Abstract

Vector-borne diseases transmitted by mosquitoes are considered a significant public health problem worldwide. Aedes aegypti is one of the mosquito species responsible for transmitting these diseases. One environmentally friendly method of vector control is the use of microbial agents such as Bacillus species. This study aimed to explore investigate indigenous entomopathogenic bacteria of Bacillus species isolated from A. aegypti larvae. Larvae samples were collected from breeding sites of A. aegypti. All isolates underwent screening and affirmation confirmation tests to assess their larvicidal toxicity against A. aegypti larvae. Phenotypic characterizations and molecular identifications were conducted to determine the species of the Bacillus isolates based on similarity index and percent identity (%ID). Phylogenetic trees were used to compare the isolates with other Bacillus species. The results revealed 120 isolates of Bacillus species from A. aegypti larvae samples. Among them, three isolates (LS3.3, LS9.1, and LSD4.2) exhibited the highest larvicidal toxicity in the confirmation test, resulting in larval mortality rates of 100%, 96.7%, and 100%, respectively, after 48 hours of exposure. Molecular identifications, showed that LSD4.2 had a 99.16% ID with Bacillus velezensis, LS3.3 had a 98.22% ID with Bacillus mojavensis, and LS9.1 had a 99.93% ID with Bacillus subtilis. These three bacteria from the Bacillus genus have been reported to offer significant benefits to humans.

Abstract

Biofertilizer application can potentially reduce chemical residues in soil from synthetic fertilizers. This study investigates the effects of varying doses and application frequencies of biofertilizers on the growth, productivity, and Relative Agronomic Effectiveness (RAE) of Glycine max L. A Completely Randomized Design was employed, with 9 treatment combinations and 2 control groups (positive and negative), each replicated three times. The biofertilizer doses tested were 5, 10, and 15 ml per plant (B5, B10, B15), and application frequencies included 1 week after planting (WAP) (F1), at 1 and 4 WAP (F2), and at 1, 4, and 8 WAP (F3). The negative control received no biofertilizer, while the positive control received NPK (5 g/plant). Plant growth was assessed by measuring height and leaf number, and productivity was evaluated based on the number of pods, pod weight, dry seed weight, and weight per 100 seeds. Data were analyzed using one-way ANOVA. The results indicated that the treatment with 10 ml of biofertilizer applied twice (B10F2) significantly improved the growth and productivity of Glycine max L., achieving an RAE value of 169.70%.

Abstract

Komunitas peternak sapi perah di Junrejo, Batu menghadapi tantangan dalam meningkatkan produktivitas dan kualitas susu akibat rendahnya kesehatan ternak dan keterbatasan akses terhadap teknologi pakan yang efisien. Program pengabdian masyarakat ini bertujuan untuk meningkatkan pemahaman dan keterampilan peternak dalam memanfaatkan probiotik dan Single Cell Protein (SCP) sebagai pakan tambahan, yang diharapkan dapat meningkatkan kualitas kesehatan dan produktivitas ternak. Program ini menggunakan pendekatan partisipatif yang melibatkan pelatihan langsung dan sesi diskusi untuk memastikan penerapan yang efektif dan berkelanjutan. Probiotik dan SCP berperan dalam memperbaiki mikrobiota usus ternak, meningkatkan efisiensi pencernaan, dan memberikan sumber protein berkualitas tinggi. Hasil pelatihan menunjukkan bahwa setelah mengikuti kegiatan ini, seluruh peserta (100%) memahami dasar-dasar probiotik dan SCP, serta mampu menerapkannya secara mandiri di peternakan mereka. Di samping itu, penggunaan molase sebagai media perbanyakan probiotik mendukung prinsip ekonomi sirkuler, mengurangi limbah produksi gula, dan menciptakan solusi pakan yang ramah lingkungan. Program ini berhasil meningkatkan kapasitas komunitas peternak lokal, sehingga dapat berkontribusi terhadap kesejahteraan ekonomi mereka melalui peningkatan hasil susu dan biaya operasional yang lebih efisien.

Abstract

Bioindustries often involve biochemical processes that occur at higher temperatures. However, most proteins, including enzymes, lose their structural integrity and functionality at higher temperatures. Thus, thermostable enzymes from thermophilic microorganisms are best suited candidates for successful bioprocessing under such conditions. Indonesia is one of the best study sites for performing bioprospecting of thermostable enzyme-producing thermophilic microorganisms due to the numerous hot springs. To explore the biodiversity of thermophilic microorganisms with potential industrial applications, we isolated and characterized thermophilic bacteria from the Cangar hot spring, Batu, East Java, Indonesia. One isolate (CGR-1) showed growth at 60°C and was identified as Bacillus subtilis subsp. inaquosorum based on 16s rRNA gene sequencing followed by bioinformatic analysis. This is the first report on the isolation of Bacillus subtilis subsp. inaquosorum CGR-1 from Indonesia, especially from a hot spring environment. This isolate showed cellulolytic and amylolytic activity at 50°C, which would encourage further exploration on the industrial and environmental applications.

Abstract

Indonesia, as a megabiodiverse country, possesses a wealth of beneficial microorganisms, including bacteria and fungi capable of producing antimicrobial compounds. Over the past decade, extensive research has been conducted in Indonesia to explore and screen the antimicrobial activities of bacteria and fungi. These microorganisms have been isolated from various ecosystems, such as terrestrial, maritime, extreme environments like hot springs and sand dunes, as well as from animals and plants hosts. Most studies have focused on the antimicrobial activity of crude metabolite extracts, which demonstrate inhibitory effects against clinically significant pathogens, including Methicillin-resistant Staphylococcus aureus, multidrug-resistant Escherichia coli, and Salmonella typhi. This review systematically synthesizes findings from peer-reviewed literature, detailing isolation strategies, antimicrobial screening techniques, and the bioactivities reported. By consolidating current knowledge, it aims to elucidate the potential of Indonesia’s microbial resources for developing novel antimicrobial agents and promoting their sustainable exploitation within the context of global health challenges.

Abstract

The amylase is an enzyme that widely used in industry. In this research, amylase was isolated from endosymbiont bacteria (EKA 4) of cockroach Pycnoscelus surinamensis from household garbage dump Surabaya. This research aims to identify bacteria EKA 4, produce and characterize amylase from it at various temperature, pH and effect of metal ions. Amylase screening assay was done by imoculating the isolate into medium contain 2% of starch then incubated for 24 hours. Detection of amylase was done qualitatively by measured presence of halozone and quantitatively using DNS method. 16S rRNA analysis found that bacteria EKA 4 has similarity to Bacillus subtilis with 100% of percent identity. Result showed that optimum activity of amylase from B. subtilis were at 40°C, pH 6 and could use ion CaCl2 as a cofactor in a reaction. Thus, utilization of amylase from endosymbiont P. surinamensis could be as an alternative source in big industries.

Abstract

Kelurahan Ampel Surabaya, merupakan kawasan yang potensial untuk dikembangkan dari sektor pariwisata dan budaya. Tantangan yang dihadapi saat ini adalah pengelolaan kawasan yang kurang asri dan berwawasan lingkungan. Kurangnya pemberdayaan lahan sempit untuk bercocok tanam yang selaras dengan alam juga turut mempengaruhi kondisi lingkungan di kawasan ini.  Salah satu upaya untuk mengatasi masalah ini adalah dengan memberdayakan masyarakat sekitar untuk membantu mengelola kawasan yang asri dan berwawasan lingkungan.  Tujuan dari pengabdian masyarakat ini adalah memberdayakan ibu-ibu kader PKK di Kelurahan Ampel agar dapat bercocok tanam dengan memanfaatkan lahan sempit di sekitar rumah menggunakan pupuk hayati yang ramah lingkungan.  Metode yang digunakan adalah memberikan pelatihan meliputi aspek pengetahuan dan keterampilan. Aspek pengetahuan meliputi pemberian materi tentang pemanfaatan dan konsep pupuk yang ramah lingkungan. Aspek keterampilan, meliputi bagaimana cara membuat pupuk hayati serta cara mengaplikasikannya ke tanaman budidaya, dalam hal ini tomat, terong, cabe rawit, dan cabe merah. Evaluasi dilakukan melalui pemberian pretest, postest untuk melihat keterserapan materi, serta kuisioner untuk mengevaluasi kegiatan secara keseluruhan. Hasil evaluasi menunjukkan bahwa rerata nilai pretest adalah 59; rerata nilai postest adalah 79. Tingkat kepuasan peserta terhadap kegiatan ini adalah 100 % sangat puas (pengabdian masyarakat sesuai kebutuhan mitra), dan 95% sangat puas ; 5 % puas (pelatihan pembuatan pupuk hayati dapat meningkatkan pengetahuan, dan keterampilan, serta kesadaran masyarakat untuk turut menciptakan kawasan asri menggunakan pupuk yang ramah lingkungan (pupuk hayati).
