Abstract

Efficacy and quality of medicinal plant extracts depend on chemical composition therein. Therefore, to ensure its efficacy orquality, the chemical composition of extracts have to be analyzed based on its chemical marker or its chemical pattern. This study tried to develop a method for extracts quality assay based on infrared spectrum in fingerprint region and extracts inhibition activity to xanthine oxidase. Five extracts from kunyit (Curcuma domestica), temulawak ( C xanthorriza), jahe (Zingiber officina/e), temukunci (Boesenbergia pandurata), and cabe jawa (Piper retrofractum) were tested to inhibit xanthine oxidase activity. The bioactive data of extracts and its infrared spectrum was analyzed using PCA and PLS-DA (partial least squarediscriminant analysis). Almost all extracts that were tested showed inhibition activity to xanthine oxidase activity, except the extract of jahe. The result showed that PCA can group all of extract into each of their region. Meanwhile, PLS-DA has shown to be a good prediction model for classifying extracts based on fingerprint region of its mR Spectrum and its biological activity. Abstract

Mangroves are a source of traditional medicine that can be used as a source of bioactive compounds. With the conversion of mangrove ecosystem into another designation led to the extinction of mangrove ecosystems. Therefore we need a good management of natural resources. In natural resource management, biodiversity information is needed to sustain the species utilization, exploration potential of the species and their biological and ecological monitoring, policy making, and for the development of biotechnology innovation. Research center of IPB Biopharmaca (Institute for Research and Community Services of Bogor Agricultural University) has the mandate to conduct research from upstream to downstream in the medicinal field. This study develops Indonesian mangrove biodiversity database for Biodiversity Informatics. Biodiversity informatics (BI) is the development of computer-based technologies for the management of biodiversity information. BI can be used to improve the knowledge management (knowledge management), exploration, analysis, synthesis, and interpretation of data ranging from the level of genomic biodiversity, species level to the ecosystem level. From the results of this study are expected data, information and knowledge of natural wealth mangroves can be managed properly so that the preservation of natural resources can be properly maintained and can be used in particular to the field of medicinal studies.

Abstract

Tabat Barito (Ficus deltoidea) is known as a plant that has potency as an antioxidant because of containing a significant phenolic compound. In this study, we performed metabolite profiling on F. deltoidea leaves by UPLC-QTOF-MS/MS to identify its phenolic compounds. Extraction by maceration and ultrasonication techniques with methanol (MM and UM) and ethanol (ME and UE) were used to extract the F. deltoidea metabolites. About 70 metabolites were identified by using UPLC-QTOF-MS/MS in negative ion mode. The amounts of metabolites found in each extract were different, i.e., 45 metabolites in MM, 64 metabolites in UM, 42 metabolites in UE and 41 metabolites in ME. MS/MS could further tentatively identify 16 metabolites. The identified compounds belonged to the class of flavonoids and phenolic acid. Also, we conducted an antioxidant activity by using DPPH method on each extract to determine its potency as an antioxidant. The highest antioxidant activity was exhibited by UM extract (IC50 71.93 ppm) may be due to the number of metabolites in UM extract which was higher than the other extract based on the detected metabolites.

Abstract

In this paper, we described the use of a light emitting diode (LED)-based photometer and chemometric analysis for quality control of king of bitter or sambiloto (Andrographis paniculata) raw material. The quality of medicinal plants is determined by their chemical composition. The quantities of chemical components in medicinal plants can be assessed using spectroscopic technique. We used an “in house” photometer to generate spectra of sambiloto. The spectra were analyzed by chemometric methods, i.e. principal component analysis (PCA) and partial least square discriminant analysis (PLS-DA), with the aim of herbal quality classification based on the harvesting time. From the results obtained, based on thin layer chromatography analysis, sambiloto with different collection times (1, 2, and 3 months) contained different amounts of active compounds. Evaluation of sambiloto, using its spectra and chemometric analysis has successfully differentiated its quality based on harvesting time. PCA with the first two PC’s (PC-1 = 60% and PC-2 = 35%) was able to differentiate according to the harvesting time of sambiloto. Three models were obtained by PLS-DA and could be used to predict unknown sample of sambiloto according to the harvesting time

Abstract

Looking by the lack of information about medicinal plants and not good enough effort by the member of PKK in optimizing swamedication with medicinal plants, IbM held three types of training about swamedication using medicinal plants and monitoring the program. The purpose of this program is to improve the skills and knowledge of every member of PKK about medicial plants, grow the plants at limited space and gain the benefits from medicinal plants for inhabitant. The first training was about improving knowledge about medicinal plants and distribute seedling of medicinal plants which can plant at house yards. The second training was about production techniques of fourinds traditional medicine: jamu beras kencur, kunyit asam, galohgor and instant type of beras kencur. The last training was about techniques for organic compost from household waste. Process of evaluating and monitoring held after all the training done, then there was interview session and held a competition of producing jamu. The evaluation shows there was some improvement in consumption of beras kencur and kunyit asam families and inhabitant. Also, there was indication of transferred information from aenoughtrained member to not-trained member in informal ways. Overall, all of the programs has done with good achievement. This programs already made a significant improvement in public information about medicinal plants.

Abstract

Longer immunity to Severe Acute Respiratory Syndrome-CoronaVirus-2 (SARS-CoV-2) infection is thought to occur through memory cellular responses by activity of specific T lymphocytes. However, most patients with Coronavirus disease-19 (Covid-19) experienced a decrease in the number of T lymphocytes or lymphopenia. Agents that help maintain T cell counts such as Curcumin appear to have played an important role during the Covid-19 pandemic. Curcumin is known to provide a balance between T cell effectiveness and T cell autoaggressiveness, as well as restoring memory T cell function as observed in tumor-induced mice. The mixture of 4 herbal extracts of empon-empon which is commonly used as herbal medicine, namely temulawak, ginger, lemongrass, and turmeric, is thought to have the same effect as curcumin. This is known from the tracing of a plant-protein-compound database which shows that there are not many compounds other than curcumin that can modulate T cells. It is necessary to study the role of Curcumin and a mixture of 4 herbal empon-empon in modulating T cells in cases of infection by the SARS-Cov-2 antigen.

Abstract

The quality of medicinal plants, such as Guazuma ulmifolia (jati belanda, JB), affects the quality of the herbal material derived from them, and can be determined using image analysis. The objective of this study is to investigate the possibility of using an image-generated spectrum and chemometrics as a method for quality control of Jati belanda leaves. Three different quality levels of JB leaves were determined, based on their harvesting time, and confirmed by total flavonoid content analysis. The images of JB samples were collected and reconstructed as a reflection spectrum using the Wiener estimation. The reconstructed spectrum had a goodness-of-fit coefficient of 0.9576 and a root-mean-square-error (RMSE) of 36.65%, compared to the experimental spectrum. Principal Component Analysis (PCA) was used to classify the JB reconstructed spectrum based on its quality. A score plot of two PCs that represented 98% variance was able to group the JB spectrum. Further analysis using Partial Least Squares-Discriminant Analysis (PLSDA) showed that the method can result in around 90% prediction success rate with external validation. This study indicates that image analysis and chemometrics could be used as quality control methods for herbal material.

Abstract

Tirosinase atau fenol oksidase adalah enzim utama yang terlibat dalam biosintesis melanin. Untuk menghindari produksi melanin secara berlebihan pada lapisan epidermal, maka dicari senyawa yang mampu menghambat tirosinase sehingga dapat digunakan sebagai bahan pemutih kulit. Inhibitor enzim tirosinase dapat diperoleh dari senyawa bahan alam diantaranya, polifenol, kumarin, stilben sebagai pengganti senyawa sintetik. Tirosinase telah diketahui struktur molekular sehingga dapat diketahui mekanisme kerjanya melaui uji in-silico dan pembuktian secara in-vitro. Penelitian ini digunakan untuk mendeteksi keefektifan gabungan dari ekstrak etanol pulosari (Alyxia reinwardtii) dan secang (Sappan lignum) sebagai inhibitor tirosinase. Dari hasil in-silico pengujian aktivitas inhibisi tirosinase menggunakan software MOE 2008 menunjukkan bahwa dalam ekstrak etanol dari secang yaitu senyawa brazilin dan rhamnitin berturut-turut memiliki nilai ∆G -15.6582 kkal/mol, -13.3378 kkal/mol dengan inhibisi 10.021 μM, 8.331 μM dan Hdon-acc 6, 8. Sedangkan dalam ektrak etanol dari pulosari dengan senyawa scopoletin dan zhebeiresinol berturut-turut memiliki nilai ∆G -12.1661 kkal/mol; -13.8982 kkal/mol dengan inhibisi 7.279 μM; 9.104 μM dan Hdon-acc 5 dan 6. Sedangkan senyawa parameter L-DOPA dan pembanding asam kojat berturut-turut memiliki nilai ∆G -9.8247 kkal/mol; -8.8047 kkal/mol dengan inhibisi 5.592 μM; 4.976 μM dan Hdon-acc 3; 3. Dari pembuktian secara in-vitro menunjukkan bahwa uji aktivitas inhibisi tirosinase berturut-turut dari secang (Sl), pulosari (Ar), gabungan Sl dan Ar dengan pembanding asam kojat memiliki nilai IC50 berturut-turut 797.090 ppm, 1962.934 ppm, 571.352 ppm dan 93.557 ppm. Sehingga dari hasil in-silico dan in-vitro disimpulkan bahwa penggabungan antara pulosari dan secang memiliki tingkat IC50 lebih baik dibandingkan pemberian masing-masing ekstrak.

Abstract

Herbal medicines become increasingly popular all over the world for preventive and therapeutic purposes. Quality control of herbal medicines is important to make sure their safety and efficacy. Chromatographic fingerprinting has been accepted by the World Health Organization as one reliable strategy for quality control method in herbal medicines. In this study, high-performance liquid chromatography fingerprint analysis was developed as a quality control method for glucofarmaka antidiabetic jamu. The optimum fingerprint chromatogram were obtained using C18 as the stationary phase and linear gradient elution using 10–95% acetonitrile:water as the mobile phase within 60 minutes of elution and detection at 210 nm. About 20 peaks were detected and could be used as fingerprint of glucofarmaka jamu. To evaluate the analytical performance of the method, we determined the precision, reproducibility, and stability. The result of the analytical performance showed reliable results. The proposed method could be used as a quality control method for glucofarmaka antidiabetic jamu and also for its raw materials.

Abstract

Curcuma aeruginosa is known as one of the components of herbal medicine with various biological activities. This research aims to identify the metabolites content of C. aeruginosa related to their biological activities using LC-MS fingerprint combined with chemometrics. C. aeruginosa from 3 areas in Java were collected and macerated with ethanol and then analyzed with LC-MS. Along with this analysis, the antioxidant activity of all samples was determined using CUPRAC method, and the toxicity was determined using Brine Shrimp Lethality Test (BSLT), and chemometric method was used Principle Component Analysis (PCA) and Partial Least square (PLS). Metabolites profiles showed 175 predicted compounds, in which the dominant compounds are from the sesquiterpene of Curcuma genus. The PCA metabolites profiles can separate the samples by their location of origin. Interpretation of the correlation between metabolites profiles and their bioactivities was determined using PLS technique. The results showed that the toxicity of samples was exerted by compounds with ion mass of 312.28 and 248.15, which have the highest antioxidant and toxicity potentials. Compounds with ion mass of 248.15 were predicted to be 9-Oxo-neoprocurcumenol, 7α,11α,-Epoxy-5β-hydroxy-9-guaiaen-8-one, Curcumenolactone A, or Curcumenolactone B. While compound with ion mass of 312.28 was predicted to tetrahydro-bisdemethoxycurcumin.
