Abstract

This research aimed to observe quantitatively the effect of temperature and pH on protease activities from thermophilic bacteria collected from Lejja—Hot springs.  Spectrophotometry on the casein substrate was used to test the proteolytic activity of the crude protease.  Mixtures of enzyme and casein were incubated at various temperatures and pH for 20 minutes. The absorbance of tyrosine from protein hydrolysis was determined by spectrophotometry on λ 280 nm. Temperature and pH impacted on protease activity were determined at temperature (60; 65; 70; 75; 80; 85 and 90oC) and pH (6.0; 6.5; 7.0; 7.5; 8, 0; 8,5 and 9,0). Results showed that treatment of temperature variations and pH had a significant effect on protease activity. Crude extract of Bacillus licheniformis protease showed that the highest activity at 80oC and pH 7.5 was 0.1303 unit/ml/minute. Bacillus stearoformis showed the highest enzymatic activity of the protease at 85oC, and pH 7.5 was 0.1226 Unit/ ml/minute. In comparison, Bacillus coagulans reached optimum activity at 75oC, and pH 7.5 was 0.2052 Unit/ml/minute. Isolates of Bacillus licheniformis, B. coagulans and B. stearoformis are bacteria that produce thermostable protease enzymes that can be developed as a source of genes and as a producer of the enzyme itself.

Abstract

Kegiatan pengabdian kepada masyarakat ini bertujuan untuk meningkatkan literasi digital sivitas akademika Politeknik Penerbangan Indonesia Curug (PPIC) melalui penyelenggaraan workshop edukatif bertema keamanan siber, khususnya pada isu serangan phishing dan teknik mitigasinya. Pendekatan pembelajaran yang digunakan mengacu pada prinsip andragogi Malcolm Knowles, dengan mengedepankan relevansi materi, partisipasi aktif peserta, serta orientasi pembelajaran pada pemecahan masalah nyata. Kegiatan diikuti oleh 75 peserta yang terdiri dari tenaga pengajar, pegawai, dan taruna, dengan metode evaluasi menggunakan kuesioner skala Likert dan pemetaan ke dalam enam prinsip andragogi. Hasil evaluasi menunjukkan skor tinggi pada dimensi readiness to learn, orientation to learning, dan self-concept, yang mencerminkan kesiapan dan komitmen peserta dalam menerapkan hasil pembelajaran. Kegiatan ini juga relevan secara strategis, mendukung prioritas nasional dalam Program Asta Cita, sejalan dengan visi Telkom University dan peta jalan Center of Excellence Artificial Intelligence for Learning and Optimization (CoE AILO), serta berkontribusi pada pencapaian Sustainable Development Goals (SDGs). Temuan ini menunjukkan bahwa pendekatan andragogi efektif untuk kegiatan literasi digital di lingkungan pendidikan vokasi dan dapat direplikasi untuk penguatan ketahanan digital kelembagaan.

Abstract

This research examines how women struggle in the film Secret Superstar by director Advait Chandan which will be presented using the study of liberal feminism theory. The object that is the focus of this research is the resistance of the main character, a young girl named Inisa Malik who dreams of becoming a famous singer amid the helplessness experienced by her and her mother because her father claims that women are just a useless burden. This film tells how Insia struggles to make her dream come true with her father's strict circumstances and strictly forbids Insia from becoming a singer. The purpose of this study is to analyze the dialogue in the film to find out how the resistance is carried out by female characters who do not have freedom in living their lives and must live in fear, injustice and powerlessness due to the views of the father who thinks that only he has the right to regulate and make decisions. for the lives of the people in his family. The method used in this study is a qualitative method based on dialogue text selected in the film Secret Superstar as the data to be analyzed.   Pada penelitian ini mengkaji mengenai bagaimana perjuangan perempuan dalam film Secret Superstar karya sutradara Advait Chandan yang akan disajikan dengan menggunakan kajian teori feminisme liberal. Objek yang menjadi fokus dalam penelitian ini ialah resistensi dari tokoh utama seorang remaja perempuan bernama Inisa Malik yang memiliki mimpi untuk menjadi seorang penyanyi terkenal ditengah ketidakberdayaan yang dialami ia dan ibunya karena sang ayah yang mengklaim bahwa perempuan hanyalah sebuah beban yang tidak berguna. Film ini menceritakan bagaimana perjuangan Insia yang berusaha mewujudkan impiannya dengan keadaan sang ayah yang bersifat keras dan sangat melarang Insia menjadi seorang penyanyi. Tujuan dari penelitian ini adalah untuk menganalisis dialog dalam film untuk mengetahui bagaimana perlawanan yang dilakukan oleh tokoh perempuan yang tidak memiliki kebebasan dalam menjalani kehidupannya dan harus hidup dalam ketakutakan, ketidakadilan serta ketidakberdayaan akibat pandangan sang ayah yang menganggap bahwa hanya ia yang berhak mengatur dan mengambil keputusan untuk kehidupan orang orang dalam keluarganya. Metode yang digunakan dalam penelitian ini ialah metode kualitatif berbasis teks dialog yang dipilih dalam film Secret Superstar sebagai data yang akan akan dianaliis.

Abstract

Abstract: Bagasse is a solid waste from the sugar cane milling process in the sugar industry. The waste can be used as an ingredient in making silica gel which functions as an adsorbent. This can be done by changing its chemical structure composition to increase its role and function. This study aims to determine the characteristics of silica gel synthesized from bagasse (Saccharumofficinarum L.) and modified using diphenylcarbazone as much as 0.24 g with sol-gel technique. The characterization results obtained from Fourier transform infrared (FTIR) analysis of diphenylcarbazone modified silica gel, namely the absorption of Si-OH, Si-O-Si groups supported by the appearance of C꞊O, C꞊N, NH, and N꞊N groups. Characterization using x-ray diffraction (XRD) showed a change in amorphous silica to crystals, with the results obtained showing a crystal size of 41.468 nm.Abstrak: Ampas tebu merupakan limbah padat yang berasal dari proses penggilingan tebu pada produksi gula. Limbah tersebut dapat dimanfaatkan sebagai bahan pembuatan silika gel yang berfungsi sebagai adsorben. Hal tersebut dapat dilakukan dengan mengubah komposisi struktur kimianya agar dapat meningkatkan peran dan fungsinya. Penelitian ini bertujuan untuk mengetahui karakteristik silika gel yang disintesis dari ampas tebu (Saccharumofficinarum L.) dan dimodifikasi menggunakan difenilkarbazon sebanyak 0,24 gr dengan teknik sol-gel. Diperoleh hasil karakterisasi dari analisis fourier transform infrared (FTIR) terhadap silika gel termodifikasi difenilkarbazon menunjukkan adanya serapan gugus Si-OH, Si-O-Si yang didukung dengan pembentukan gugus C꞊O, C꞊N, N-H, dan N꞊N. Karakterisasi menggunakan difraksi sinar-x (XRD) menunjukkan perubahan silika amorf menjadi kristal dengan hasil yang diperoleh memperlihatkan ukuran kristal sebesar 41,468 nm.

Abstract

High egg consumption in Indonesia, particularly in South Sulawesi, presents a significant opportunity to utilize eggshells as precursors for hydroxyapatite. This study aims to characterize hydroxyapatite derived from chicken eggshells and to evaluate the absorption properties of chitosan-hydroxyapatite composites for methylene blue removal. The precipitation method was used for hydroxyapatite synthesis, as it produces hydroxyapatite with a high level of purity and low contamination. X-ray diffraction (XRD) analysis revealed that eggshells contain approximately 30% CaCO₃. The XRD diffractogram of hydroxyapatite from eggshells showed a peak at 2θ = 31.94º. Fourier-transform infrared (FTIR) spectroscopy identified key functional groups in hydroxyapatite, including hydroxyl (-OH) at 3434 cm⁻¹, carbonate (-CO₃²⁻) at 1421 cm⁻¹, phosphate bending (-PO₄³⁻) at 565 cm⁻¹, and phosphate stretching (-PO₄³⁻) at 1035 cm⁻¹. The chitosan-hydroxyapatite (Cs-HAp) composite showed the incorporation of chitosan functional groups, including amine (-NH) at 1577 cm⁻¹ and methylene (-CH) at 2928 cm⁻¹. The chitosan-hydroxyapatite composite demonstrated effective removal of methylene blue dye, with an absorption efficiency of 96% at a Cs-HAp ratio of 2:2 within 180 minutes. Based on the high adsorption efficiency, the chitosan-hydroxyapatite composite shows great potential for absorbing methylene blue.

Abstract

Purebred chicken eggshell is a promising natural source because it contains high calcium carbonate (CaCO3), which is used as a hydroxyapatite (HAp) precursor, with natural abundance and biodegradability, making it an eco-friendly alternative to synthetic materials. HAp is relevant for dye removal because of its high adsorption capacity, chemical stability, and ability to interact with dye molecules through ion exchange and surface interactions with the material. The composite adsorbent of Hap-zeolite is effective for wastewater treatment based on the result of HAp-zeolite composite's adsorption capacity against methylene blue. Methylene blue is one of the dyeing wastes originating from the textile industry. The method of synthesis of Hap used precipitation. The FTIR characterization results showed OH groups at wavenumber 3434 cm-1, CO32- group with wavenumber 1421 cm-1, PO43- bending group with wavenumber 565 cm-1, and PO43- stretching at wavenumber 1035 cm-1. The highest peak XRD yield at an angle of 2θ=34.04º corresponds to a hexagonal crystal formed. The adsorption process is carried out with a HAp-zeolite mass ratio of 4:2 (w/w), with contact times for 180 minutes, resulting in the best adsorption efficiency around 99.99% measured via UV-Vis spectrophotometry. The results indicate that HAp-zeolite composites are highly effective for methylene blue removal and have potential application in wastewater treatment.
