Abstract

The effect of chitosan  on micropropagation of Dendrobium lasianthera J.J.Sm was observed. The media used was Vacin and Went (VW) modified with the addition of coconut water and given chitosan with several concentrations. The study was conducted at the Plant Tissue Culture Laboratory of the Department of Biology, Faculty of Mathematics and Natural Sciences, Cenderawasih University for 6 months, from April to September 2019. Experiments using a Completely Randomized Design (CRD) of 8 treatments with 3 explants in each bottle for total 3 bottle as a replication. The total explants used in this experiment was 72 explants. The concentration of chitosan were 0% , 1%, 5%, 10%, 15%, 20%, 40% and 50% separately. Observation data were analyzed using ANOVA, if there were significant differences followed by DMRT at the 5% level. Observation variables included number and length of roots, number and length of leaves. The results showed that giving chitosan 10% had a maximum effect on the number of roots and number of leaves, 6.67 and 4.67. As for the highest leaf length of 17.43 mm  and the longest root length 19.21 mm were in the media with the addition of chitosan concentration of 15%. The observations showed that chitosan  had a positive influence on the growth and development of orchids D. lasianthera plantlet  in vitro.Keywords : Chitosan, orchid, Vacin and Went, plantlet.

Abstract

Orchid found as one of medicinal plant in some areas, especially Dendrobium. Some species were investigated its bioactive compound, and antibacterial activity, but the information about antibacterial activity of Papuan Dendrobium species is still limited. The aims of this research were to determine the phytochemical constituents and the antibacterial activity of ethanolic extract and fraction of Dendrobium species from Papua against Escherichia coli and Staphylococcus aureus. There were three species of Dendrobium namely: D. spectabile, D. violaceoflavens, and D. antennatum used in this study. Dried simplisia was macerated using ethanol, then tested for the phytochemical content. Total ethanolic extract was fractionated with three different solvents, ethanol, ethyl acetate and hexane. Disc diffusion assay was used to examine the antibacterial activities of the total extract and the fractions. Total extract were positive for flavonoid, alkaloid and tanin, except D. spectabile stem extract only alkaloid and tannin.  No saponin was found in the plants. The ethyl acetate fraction of leaves of D. spectabile showed the highest antibacterial activity against S. aureus with diameter of inhibition zone 20.54 + 1.47 mm (strong category). Other plant extract and fraction tested had  moderate antibacterial activities against both E.coli and S aureus  with diameter zone  6-9 mm. The present work indicates that the ethyl acetate fraction of D. spectabile leaves is potential to be developed in antibacterial drug design research.  Key words: antibacterial activity; Papua; Dendrobium; E. coli; S. aureus.

Abstract

Anggrek Macan (Grammatophyllum scriptum (Lindl.) Bl.) merupakan salah satu tanaman yang digemari di kalangan masyarakat sebagai tanaman hias karena bunganya yang indah. Aggrek macan memiliki senyawa metabolit sekunder alkaloid, flavonoid dan tannin yang dapat digunakan sebagai pengobatan tradisional. Namun, penelitian mengenai Anggrek Macan di bidang farmakologi masih sangat terbatas. Tujuan penelitian ini adalah mengetahui kandungan senyawa kimia yang terdapat dalam ekstrak herba Anggrek Macan secara LC-MS/MS serta nilai LC₅₀ dan konsentrasi dari ekstrak herba Anggrek Macan yang paling efektif membunuh larva udang (Artemia salina Leach.). Metode yang digunakan dalam penelitian ini adalah pengambilan sampel dari kebun Anggrek yang ada di Lingkungan FMIPA Universitas Cenderawasih Papua-Indonesia, selanjutnya sampel dibuat dalam bentuk simplisia, diekstraksi dengan etil asetat. Ekstrak selanjutnya dianalisis dengan LC-MS/MS dan dari ekstrak tersebut juga dilakukan uji sitotoksik dengan metode BSLT (Brine Shrimp Lethality Test). Hasil penelitian menunjukkan bahwa Anggrek Macan (Grammatophyllum scriptum (Lindl.) Bl.) mengandung 2′,6′-Dihydroxy-4′-methoxy-dihydrochalcone, Apigenin-7-O-α-L-rhamnose(1→4)-6"-O-acetyl-β-D-glucoside, Kushenol H, Rubrofusarin, 3-Hydroxy baicalein dan Luteolin yang diperoleh dari hasil analisis menggunakan Liquid Chromatography tandem Mass Spectrometry (LC-MS/MS). Ekstrak herba Anggrek Macan memiliki aktivitas sitotoksik dengan nilai LC₅₀ 150,96 ppm dengan tergolong kurang toksik.
