Abstract

Bacillus subtilis, a causative agent of foodborne illness in bread, cakes, cereals, and cheese leading to symptoms like diarrhea and nausea, was investigated for its role in nosocomial infections, including endocarditis, bacteremia, septicemia, and meningitis. The research focused on three genes: codY, narH, and ureC, which respectively contribute to bacterial survival through biofilm and spore formation, acid resistance, and adaptation to anaerobic conditions. The objective of this study was to evaluate the codY, narH, and ureC genes using the Gradient PCR method for Bacillus subtilis detection in bread and cheese through Real-Time PCR. During the research, bacterial growth was observed with an OD600 of 1.438 in Tryptic Soy Broth (TSB), and colonies of medium size, smooth, cream-coloured, and round shape were successfully isolated on Tryptic Soy Agar (TSA). The DNA template used had a 71.5 ng/µL concentration with an A260/280 ratio of approximately 1.830. The annealing temperature for Gradient PCR used was 53-62°C. The primers successfully amplified codY (175 bp), narH (222 bp), and ureC (153 bp) gene amplicons. The optimal annealing temperature for the primers used was 60°C, as indicated by the presence of a single bright band in electrophoresis. Using these three different genes, testing can also be conducted with the Multiplex PCR method. The next step involves developing a detection kit using optimized primers and annealing temperatures for the identification of Bacillus subtilis in bread and cheese samples using Real-Time PCR.

Abstract

Two xanthone derivatives, garciniaxanthone E (1) and 12b-Hydroxy-des-D-garcigerrin A (2) have been isolated from ethyl acetate extract of the tree bark of Garcinia dulcis. The Ultraviolet (UV), Infrared (IR), Nuclear Magnetic Resonance (NMR), and Mass Spectrometry (MS) data analysis elucidated the structure of the isolated compounds. This study represents the first evaluation of compounds 1 and 2 in terms of their efficacy against receptor tyrosine kinases. The results showed that compound 1 exhibited weak activity with 12% inhibition against Insulin Receptor (InsR), while compound 2 showed moderate activity with 29% inhibition against epidermal growth factor receptor (EGFR). A molecular docking study targeting EGFR-TK suggests that the hydroxyl group at C-4 on compound 2 can be demolished to raise the inhibitory activity in future research.

Abstract

This study aims to analyse the students’ mental model of students in SMA 42 Jakarta in the implementation of Learning Cycle 8E learning model on salt hydrolysis learning. This research was conducted in first semester of 2016/2017 academic year. There are 36 students of year XI in MIPA 5 was involved in the study. This qualitative research was conducted with the data collection of writing-drawing technique, in-depth interviews, classroom observation, student journal reflective. The Miles and Huberman data analysis was employed with quality standards of data reduction, display data, and verification. Class observation conducted before and after the learning Cycle 8E was implemented for understanding, the characteristics of students and classroom learning environment. The interview technique was conducted to have deep understanding the students’ mental model. The result showed that the students developed understanding on the concept of hydrolysis reaction process and pH of salt solution which were generated categories of understanding, not understanding, and misconception. The three categories are seen from the four stages of the learning Cycle 8E which are explore, elaborate, extend, and explain. students’ understanding and mental model development are influenced by the learning environment and prior knowledge. This model also has developed students’ soft skills of empathy communication, critical thinking and collaboration skills. Abstract

This study aims to produce a plastic pyrolysis learning resource in the form of books based on Education for Sustainable Development. The development of ESD-based learning resources is done by Borg & Gall (2006) research and development method which is then adjusted into three stages: needs analysis, development stage, and pilot phase. Learning resources produced by reading books on plastic waste issues and handling through a chemical approach, presented with a magazine display, using communicative language, and adapted to chemistry teaching materials class XI SMA of curriculum 2013. Percentage of test results feasibility by material and Language experts is 89.63%, while the results of feasibility tests by media experts amounted to 92.23%, with each reliability of 0.8 and 0.77. The results of ESD-based learning by small-scaled learners are 85% and large scale is 91.44%. Interpretation of assessments of learning resources by experts and learners as a whole has very good criteria for each assessment indicator. Therefore, it can be concluded that the ESD-based learning source developed has been feasible to be used as a reading book to support chemistry learning. Abstract

This study aims to determine the effect of REACT strategy on the chemical literacy of 10th grade students on learning redox reactions. The survey conducted in the even semester of the 2018/2019 academic year at one of public senior high school in Jakarta. The method used in this research was Quasi Experiment with Posttest Only Control Group Design. The sample used in this study was obtained through a purposive sampling technique to get two classes, X MIA 2 as an experimental class applied REACT strategy, and X MIA 1 as a control class applied active learning. The data was taken based on pretest and posttest score from using chemical literacy instruments. The results showed that the average posttest score of experimental class (67,324) was higher than the control class (54,030). Based on the calculation of the t-test obtained the value of count (3.827) and table (1,668) at the significance level of 5%. Therefore, the value of count > table, Then H0 is rejected and H1 is accepted. So, it can be concluded that the use of REACT strategy has a positive effect on student’s chemical literacy on redox reaction material. Keywords Chemical literacy, redox reactions, REACT strategy

Abstract

The interest of students in learning chemistry is one of the factors that increases the learning process that can be improved through Learning Cycle 5e learning and the use of mystery topics in learning. Therefore the purpose of this study is to analyze the interest of chemistry learning students through the application of Learning Cycle 5E using mystery stories to acid-base learning. This study was conducted at SMAN 61 Jakarta in the even semester of the 2018/2019 academic year with the research subjects consisting of 32 people of class XI MIPA 3. The study was conducted using qualitative analysis methods by collecting data through observation, reflective journals, student interviews and interest questionnaire. Learning Cycle 5E learning consists of 5 stages, namely engagement, exploration, explanation, elaboration and evaluation. Mystery Scenario used in the elaboration stage to expand students' understanding and make students interested in learning. The results of the analysis show situational interest, pleasure for the study of chemistry and self-efficacy in chemistry in chemistry formed during learning. Abstract

Zat aditif merupakan bahan yang ditambahkan ke dalam produk, khususnya makanan dan minuman, untuk meningkatkan kualitas, memperpanjang daya simpan, memperbaiki rasa, warna, tekstur, atau penampilannya, serta menjaga keamanan produk. Zat aditif umumnya digunakan dalam proses pengolahan makanan untuk meningkatkan mutu atau mencegah kerusakan pada produk. Kegiatan Pengabdian Masyarakat ini merupakan tanggung jawab serta peran aktif dosen melaksanakan salah satu kegiatan dari Tridarma Perguruan Tinggi untuk memberikan edukasi dan sosialisasi tentang bahaya zat aditif pada makanan serta identifikasi untuk menumbuhkan kesadaran masyarakat terhadap pentingnya memilih asupan makanan dalam tubuh. Metode pengabdian yang dilakukan adalah penyuluhan dan demonstrasi yang melibatkan 1 orang guru Labschool Cirendeu, 1 orang guru Labschool Kebayoran, 3 orang guru SD Labschool Cibubur, 10 orang guru SMA Labschool Cibubur, 5 orang guru SMP Labschool Cibubur, 6 orang mahasiswa Prodi Kimia UNJ sebagai khalayak sasaran dengan tujuan memberikan pemahaman kepada guru dalam mengetahui kandungan dari zat aditif makanan yang berbahaya. Hasil kegiatan PKM yaitu terjadi peningkatan pengetahuan dan keterampilan guru dalam menguji adanya zat aditif pada makanan dan minuman.

Abstract

Latar Belakang: Penurunan angka stunting di Indonesia dari 27,7% menjadi 21,6% terjadi di tahun 2022. Namun, nilai tersebut masih diatas ambang batas yang ditetapkan oleh WHO yaitu dibawah 20%. Oleh karena itu, perlu adanya kegiatan untuk meningkatan pemahaman, keterampilan, dan kreativitas kader Posyandu mengenai pencegahan stunting dalam pemenuhan gizi ibu hamil dan bayi melalui pembuatan yoghurt sederhana dan mudah, karena yoghurt menjadi salah satu makanan sumber protein tinggi. Metode: Pendampingan pembuatan yoghurt telah dilakukan oleh tim PkM KKN UNJ kepada kader Posyandu Cempaka, Kelurahan Larangan Selatan, Kecamatan Larangan, Kota Tangerang. Terdapat tiga tahap pelaksanaan yaitu (1) sosialisasi informasi; (2) pelaksanaan pedampingan; dan (3) evaluasi. Hasil: Semua peserta mitra sangat bersemangat dan memiliki antusiasme tinggi selama kegiatan berlangsung. Berdasarkan hasil angket umpan balik kegiatan, peserta mitra memperoleh wawasan dan pengetahuan baru (55,56%), serta informasi dan pengetahuan tentang yoghurt akan peserta mitra berikan dan sampaikan kepada masyarakat sekitar lingkungannya (88,89%). Kesimpulan: Kader Posyandu Cempaka Kelurahan Larangan Selatan Kota Tengerang memperoleh ilmu baru tentang pembuatan yoghurt sehat, mudah dan sederhana serta peserta mitra berkomitmen untuk berbagi ilmu kepada masyarakat sekitarnya dalam rangka pencegahan stunting.

Abstract

Yersinia enterocolitica is a pathogenic bacterium with the ability to survive and multiply in food in a low-temperature environment that can cause death in humans. In previous studies, the optimum annealing temperature of ymoA, ystA, and ail gene primers with amplicons of 185 bp, 123 bp, and 192 bp, respectively, was successfully found. This study aims to develop a pathogenic bacteria detection kit with confirmation, sensitivity, and specificity of myfA and ystA primers in detecting Yersinia enterocolitica bacteria quickly and accurately using the real-time Polymerase Chain Reaction method. The results showed that myfA and ystA primers have optimum annealing temperatures at 60°C with amplicon lengths of 181 bp and 123 bp, respectively. Primer myfA was able to amplify the target with real-time PCR at Ct 12.07±1 and Tm 81±1°C, while the ystA primer at Ct 12.38±1 and Tm 83±1°C. myfA and ystA primers were also able to distinguish target and non-target bacteria based on Ct or Tm. The designed primers successfully detected Yersinia enterocolitica bacteria with the smallest concentration of 0.000439 ng/µL equivalent to 7.024 × 102 CFU. The detection limit obtained is smaller than the contamination threshold set by the Food and Drug Administration (BPOM). Primer myfA and ystA Yersinia enterocolitica also successfully detected the target bacteria in cabbage and lettuce samples artificially. Based on these results, myfA and ystA primers successfully detected Yersinia enterocolitica in vegetable samples using real-time PCR quickly, sensitively, specifically, and accurately.

Abstract

Makanan adalah kebutuhan vital, dengan kriteria utama adalah keamanan, kualitas, dan nilai gizi. Untuk memastikan keamanan makanan, diperlukan metode deteksi yang cepat dan akurat, terutama untuk mendeteksi bakteri patogen penyebab keracunan makanan. Vibrio parahaemolyticus merupakan bakteri patogen yang banyak ditemukan pada makanan laut. Penelitian ini bertujuan untuk mengembangkan metode deteksi cepat V. parahaemolyticus dengan menargetkan gen toxR2 menggunakan Gradient Polymerase Chain Reaction. Gen toxR dipilih karena fungsinya sebagai pengatur penting gen virulensi. Tahapan yang dilakukan meliputi desain primer, penyiapan sampel bakteri dari biakan murni, dan uji amplifikasi menggunakan PCR Gradien. Hasil uji amplifikasi menunjukan bahwa pasangan primer toxR2 berhasil mengamplifikasi pada suhu 58-62°C dengan dihasilkan pita berukuran 137 bp. Berdasarkan hasil tersebut, dapat disimpulkan bahwa PCR Gradien dengan primer toxR2 dapat diaplikasikan untuk mengembangkan alat deteksi V. parahaemolyticus dengan dilakukan uji lanjutan seperti uji konfirmasi, uji spesifisitas, uji sensitivitas, dan uji pada pangan menggunakan Real-Time Polymerase Chain Reaction.
