Abstract

The SARS-CoV-2 coronavirus outbreak has resulted in severe pneumonia, even death (COVID-19). ADP-Ribose phosphatase (ADPR), a highly conserved macrodomain of this virus, was appropriate for viral RNA replication and transcription. According to studies, quercetin suppresses the main protease and 3-chymotrypsin and papain-like proteases, exhibiting antiviral efficacy against SARS CoV-2. However, quercetin analogs to ADPR have yet to be investigated. This study aims to obtain candidate compounds for ADPR based on binding energy, interaction mode, and binding stability using docking and molecular dynamics (MD) studies.The native ligand (AMP) has estimated binding energy based on docking results of -7.35 kcal/mol. Quercetin analogs, lig_C00013871 (Quercetin 3-(2''-galoylrutinoside), lig_C00006532 ([5',5']-Bisdihydroquercetin), and lig_C00013874 (Quercetin 3-(2G-(E)-p-coumaroylrutinoside) has more negative binding energy, with estimates of -9.43, -9.26, and -8.98 kcal/mol, respectively. These results align with binding energy estimates based on MM-GBSA of -14.76, -29.39, -34.90, and -42.79 kcal/mol for AMP, lig_C00006532, lig_C00013871, and lig_C00013874, respectively. According to the MD simulation, lig_C00006532 and lig_C00013874 will be more effective in stabilizing the ADPR complex. Finally, these two analogs are potential candidate compounds as ADPR inhibitors of SARS CoV-2.

Abstract

Ikan Belanak (Mullet) is a small pelagic fish that lives in shallow coastal waters. This type of fish lives in the coastal area of Lembo Village, North Konawe Regency. The objective of this study was to characterize physicochemicals and determine the fatty acid profile of mullet oil, which is a source of fish oil that contains unsaturated fatty acids that are beneficial to health. The reflux process is used to remove mullet flesh, which yields fish oil. The findings demonstrated that the physicochemical properties of mullet oil varied. These properties included the acid number, peroxide number, iodine number, and saponification number, which were, respectively, 0.81 mg KOH/g, 8.30 meqO2/kg, 50.98 I2/100 g, and 109.07 mg KOH/g. The fatty acid profile of mullet oil is dominated by palmitic acid, cis-9-oleic acid and DHA. Based on the test results, mullet oil can be used as edible oil according to SNI and IFOS standards.

Abstract

ABSTRACT. Etlingera is a genus known for its potential medicinal properties. Etlingera rubroloba A.D. Poulsen is a recently identified species within the Etlingera genus found in Southeast Sulawesi. This study aims to isolate and identify secondary metabolite compounds from the methanol extract of the E.rubroloba A.D. Poulsen rhizome and its activity as an antimicrobial and antioxidant. The compound was isolated by using Thin Layer Chromatography (TLC), Vacuum Liquid Chromatography (VLC), and Radial Chromatography (RC) techniques. Isolated compounds were identified by 1H and 13C NMR spectroscopy techniques and compared the data with reference. Antimicrobial tests were carried out on Escherichia coli, Streptococcus mutans, and Candida albicans by broth microdilution method. The antioxidant test was carried out using the 1,1-dipheny1-2-picrylhydrazyl (DPPH) method. Four isolated compounds were successfully isolated : stigmasterol (1); 1,7-diphenyl-6-hepten-3-on; (2); and 1,7-diphenyl-heptan-3-on (3); and sinapyl alcohol diacetate (4). Antimicrobial activity test results extract were active against all bacteria and C. albicans. The results of antioxidant activity test showed that extract, compound 2, 3, and 4 had strong and medium antioxidant activity with IC50 values at 75.05 μg/mL; 201.4 μg/mL; 207.88 μg/mL, and 190.59 μg/mL, respectively. Abstract

Hypertension is one of the most prevalent non-communicable diseases among older adults and has the potential to cause serious complications such as stroke and heart disease. This community service activity aimed to improve the knowledge, awareness, and healthy behaviors of older adults through health education and routine health screenings at the Elderly Posyandu of UPTD Puskesmas Mokoau. The methods included Body Mass Index (BMI) assessment, blood pressure measurement, health counseling, and educational sessions on hypertension prevention and control. A total of 20 older adults participated in the program. The results showed that 40% of participants were overweight or obese, and 50% were classified as having stage 2 hypertension. These findings indicate a strong relationship between nutritional status and hypertension risk. Health education was found to increase awareness regarding healthy lifestyle practices, the importance of regular blood pressure monitoring, and the management of risk factors. This activity demonstrates that collaboration between the Community Health Center, the Elderly Posyandu, and healthcare workers plays a significant role in promotive and preventive efforts to address hypertension among older adults.

Abstract

Tuberculosis (TB) is an infectious disease that is still a global health problem, caused by the bacterium Mycobacterium tuberculosis, which is acid-resistant and mainly attacks the lungs, but can also attack other organs. This disease can affect people of all ages, including babies, children, adults, and the elderly. TB transmission occurs through the air from people who cough or sneeze, with the main factors affecting public health being environmental factors and behaviours, such as uncovered coughing and poor sanitation. TB cases in Southeast Sulawesi Province have shown a significant upward trend in recent years, with the incidence rate continuing to rise every year. The success of TB treatment is influenced by a variety of factors, including medication adherence, which is a major challenge to avoid the emergence of drug resistance, such as TB-MDR. The Government of Indonesia has issued policies and focuses on reducing TB incidence through education and socialisation to encourage active community involvement in TB control to achieve the national target by 2030.
