Abstract

Selektifitas alat tangkap menjadi permasalahan utama dalam menjaga kelestarian sumberdaya ikan, karena semua nelayan cenderung memperkecil ukuran mata jaring untuk menyikapi semakin sedikitnya sumberdaya ikan. Keterbatasan sumberdaya ikan dan meningkatnya biaya operasi penangkapan merupakan dilema yang harus dihadapi dengan melakukan operasi penangkapan ikan yang efektif dan selektif sesuai dengan prinsip kelestarian sumberdaya ikan. Penelitian bertujuan untuk mengetahui prosentase gaya extra bouyancy dan bukaan mata jaring purse seine sebagai indikator efektifitas dan keramahan terhadap lingkungan. Penghitungan efektifitas operasi penangkapan diawali dengan analisa teknis jaring purse seine seperti shortening, hanging ratio, bukaan mata jaring, luas jaring, gaya apung dan gaya tenggelam jaring.  Hasil penelitian menunjukkan bahwa kecepatan kapal di bawah 6 knot kurang efektif untuk menangkap ikan karena banyak ikan yang lolos dan beberapa kali mengalami kegagalan operasi penangkapan. Kecepatan efektif penangkapan sekitar7,1 knot dengan hasil tangkapan ikan1000 kg dan waktu operasi panangkapan sebesar 2.853 detik. Kecepatan operasi penangkapan di atas 8 knot sudah tidak efisien lagi karena terjadi lonjakan konsumsi bahan bakar yang cukup signifikan dan hasil tangkapan cenderung tetap. Efektifitas waktu operasi penangkapan dicapai pada setting yang ketiga dengan nilai sekitar 0,0476 dan hasil tangkapan sebesar 1300 kg.Kata Kunci: bouyancy, jaring, efektifitas, selektifitas

Abstract

The Jaguar cichlid, Parachromis managuensis (Günther, 1867), is native to Central America, with introduc­tions reported from West Java and Central Java Provinces of Indonesia. On 7-8 January 2019, sixteen specimens of P. managuensis were collected from Karangkates, the largest hydropower reservoir in East Java Province of Indonesia. A description of the morphological characters of specimens is provided.

Abstract

Ikan maru merupakan salah satu ikan hias karnivor yang bernilai tinggi, mempunyai dua varian warna yaitu jenis yellow maru dan red maru serta menampilkan corak yang beragam pada size yang berbeda. Dengan adanya pertimbangan demikian, tujuan dari penelitian ini ialah bagian tahapan yang harus dilaksanakan apabila pelaku budidaya ingin membuat langkah untuk domestikasi. Penelitian ini menyatakan bahwa tiap tingkatan suhu sangat mempengaruhi kadar glukosa darah ikan yang mengindikasikan bahwa ikan tersebut mengalami stress, biasanya stress pada ikan dapat diakibatkan perubahan lingkungan yaitu suhu atau akibat beberapa perlakuan misalnya akibat pengangkutan atau transportasi. Glukosa darah pasca stress memperlihatkan fluktuasi menurun, tinggi dan menurun kembali ini dapat dilihat pada suhu 29 ºC menghasilkan 115,2 mg/dL – 117,8 mg/dL dapat menurunkan glukosa darah. Kortisol juga dapat berdifusi keluar dari ikan, terutama melintasi insang, dengan difusi pasif bentuk bebas.

Abstract

Lempuk fish (Gobiopterus sp.) is one of the local species in Ranu Grati. The high level of Lempuk fishing and the low level of awareness of Grati community on the importance of conservation and domestication efforts, as well as the taxonomy of the fish itself have threatened the population. Therefore, long-term efforts should be undertaken to preserve and identify the origin of Lempuk fish by conducting a molecular phylogenetic study based on COI. This study aimed to determine, analyze and describe the kinship and phylogenetic relationships of Lempuk fish (Gobiopterus sp.). This study used a descriptive method, starting from the specimen collection, extraction by the Phenol-Chloroform method and amplification by the PCR method, to the screening process. The data analysis method used the MEGA 5 application. Phylogenetic study used the Maximum Likelihood method with a bootstrap value of 1.000 times. The results showed that the intra-population genetic distance of Lempuk fish is low at 0.00%. Genetic distance between the population of Lempuk fish is relatively high with G. brachypterus from Malaysia of 22.4%, Central Java of 21.5%, and India of 21.4% and with G. lacustris from Malaysia of 22.5%, the Philippines of 21.6% %, Thailand of 19.7%, and with G. chuno and G. semivestitus of 21.2% and 23.6% accordingly. The phylogenetic study obtained showed the formation of a separate group that is different between Lempuk fish group and other Gobiopterus fish, supported by a bootsrap value of 99%.

Abstract

The supplying of live fish broodstock is routinely delivered by water. However, the traditional transportation of live fish with water results in small volume of transportation. Therefore, waterless transportation with different filler medium of waterless live fish transport is considered an alternative strategy. This research aimed to evaluate the effect of different filler medium of waterless live fish transportation on survival rate of African catfish (Clarias sp.) broodstocks. Brooders with mean weight 116 ±10,25 g were packed at 5 fish/bag for simulated 15h transportation. The bags, each 40x25x15 cm3, were filled with different treatment of medium; Sponge, newspaper, cloth, and sawdust at 3 cm thickness of sponge medium and ½ kg for newspaper, cloth and sawdust medium per bag. One bag without medium was designed as a control. Mortality, temperature, Humidity and weight gain loss were monitored throughout the experiment. After simulation, mortality, pH, DO and temperature were monitored in fiber pond over 14 days. Effect of different treatment of medium was significantly different (p < 0.05) with highest survival (100%) at sponge and cloth medium, and lowest survival (24 %) at sawdust medium. Humidity of newspaper medium and weight gain loss of sawdust medium significantly decreased and no significantly different in temperature parameter after simulated 15h transportation. It was concluded that sponge and cloth medium might be applied as a filler medium for waterless live fish transportation.

Abstract

Study aims to analyze the genotoxic potential of glyphosate-based herbicide on common carp (Cyprinus carpio L) using Micronucleus and hematological assay. The concentration of Glyphosate-based herbicide in this study was 0 ppm, 1.35 ppm, 1.8 ppm, 2.4 ppm, 3.2 ppm, 4.2 ppm, 6.5 ppm, and 8.7 ppm. Administration of herbicide base on modification of published methods with 96 hours of exposure. This research result has shown that the average number of micronuclei was increased simultaneously with increasing the concentration of herbicide exposure. There are also other types of cell nucleus abnormalities, namely: blebbed, lobed, notched, and binuclear. In the treatment of 0 ppm of herbicide shown blebbed nuclei are 8 ‰, lobed nuclei are 6.6 ‰, notched nuclei are 10 ‰, binuclei is 4 ‰. From the research results, it can be concluded that the LC50-96 hours exposure of the herbicide isopropylamine glyphosate in carp (Cyprinus carpio L.) was obtained at a concentration of 8.57 ppm. Based on the evaluation on hematology, it was found that there was a decrease in the number of erythrocytes, hematocrit, and hemoglobin of fish blood, whereas the number of leukocytes, micronuclei, and other abnormal micronuclei showed an increase along with the increase in the dose of exposure to the herbicide isopropylamine glyphosate, which indicates a genotoxic effect.

Abstract

Red ginger (Zingiber officinale var. Rubrum) as one of the Indonesian spices has so many important roles, especially in the health sector as a medicinal plant, which has many active compounds including phenols, alkaloids, flavonoids, steroids and tannins. From some researches before, we can know that plants which grow in different places or locations have different tolerances generally. That statements can show that they have different content in their constituent metabolites definitely. Therefore, phytochemical analysis is needed to analyze the content of each red ginger (Z. officinale var. Rubrum) from three locations such as Red ginger (Z. officinale var. Rubrum) from Batu, Malang (RGB), Plaosan, Magetan (RGP) and Simalungun, Medan (RGS) to analyze each content its contains. Also, perform LC50 was needed for analyzing its effect on Zebra fish (Danio rerio) as a model fish. For this research, we use an experimental study with a Completely Randomized Factorial Design (CRFD) method that has two independent variable such as different dosage and type of red ginger (Z. officinale var. Rubrum). In this study, from five dosages (10 mg/l; 20 mg/l; 30 mg/l; 40 mg/l; 50 mg/l) and three types of red ginger (Z. officinale var. Rubrum) treatment, it found that the highest mortality and lowest survival rate were at the highest dosage (50 mg/l) in RGB and extreme increase or decrease in the curve is found in RGP treatment.

Abstract

Transgenik pada ikan lele dumbo (Clarias spp) dengan menggunakan metode elektroporasi pada sperma ikan sebagai media transfer gen masih belum pernah dilakukan di Indonesia, sehingga perlu dilakukan percobaan pemberian tegangan pada sperma ikan lele dumbo tersebut. Tujuan dilakukan penelitian ini adalah untuk mengetahui pengaruh stress kejutan listrik (V) terhadap (motilitas) sperma dan daya fertilisasi sperma ikan lele dumbo. Penelitian ini dilaksanakan di Laboratorium Breeding (Budidaya Perairan) Fakultas Perikanan dan Ilmu Kelautan serta di Laboratorium Sentral Ilmu Hayati (LSIH) Universitas Brawijaya Malang, Jawa Timur mulai bulan Januari sampai Juni 2009. Rancangan percobaan yang digunakan yaitu Rancangan Acak Lengkap (RAL) dengan 5 perlakuan 3 kali ulangan dan 1 kontrol. Sebagai perlakuan adalah perbedaan pemberian stress kejutan listrik dengan Gene pulser pada tegangan: A (40 V), B (160 V), C (280 V), D (400 V), E (520 V. Hasil penelitian menunjukkan bahwa pemberian stress kejutan listrik dengan tingkat voltase yang berbeda berpengaruh nyata terhadap motilitas sperma namun tidak berpengaruh nyata terhadap daya fertilisasi dan viabilitas sperma serta daya tetas telur. Berdasarkan analisis polynomial orthogonal didapatkan hubungan antara perlakuan tegangan dengan hasil motilitas sperma berbentuk linier dengan persamaan y=39,198-0,04x dengan R2=0,933 dan r=0,966. Semakin tinggi tegangan yang diberikan maka semakin rendah motilitas sperma yang dihasilkan. Dari hasil penelitian ini dapat disarankan bahwa elektroporasi pada sperma ikan lele dumbo (Clarias spp) sebagai media transfer gen sebaiknya dilakukan pada tegangan 40 Vcm-1 dengan satu kali kejutan dan lama kejutan 0,05 ms.Kata kunci: Clarias spp, sperma, elektroporasi, tegangan, transgenik

Abstract

Sperm cells can be used as a medium to insert foreign DNA into the egg. Electroporation humanized renilla reformis Green Fluorescent Protein (hrGFP) gene as a reporter gene is very important to know the effectiveness of gene transfer. Research is aimed to understand the effectiveness rate of hrGFP gene reporter to detect the successful gene transfer process based on the result of observation with convocal microscope over transgenic sperm level, embryo and larva of Carp Fish. Result of trans-genesis of hrGFP as sperm-mediated gene reporter must be accurately detected. Based on the result of observation with convocal microscope, it is shown that hrGFP enters the core level, indicates by PI pigmentation and thereby, hrGFP gene reporter can provide information about hrGFP fluorescence starting from sperm, embryo and larva of Carp Fish.Keywords: Carp Fish, effectiveness, gene reporter, hrGFP, sperm fluorescence.
