Abstract

As many as 50 million people worldwide are currently experiencing a deficiency of vitamin A and about 250000-500000 malnourished children in developing countries become blind each year from vitamin A deficiency, with the highest prevalence in Southeast Asia and Africa which will result in visual impairment, and increased child mortality and pregnant women. Melon can be one of the solutions for higher productivity and fruit may contain carotene (pro-vitamin A) with varying levels. In the years 2009-2011 Genetics laboratory, faculty of Biology UGM has produced a new melon cultivar called Melodi Gama 1 (MG 1). MG 1 phenotype has a rounded character with rather thick rind and hard, green and fleshy with a smooth orange flesh color. The purpose of this research was to study and analyze the nutritional content of MG 1 compared to commercial cultivars from multilocation and multiseason cultivation. The methods were used vitamin A quantitative analysis by extraction and spectrophotometer, whereas vitamin C in quantitative analysis using the iodine titration method. The results obtained in the analysis of total carotene in the dry season for the MG 1 has the highest value compared with 4 other commercial cultivars, it was 706.61 μg/100gr for cultivation in Yogyakarta and 292.16 μg/100gr for cultivation in Magetan. In the rainy season total carotene MG 1 was 59.88 μg/100gr for cultivation in Yogyakarta, and 20.76 μg/100gr for cultivation in Magetan. The results of the quantitative analysis of vitamin C MG 1 in the dry season was 7.50 mg/100gr for cultivation in Yogyakarta and 16.50 mg/100gr for cultivation in Magetan, while the rainy season was 8.55 mg/100gr for cultivation in Yogyakarta and 6.75 mg/100gr for cultivation in Magetan. Based on these results it can be concluded that the content of Vitamin A and C from Melodi Gama 1 cultivar is higher in the dry season than in the rainy season.

Abstract

Strawberry is fruit commodities which originating from the member of Rosaceae and has been widely cultivated in several countries of the world including Indonesia. One of the strawberry cultivars which consumed and cultivate in Indonesia is the strawberry Festival (Fragaria x ananassa D. var. Festival), which was developed in the area of Banyuroto, Magelang. In order to improve the quality and quantity of strawberry plants, polyploidization using colchicine has been employed in previous studies. The objectives of this research were study the chromosome character which include number, size, shape, and karyotype of normal strawberries and treatment strawberries with colchicine (0.05% 36 hours of root and leaf induction, 0.05% 24 hours of root and leaf induction, 0.05% 36 hours of leaf induction, 0.01% 36 hours of root and leaf induction). The results of this research shown that chromosome number difference and degree of ploidy between Fragaria x ananassa var Chandler in America with Fragaria x ananassa var. Festival in Indonesia due to natural mutation. While Festival strawberry control and treatment colchicine have same number chromosome is 2n=4x=28M and there are a difference long sleeve and short sleeve chromosome that may affect the content of chromosome expression and phenotype character are produced. The analysis of flow cytometry showed that treatment strawberry with colchicine has two peaks with intensity fluorescence equal to strawberry control so that it can be interpreted that the ploidy number of strawberries treatment with colchicine is tetraploid.

Abstract

A random amplified polymorphic DNA (RAPD) marker linked to powdery mildew resistance gene (Pm-I) in melon PI 371795 was reported. However, the RAPD marker has problem in scoring. To detect powdery mildew resistance gene (Pm-I) in melon accurately, the RAPD marker was cloned and sequenced to design sequence characterized amplified region (SCAR) markers. SCAPMAR5 marker derived from pUBC411 primer yielded a single DNA band at 1061 bp. Segregation of SCAPMAR5 marker in bulk of F2 plants demonstrated that the marker was co-segregated with RAPD marker from which the SCAR marker was originated. Moreover, results of SCAR analysis in diverse melons showed SCAPMAR5 primers obtained a single 1061 bp linked to Pm-I in resistant melon PI 371795 and PMAR5. On the other hand, SCAPMAR5 failed to detect Pm-I in susceptible melons. Results of this study revealed that SCAR analysis not only confirmed melons that had been clearly scored for resistance to Pm-I evaluated by RAPD markers, but also clarified the ambiguous resistance results obtained by the RAPD markers.   Key words: Cucumis melo L., Pm-I, RAPD, SCAPMAR5

Abstract

The cultivation of strawberry needs of technological innovation to gain competitive advantage strawberry fruit quality and quantity. This is motivated because the decline in the production of strawberries in the area and quality of strawberry fruit is still low. Since 2012 until now, Laboratory of Genetics has identified and developed the character of phenotype and genotype of strawberry plants in Agro Banyuroto, Village Banyuroto Sawangan District of Magelang regency. Character genotypes studied through the identification of ploidy with cytogenetic approach and apply poliploidisasi technology that aims to double the number of sets of chromosomes in order to obtain a plant that has the character of superior phenotype compared with the control. The purpose of this activity was to awaken the capacity of communities that were able to develop and utilize natural resources to meet human needs while maintaining the environmental implementing action plans that lead to sustainable development in a sustainable manner. The method was executed in this activity was the cultivation of strawberries by exploiting natural resources Banyuroto village environment that was using bamboo, strawberry cultivation techniques with vertikultur systems. Moreover, the carrying capacity was also supported by the promotion of village tourism as central region Banyuroto strawberry cultivation of strawberries superior to tourists every week, promotion through Online system (in the network) by utilizing Information Technology and Computing and community empowerment with the diversification of food made from strawberries. The end result of this activity was obtained hallmarks of crops polyploidy were plant growth faster, the ability of the formation of stolons and nursery faster, leaf area and stem diameter was wider, the stem stout and roots were longer, the fruit produced was greater and a sweeter taste with more red fruit color. Innovation strawberry crop is expected to be followed by the cultivation of sustainable optimal as possible so that the results obtained are able to improve the welfare of farmers in Banyuroto strawberries.

Abstract

Obesity is one of major nutritional problems in the world. Obesity is very dangerous, especially when concentrated in the abdomen, because it is closely linked to various diseases such as diabetes, hypertension, heart disease, which can to causing death. This study aims to identify the nature and distribution of ghrelin on gastric endocrine cells in the obese rat (Rattus norvegicus) by using immunohistochemical techniques. The results will strengthen the understanding of the role and function of ghrelin as an alternative therapeutic target on obesity. The research used gastric tissues of ten obese and control rats which were stained with avidin-biotin-peroxidase complex method of immunohistochemistry. The results showed the existence of two types of ghrelin-producing cells (open and closed types) on the gastric mucosa of control rats, and only one type of ghrelin producing cells (open type) in obese rats. The intensity of ghrelin immunoreactive positive cells was detected weak in obese rats, but very strong in control rats. Ghrelin endocrine cells mainly distributed in the basal part of the gastric mucosa of the fundus parts, with a very small number in obese rats, but highly abundant in control rats. This study confirmed the decrease of the ghrelin synthesis and secretion in obese rat (Rattus norvegicus) at the cellular level. The decrease of ghrelin synthesis is characterized by a reduction on the number of ghrelin producing cells, the disappearance of the close type of ghrelin producing cells, and the low activity of protein synthesis in the ghrelin producing cells. Ghrelin endocrine cells distributed mainly in the basal part of the gastric mucosa, especially in the fundus parts.

Abstract

Lahan kritis berkapur, karst, memberikan cekaman abiotik pada tanaman karena hambatan hidrasi dan nutrisi. Asam absisat (ABA) adalah hormon yang diekspresikan tumbuhan pada kondisi cekaman abiotik. CmBG1 merupakan salah satu gen peregulasi hormon ABA pada melon yang akan terakumulasi saat tumbuhan mengalami cekaman. Tujuan penelitian ini adalah mendeskripsikan pengaruh lahan kritis karst terhadap ekspresi gen CmBG1 secara kualitatif dan kuantitatif melon hasil turunan kultivar TACAPA, yaitu kultivar PT dan AT yang ditanam pada medium tanah karst dari wilayah Agroekosistem II dan III Yogyakarta (Gunungsewu, Dlingo, maupun Sentolo). cDNA library diperoleh dari reverse transcription isolat RNA. cDNA diamplifi kasi dengan primer spesifi k, kemudian dispektrofotometri pada λ260 nm untuk mengetahui konsentrasi gen CmBG1. Ekspresi gen dianalisis dengan real time PCR dengan gen referensi Cm-actin. Uji kualitatif dilakukan dengan elektroforesis gel agarosa 1,5%. Hasil penelitian menunjukkan gen CmBG1 terdeteksi dengan ukuran ±1258 bp pada kultivar PT dan AT. Konsentrasi gen CmBG1 melalui spektrofotometri menunjukkan semua kultivar yang ditanam pada media kontrol memiliki konsentrasi yang lebih rendah bila dibandingkan media tanam dengan perlakuan lahan kritis baik Gunungsewu, Dlingo, maupun Sentolo. Hasil ini sama dengan uji ekspresi gen CmBG1 menggunakan analisis kuantitatif real time PCR. Karsts critical land gives abiotic stresses in plants because of hydration and nutrition disturbances. Abscisic acid (ABA) is a hormone that expressed in the plant in abiotic stress conditions. CmBG1 is one of the regulatory genes encoding hormone ABA in melon plants which is accumulated in stress condition. The purpose of this study was to describe the infl uence of karsts critical land on the genes expression of CmBG1 melon cultivars PT and AT qualitatively and quantitatively. The plants were grown in medium karst land of Agroecosystems II and III of Yogyakarta (Gunungsewu, Dlingo, and Sentolo). Total RNA was extracted from leaf tissue then Reversed Transcriptase (RT-PCR) to collect cDNA library. cDNA was amplifi ed using specifi c primer. Spectrophotometry was conducted in λ260 nm and electrophoresis run in 1.5% agarose gel. Control band and reference gene chosen in Real Time PCR was Cm-Actin. CmBGI band (± 1258 bp) was showed both on PT and AT. Cm-actin was showed band of DNA as ± 445 bp. CmBGI gene concentration in critical land medium treatment which is given greater stress on melons are higher than normal condition. This suggests that the CmBGI gene is expressed more in cultivar PT and AT melons when they are grown under stress condition. This result show similarly when using real time PCR.

Abstract

Blubuk Hamlet, Kulon Progo is a rural area with great potential in agriculture and ecotourism. However, the community faces challenges in maximizing this potential due to limited knowledge. This community service program aims to empower the local community through waste utilization to have economic value. The program uses various methods, including training, participatory mapping, and focus group discussions (FGDs). The main activities include maggot cultivation for fish feed production, biodiesel production from used cooking oil, eco-enzyme production from organic waste, and ecotourism planning. The results showed that 80% of training participants successfully understood maggot cultivation techniques, collectively produced 3 kg of maggots, and produced 1 kg of fish pellets in two months. Eco-enzyme production from household organic waste reduced waste by 40%, with 85% of participants using the product as a natural cleaner and fertilizer. Biodiesel production produced 2 liters of biodiesel from 5 liters of used cooking oil, which was tested and used as an alternative fuel. This program successfully introduced the concept of community-based ecotourism management and increased public understanding of the economic potential that can be developed from local natural resources.

Abstract

Sidomulyo Village, Pengasih, Kulon Progo Regency in Special Region of Yogyakarta is one of the agrotourism areas that produces a lot of high local food potential. This area is located in the right on the edge of the Menoreh hills with popular local fruit and vegetable cultivation production centers. In addition, local wisdom is still very strong which is felt in every behavior and activity of the people who like to gardening and growing crops in the fields. Some of the superior regional potentials food to be developed are cassava, clove wood, petai and vegetables. Sidomulyo has natural and local potential as well and most of the population opens activities by establishing Micro, Small and Medium Enterprises (MSME). However, this potential requires scientific and technological innovation because it was necessary to compete with production from other regions, lack of innovation and marketing regarding the quality and quality of the products produced. Therefore, there are required to carry out assistance, guidance and training on cooperative relations from farmers with academia, industry and government in order to improve the quality and quality of Indonesian crops through empowering the MSME community and digitizing products. The objectives of this program are to provide intensive and efficient community assistance and empowerment in stages and sustainably, optimizing business opportunities for the Sidomulyo community by utilizing local village potential, increasing marketing of local village products through digital transformation of MSME activities, building village independence through industrial partnership networks to strengthen the potential of a civilized and prosperous Agrotourism village. This program is able to provide information on business opportunities through utilizing local village potential, equipping village communities with digital information systems to market MSME results.

Abstract

Corn is one of the main agricultural products in Tobadak Village. The people in Tobadak Village mainly use the corn kernels to feed their farm animals and then burn or throw away the cob which can be a problem to the environment, especially its soil and air around it. If we can correctly utilise and process its cob, it can be useful to the environment and reduce the pollution around it. One of the products that we can make using this corn cob is straw mushroom cultivation. It does not take long to cultivate this straw mushroom. It takes around 2 weeks to harvest and after that, we can enjoy the benefits. This activity consists of several methods, which are counselling, hands-on practice, society mentoring, monitoring and evaluation. This activity aims to reduce the agricultural waste and improve their knowledge and skill in processing this agricultural waste, especially corn cob to make and cultivate straw mushrooms.

Abstract

Sulobaja Village, located in Tobadak District, Mamuju Tengah Regency, faces significant challenges in managing household organic waste, which is often discarded indiscriminately or burned, leading to environmental pollution and reduced water quality. This community service program aimed to enhance public awareness and skills in organic waste management through the production of eco-enzyme, a liquid derived from fermenting organic waste that can be used as a natural cleaner, liquid fertilizer, and pesticide. The program included several stages: initial observation, socialization, hands-on practice, evaluation, and follow-up. Socialization was conducted through educational sessions at the Sulobaja Village Hall, followed by practical training on producing eco-enzyme using household organic waste, molasses, and water. After one month of fermentation, the resulting eco-enzyme exhibited a dark brown color and a distinct acidic aroma, indicating a successful fermentation process. Participants showed a high level of enthusiasm, with over 70% expressing their willingness to continue production independently. The program successfully reduced household organic waste, improved soil fertility, and decreased the use of chemical-based household products. Furthermore, it encouraged the establishment of small production groups to ensure program sustainability. The results demonstrate that eco-enzyme production can serve as a practical and sustainable solution for organic waste management while providing economic and environmental benefits to the community. This program is expected to be replicated in other villages as a model for environmentally friendly waste management.
