Abstract

Rubber seeds have no dormancy, germinate rapidly and fungal growth is the limiting factor influencing storage period of seed viability. Solution of Polyethylene glycol 6000 (PEG 6000) have osmotic potential and osmotic adjustment that is used as a coating material is expected to be a preservative during storage. The purpose of this study was to test the effect of polyethylene glycol 6000 in inducing secondary dormancy by reducing the seeds germination and fungus growth to maintain the seed viability in storage period and to test seed viability and vigor after storage period. First stage of the study was using nonfactorial completely randomized design with four levels of polyethylene glycol with four replications, namely:PEG 6000 (w / v): 0%, 15%, 30%, 45%; The second step, was germination phase in germination tub after the storage period. The first study showed that concentration of 15% PEG 6000 to 45% were able to induced secondary dormancy by delaying seed germination from 0.67% to 0.33%, that significantly different from the 0% PEG 6000 (17.67%) that was able to reduced fungal seed from 16.33 to 26.33% andwas not significantly different in each treatment. The second study showed that PEG 6000 can maintain viability and vigor of seeds with seed germination above 97%, seedlings height 22.40 cm and number of leaves was 7.16. PEG 30% is the best concentration to increase the shelf life period of rubber seed andmaintained morphological characteristics after storage period. The higher the concentration of PEG 6000 the higher were the seed viability during shelf. The morphology and growth of the seedlings, had the same adaptability to the tub germination in planting medium.

Abstract

Rubber seed will loose its storability in a short time. Seed germination and fungal attack were factors that barrier the storage period. The research aim was to test the ability of the potential osmotic solution and fungicides to reduce seed germination. Completely randomized design with two factors and three replications, i.e: PEG 6000 (w/v): 0%, 15%, 30%, 45%, 60% (potential osmotic solution) and fungicide (active ingredients were pyraclostrobin + metiram ) (g / 1 kg seed): 0 g, 10 g, 20 g, 30 g and 40 g, were applied. The results showed that PEG 30% can inhibit seed germination up to 9.07% and 37.47% and fungicides 40 g/1kg can reduce fungal attack during storage of 12 and 16 days.  Combination of PEG-6000 30% and fungicides 40g / 1 kg could maintain seed storability by pressing the seeds germination up to 10.67% and fungal attacks up to 18.00% during storage of 16 days with 96.80% germination.

Abstract

Preliminary studies in Screening of Potato (Solanum tuberosum L.) as a result of drought stress (in vitro). This study aim was to determine the limits of potato (Solanum tuberosum L.) to drought stress conditions in vitro using PEG. This research was conducted at the Tissue Culture Laboratory Unit / Laboratorium Kultur Jaringan UPT/Balai Benih Induk Hortikultura Dinas Pertanian Propinsi Sumatera Utara, Jl. Karya Jasa No. 6 Gedung Johor Medan in March 2012 to June 2012. The Completely Randomized Design (non factorial) was used with PEG concentrations of five levels, namely: P1 = 10%, P2 = 20%, P3= 30%, P4 = 40% and P5 = 50% with 5 replications. The results showed that increasing the concentration of PEG will reduce the percentage of plantlets survival, reduce plant height, number of roots and number of nodes. Treatment of 50% PEG resulted in the death of potato cuttings due to drought stress.
