Abstract

Abstract

Asiaticoside, a bioactive compound derived from Glycyrrhiza glabra (licorice root), has garnered attention for its potential as a hypolipemic agent. This study explores the lipid-lowering effects of asiaticoside, focusing on its mechanism of action and pharmacological properties. In silico analysis showed a high activation probability (pa=0.934) for asiaticoside, indicating its strong potential to interact with targets involved in lipid metabolism, such as PPAR-α, and reduce blood lipid levels. Additionally, its low inhibition probability (pi=0.003) suggests minimal risk for non-specific biological interference, making it a promising candidate for therapeutic use with a favorable safety profile. Asiaticoside’s antioxidant and anti-inflammatory properties further enhance its potential for cardiovascular protection by reducing oxidative stress, a key factor in atherosclerosis. While preliminary in silico results are promising, further in vitro and in vivo studies are required to confirm its efficacy, safety, and pharmacokinetics. Overall, asiaticoside represents a promising natural agent for managing hyperlipidemia and related metabolic disorders, offering an alternative to conventional synthetic therapies.

Abstract

This study evaluates the ADMET (Absorption, Distribution, Metabolism, Excretion, and Toxicity) profile of nuciferine to assess its potential as a drug candidate. Based on in silico analysis, nuciferine demonstrates favorable oral absorption and extensive systemic distribution, including its capability to cross the blood-brain barrier into the central nervous system. Regarding metabolism, the compound acts as a substrate for CYP2D6 and CYP3A4 enzymes while serving as an inhibitor of CYP1A2 and CYP2D6, which may lead to drug-drug interactions. In terms of excretion, nuciferine displays moderate clearance values and utilizes renal transporter OCT2. However, toxicological evaluations suggest potential risks, including mutagenicity (positive AMES test), hepatotoxicity, and inhibition of hERG II potassium channels, which are associated with cardiotoxicity risks. Despite its favorable bioavailability and distribution properties, concerns about chronic toxic effects and metabolic interaction risks underscore the need for careful consideration in its development. Further research is necessary to validate its safety and efficacy for therapeutic applications.

Abstract

The estrogen receptor β (ERβ) molecule is one of the target proteins in cancer treatment. The type of ERβ molecule in the protein data bank (PDB) determines the value of most favored regions and MolDock score. The purpose of this study was to determine the effect of most favored regions of ERβ molecules on MolDock score using Tamoxifen ligand. There are 37 molecules that have ERβ protein, 7 molecules were selected to be analyzed for the value of most favored regions and MolDock score, based on the r table value, pearson correlation value 0.548 < 0.754 (r table). This is also the same when viewed from the significance value of 0.203 > 0.05. It can be concluded that there is no correlation between the value of most favored regions and MolDock score.

Abstract

Background: The prevalence of breast cancer continues to increase, it is the second leading cause of death after lung cancer. Estrogen receptor beta (ERβ) has an important role in breast cancer pathology, activation of Akt pathway will trigger E3 ubiquitin ligase murine double minute 2 (MDM2) to ERβ, which will increase the risk of breast cancer. Thus, both proteins have potential as therapeutic agents in breast cancer drug development. This study aims to find breast cancer drug candidates from natural products and compare the effectiveness of these compounds with Doxorubicin and Paclitaxel.Method: ERβ (PDB ID: 3OLS) and MDM2 (PDB ID: 1T4E) proteins were combined using ClusPro 2.0. Doxorubicin and Paclitaxel ligands were obtained from PubChem, there are 842 natural products obtained from the ZINC database, when the energy minimization is reduced to 839 natural products. Virtual screening between proteins and ligands was performed using PyRx 8.0, followed by analysis of amino acid residues resulting from interactions between proteins and ligands using protein interaction calculator (PIC) and protein ligand interaction profiler (PLIP). Results: Ergoloid compounds have the lowest binding affinity compared to doxorubicin and paclitaxel compounds, and are able to interact strongly with the ERβ-MDM2 Protein as determined from the results of interactions between proteins and ligands using PIC and PLIP.Conclusion: Ergoloid compounds can interact well with ERβ-MDM2 Protein. Thus, it can be used as a breast cancer drug candidate in the future. In vitro, in vivo, and biochemical testing needs to be done to confirm this discovery

Abstract

Antibiotic resistance in gonorrhoea cases and patients is detrimental. Amidst the rising cases of gonorrhoea caused by Neisseria gonorrhoeae, Hygrophila auriculata (Schum.) Heine has activity as an antibiotic for the disease. The penicillin-binding protein 2 (PBP2) is a transpeptidase that catalyses the formation of cross-bridges on bacterial cell wall peptidoglycan, which will be the target of this plant. The purpose of molecular docking study is to see the binding affinity, compounds in Hygrophila auriculata (Schum.) Heine and ceftriaxone which is used as a comparison drug, will be targeted at PBP protein. Discovery studio visualizer v21.1.0.20298 was used for PBP2 protein preparation and visualisation. DoG Site Scorer was used to predict ligand binding sites on PBP2 proteins. PyRx 0.8 was used for virtual screening, validating of the docking method, and ligand preparation. Compounds in Hygrophila auriculata (Schum.) Heine as ligands were derived from MPDB 2.0 and the following PubChem codes; apigenin CID 5280443, luteolin CID 5280445, allagic acid CID 5281855, gallic acid CID 370, quercetin CID 5280343, lupeol CID 259846, lupenone CID 92158, betulin CID 72326, stigmasterol CID 5280794, and comparator drug ceftriaxone CID 447043. The binding affinity of ellagic acid -9,8 from Hygrophila auriculata (Schum.) Heine was lower than ceftriaxone -9,4 on the target PBP2 protein. Some of the amino acid residues that appear in protein-ligand docking include: ALA A:310, THR A:500 and 347, LYS A:313, and SER A:362. These amino acid residues owned by the PBP2 protein serve as the bonding bridge. Ellagic acid, the compound has potential as an antibiotic in gonorrhoea. Further testing and studies are needed to strengthen the evidence of the findings in this study

Abstract

This community service program aimed to increase adolescents' awareness and knowledge regarding the role of Vitamin A (Retinol) in preventing myopia, particularly in rural areas such as Gembleb Village, Trenggalek. The activity was conducted on Saturday, May 3, 2025, in collaboration with the Government of Gembleb Village and supported by the Institute for Research, Community Service, and Publication (LP3M) of Universitas Kadiri. A total of 15 adolescents participated in the educational intervention, which included a pre-test, a lecture session, and a post-test evaluation. The results showed a significant improvement in participants’ knowledge about myopia and the importance of Vitamin A for eye health. Pre-test scores averaged lower compared to post-test results, with an average increase of 20–25 points. This finding suggests that structured education can effectively enhance health literacy among youth, particularly regarding nutritional prevention of visual disorders. These outcomes underscore the importance of continued community-based health education programs, especially in underserved areas, as a strategic effort to reduce the prevalence of preventable eye diseases
