Abstract

Endophytic bacteria are beneficial microorganisms that interact with host plants without causing disruption or damage to the host. Some studies suggest that certain endophytic bacteria can produce chemical compounds that have an effect on health, especially endophytic bacteria isolated from medicinal plants. Green betel (Piper betle L.) is a medicinal plant that has been used for years and has many benefits. The purposes of this study are to isolating and screening of endophytic bacteria from green betel against four pathogenic bacteria (Staphylococcus aureus, Escherichia coli, Bacillus cereus and Salmonella enteritidis). The number of endophytic bacteria that has been isolated are 14 isolates. Based on the screening results, three isolates of endophytic bacteria have potential activity (characterized by the formation of inhibition zone) against S. aureus. The inhibition zone may indicate that those isolates produce compounds that have antibacterial effects. Those isolates are AS1, BS1 and BS2. The biggest inhibition zone showed by BS1, so it can be concluded that BS1 is the most potential isolate as a novel source of antibacterial compound.

Abstract

Antioksidan diperlukan untuk menangkal radikal bebas dalam tubuh serta stress oksidatif yang mendorong proses penuaan dan berbagai penyakit degeneratif. Portulaca grandiflora Hook. diketahui memiliki potensi sebagai antioksidan. Perbedaan bagian-bagian tanaman dapat menyebabkan perbedaan kandungan fitokimia, yang kemudian dapat mempengaruhi aktivitas antioksidannya. Penelitian ini bertujuan mengukur kandungan total fenolik dan flavonoid pada organ daun, batang tua, batang muda, dan bunga dari tanaman krokot (Portulaca grandiflora Hook.) yang diekstrak menggunakan etanol 96% serta menentukan aktivitas antioksidan dari ekstrak tersebut dengan metode DPPH. Hasil penelitian menunjukkan bahwa setiap organ tanaman krokot mengandung konsentrasi senyawa fenolik dan flavonoid yang berbeda. Kandungan total fenolik dan flavonoid paling tinggi terdapat pada bagian daun (113.26 ± 3.85 mg GAE/g dan 97.99 ± 1.28 mg QE/g), tetapi aktivias antioksidan tertinggi terdapat pada bagian batang tua dengan nilai IC50 122.15 ± 1.30 ppm (tergolong antioksidan sedang). Kata kunci: Antioksidan, Fenolik, Flavonoid, Portulaca grandiflora Hook.

Abstract

Exploration of bioactive compounds from endophytic bacteria as novel source of antibacterial need to be explored, as an alternative to substitute synthetic antibiotics. Based on previous studies, DM6 was a potential isolate that showed antibacterial activity. The isolate has been identified as Brevibacillus sp. based on 16S rRNA analysis. This study aimed to identify antibacterial compounds produced by DM6 isolate using GC-MS. The GC-MS results showed that the chloroform extract of DM6 isolate contained : 1-Octadecene (CAS).alpha.-Octadecene, Hexanedioic acid, dioctyl ester (CAS) Dioctyl adipate, Phenol, 3,5-bis(1,1-dimethylethyl)- (CAS) 3,5-Di-tert-butylphenol, and 1,4-diaza-2,5-dioxobicyclo[4.3.0]nonane. Adipic acid/hexanedioic acid and phenols are also known to contained in the leaves of Coleus scutellariodes. This indicated that endophytic bacteria were able to synthesize antibacterial compounds which similar with the host plant.

Abstract

Butterfly pea (Clitoria ternatea L.) has been widely used as a healthy drink as well as traditional medicine. This study aimed to test the antioxidant activity of C. ternatea aqueous extract based on its ability to inhibit lipid peroxidation (in vitro) using different parts and extraction methods. Extraction was carried out on flowers, leaves, and roots. The leaves and roots were extracted by boiling technique. Flowers were extracted in 3 ways: boiling, maceration, and brewing. Antioxidant activity was determined based on phenolic content, flavonoid content, and antioxidant test using the TBA method. The highest phenolic and flavonoid levels were shown by the aqueous extract of C. ternatea leave of 57.51 mg GAE/g and 1.50 mg EK/g, respectively. The highest levels of flavonoid were from flower parts treated with boiling technique (0.88 mg EK/g), followed by brewed flowers and macerated flowers. The highest antioxidant activity was obtained from flowers that were boiled for 30 minutes with a percentage inhibition of 94%. Based on the method of extraction and part of the plant, all extracts of C. ternatea have similar antioxidant potential.

Abstract

The potential of black soldier fly (Hermetia illucens (Linnaeus), BSF) maggots as the source of biopharmaca, has not been extensively studied. This research aimed to identify bioactive compunds in BSF maggot extract that potentially activate antioxidant signaling mechanism. BSF maggots fed with vegetable waste were extracted based on maceration method with water, methanol, and acetone as solvents. Forty bioactive compounds were identified by liquid chromatography-mass spectrometry: 15 were in the aqueous extract, 13 were from methanolic extract, and 12 were in the acetone extract. Most of those compounds (90%) were having high bioavailability score (= 0.55 or more) and relatively low toxicity (500 mg/Kg<LD50<5000mg/Kg BW). Molecular docking predicted that there were 26 bioactive compounds potential to activate cellular antioxidant signaling through activation of NRF2 transcription factor, better than the commercial NRF2 activator. The aqueous extract compound with PubChem CID: 73775828 was the best one that inhibited NRF2 signaling by binding to Keap-1 protein (PDB ID: 6FFM) with Gibbs free energy (ΔG) = -6.08 Kcal/mol and dissociation constant (Kd) = 3.58 í— 10-5 µM. Whereas inhibition of inflammation via NF-kappa B signaling was shown by an aqueous extract compound verpacamide A which bound inhibitor NF-kappa B kinase (IKK2) (PDB ID: 4KIK) with ΔG = -5.024 kcal/mol; Kd = 0.207 í— 103 µM. This potency was better than that of aspirin. In conclusion, BSF maggot extracts are source of biopharmaca with potential cellular antioxidant and anti-inflammatory activity.

Abstract

Kesehatan masyarakat di pedesaan Indonesia seringkali terhambat oleh pola konsumsi pangan yang tidak seimbang, dengan ketergantungan tinggi pada karbohidrat dan asupan protein nabati yang kurang optimal. Salah satu solusi potensial adalah peningkatan konsumsi sari kedelai, yang kaya akan protein, vitamin, dan mineral. Sari kedelai dapat membantu memperbaiki gizi dan mengurangi risiko penyakit degeneratif. Desa Sukamaju di Kecamatan Cigudeg, Kabupaten Bogor, memiliki potensi untuk mengembangkan produksi sari kedelai, tetapi pemanfaatannya belum maksimal. Program ini bertujuan untuk meningkatkan gizi masyarakat dan memberdayakan ekonomi lokal melalui pendirian rumah produksi sari kedelai. Kegiatan melibatkan kader Posyandu dan PKK melalui sosialisasi dan pelatihan. Metode yang digunakan meliputi pengumpulan data melalui kuisioner pre-test dan post-test untuk menilai pengetahuan sebelum dan setelah pelatihan. Hasil pelatihan menunjukkan peningkatan pemahaman masyarakat tentang sari kedelai, meskipun awalnya pengetahuan mereka terbatas. Program ini berhasil meningkatkan minat dan pengetahuan mengenai sari kedelai, serta berpotensi untuk menjadi model bagi desa lain. Kesimpulannya, program ini memberikan manfaat ganda dengan meningkatkan gizi masyarakat dan mendukung ekonomi lokal, meskipun masih ada tantangan dalam mendorong produksi dan konsumsi sari kedelai secara mandiri.

Abstract

Red ginger (Zingiber officinale var. rubrum) is traditionally recognized for its medicinal properties, largely due to its antioxidant potential attributed to secondary metabolites such as flavonoids. This study aimed to evaluate the total flavonoid content and antioxidant capacity of ethanol and water extracts of red ginger from Sukabumi, specifically the Jahira 1 variety, as well as identify bioactive compounds. Extraction was carried out using the maceration method. The total flavonoid content was determined using the aluminium chloride colorimetric method, while the antioxidant capacity was assessed via the ferric reducing antioxidant power (FRAP) assay. Results showed that the ethanol extract had higher total flavonoid content and antioxidant capacity, with values of 5.99 mg quercetin equivalent (QE)/g and 473.93 µM Trolox equivalent (TE)/g, respectively. In contrast, the water extract exhibited 3.73 mg QE/g and 206.75 µM TE/g. Further LC-MS/MS analysis of the ethanol extract identified 26 compounds, including phenolics and terpenoids, which contribute to the antioxidant capacity of red ginger from Sukabumi.

Abstract

Dental caries are caused by plaque formation resulting from biofilm accumulation on tooth surfaces. The bacterium Streptococcus mutans plays a crucial role in biofilm formation, partly through the production of glucosyltransferase, which catalyzes biofilm synthesis. Flavonoids are secondary metabolites commonly found in plants, known for diverse biological properties, including antibiofilm effects. This study aimed to screen the potential of flavonoid compounds as antibiofilm agents through inhibition of glucosyltransferase using an in-silico approach. A total of 87 flavonoid compounds obtained from the ZINC database were evaluated via molecular docking methods. Screening results based on binding free energy (ΔG) values, analyzed using the PyRX-Virtual Screening Tool, indicated that 36 compounds had potential to inhibit glucosyltransferase. Further molecular docking using AutoDock Vina identified nine compounds with ΔG values more favorable than the natural ligand of glucosyltransferase (maltose). Molecular interaction analysis using LigPlot+ and PyMOL revealed that taxifolin, gallocatechin, and sakuranetin interacted with three catalytic residues of the enzyme, whereas the remaining six compounds interacted with two catalytic residues. Liquiritigenin exhibited the lowest ΔG (-7.0 kcal/mol) and an inhibition constant (Ki) of 7.39 µM, indicating high affinity for glucosyltransferase. This compound formed two hydrogen bonds and four hydrophobic interactions, engaging two catalytic residues of the enzyme, Asn481 and Trp517. These findings highlight the potential of flavonoids as antibiofilm agents via glucosyltransferase inhibition. Further experimental validation through in vitro studies is necessary to confirm these in-silico findings.

This study evaluates the effects of fish feed formulated with BSFL and tofu waste on the growth performance, proximate composition, and fatty acid profile of red tilapia (Oreochromis niloticus). Experimental diets included: commercial feed, dried BSFL, and a combination of BSFL:tofu waste. Fish were reared for eight weeks under controlled conditions. Treatment with BSFL:tofu waste exhibited superior growth performance, with the highest weight gain (102.88%) and length gain (33.75%). Proximate analysis revealed significant variations across treatments, with a combination of BSFL:tofu waste achieving the highest caloric value (110.71 cal/g) and fat content (3.75%) while maintaining protein levels comparable to commercial feed. Fatty acid analysis showed a combination of BSFL:tofu waste enhanced total fatty acid content (77.92%), predominantly polyunsaturated fatty acids (PUFAs), increasing from 37.34% in commercial feed to 47.04%. Essential fatty acids, including DHA and linoleic acid, were elevated in a combination of BSFL:tofu waste, indicating improved nutritional quality. Cholesterol levels remained unaffected among treatments. These findings suggest that combining BSFL and tofu waste can enhance fish feed quality, optimize growth performance, and improve the nutritional value of red tilapia. Future research should explore refining feed formulations to maximize nutrient stability and sustainability for broader aquaculture applications.

Abstract

Program pelatihan dan sosialisasi mengenai pemanfaatan SIJAKA (sirih merah, jahe merah, dan kayu manis) sebagai minuman fungsional antidiabetes bertujuan untuk meningkatkan pemahaman dan praktik masyarakat dalam mengelola diabetes secara alami. Desa Sukamaju yang terletak di Kecamatan Cigudeg, Kabupaten Bogor memiliki peluang untuk mengembangkan produksi minuman fungsional berbahan dasar SIJAKA, namun potensi ini belum dimanfaatkan secara optimal. Program ini dirancang untuk meningkatkan taraf kesehatan masyarakat serta mendorong pertumbuhan ekonomi lokal melalui pendirian fasilitas produksi minuman fungsional SIJAKA. Pelaksanaan kegiatan melibatkan kader Posyandu dan PKK melalui kegiatan penyuluhan dan pelatihan. Metode yang digunakan mencakup pengisian kuisioner pre-test dan post-test guna mengevaluasi tingkat pengetahuan peserta sebelum dan sesudah pelatihan. Hasil pelatihan menunjukkan adanya peningkatan pemahaman masyarakat terkait minuman fungsional SIJAKA, meskipun sebelumnya pengetahuan mereka masih terbatas. Pelatihan ini juga mengungkapkan bahwa 79,31% peserta mengenal sirih merah dan jahe merah sebagai herbal potensial, sedangkan 72,41% mengetahui manfaat kayu manis. Hasil ini menunjukkan adanya peningkatan kesadaran akan potensi herbal dalam mengelola diabetes. Selain itu, mayoritas peserta (92,11%) menunjukkan ketertarikan yang tinggi terhadap pelatihan dan ingin lebih memahami manfaat serta cara pembuatan SIJAKA. Program ini terbukti mampu meningkatkan antusiasme dan wawasan masyarakat terhadap SIJAKA, serta memiliki potensi untuk dijadikan sebagai contoh bagi desa-desa lainnya. Secara keseluruhan, program ini memberikan dua manfaat utama, yaitu peningkatan kualitas kesehatan masyarakat dan penguatan ekonomi lokal, meskipun masih terdapat kendala dalam mendorong masyarakat untuk memproduksi dan mengonsumsi SIJAKA secara mandiri.
