Abstract

Children living in orphanages are generally vulnerable to close-contact transmission diseases, including head lice infestation. The clinical manifestation of head lice infestation was relatively mild, such as itchy scalp, and hence it is often overlooked as a community health issue. Education plays an essential role in preventing re-infection and eradicating head lice infestation. The treatment options include chemical treatment using pediculicides and physical management by shaving the head and manually removing the parasite. This community service activity aimed to eradicate head lice from Yayasan Griya Asih Orphanage. A series of three activities were conducted at Yayasan Griya Asih Orphanage in Central Jakarta. Each activity was done around one month apart to evaluate the effectiveness of treatment. Firstyear medical students were recruited to participate in these activities to familiarize them with real-world community health problems. Student volunteers helped applied pulse doses of Permethrin 1% shampoo and provided health education activity. Fiftyfive Yayasan Griya Asih Orphanage residents joined this activity from mid-October until December 2022. Residents were instructed not to share their personal items to prevent re-infection. Health education activities were done using one-to-many followed by a one-on-one format. In conclusion, head lice infestation screening is essential in crowded environments, and permethrin 1% pulsed dose followed by personalized health education activities and pest control effectively broke the disease transmission chain and eradicated the head lice infestation.

Abstract

Abstract

Background: Parkinson's disease (PD) is a progressive neurodegenerative disorder with an increasing prevalence. Oxidative stress is known to contribute to the development of PD through lipid peroxidation, which causes damage to the substantia nigra pars compacta. Lipid peroxidation produces malondialdehyde (MDA), which is known to be elevated in PD patients. Up till now, there is no cure for PD, and the available therapies are only symptomatic. The secretome from Mesenchymal Stem Cell (MSC) has antioxidant and neuroprotective components, making it a potential therapeutic agent that may slow the progression of PD. Objective: This study seeks to determine the effect of MSC secretome on oxidative stress in rotenone-induced PD rats. Methods: This research was an in-vivo experiment conducted with 30 male Sprague Dawley rats, divided into sham control, rotenone (+) secretome (-), and rotenone (+) secretome (+) groups. Rotenone (2.75 mg/kgBW) was administered for seven days to induce a PD model. Secretome administration (1 mg/ml) was carried out on days 3, 5, and 7. MDA levels were determined using the sandwich ELISA method. Results: This study found no significant difference in MDA levels among the three groups (p = 0.203). The sham control group had the lowest MDA 1.32(0.53) nmol/mL, followed by the rotenone (+) secretome (+) group 1.56(0.33) nmol/ml, and the highest MDA was observed in the rotenone (+) secretome (-) group 1.88(0.14) nmol/mL. Conclusion: Administration of MSC secretome did not significantly cause changes in plasma MDA levels of rotenone-induced Parkinson Disease rats.

Abstract

Background: Parkinson's disease (PD), a chronic progressive neurodegenerative disease, threatens much of the world's increasingly aging population and has no promise for disease-modifying drugs, only symptomatic treatment. One potential biomarker correlates PD to the disruption of cholesterol metabolizing processes that results in formation of cytotoxic bile acids (BA) which induces mitochondrial dysfunction that leads to the decrease of dopaminergic neurons in various PD models. Objective: This study aims to determine the effect of rotenone-induction as well as MSC-Secretome administration on plasma-total cholesterol levels in rat models.. Methods: This was an experimental research using 30 male Sprague Dawley rats divided into sham control, rotenone (+) secretome (-) and rotenone (+) secretome (+). Blood was withdrawn on day 0 and day 8 and analyzed under colorimetry and spectrophotometry for its plasma-total cholesterol concentration. Results: The Tukey Post-Hoc Test showed that there was no significant difference between the sham control and rotenone (+) secretome (-)  (p = 0.073), plasma-total cholesterol level is lower in rotenone (+) secretome (-) than in the sham control. Furthermore, we were unable to prove for a significant difference between the rotenone (+) secretome (-) and rotenone (+) secretome (+) group (p= 0.234), even though in average the plasma-total cholesterol level is lower in the rotenone (+) secretome (+)  group. Conclusion: Induction of rotenone followed by an intervention of MSC-Secretome has no effect on the change in plasma-total cholesterol levels on rat models.

Abstract

Abstract

HIGHLIGHTS KM score is a reliable method to help determine the diagnosis of cervical dysplasia and squamous cell carcinoma. KM score cannot be used to differentiate infection status in cervical dysplasia and squamous cell carcinoma.   ABSTRACT Objective: Cervical cancer is the second most common malignancy in women, with 604,127 new cases reported globally in 2020. Persistent high-risk human papillomavirus (HPV) infection is the primary cause. This study aimed to evaluate the relationship between stromal inflammatory cell infiltration using the Klintrup-Mäkinen (KM) score and HPV infection status in cervical dysplasia and squamous cell carcinoma (SCC). Materials and Methods: A cross-sectional study was conducted using secondary data from formalin-fixed paraffin-embedded (FFPE) cervical tissue samples diagnosed as dysplasia or SCC at Atma Jaya Catholic University of Indonesia from 2014 to 2022. HPV status was determined using PCR-based assays, while stromal inflammatory cell infiltration was assessed microscopically at 100× magnification and graded according to the KM scoring system by two blinded observers. Statistical analyses, including Chi-square and Spearman’s correlation tests, were performed using STATA/IC 15, with p < 0.05 considered significant. Results: A total of 38 samples were analyzed, comprising 20 (52.63%) cases of dysplasia and 18 (47.37%) cases of SCC. HPV positivity was identified in 70% of dysplasia and 94.44% of SCC cases. KM scores of 2/3, indicating moderate to severe inflammatory infiltration, were significantly more frequent in SCC (77.78%) compared to dysplasia (20%), showing a strong association with pathological diagnosis (p = 0.0019). However, no significant correlation was observed between KM score and HPV status in either lesion type. Conclusion: The KM scoring method provides a reliable and cost-effective approach for assessing stromal inflammatory cell infiltration in cervical lesions, aiding histopathological diagnosis, although it does not predict HPV infection status.
