / mm3, treatment C was 308,533 cells / mm3 and treatment D was 318,400 cells / mm3. Furthermore, based on the results of Immunohistochemistry, the results of handling A DAB value of 15.1%, handling B DAB value of 31.7%, handling C DAB value of 53.5% and at the time of treatment D DAB value of 47.5%. From this study it can be concluded that the treatment of probiotics affects erythrocytes and leukocytes and can increase the expression of NF-KB as an immune response in koi fish (Cyprinus carpio) infected with Myxobolus sp. The optimal dose of probiotics is at a dose of 0.55 ml

Abstract

Seiring berkembangnya sektor budidaya ikan koi, banyak ditemukan permsalahan dalam prosesnya. Berbagai macam permasalahan tersebut yaitu penyakit, penurunan daya dukung air dan kendala pemasaran ditengah masa pandemi Covid 19 ini. Guna mengatasi permasalahan yang maka diperlukan penerapan teknologi untuk meningkatkan potensi budidaya ikan koi. Penerapan teknologi yang diterapkan harus mudah dipahami dan berkelanjutan sehingga efektif dan efisien dalam penerapannya. Tujuan Pengabdian Kepada Masyarakat Doktor Mengabdi ini adalah transfer paket teknologi Microbubble dan Internet of Things (IoT) pada kolam budidaya ikan koi untuk meningkatkan potensi kelompok masyarakat. Selain itu, juga dilakukan pelatihan dan pendampingan terkait penerapan paket Microbubble dan Internet of Things (IoT). Metode pelaksanaan kegiatan Pengabdian Kepada Masyarakat Doktor Mengabdi meliputi, memberikan pelatihan dan pendampingan mengenai penerapan ilmu pengetahuan dan teknologi (IPTEK) dengan menggunakan Microbubble berbasis Internet of Things (IoT) dalam mendukung budidaya ikan dengan sistem intensif dan mengatasi permasalahan fluktuasi kualitas air yang selama menjadi kendala. Teknologi Microbubble berbasis Internet of Things (IoT) yang diterapkan telah disesuaikan dengan kebutuhan kolam, tipe kolam serta memudahkan dalam pemeliharaannya. Pelaksanaan kegiatan diikuti oleh seluruh anggota tim pengabdian dan anggota mitra kegiatan. Berdasarkan hasil pelatihan dan pendampingan yang telah dilakukan terdapat peningkatan Sumber Daya Manusia, dilihat dari meningkatnya pengetahuan dan pemahaman anggota mitra dalam penerapan paket teknologi yang diberikan dan mendukung peningkatan produksi budidaya ikan koi yang berkelanjutan

Abstract

Cantang grouper (Epinephelus sp.) is a fish with a high economic value and has been widely cultivated. VNN disase, caused by Betanodavirus infection, can lead to mass mortality of cantang grouper. Precautions considered effective in preventing Viral nervous necrosis (VNN) disease include vaccination. This study aimed to determine the effect of administering recombinant Chlorella vulgaris protein nanovaccine based on chitosan hybrid nanoparticles in increasing the immune response of cantang grouper infected with VNN. The study consisted of five treatments: K+ (cantang grouper infected with VNN), K- (healthy cantang grouper), T1 (healthy cantang grouper given 33 µl of nanovaccine and then challenged with VNN), T2 (healthy cantang grouper given 66 µl of nanovaccine and then tested with a VNN challenge), and T3 (healthy cantang grouper given 112 µl of nanovaccine and then tested with a VNN challenge). The parameters observed were haematology, nanovaccine Relative percent survival (RPS), and Survival rate (SR) levels. The study results showed that administering the nanovaccine increased the immune response of the fish, as indicated by an increase in haematology response, RPS, and SR in cantang grouper compared to the treatment without nanovaccine administration. Abstract

Viral Nervous Necrosis (VNN) and Vibriosis are contagious diseases that can affect grouper fish from the larval to adult stages and cause significant economic losses for farmers. The owner brought the sunuk grouper to BKIPM Mataram to determine the fish's health. Since the grouper arrived at the floating net cage, the owner reports it has sustained bodily injuries. The grouper is separated from the healthy fish and placed in the area where the sick fish are. During maintenance, the fish exhibits a decrease in appetite, wounds on the mouth, operculum, and body from the head to the tail, low mobility, and swimming upside-down. The physical examination revealed that the fish was swimming limply in an inverted position and had lesions on various body parts. According to virological and bacteriological laboratory tests, the sunuk grouper infected with Vibriosis and VNN can be affected by the condition of fish that have been sick since they first arrived. Also, stress, contact with infected fish, and a decreased appetite result in sunuk grouper contracting the disease. Due to a disease, a fish's immune system does not function optimally. This condition makes sunuk susceptible to several infectious diseases.

Abstract

The goldfish (Carassius auratus) has become one of the most sought-after ornamental fish today. The continuously rising market demand for goldfish as pets and decorative objects has driven the growth of the goldfish (Carassius auratus) farming industry. However, fish farmers still face difficulties in controlling the health of the fish and the quality of their water. The aquatic environment plays a crucial role in the growth of goldfish (Carassius auratus). Therefore, environmental management and fish health are essential in goldfish (Carassius auratus) farming. After the PKM Team analyzed the existing issues, the activities carried out to address these issues include the implementation of an Environmentally Friendly Recirculating Aquaculture System (RAS) in the goldfish (Carassius auratus) farming ponds. In this PKM program, water filtration will consist of mechanical filters, biological filters, chemical filters, and UV light. The results of the community service activities conducted by the PKM Team with their partners include increased knowledge, improved skills, increased production capacity, and increased profitability. Abstract

The Porong River is one of the rivers that faces high anthropogenic impacts, such as surfactant waste from detergents. The hematology of Javanese carp (Barbonymus gonionotus) serves as a sentinel organism that can be used as a biomarker for the waters of the Porong River. The aim of this study is to analyze the water quality status of the Porong River polluted with surfactants using the STORET method, as well as its impact on the hematology of Javanese carp using regression and correlation analysis. The research employed a descriptive survey method. The results of the study indicated that the Porong River was categorized as moderately polluted. The average highest surfactant concentration reached 7.2 mg/L, leading to an increase in TSS to 107.95 mg/L and a decrease in DO to 1.80 mg/L. Surfactants exhibited a negative correlation with erythrocytes (91.2%), hematocrit (47.7%), and hemoglobin (60.6%), indicating that the higher the surfactant concentration, the lower the number of erythrocytes, hematocrit levels, and hemoglobin levels in Javanese carp. Conversely, there was a positive correlation between surfactants and leukocytes (56.7%) and blood glucose (66.7%), suggesting that the higher the surfactant concentration, the higher the number of leukocytes and blood glucose levels in Javanese carp.

Abstract

Berdasarkan data BPS (Badan Pusat Statistik) pada tahun 2021, hasil produksi perikanan tangkap di perairan NTT (Nusa Tenggara Timur) mencapai 190.594 ton dan berpotensi naik setiap tahunnya, sehingga diperlukan pemetaan zona potensi penangkapan ikan di Selat Ombai. Metode yang digunakan adalah metode pembobotan untuk melihat area yang berpotensi dan tidak berpotensi menjadi zona penangkapan ikan. Waktu penelitian dilakukan selama bulan September hingga Oktober 2023 di Perairan Selat Ombai. Data yang digunakan adalah data sekunder Suhu Permukaan Laut (SPL) dan klorofil-a  dari citra Aqua-MODIS Level 3 dengan pendekatan pengindraan jauh dan sistem informasi geografi.Waktu penelitian dilakukan selama bulan September hingga Oktober 2023 di Perairan Selat Ombai. Perbedaan peta zona potensi penangkapan ikan dapat dilihat dari parameter suhu permukaan laut, klorofil-a, musim dan sumber data yang diperoleh. Persebaran area potensi tangkapan ikan terjadi di area pesisir pantai di Pulau Timor dan Pulau Flores. Selama tahun 2022, pada bulan Februari hingga Oktober 2022 merupakan waktu dengan zona potensi penangkapan ikan sangat kuat, sedangkan pada bulan Januari, November dan Desember merupakan waktu dengan zona potensi penangkapan ikan yang sangat lemah.   According to BPS (Central Statistics Agency) data in 2021, catch production in NTT (East Nusa Tenggara) waters reached 190.594 tons and is potentially rising every year, so it is necessary to map potential fishing zones in the Ombai Strait. The method used is a weighting method to see areas that have and do not have the potential to become fishing zones. The data used is secondary data on Sea Surface Temperature (SST) and chlorophyll-a from Aqua-MODIS Level 3 imagery using a remote sensing approach and geographic information system. The research was conducted from September to October 2023 in the waters of the Ombai Strait. Differences in fishing potential zone maps can be seen from parameters such as sea surface temperature, chlorophyll-a, seasons, and data sources obtained. The spread of potential fishing areas occurred in the coastal areas of Timor Island and Flores Island. During 2022, February to October 2022 was a time with a very strong fishing potential zone, while January, November, and December had very weak fishing potentials.

Abstract

Pemeliharaan ikan lele yang dilakukan pembudidaya di Pokdakan ROI Lele sebagian besar tidak melakukan upaya pengelolaan kualitas air seperti, filtrasi, sirkulasi maupun penggantian air (water exchange). Hal ini menyebabkan penurunan mutu air sebagai akibat dari penumpukan sisa pakan maupun feses ikan serta sisa hasil metabolisme lainnya. Lingkungan perairan merupakan suatu hal yang sangat berperan penting dalam pertumbuhan ikan lele (Clarias sp.). Oleh karena itu, perlu adanya manajemen lingkungan dan kesehatan pada pemeliharaan ikan lele (Clarias sp.). Keberhasilan suatu usaha budidaya sangat erat kaitannya dengan kondisi lingkungan yang optimum untuk kelangsungan hidup dan pertumbuhan ikan yang dipelihara. Setelah Tim PKM Mengidentifikasi pemecahan masalah yang ada, maka kegiatan pemecahan atau pengembangan yang dilakukan harus sesuai dengan kebutuhan yakni dengan menerapkan teknologi Recirculating Aquaculture System (RAS) pada kolam budidaya ikan di Pokdakan ROI Lele.  Dalam penerapan sistem RAS, penggunaan filtrasi air sangat menentukan keberhasilan sirkulasi air. Pada progam PKM ini akan menggunakan Filtrasi air terdiri dari filter mekanis, filter biologi dan filter kimia. Hasil dari kegiatan pengabdian masyarakat PKM terhadap mitra adalah Pengetahuannya meningkat, keterampilannya meningkat, kapasitas produksi meningkat dan keuntungannya meningkat.

Abstract

Water quality is an important factor in aquaculture activities, including goldfish (Carassius auratus) farming. One approach to improving water quality is the use of nanofilters. This study aims to evaluate the performance of nanofilters made from Sargassum sp. with the addition of copper oxide nanoparticles. The method used involves the fermentation of bacterial cellulose from Acetobacter xylinum using Sargassum sp. extract as a medium, followed by homogenization and the addition of CuO-NPs at concentrations of 0.5%, 1%, and 1.5%, and subsequent oven drying. The nanofilter membrane will then be analyzed using SEM, XRD, and FTIR to characterize its properties. Performance tests will assess the quality of water used in goldfish farming, including pH, TOM, TDS, and TSS after treatment with the nanofilter. Morphological results show a rougher and denser surface with dispersed CuO-NPs. Characterization reveals cellulose I with crystallinity values ranging from 86.80% to 90.20%, with functional group peaks at 3145, 2927, and 1735 cm⁻¹ indicating cellulose characteristics, and a peak at 424 cm⁻¹ indicating Cu-O bonds. Statistical analysis of performance tests on the water quality of goldfish farming in the F-test shows significant differences (p
