Abstract

Penelitian ini bertujuan untuk mengetahui perbedaan pertumbuhan dari beberapa genotipe bibit asam gelugur. Penelitian ini dilaksanakan di lahan Sentra Agribisnis Jalan A.H. Nasution No. 1 A Kecamatan Medan Johor Kelurahan Pangkalan Mansur Kota Madya Medan dan Laboratorium Fisiologi Tumbuhan Fakultas Matematika dan Ilmu Pengetahuan Alam Universitas Sumatera Utara Medan, dari bulan Mei sampai dengan Oktober 2016.  Parameter yang diamati adalah tinggi tanaman, diameter batang, jumlah daun, ukuran daun, bentuk daun, tepi daun, ujung daun, pangkal daun dan warna daun. Hasil penelitian menunjukkan bahwa  genotipe Malaysia, Deli Serdang, Langkat dan Kota Cane perbedaan nyata terhadap tinggi tanaman, jumlah daun, diameter batang dan ukuran daun serta memiliki kesamaan pada karakter bentuk daun (oblong), tepi daun (rata), ujung (acuminate) dan pangkal daun (cuneate), warna daun tua (hijau tua). Genotipe Malaysia memiliki rataan tertinggi pada tinggi tanaman (10,82 cm), diameter batang (3,04 mm), jumlah daun (10 helai), ukuran daun (panjang 78,70 mm dan lebar 38,30 mm). Abstract

The purpose of this research was to determinate the growth response of embryogenic callus from some varieties of soybean (Glycine max (L.) Merrill) on hypoxic and in vitro condition. The research was conducted at The Tissue Culture Laboratory of Agriculture Faculty in University of Sumatera Utara on April until December 2016. This research had two stages, induction of embryogenic callus and analysis metabolism of callus after hypoxic condition with t-test. This analysis used factorial Completely Randomized Design with two factors. The first factors were varieties of soybean and the second factors were combination of Plant Growth Regulator (PGR) (5 mg/l 2,4-D + 1 mg/l BAP, 10 mg/l 2,4-D + 1,5 mg/l BAP, and 15 mg/l 2,4-D + 2 mg/l BAP). The result showed the varieties, combination of PGR, and interaction of varieties and PGR give significant effect to weight callus. The result of T-test showed that in hypoxic condition, POD enzyme exercise on Gepak Kuning’s callus in 5 mg/l 2,4-D + 1 mg/l BAP was different before and after hypoxic condition. Abstract

Hasil penelitian menunjukkan bahwa dari kesembilan marka SSR mampu mendeteksi total alel sebanyak 57 alel dengan kisaran 4 - 10 alel per primer dengan nilai Polymorphic Information Content (PIC) diperoleh berkisar antara 0.55 - 0.82. Jumlah alel pada 4 berkisar antara 2.3 – 3.67 per populasi dengan nilai persentase lokus polimorfik diperoleh berkisar antara 36% - 100%, primer yang terbaik pada penelitian ini yaitu mEgCIR3555 (10 alel per primer) dengan nilai PIC sebesar 0.82 dan mEgCIR0782 (8 alel per primer) dengan nilai PIC sebesar 0.81. Analisis dendogram keempat populasi yaitu E. oleifera Suriname, E. oleifera Brazil dengan hibrida interspesifiknya (BC1 Suriname dan BC1 Brazil) terbagi ke dalam 3 group besar dengan jarak genetik berkisar antara  0.04 - 0.98, keragaman genetik yang dapat dijelaskan oleh marka SSR pada penelitian ini sesebar 57.29%.Kata kunci: E. oleifera, Hibrida Interspesifik, Kekerabatan Genetik, Keragaman Genetik,                      Simple Sequence Repeats.

Abstract

This reserach aimed to identify the morphological chracteristics and phylogenetic relationship of asam gelugur in three North Sumatera district. The research was done at Deli Serdang district, Langkat district and Serdang Berdagai district in Mei to September 2016 with survey method using asam gelugur descriptor of IPGRI. Sampling technique using accidental sampling. Parameters observed were tree height, trunk height, trunk circumference, trunk surface, crown shape, young leaf colour, mature leaf colour, leaf blade size (lenght and widht), leaf blade shape, leaf base shape, leaf apex shape, leaf blade margin, leaf venation appearance, flower size, position of flowers, sepal colour, petal colour, fruit shape, number of fruit segments, fruit diameters, fruit weight, fruit size, fruit skin thickness, mature fruit colour, seed lenght, seed widht, number of seeds per fruit, seed shape, seed coat colour. This result of research showed that in tree North Sumatera district there were 21 accession of asam gelugur been identified. The result of morphologycal characteristics for crown shape (pyramidal and elliptic), trunk surface (smooth, rough, very rough), leaf shape (elliptic, oblong, lanceolate), flower shape (female flower and hermaphrodite flower), fruit shape (round, flattened, ovoid).   Keywords : garcinia atroviridis, identification, morphologycal-characteristics

Abstract

The aim of the research was to determine the effect of cytokinin on the scion compatibility on the grafting union of Garcinia atroviridis. The research was conducted in Gelugur Jaya Nursery, Dusun II Bahorok, Langkat District from February 2017 to July 2017. The randomize block design (RBD) was used with 2 factors; the first factor was the rootstock type i.e; hermaphrodite rootstock and female rootstock; the second factor was the cytokinin (0 ppm, 2 ppm, 4 ppm). The result showed that the cytokinin significantly affected the number of leaves. The cytokinin with 4 ppm showed the highest number of leaves. The rootstock was not affected the whole parameters.

Abstract

This reserach aimed to identify the morphological chracteristics and phylogenetic relationship  of   G. atroviridis and a conservation material in three districts of North Sumatra The research was conducted at Asahan, Batubara and Serdang Berdagai district from Februari to mei 2017 by survey method. G. atroviridis samples founded in three districs were identified and  characterzed based on International Plant Genetic Resources Institute (IPGRI) standard. Sampling technique using accidental sampling. Parameters observed were fruit shape, number of fruit segments, fruit diameters, fruit weight, fruit size, fruit skin thickness, mature fruit colour. The result showed that there were 18 accession of G. atroviridis found in three districts. Morphologycal characteristic of G. atroviridis was  fruit shape (round, flattened, ovoid,). In fruit morphology showed that the 18 individuals analyzed were divided into 4 groups on a distance scale of 0,796 with a similarity rate of about 79-85.

Abstract

The research aimed to get The Parameter genetic of the generation  crossed F4 Soybean Detam-2  and Grobogan . The research was conducted at experimental field of college of Agriculture USU in March 2017 up to August 2017. The parameters observed were : the days of flowering, the time of harvested, the plant height, the number of primary branches,the number seeds  per plant, the weight of 100 seeds. The result showed that there were nineteen sample of genotype that have the character of time early ripening and high production Soybean plant F4 each row of the best result of crossed.

Abstract

The oil palm plant is a one-piece plant included in Palmae's family. Molecular marking applications can be used to improve efficiency in analyzing genetic relationship, gene mapping, and marker-assisted selection (MAS) in oil palm plants. This study aims to determine the visualization of the amplification of DNA of Origin of Oil Palm (Elaeis guineensis Jacq.) Germplasm PT. Socfindo Based on Three SSR (Simple Sequence Repeats) Primers. The material analyzed is the 14 origin of oil palm originating from the germplasm of PT. Socfindo. The results showed the size of DNA bands that varied from 138 bp to 308 bp. The lowest band size was found on the FR-0782 primers, namely 138-211 bp and the polymorphic percentage was 60%. Key words: oil palm, molecular markers, SSR markers
