Abstract

Bananas are one of the horticultural crops of the fruit group that have quite high social and economic value. One type of banana that is very popular is the pisang kepok. The main obstacle in pisang kepok  production is the availability of seedlings. The above constraints can be overcome by propagation of banana seedlings using tissue culture techniques by adding colchicine to the tissue culture media. Colchicine is often used to improve plant quality. This study aims to determine the effect of adding colchicine to the media on the growth and formation of pisang kepok abu plantlet. The study was conducted from November 2019 - February 2020 at the MTC Laboratory, PT Great Giant Pineapple PG 4, East Lampung. The concentration of colchicine added to the media was 0.1%. The growth and development of plantlets produced from media with colchicine treatment will be compared with plantlets from the control group. The tests were repeated 10 times. The results showed that the growth and formation of pisang kepok abu plantlets from the treatment of colchicine were lower than the growth and formation of the pisang kepok plantlets in the control group.

Abstract

The world community, including Indonesia much like bananas because it has a relatively low price and good nutritional content. Banana kepok is a type of banana favored by the public because it tastes good and sweet. However, the obstacle that is often faced in kepok bananas is the difficulty in providing quality banana seeds quickly. Therefore, alternative technology is needed through tissue culture techniques. The purpose of this study was to determine the effect of adding breech flower tuber extract in culture media on the formation of banana plantlets of yellow kepok cultivar and to obtain scientific information about the growth profile of yellow kepok plantlets due to the addition of breech flower tuber extract to culture media. This research was conducted at the MTC Laboratory, PT. Great Giant Pineapple PG 4 Labuhan Ratu, East Lampung from December 2019 - April 2020 using a Completely Randomized Design (CRD) with ten replications consisting of the addition of 10% breech flower tuber extract as the primary treatment, 0.1% pure colchicine as a positive control, and without addition as a negative control. The results showed that the addition of 10% breech flower tuber extract in the culture media caused the growth of yellow kepok banana plantlets to be slower than the addition of 0.1% colchicine. Then in the media, with the addition of 10% breech flower tuber extract, the number of shoots, shoot height, number of better root and leaf area than the media without expansion, but 0.1% colchicine still showed the best results.

Abstract

Hyperglycemia is an early symptom of diabetes caused by the pancreas not being able to produce enough insulin. This condition can disrupt blood glucose metabolism, thereby damaging organs in the body such as the pancreas. Hyperglycemia can be treated using traditional plant medicines. Songgo Langit leaves (Tridax procumbens L.) are reported to have secondary metabolite compounds that have potential as antidiabetic drugs. This study aims to determine the effect of administering Songgo Langit leaf extract on blood glucose levels and changes in the histopathological structure of the pancreas in hyperglycemic mice induced by alloxan. Using Complete Random Design, mice were divided into 5 groups of 5 mice each. Group K (-) was a negative control (not treated), group K (+) was a positive control (only induced by alloxan), group P1 was induced by alloxan and extract of 0.45 mg/g BW, group P2 was induced by alloxan and extract 0.9 mg /g BW, and group P3 was induced by alloxan and extract 1.8 mg/g BW for 14 days. Glucose level data were analyzed using ANOVA and post hoc test with LSD. Pancreatic histopathological structure data were analyzed using the Kruskal-Wallis test and the Mann-Whitney post hoc test. The results showed that the treatment of ethanol extract of Songgo Langit leaf at doses P1, P2, and P3 were significantly able to reduce blood glucose levels by 61.70%, 65.11%, and 74.79% and was able to repair damage to pancreatic histopathology of mice induced by alloxan.

Abstract

Salah satu upaya dalam peningkatan ketahanan pangan dan gizi keluarga yang dianjurkan pemerintah adalah pemanfaatan sumberdaya yang tersedia di lingkungan, melalui pemanfaatan lahan pekarangan. Strategi penataan lahan dan pemilihan komoditas (tanaman) yang tepat diperlukan dalam pemanfaatan lahan pekarangan yang sempit dan terbatas. Hasil survei kami, menunjukkan bahwa sebagian besar ibu-ibu di kota Bandar Lampung belum mendapat pengetahuan yang cukup tentang pemanfaatan pekarangan tersebut. Ibu-ibu belum pernah mendapat pengetahuan tentang model-model penatahan lahan pada pekarangan sempit perkotaan dan pemilihan jenis tanaman yang tepat. Oleh sebab itu, pada pengabdian ini dilakukan kegiatan penyuluhan yang bertujuan memberikan pengetahuan tentang berbagai model penataan lahan pada pekarangan sempit perkotaan dan strategi pemilihan tanaman yang tepat sebagai upaya untuk meningkatkan kesehatan dan kesejahteraan keluarga. Kegiatan penyuluhan dilakukan secara ceramah dan diskusi, serta demontrasi. Kegiatan ini diikuti oleh 23 peserta yang terdiri dari ibu-ibu anggota majlis Taklim An-Nisa lingkungan Swadaya 8 dan 9, Gunung Terang, Langkapura, Bandar Lampung. Evaluasi kegiatan dilakukan dengan pretest dan postes, serta observasi langsung pada peserta melalui lembar observasi. Hasil postes menunjukkan terdapat peningkatan rata-rata skor sebesar 15,2, meningkat 22,42% dibandingkan rata-rata pretes. Sedangkan berdasarkan observasi selama kegiatan berlangsung, menunjukkan bahwa peserta selalu memperhatikan penjelasan dan terlibat dalam diskusi.

Abstract

Colchicine can inhibit spindle formation so that doubled chromosomes fail to separate during anaphase cytologically to induce mutations that produce polyploid plants. Yellow kepok banana is a triploid banana. The supply of yellow kepok bananas is hampered by conventional cultivation methods and the limited availability of quality seeds. This study aims to determine the effect of adding colchicine compound to tissue culture media on the mitotic index, chromosome number, and chromosomal abnormalities in yellow kepok bananas and the formation of polyploid plantlets. This study was included in the experimental study by conducting mitotic preparation and observing morphology after the test sample was induced by colchicine. The results of the research that has been done show that the addition of the compound colchicine to the tissue culture medium of yellow kepok banana causes a decrease in the mitotic index, an increase in the number of chromosomes, the presence of chromosomal abnormalities, the formation of polyploid plantlets, and an increase in the number of roots, but there is a reduction in root length.

Abstract

Dengue fever is one of the viral diseases transmitted through arthropods and is still a significant health problem worldwide. Dengue virus is transmitted by Aedes aegypti mosquitoes from one human to another through blood sucking. These mosquitoes usually live and lay eggs in habitats close to stagnant water. The usual efforts to control Ae. aegypti are using natural and synthetic chemicals. Compounds to control Ae. aegypti eggs are called ovicides. One of the natural ingredients that is thought to contain natural chemicals that have the potential to be used as an ovicide is cherry leaves (Muntingia calabura L.). This study aims to determine the potential of the ethanol extract of cherry leaves (M. calabura L.) as an ovicide against Ae. aegypti. Five treatments and four replications were used. The results of this study indicate that the ethanol extract of cherry leaves (M. calabura L.) can potentially be an insecticide for Ae. aegypti at a concentration of 1% and an exposure time of 72 hours.

Abstract

Tujuan dari penelitian ini yaitu mengetahui ekstak etanol daun kenikir (Cosmos caudatus Kunth) terhadap penyembuhan luka sayat pada mencit (Mus musculus L.) dengan konsentrasi yang berbeda yaitu 10%, 15%, 20%, 25%. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL), dengan 6 perlakuan dan 4 kali pengulangan. Terdiri dari K+ dengan povidone iodine, K- dengan Aquades, P1 dengan ekstrak etanol daun kenikir 10%, P2 15%, P3 20% dan P4 25%. Pemberian perlakuan dilakukan pada pagi dan sore hari selama 14 hari. Hasil analisis yang digunakan yaitu uji homogenitas, ANOVA dan uji lanjut BNT pada taraf 5%. Hasil penelitian ini menunjukkan bahwa ekstrak etanol daun kenikir dapat mempercepat penyembuhan pada luka sayat yang dapat dilihat dari gambaran hasil pengamatan histopatologi kulit mencit.

Abstract

Muli banana peels contain flavonoids, saponins, and tannins that foster the process of wound healing. The purpose of this study was to determine the effect of ethanol extract in muli banana peels (Musa acuminata) on cut wounds healing process in mice (Mus musculus). The study design was the completely randomized design by using 24 male mice which were divided into 6 groups, namely (K+, K-, P1, P2, P3, and P4). Each treatment consisted of 4 mice. All groups of mice were treated with a 1 cm cut wound on the back. The K+ group as the positive control group were applied with povidone-iodine, the K- group as the negative control group were applied with distilled water, the P1 group was applied with a concentration of 10% extract, the P2 group with 20% extract, the P3 group with 30% extract, and the P4 group with 40% extract for 14 days. On the 15th day, histological slides were made by taking 1 sample from each group. The results showed that the ethanol extract of muli banana peels played a role in the cut wound healing process. Uji Efektivitas Ekstrak Etanol Kulit Buah Pisang Muli (Musa Acuminata) Terhadap Penyembuhan Luka Sayat Pada Mencit (Mus Musculus L.)Abstrak: Kulit buah pisang muli mengandung senyawa flavonoid, saponin dan tanin yang memiliki peranan dalam proses penyembuhan luka. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh pemberian ekstrak etanol kulit buah pisang muli (Musa acuminata) terhadap proses penyembuhan luka sayat pada mencit (Mus musculus). Rancangan penelitian yang digunakan adalah rancangan acak lengkap, menggunakan 24 ekor mencit jantan yang dibagi menjadi 6 kelompok yaitu (K+, K-, P1, P2, P3, P4) dengan masing- masing perlakuan terdiri dari 4 mencit. Semua kelompok mencit diberi perlakuan luka pada punggung sepanjang 1 cm. Kelompok K+ sebagai kontrol positif dioleskan povidone iodine, kelompok K- sebagai kontrol negatif dioleskan aquades, kelompok P1 dioleskan ekstrak konsentrasi 10%, kelompok P2 dioleskan ekstrak konsentrasi 20%, kelompok P3 dioleskan ekstrak konsentrasi 30%, dan kelompok P4 dioleskan ekstrak konsentrasi 40% selama 14 hari. Pembuatan preparat histologi dilakukan pada hari ke 15 dengan mengambil 1 sampel dari tiap kelompok. Hasil penelitian menunjukan bahwa ekstrak etanol kulit buah pisang muli berperan dalam proses penyembuhan luka sayat yang dilihat dari gambaran histologi lebih baik dibandingkan kelompok kontrol.

Abstract

This research aimed to determine the mitotic abnormalities, changes in the number of chromosomes, mitotic index, and the number and length of kepok abu banana plantlet roots due to administration of 0.1% colchicine in tissue culture media. This research was performed at the Botanical Laboratory of the Biology Department, Faculty of Mathematics and Natural Sciences, UNILA. This research compared the cytology by employing the Squash method between root tip cells of kepok abu banana plantlets after administering 0.1% colchicine and the control group. The researchers analyzed the data descriptively and presented them in tables and bar charts. The results showed that the administration of 0.1% colchicine resulted in mitotic abnormalities. It also increased the number of chromosomes in the controls 2n=3x and 2n=3x+4. The mitotic index decreased, and the average number of roots was lower. However, the size was longer compared to the control group.

Abstract

Colchicine is often used in plant breeding to induce mutations and produce polyploid plants with superior traits. Cytologically polylpoid plants have variations in shape, number of chromosomes and larger cell sizes. The use of colchicine in kepok batu bananas is expected to produce polyploid banana kepok batu plants. The purpose of this study was to determine the effect of colchicine compounds on mitotic abnormalities, changes in chromosome number, decreased mitotic index, and the number and length of roots in banana kepok batu plantlets formed on tissue culture media. The research was carried out at the Botanical Laboratory, Department of Biology, FMIPA Unila. This study compared cytology with the squash method between root tip cells of banana kepok batu plantlets after the addition of 0.1% colchicine with controls. Data were analyzed descriptively and displayed in the form of tables and bar charts. The results showed that administration of 0.1% colchicine resulted in mitotic abnormalities (mitotic abnormalities) and increased the number of chromosomes in the control 2n=3x or 33 and 0.1% colchicine treatment, namely 2n=3x+6 or 39. The mitotic index decreased, but averaged – the average number of roots was more and their size was longer than the control. From these results it can be concluded that the administration of 0.1% colchicine was able to induce polyploid cells in banana kepok batu plantlets.AbstrakKolkisin sering digunakan dalam pemuliaan tanaman untuk menginduksi mutasi dan menghasilkan tanaman poliploid dengan sifat unggul. Secara sitologi tanaman polilpoid memiliki variasi bentuk, jumlah kromosom serta ukuran selnya lebih besar. Penggunaan kolkisin pada pisang kepok batu diharapkan dapat menghasilkan tanaman pisang kepok batu poliploid. Tujuan penelitian ini untuk mengetahui pengaruh senyawa kolkisin terhadap abnormalitas mitosis, perubahan jumlah kromosom, penurunan indeks mitosis, serta jumlah dan panjang akar pada planlet pisang kepok batu yang terbentuk pada media media kultur jaringan. Pelaksanaan penelitian di Laboratorium Botani Jurusan Biologi FMIPA Unila. Penelitian ini membandingkan sitologi dengan metode squash antara sel ujung akar planlet pisang kepok batu setelah penambahan kolkisin 0,1% dengan kontrol. Data dianalisis secara deskriptif dan ditampilkan dalam bentuk tabel serta diagram batang. Hasil penelitian menunjukkan bahwa pemberian kolkisin 0,1% memunculkan kelainan mitosis (abnormalitas mitosis) serta meningkatkan jumlah kromosom pada kontrol 2n=3x atau 33 dan perlakuan kolkisin 0,1% yaitu 2n=3x+6 atau 39. Indeks mitosisnya menurun, namun rata – rata jumlah akar lebih banyak dan ukurannya lebih panjang dibandingkan dengan kontrol. Dari hasil tersebut dapat disimpulkan bahwa pemberian kolkisin 0,1% mampu menginduksi sel – sel poliploid pada planlet pisang kepok batu.
