Abstract

There are obstacles in the aquaculture business that is still often facedbyfarmers, one of the challengesisthe high price of commercial feed. Feed as an energy source for growth is a reasonably high-cost component in production that reaches 40-89 %. In the control of food for efficiency, it could be done by the fasting method. This research was aimedtoanalyze the effect of periodic fastedofgiant prawn (Macrobrachium rosenbergii de Man, 1879) against growth and feed efficiency. This research was conducted at IBAP Installation, Probolinggo District - East Java Province. The experimental method used completely randomized design with five treatments and three replications, such as; P0: feeding without fasting (as control), P1: a-day fasted, a-day feeding, P2: a-day fasted, two-days feeding, P3: a-day fasted, three-day feeding, P4: a-day fasted, four-days feeding, P5: a-day fasted, five-day feeding. The results showed that periodic fasted of M. rosenbergii no significant effect on the daily growth rate, absolute weight growth, and feed efficiency. However, by looking at the rate of growth, thisfasting methodcouldbe used as an alternative to saving feed. The P5 showed the most effective in increasing daily growth rate, absolute weight growth, and effectiveness of giant prawn feeding. The strategy of periodic fasted to increase the production of giant prawns should consider the other supporting factors such as ideal water quality (pH, temperature, and DO), proper stocking density, and feeding periods on schedule.

Abstract

Cantikgrouper is the hybridization results grouper or cross-breeding between Epinephelus fuscoguttatus as a female and Epinephelus microdon as a male. The main barriers faced in the development of this commodity is still low levels of spawning up to seeding grouper. Based on the background, this study aimed to investigate optimum temperature observations against the rate of embryonic development Epinephelus sp.larvae. This study used the results of artificial spawning eggs. The fertilized eggs were incubated on six pieces of the container temperature treatment;each treatment there was repeated three times. The incubation temperature was kept on (A) 21-22°C; (B) 23-24°C; (C) 25-26°C; (D) 27-28°C; (E) 29-30°C; (F) 31-32°C. Results showed that eggswere incubated at a temperature of 21-22 ℃ embryonic development to a halt in the blastula, and temperature 23-24°C stalled on phasemyomere embryos. The low-temperature incubation period lasts a long time. Temperature 25-26°C needed 18 hours 6 minutes by 8.33% abnormality rate. Temperature 27-28°C needed 16 hours to hatch witha degree of abnormality of 7.6%. Temperature 29-30°C needed 15 hours 1 minute for the hatch tothe degree of abnormality of 5.33%. The 31-32°C temperature needed 14 hours 6 minutes to hatch witha degree of abnormality of 17.3%. The limits of tolerance for the incubation of the eggs ofcantik grouper (Epinephelusspp.) were 26-32°C.The best temperature of each treatment were obtained at a temperature of 29-30°C. Based on our results, it concluded that the changing temperature affected how long eggs could hatch.

Abstract

Fish utilize energy from carbohydrate sources in small amounts because of the low secretion of the enzyme amylase for digestion. One alternative is to increase the availability of exogenous digestive enzymes by utilizing bacteria from the digestive tract that have amylolytic activity. This study aims to isolate amylolytic bacteria in the digestive tract of Red Snapper as a probiotic candidate. This study used several methods, namely amylolytic test on starch, test for resistance to acid conditions (pH 3), observation of bacterial growth for 30 hours, antagonistic test against pathogenic bacteria, test for attachment to the stomach, gram staining test, and pathogenicity test on shrimp. Screening of amylolytic bacteria in the digestive tract of red snapper produced five isolates, namely A1, A2, A3, A4, and A5. From the results of the amylolytic test, only three isolates were further tested, namely A1, A2, and A4. The three isolates were able to survive in acidic conditions of pH 3 and pH 7 for eight hours with density values (OD) in isolates A1 (0.630), A2 (0.597), and A4 (0.601). Isolates A1, A2, and A4 respectively produced antagonistic values with inhibition zones of 8.4 mm, 7.5 mm, and 10.3 mm. Next. the ability to attach bacteria to isolates A1 (6.6 x 102 cfu/ml), A2 (11.8 x 102 cfu/ml) and A4 (16.5 x 102 cfu/ml). After that, the pathogenicity test was carried out aimed at proving that the isolate was not pathogenic to the host. Based on the results of the study, there were three bacterial isolates (A1, A2, and A4) in the digestive tract of red snapper that had met the requirements for probiotic candidates in fish/shrimp and the best was A4 isolate.
