Abstract

This study aims to determine the content of nickel (Ni) and iron (Fe) as well as histopathological analysis of marine fish in Obi Island waters as a bioindicator of pollution. Besides, water quality conditions were carried out in-situ and ex-situ observations. The parameters observed were temperature, brightness, salinity, pH, dissolved oxygen, nitrate, orthophosphate, ammonia, iron (Fe), and nickel (Ni). The results showed the temperature range between 26.48 ℃ to 27.99 ℃ below the quality standard or low temperature. The brightness of the relationship between 12 m and 13 m is under quality standards. The salinity range between 31.01 ppt to 32.13 ppt below the quality standard. The pH range is from 8.6 to 8.7 in high or alkaline conditions. Ammonia range between 0.4 mg / L to 0.7 mg / L exceeds the quality standard. The range of nitrate between 0.009 mg / L to 0.012 mg / L exceeds the quality standard. The range of phosphate between 0.016 mg / L to 0.019 mg / L exceeds the quality standard. The DO range between 3.68 mg / L to 3.77 mg / L lower than the quality standard. The metal range of 0.6 mg / L to 0.9 mg / L exceeds the quality standard. The range of Ni metal between 0.06 mg / L to 0.09 mg / L exceeds the quality standard. Histopathological analysis showed that the liver had a hemorrhage, degeneration of blood vessels, vacuolate degeneration, necrosis, or cell death. The muscles experience edema, degeneration of muscle fibers, atrophy of muscle bundles, vacuolar degeneration of muscle Bundles, hemorrhage, infiltration of lymphocytes, and necrosis. The intestine experience infiltration of lymphocytes, melanomacrophages, and necrosis. While P. tayenus fish ovaries showed necrosis structure oocytes. This research can be a reference for warning of heavy metal pollution in Obi Island waters, binding to the nature of heavy metals that can accumulate in fish tissue.Keywords: Water quality; Heavy Metal; Pollution; Histopathological; Obi Island.

Abstract

The potential of aquaculture in North Maluku Province, especially the Obi islands, has not been maximally utilized. One commodity that can be developed is Whiteleg shrimp. This commodity is a major aquaculture commodity with important economic value. Analysis of the suitability of waters for Whiteleg shrimp cultivation is an early stage of activity that greatly determines the success of cultivation. Therefore, this study aims to assess the suitability of waters for the development of Whiteleg shrimp aquaculture on the island of Obi. This study was conducted in the waters of Soligi, Kawasi, Baru, Akegula, Laiwui, Buton, Jikotamo, Sambiki, and Anggai of Obi Island, South Halmahera Regency. Water quality parameters observed were temperature, brightness, depth, current speed, salinity, pH, dissolved oxygen, nitrate, and phosphate. Each parameter was then analyzed by land suitability. The results showed that the waters of Obi Island have the potential for the development of Whiteleg shrimp cultivation based on the observed water quality parameters. This study concludes that the waters of Obi Island, South Halmahera Regency, North Maluku Province can be used to develop Whiteleg shrimp farming. The location of the waters has the criteria of suitability is very suitable for the cultivation of Whiteleg shrimp. Keywords : Penaeus vannamei, Obi Island, water quality.

Demand for this commodity is quite high, as reflected in the increasing volume of exports each year. Fulfillment of these demands is obtained from the production of cultivation. Generally farmers get natural seaweed seedlings, namely cuttings from existing seaweed. The continuous use of seeds from nature can cause deterioration in the quality and quantity. Handling the problem of quality deterioration from seaweed seeds originating from nature, can be overcome by multiplying the seeds through tissue culture methods in vitro. In term of tissue culture techniques method, the most important thing to note is environmental parameters. An environment parameter that changes suddenly such as salinity can inhibit the growth of seaweed. Thus, this work is attampting the different salinity treatment on seaweed explants K. alvarezii. This study aims at determining (weight) the explants of K. alvarezii with a comparison of the different salinity levels in the in vitro tissue culture method. The method used in this study was a completely randomized design (CRD) with the different salinity treatments namely 30, 31, 32, 33, and 34 ppt. The results showed that the different salinities influenced the growth rate of K. alvarezii seaweed explants with the best explant growth at the salinity of 31 ppt, while the lowest growth value was obtained at 34 ppt

Abstract

ABSTRAK Penelitian ini bertujuan untuk mengisolasi dan estimasi kepadatan koloni bakteri Vibrio spp. pada bulan September - Januari 2023 di tambak Balai Budidaya Laut dan Payau (BBLP) Desa Porniti (Lokasi 1) dan Unit Pelaksana Teknis Daerah (UPTD) Dinas Kelautan dan Perikanan Desa Tuada (Lokasi 2) Kabupaten Halmahera Barat. Pengambilan sampel dilakukan dengan metode survei dan sampling langsung pada kedua tambak. Pengukuran kualitas air dilakukan secara insitu terhadap suhu, salinitas, DO,suhu, dan pH. Isolasi dan kepadatan bakteri dihitung pada organ hepatopancreas udang dan media air. Kepadatan bakteri Vibrio dianalisis menggunakan uji t sedangkan data lainnya dianalisa secara deskriptif dan ditampilkan dalam tabel dan grafik. Hasi analisis menunjukan bahwa kepadatan bakteri Vibrio spp pada lokasi 1 mencapai rata-rata 3,0x102 CFU/g dan media air mencapai rata-rata 1,2x103 CFU/mL sedangkan pada lokasi 2 mencapai rata-rata 4,2x102 CFU/g dan media air mencapai hingga 1,6x103 CFU/mL. Kedua lokasi ditemukan bakteri Vibrio spp. Pada organ hepatopancreas dan media air. Kepadatan bakteri tertinggi diperoleh di lokasi kedua.Kata kunci:  Tambak, vaname, Vibrio, Densitas, HepatopankreasABSTRACTThis study aims to isolate and estimate the colony density of Vibrio spp. bacteria in September - January 2023 in the ponds of the Marine and Brackish Aquaculture Center (BBLP) Porniti Village (Location 1) and the Regional Technical Implementation Unit (UPTD) of the Marine and Fisheries Service Tuada Village (Location 2) West Halmahera Regency. Sampling was conducted using survey and direct sampling methods in both ponds. Water quality measurements were carried out in situ on temperature, salinity, DO, temperature, and pH. Isolation and density of bacteria were calculated in the shrimp hepatopancreas organ and water media. Vibrio bacteria density was analyzed using t test while other data were analyzed descriptively and displayed in tables and graphs. The analysis showed that the density of Vibrio spp bacteria at location 1 reached an average of 3.0x102 CFU/g and water media reached an average of 1.2x103 CFU/mL while at location 2 reached an average of 4.2x102 CFU/g and water media reached up to 1.6x103 CFU/mL. Both locations found Vibrio spp. bacteria in the hepatopancreas organ and water media.
