Abstract

ABSTRAK Jamur endofit dapat diartikan sebagai mikroba yang hidup berkoloni dalam jaringan internal makhluk hidup seperti pada daun tanpa menyebabkan efek yang merugikan secara langsung pada organisme tersebut. Jamur endofit mampu menghasilkan enzim ekstraseluler, salah satunya enzim protease. Penelitian ini bertujuan untuk mengidentifikasi jamur endofit penghasil enzim protease yang hidup pada daun mangrove Avicennia marina di Pantai Neolbaki. Metode yang digunakan adalah eksperimental dengan dua perlakuan yaitu daun tua dan daun muda mangrove A. marina. Data karakterisasi morfologi secara makroskopis dan mikroskopis dianalisis secara kualitatif sedangkan data uji aktifitas protease dari jamur endofit dengan metode plug agar berupa hasil perhitungan zona bening secara kuantitatif dan dideskripsikan dalam bentuk gambar dan tabel. Berdasarkan data penelitian ditemukan bahwa dari daun tua dan daun muda mangrove A. marina yang ada di pantai Noelbaki menghasilkan 12 total isolat hasil isolasi. Dari total 12 isolat diperoleh 5 isolat jamur endofit penghasil enzim protease ekstraseluler yang terbagi atas 3 isolat dari daun tua dan 2 isolat dari daun muda. Pembentukan zona aktifitas proteolitik tertinggi pada daun tua A. marina dalam uji enzim protease ekstraseluler terdapat pada kode isolat DT Am 05 OLL dengan diameter zona proteolitik sebesar 2,1 mm. sedangkan untuk daun muda A. marina aktivitas tertinggi terdapat pada kode isolat DM Am 02 OLL dengan diameter zona bening sebesar 1,9 mm. ABSTRACT Endophytic fungi can be defined as microbes that live in colonies in the internal tissues of living things such as leaves without causing direct adverse effects on these organisms. Endophytic fungi are able to produce enzymes, one of which is the protease enzymes. This study aims to identify Endophytic fungi that produce protease enzymes that live on mangrove leaves of Avicennia marina in Noelbaki Beach. The research used an experimental method with two treatments, namely old and young leaves of the A. marina mangrove. The macroscopic and microscopic morphological characterization data were analyzed qualitatively, while the protease activity test data of endophytic fungi using the plug agar and the clear zone were analyzed quantitatively, then described in the form of figures and tables. The founding of the research of the old and young leaves of A. marina from the Noelbaki beach had been isolated a total of 12 isolates of endophytic fungi. Of the 12 isolates, 5 isolates were shown to produce extracellular protease enzymes, consisting of 3 old leaves isolates and 2 young leaves isolates. In the extracellular protease enzyme test, the proteolytic activity zone was confirm as a clear zone was found to be the highest in the old leaves of A. marina found in the isolate code DT-Am-05-OLL with a proteolytic zone diameter of 2.1 mm. As for young leaves of A. marina, the highest activity was found in the DM-Am-02-OLL isolate code with a clear zone diameter of 1.9 mm. Abstract

The revitalization of local wisdom through the processing of various types of rhizomes by housewives in Oelnasi village into traditional herbal medicine/drinks has good potential for development. Housewives and village women do not have knowledge about this. The high level of health problems but the community's minimal purchasing power for chemical medicines from pharmacies due to constraints due to the family's economic incapacity has become an urgency for housewives and village women to be skilled at processing rhizomes into herbal medicines. This activity aims to increase the knowledge of Oelnasi village women regarding the properties of rhizomes, skills in selecting rhizome types, processing management, hygiene of herbal medicine products with the opportunity to become a permanent business so that marketing can be done online. The methods used in implementing training are: (1) consolidation with partners; (2) lecture; (3) mentoring; (4) evaluation through giving questionnaires to training participants regarding the implementation of activities based on LIKERT with the aim of knowing the results of activities that are measured qualitatively. The results of PKM activities show that partners are able to select good quality types of rhizomes, are able to process rhizomes into herbal medicine by implementing hygiene protocols during the production period, and are able to market herbal medicine products via social media (Facebook, TikTok and WhatsApp) using existing devices. The marketing of herbal medicine products processed by partners has been marketed through these three social media, however this has only reached a percentage of 50% because 6 out of 12 partner members are still not digitally literate on social media. Partners produce herbal medicine, market it using digital platforms, but this is not yet optimal due to constraints on human resources, motivation, family and community support.

Abstract

The utilization of medicinal plants has become one alternative for communities to treat illnesses. This is because the use of medicinal plants, besides being relatively affordable, also does not cause side effects compared to the use of modern medicine or chemical-based drugs. This study aims to develop a reference book of medicinal plants in Rote Tengah Sub-district, Rote Ndao District. The development model used in this research is the ADDIE model (analysis, design, development, implementation, evaluation). The research results obtained from material expert testing showed a validity of 94.6%, design expert testing showed 95.5%, and media expert testing showed 100%. Overall, the percentage of assessments from all three experts indicates a value of 96.7% with a qualification of very valid (highly appropriate). This indicates a positive response to the developed medicinal plant reference book, which is deemed suitable for use as a learning resource. This is because the developed medicinal plant reference book has its own advantages, where the illustrations in the reference book are made with attractive designs, thus attracting students' attention and making it easier to understand

Abstract

Studi etnobotani berguna untuk menganalisis pemanfaatan jenis dan bagian tumbuhan mangrove secara kuantitatif berdasarkan indeks signifikansi budaya (Index of Cultural Significance) masyarakat pesisir. Penelitian dilakukan di area mangrove Taman Wisata Mangrove Kelurahan Oesapa, Pantai Manikin, Pantai Sulamanda Desa Mata Air, Desa Kelapa Tinggi di Kelurahan Tarus, Desa Oebelo dan Desa Pariti. Pengumpulan data menggunakan metode survei lapangan, observasi dan teknik wawancara semi-terstruktur tentang tumbuhan, bagian tumbuhan yang digunakan dan cara pengolahan terhadap masyarakat yang ada di sekitar hutan mangrove. Hasil wawancara diperoleh bahwa terdapat 6 (enam) spesies mangrove major, yakni: Avicennia marina, Sonneratia alba, Bruguiera parviflora, Ceriops tagal, Rhizophora apiculata, dan R. mucronata, dan 1 (satu) spesies mangrove asosiasi, yakni Nypa fruticans. Berdasarkan nilai Indeks Signifikansi Budaya (ICS), R. mucronata memiliki nilai ICS tertinggi (708), yaitu sebagai indikator lingkungan (340) dan bahan bangunan (320). A. marina, nilai ICS 114, terutama pemanfaatannya untuk bahan obat (108), dan S. alba bernilai ICS 54, terutama sebagai pengganti sirih (12). Mangrove digunakan sebagai indikator lingkungan, kayu bakar, bahan bangunan, bahan obat, kegiatan pertanian, berkaitan dengan mitos, pengganti sirih, pembuatan garam, bahan perahu, pembuatan sirup dan pakan ternak.

Abstract

 Dua jenis tumbuhan berkhasiat obat dari Sabu Raijua yang memiliki nama daerah Puri raho dan Loro wawi eddu  memiliki kemiripan morfologi dengan tumbuhan brotowali, namun belum dilakukan identifikasi taksonomi dan kandungan senyawa fitokimianya. Tujuan penelitian ini adalah untuk mengidentifikasi taksonomi Puri raho dan Loro wawi eddu serta menganalisis kandungan fitokimianya (kuersetin). Penelitian ini terdiri atas 2 tahap yaitu identifikasi morfologi dan analisis penanda DNA ITS2 dilanjutkan dengan identifikasi kandungan fitokimia (kuersetin) pada ekstrak batang  menggunakan metode kromatografi lapis tipis (KLT) dan metode spektrofotometri. Identifikasi taksonomi tumbuhan Puri raho dan Loro wawi eddu menggunakan buku kunci determinasi menunjukkan bahwa kedua tumbuhan ini termasuk dalam ordo Ranunculales, selanjutnya perbandingan akar, batang dan daun dengan spesies dari genus Tinospora lainnya menunjukkan kemiripan antara Puri Raho dengan Tinospora cordifolia dan Loro wawi eddu dengan Tinospora macrocarpa namun kedua tumbuhan asal Sabu Raijua ini tidak memiliki bunga, buah dan biji sehingga dilanjutkan dengan identifikasi molekuler menggunakan penanda DNA ITS2 yang menunjukan tumbuhan Puri raho memiliki persentase identitas DNA ITS2 sebesar 100% dengan Tinospora crispa voucher Chen ZD s.n. (PE) Sequence ID: KY365661.1, sedangkan Loro wawi eddu menunjukkan persentase identitas DNA ITS2 sebesar 99,17 % dengan Tinospora smilacina voucher Gray 8798 (MO) Sequence ID: KY365675.1. Uji kualitatif kandungan kuersetin menunjukkan hasil positif dengan munculnya bercak pada plat KLT dengan Rf 0,91 pada sampel dan larutan standar kuersetin, sedangkan hasil uji spektrofotometri menunjukkan kandungan kuersetin pada ekstrak batang Puri raho sebesar 99,24 ppm dan 78,08 ppm pada ekstrak batang Loro wawi eddu.
