Abstract

Most of the plant-associated bacteria can synthesize active biological components of phytohormones such as auxin. This study aimed to examine the potency of 61 endophytic bacteria isolates from the mangroves at Kutang Beach, Lamongan in producing IAA hormone and to identify types of isolates effecting the concentration of IAA, morphological characteristics of isolates, as well as endophytic bacterial species that have the most potential to produce IAA hormone. Screening of endophytic bacteria isolates was performed using the colorimetric method and the production of IAA was carried out using the spectrophotometric method. IAA production by endophytic bacteria was analyzed descriptively and statistically. One-Way ANOVA was employed to determine the effect of the isolate type on the concentration of IAA. The most potential isolates to produce IAA hormone are identified by 16s rRNA gene marker. The screening results showed that 12 isolates of endophytic bacteria have the potential to produce IAA hormones (2.0-9.3 ppm), coded with LMG 7, 15, 31, 32, 43, 53, 54, 55, 56, 57, 62, and 63. The results of the One-Way ANOVA test suggested that the type of isolates did not affect the concentration of IAA produced by endophytic bacteria. The twelve isolates had different morphological characters and those were Gram-positive bacilli with cell sizes ranging from 1.5 µm - 3 µm. The highest concentration of IAA was produced by LMG 15 (9.3 ppm). LMG 15 was identified as Bacillus cereus strain LMG 15, having 99.33% similarity to Bacillus cereus strain IAM 12605.

Abstract

Endophytic bacteria are microorganisms that live in plant tissues and some of them contribute to nitrogen fixation for plants. This study aimed to isolate and identify endophytic bacteria from mangroves of Kutang Beach, Lamongan, which potentially as nitrogen fixing bacteria. Bacterial Isolates were used as candidates for biofertilizers. Leaves samples were taken from 10 sampling points. Bacterial isolation was initiated by sterilizing the surface of the leaves sample and grinding it aseptically. Isolation was carried out with a pour plate method on Nutrient Agar medium. Screening for endophytic bacteria's potential as N-fixing agent was carried out by growing the bacterial isolates on a semi-solid Nitrogen Free Bromothymol Blue (NFB) medium. The isolates that produced a positive reaction with a change in the color of the medium to blue were then subjected to macroscopic (shape, color, elevation, and the edge of the colony) and microscopic observations (Gram stain and bacterial cell measurements). The isolates showed the fastest change in the color of the medium were identified by the molecular marker of the 16S rRNA gene. The data obtained were analyzed descriptively. As many as 20 isolates were obtained from the mangroves of Kutang Lamongan Beach, and ten isolates of twenty potentially as nitrogen-fixing bacteria. The ten nitrogen-fixing bacteria isolates had varying macroscopic characteristics. The microscopic characteristics showed that eight isolates had Gram-positive bacilli, and two isolates were Gram-negative with varying bacterial sizes. Based on the 16S rRNA gene sequence, the most potential of nitrogen-fixing bacteria was LMG II-14 isolate and identified as Paenibacillus alvei LMG II-14 with 99.36% similarity to Paenibacillus alvei strain DSM 29 based on the NCBI database. The ten nitrogen fixing isolates that have been obtained can later be used as candidates for biofertilizer composition, especially Paenibacillus alvei LMG II-14.

Abstract

Biofertilizer application can potentially reduce chemical residues in soil from synthetic fertilizers. This study investigates the effects of varying doses and application frequencies of biofertilizers on the growth, productivity, and Relative Agronomic Effectiveness (RAE) of Glycine max L. A Completely Randomized Design was employed, with 9 treatment combinations and 2 control groups (positive and negative), each replicated three times. The biofertilizer doses tested were 5, 10, and 15 ml per plant (B5, B10, B15), and application frequencies included 1 week after planting (WAP) (F1), at 1 and 4 WAP (F2), and at 1, 4, and 8 WAP (F3). The negative control received no biofertilizer, while the positive control received NPK (5 g/plant). Plant growth was assessed by measuring height and leaf number, and productivity was evaluated based on the number of pods, pod weight, dry seed weight, and weight per 100 seeds. Data were analyzed using one-way ANOVA. The results indicated that the treatment with 10 ml of biofertilizer applied twice (B10F2) significantly improved the growth and productivity of Glycine max L., achieving an RAE value of 169.70%.

Abstract

this study aims to determine nitrogen-fixing bacteria isolates from Jenu mangrove soil, their macroscopic and microscopic characteristics, and species based on the 16S rRNA gene. nitrogen-fixing bacteria were filtered qualitatively using a selective nitrogen-free bromothymol (NFB) medium. screening results showed that six bacterial isolates from Jenu Tuban mangrove soil had potential as nitrogen -fixing bacteria, namely 1,8,10,15, and 16. the six nitrogen-fixing bacteria had varied microscopic characteristics and were gram-positive. based on the identification of the 16S rRNA, it was found that the six isolates were detected as a Bacillus cereus group with 99% query cover and 99.58% identitiy; Bacillus toyonensis with 99% query cover and 99.79% identity; Bacillus altitudinis with 99% query cover and 99.72% identity; Bacillus megaterium with 99% query cover and 99.79% identity; and Bacillus aerius with 98% query cover and 99.86% identitiy.

Abstract

The increasing demand for tomatoes has prompted farmers to use chemical fertilizers, which can lead to soil degradation and reduced fertility. This study aims to determine the appropriate dose and frequency of biofertilizer application to enhance the growth and productivity of tomato plants (Solanum lycopersicum L.). The experiment was designed using a Completely Randomized Design (CRD) with 11 treatments, including both negative and positive controls. Three variations in biofertilizer doses were tested (5, 10, and 15 ml/plant), along with three different frequencies of application: once at one week after planting (1 WAP), twice (at 1 and 4 WAP), and three times (at 1, 4, and 8 WAP). Measured parameters included soil fertility (microbial population and functional microbial quality), plant growth (plant height, leaf count, stem diameter), and plant productivity (flower count). Quality of functional microbes were analyzed descriptively. Data of microbial population, plant growth and productivity were analyzed using Kruskal-Wallis and Mann-Whitney tests with a 5% significance level. The results showed significant differences in soil microbial populations and plant height between treatments, although no significant differences were observed in stem diameter, leaf number, or flower number. The treatment with 15 ml/plant applied three times (B15F3) exhibited the highest Relative Agronomic Efficiency (RAE) compared to other biofertilizer treatments (74.63%), but it was still lower than the control (chemical fertilizer). Therefore, higher doses and/or frequencies may be needed to further evaluate the effectiveness of the biofertilizer under test.

Abstract

The purpose of this study was to determine students' perceptions of the English Development Program on students' speaking skills. English Development Program is a program established to help students learn English better, especially in terms of speaking. The subjects of this study were 4 students from Pesantren Nuris who participated in the English Development Program selected by pursosive sampling. This research uses qualitative methods, and the research instrument uses interviews. The interview results show that the majority of students have a positive perception of the English Development Program. The students believe that the English program can support them to speak English fluently. Students also have the perception that the English program is a place for students to develop their English skills, especially in terms of speaking. Not only that, students also think that the English program can make them more confident in speaking English.

Abstract

Digital posters are visually appealing and can capture students' attention more effectively than traditional teaching materials, while Canva offers a wide range of design tools and templates, allowing students to express their creativity in designing posters. Hence, digital poster creation on the Canva application is suitable as instructional media. This study aims to investigate the effect of using digital posters based on the Canva application on students’ writing ability in narrative text at SMAN 6 Pekanbaru. This study uses a quasi-experimental research design where the experimental group receives a digital poster based on the Canva application as the instructional media while the control group receives conventional instructional media. The population of this study is the tenth-grade students of SMAN 6 Pekanbaru. The purposive sampling is the type of sampling technique in this study where 36 students from X MIPA 2 as the control group and 34 students from X MIPA 4 as the experimental group. The data were collected from a pretest-posttest of writing narrative text before and after treatments. The hypothesis test was conducted using the Mann-Whitney test. The result displays the alternative hypothesis (Ha), the Asymp. Sig. (2-tailed) is 0.001<0.05. Therefore, using digital posters based on Canva application has a significant effect on students’ writing ability in narrative text at SMAN 6 Pekanbaru. It is suggested to use digital poster based on Canva application as instructional media for teachers, students, and researchers. Canva can spark creativity, improve design and writing skills, and create engaging learning materials. Â
