Abstract

Effect of ethanolic extract of cashew leaf (Anacardium occidentale L.) in the feed on the weight gain and immunity were investigated on Jawa Super Chicken. Ninety chicken were divided into 6 groups those were basal feed group and 5 different concentration of chasew leaf extract groups. Experiment was conducted for 16 days started by 2 days acclimatization after hatching. Body weight was measured every 3 days starting from 0th day. On 16th day, the chicken were sacrificed, spleen and bursa of Fabricius were collected to quantify some histological parameters such as white pulp and follicle area as well as follicle thickness of those organs. The data were analyzed by regression and one way ANOVA followed by Tuckey test. Histological analysis was performed using optilab and Image Raster software. The results showed that growth and development of immune system of chicken increased significantly as could be seen in body weight, organs weight and index, as well as histological parameters. The regression results showed that spleen and bursa of Fabricius weight influenced increasing of body weight by 64.2% and 38.4% respectively. It could be concluded that biosupplementation of cashew leaf extract in basal feed could increase growth and immunity of Jawa Super Chicken.

Abstract

Coffee fruit skin contains antioxidant compounds that can repair damaged tissues, especially those of reproductive or-gans. This study was designed to assess to what extent the metha-nolic extract of coffee fruit skin affected sperm characteristics and testicular histology in mice receiving 15% ethanol for 15 days. It used 25 male mice aged four months and employed a completely randomized design with two controls (K) and three treatments (P), namely K1 (without ethanol), K2 (15% ethanol), P1 (15% ethanol and the methanolic extract of coffee fruit skin, or MECS, at 125 mg/ kg BW), P2 (15% ethanol and MECS 250 mg/kg BW), and P3 (15% ethanol and MECS 500 mg/kg BW). These treatments were administered orally with a 1 mL disposable syringe for 15 days. The parameters observed were sperm characteristics (viability, morphology, and sperm count) and the microscopic structure of the testicles (lu-men area, diameter, and area of seminiferous tubules). These data were then analyzed using the one-way ANOVA, continued with the LSD and Duncan’s tests (P <0.05). The results showed that P2 (250 mg/kg BW) substantially improved sperm count (86.5x105±1.73 cells/ mL), sperm viability (87.26±0.05%), and the proportion of normal sperm morphology (93.33±0.004%) of mice with 15% ethanol-in-duced sperm damages. Testicular histology also confirmed improve-ments in spermatogenic cells, as evident from the lumen, diameter, and area seminiferous tubules after receiving P1 and P2. In conclu-sion, the optimum dose of the coffee skin extract for improving sperm quality and microscopic structures of mice testicles is 250 mg/kg BW.

Abstract

Consumption of alcoholic beverages can increase ROS and inflammation. In the duodenal, the consumption of alcohol can result in erosions, bleeding, and mucosal injury. Arabica coffee fruit skin contains antioxidant substances, which are expected to have a good role as an antioxidant effect. Currently, unknown effects of alcohol and extract of Arabica coffee fruit skin administration on the damage of the small intestine of mice (Mus musculus L.). This study used an experimental design, and 25 males of mice were divided into five groups of treatment for 15 days. After the treatment, the mice at necropsied and the small intestine were taken for histopathological examination using hematoxyline-eosin staining. The Data analyzed used One Way ANOVA and Duncan's tests. Histopathological observations showed a dose of 250 mg/kg BW experienced the most significant improvement damage of the small intestine of mice compared to other treatments. Histomorphometry measurement, The dose of 250 mg/kg BB showed the high of villi 272,10 ± 13,36 µm, (P<0.05), duodenal villi crypt depth 117,33 ± 30,35 µm (P<0.05) and surface area of small intestinal villi 7,66 ± 0,55 mm2 (P<0.05) was similar with negative control. The conclusion of this study is a dose of 250 mg/kg BW treatment was able to improve the histopathology and morphometry small intestine of mice that were damaged due to consumption of ethanol 15%.

Abstract

Maggot (Black soldier fly larvae) as an alternative source of protein on feed can increase fish productivity. The research aims to study maggot flour as an alternative source of protein in feed on the growth, structure of the intestine organs and skeletal muscles of Mutiara catfish (Clarias gariepinus Burchell, 1822). The research used 144 Mutiara catfish that were kept for 21 days. The research used a completely randomized design consisting of 4 groups, namely 0% maggot in feed (Control), 25% maggot in feed (P1), 50% maggot in feed (P2), 75% maggot in feed (P3). The parameters consist of the growth, the structure of intestine and skeletal muscle of Mutiara catfish. Growth observations consist of measurements of the absolute length and weight of the fish. Intestine observations consisted of length and weight, as well as tissue structure consisting of villi height, crypt depth, villi / crypt ratio and villi area area. Skeletal muscle observations consist of muscle weight and tissue structure consisting of the diameter and area of muscle fibers. Data analysis was performed with one-way anova, followed by the Duncan Test. The results showed that 75% maggot feed gave significant results on absolute weight, structure of intestine and muscle organs compared to other treatments (P <0.05). The absolute length did not provide a significant difference between treatments (P> 0.05). The conclusion shows that 75% maggot feed can be used as an alternative source of feed protein because it can increase the growth, structure of the intestine organs and skeletal muscles of the Mutiara catfish.

Abstract

Indonesia merupakan negara dengan angka kelahiran yang relatif tinggi, sehingga dibutuhkan alternatif dalam menurunkan jumlah penduduk seperti penggunaan alat kontrasepsi berbahan herbal. Penelitian bertujuan mengetahui pengaruh ekstrak etanol daun pepaya Calina terhadap kualitas sperma dan morfologi epididimis tikus wistar. Penelitian menggunakan 24 tikus wistar, dibagi menjadi 4 kelompok terdiri dari Kontrol (akuades), ekstrak dosis 100 mg/Kg BB, 200 mg/Kg BB, 300 mg/Kg BB yang diberikan menggunakan sonde lambung (1ml) selama 30 hari. Pada hari ke-31 tikus dibedah dan diambil cauda epididymis untuk dilakukan pengamatan kualitas sperma dan pembuatan preparat histologis dengan metode parafin (pewarnaan HE). Parameter kualitas sperma terdiri dari motilitas, viabilitas, jumlah, dan morfologi sperma. Parameter morfologi epididimis terdiri dari tebal sel epitel, diameter dan luas epididimis. Seluruh parameter dianalisis menggunakan uji one way Anova dan dilanjutkan uji Duncan dengan selang kepercayaan 95%. Hasil menunjukan pemberian ekstrak dosis 100-300 mg/Kg BB berbeda secara signifikan dibandingkan kontrol dengan menurunkan parameter motilitas, jumlah, viabilitas dan morfologi sperma (P<0,05). Morfologi epididimis menunjukan penurunan tebal epitel, diameter dan luas epididymis pada dosis 100-300 mg/kgBB dibandingkan dengan kontrol (P<0,05). Kesimpulan menunjukan ekstrak etanol daun pepaya Calina dosis 100 mg/kgBB memiliki potensi sebagai agen antispermatogenik ditandai dengan penurunan kualitas sperma dan morfologi epididymis tikus wistar.

Abstract

Cigarette smoke is a toxic compound that can interfere with physiological effects and damage the organ systems in the body. The effects of free radicals arising from cigarette smoke can be neutralized by consuming antioxidant compounds. The research aims to look at the protective effects of mixed juice from tropical fruits that grow in Indonesia to the histopathological picture of the kidney of wistar rats exposed to cigarette smoke. There were 35 rats used and divided into 5 treatment groups consisting of K (without treatment), K- (cigarette smoke exposed), P1 (strawberry and guava juice mixed juice 3ml), P2 (strawberry and papaya mixed juice 3ml), P3 (P3 (strawberry and apple mixed juice 3ml). In P1, P2 and P3 before being given a mixture of fruit juice, rats were exposed to cigarette smoke first. The treatment was carried out for 40 days by oral gavage, then on the 41st day the rats were dissected and their kidney organs were taken for observation and histopathological preparations. Histopathology preparations were made by the paraffin method and Haematoxylin Eosin staining. The cell damage data is then analyzed by comparing the kidney cells from the control group with the treatment group and scoring damage followed by the percentage of kidney damage. Data were statistically tested using ANOVA followed by Post Hoc LSD. The results showed that mixed tropical fruit juice could protect kidney cell damage by decreasing the percentage of hydropic degeneration, necrosis and infiltration of inflammatory cells compared with negative controls (P <0.05). There is no significant difference between P1, P2 and P3 in reducing cell damage. The conclusion of the study is that all combinations of tropical fruit juice can protect cell damage caused by free radicals from exposure to cigarette smoke in wistar rats.

Abstract

Indonesia merupakan salah satu negara dengan jumlah penduduk yang relatif tinggi, sehingga diperlukan upaya dalam menekan laju pertumbuhan penduduk seperti mengembangkan potensi obat antifertilitas berbahan dasar herbal. Daun pepaya Calina merupakan varietas tanaman pepaya di Indonesia yang memiliki beberapa kandungan fitokimia yang berpotensi sebagai agen antifertilitas. Penelitian bertujuan mengetahui potensi ekstrak etanol daun pepaya Calina sebagai agen antifertilitas terhadap Indeks Gonadosomatik (GSI) dan perkembangan folikel ovarium tikus Wistar. Penelitian dilakukan selama 30 hari menggunakan 25 ekor tikus Wistar betina umur ± 15 minggu. Perlakuan terdiri dari 5 kelompok yaitu kontrol (akuades), kontrol positif (pil kontrasepsi dosis 14,184 mg/kg BB), ekstrak etanol daun pepaya Calina dosis 200mg/Kg BB, 300 mg/Kg BB dan 400 mg/Kg BB yang diberikan secara oral gavage 1 ml. Parameter terdiri indeks gonadosomatik dan perkembangan folikel ovarium yang terdiri dari folikel primer, sekunder, de graaf, korpus luteum dan atresia. Pada hari ke 31 dilakukan pembedahan dan pengambilan ovarium pada tikus. GSI dihitung dengan membagi bobot ovarium dengan bobot badan tikus, sedangkan pengamatan folikel ovarium dilakukan dengan pembuatan preparat histologi menggunakan metode paraffin (Pewarnaan HE). Seluruh data dianalisis menggunakan One Way Anova dengan uji lanjut Duncan. Hasil menunjukan bahwa dosis 300 mg/Kg BB dapat menurunkan bobot ovarium, menurunkan indeks gonadosomatik, menurunkan jumlah folikel primer dan sekunder, menurunkan luas folikel de graaf, serta meningkatkan jumlah folikel atresia dibandingkan dengan kontrol (P<0,05). Kesimpulan penelitian adalah ekstrak etanol daun pepaya Calina dosis 300 mg/Kg BB berpotensi menjadi agen antifertilitas alami dengan cara menurunkan GSI dan perkembangan ovarium tikus Wistar.

Abstract

Land conversion around campus 4 of Ahmad Dahlan University Yogyakarta affects the diversity and abundance of pollinating insects, one of which is bees. The purpose of this study was to calculate the level of diversity and abundance of bees on campus 4 UAD and its surroundings. The sampling area consisted of 4 plots, with each plot measuring 1750 m2( 35 m x 50 m) determined using the observation method. Sampling was carried out three times, which was carried out in the morning at 07.00 – 11.00 and continued in the afternoon at 15.00 – 17.00. Bees were identified by comparing their morphological characters with identification reference books and journals. The Spearman correlation test then analyzed the bee abundance data and abiotic factors. The results showed that the level of bee diversity on campus 4 UAD and its surroundings was moderate (1 H' 3). The most abundant bee species was Xylocopa aestuan, with 118 individuals, and the least abundant was Apis mellifera, with 7 individuals. The conclusion of this study is the state of the ecosystem on campus 4 UAD and its surroundings, there is a disturbance in the form of land use change, but bees can still tolerate the disturbance.

Abstract

Cigarette smoke is a source of free radicals that cause health problems throughout the world. Indonesia is a country that has many sources of natural antioxidants in counteracting free radicals, one of which is Calina papaya leaves. The study aimed to determine the potential of Calina papaya leaves antioxidants in improving the respiratory organs of Wistar rats after being exposed to cigarette smoke. The study using 25 male Wistar rats consisted of Control (not exposed to cigarette smoke and given distilled water), KN (cigarette smoke and given distilled water), P1 (cigarette smoke and Calina Papaya Leaves Ethanolic extracts (CPLE) 100 mg/kg BW), P2 (cigarette smoke and CPLE 200 mg/kg BW), and P3 (cigarette smoke and given CPLE 300 mg/kg BW). Exposure to cigarette smoke and administration of extracts was carried out for 21 days. On day 22, the rats were euthanized and dissected to remove the lungs and trachea. Organs were prepared using the paraffin method and hematoxylin-eosin staining. Parameters consisted of histopathological observations of the lungs and trachea. All data parameters were analyzed using a one-way ANOVA test and Duncan's advanced test (p<0.05). The results showed that there was an improvement in histopathology of the lungs and trachea at a dose of 200 mg/Kg BW compared to other doses (p<0.05). The study showed that the Calina papaya leaves ethanolic extracts have the potential as a source of antioxidants in improving the respiratory organs of Wistar rats after being exposed to cigarette smoke.

Abstract

This study aims to determine the effect of Papaya Calina (Carica papaya L.) leaves ethanol extract on the blood profile of Wistar rats. The study used 20 female Wistar rats (Rattus norvegicus Berkenhout, 1769), divided into 4 groups consisting of control (aquadest), Calina papaya leaves ethanol extract at a dose of 200 mg/Kg BW, 300 mg/Kg BW, and 400 mg/Kg BW for 30 days using a 1ml gastric probe. On the 31st day, blood samples were taken from the orbital sinus using a 2 ml hematocrit pipette. Blood profile parameters include erythrocyte levels, hemoglobin, hematocrit, total leukocytes, leukocyte differential, N/L ratio, and platelets. Blood samples were analyzed using a hematology analyzer to observe the levels of erythrocytes, hemoglobin, hematocrit, total leukocytes, neutrophils, and lymphocytes as well as making a blood smear (Giemsa stain) to observe eosinophils, basophils, and monocytes. All parameters were analyzed using ANOVA test and Duncan's follow-up test with a significant level of P<0.05. The results showed that the levels of erythrocytes, hemoglobin and hematocrit were significantly different between groups with the most optimum treatment at a dose of 300 mg/Kg BW. There was a significant decrease (P<0.05) in total leukocyte levels at a dose of 400 mg/Kg BW compared to the control, but there was no significant difference in all types of leukocytes. The dose of 400 mg/Kg BW increased the N/L ratio significantly compared to the control (P<0.05). Platelet counts showed no significant difference between groups (P> 0.05). The conclusion shows that the Calina papaya leaves ethanol extract at a dose of 300 mg/Kg BW can increase the number of erythrocytes, hemoglobin, and blood hematocrit of Wistar rats. The extract at a dose of 400 mg/Kg BW affected the decrease in total leukocytes and an increase in the N/L ratio, but was still in normal condition.
