Abstract

Diatom Amphora sp. is a type of aquaculture product that has various types of metabolites with very good potential to be used as basic ingredients for drugs or other health products. Insilico analysis of extracts of Amphora sp. was carried out to predict biological activity that can be used as a basis for developing the use of aquaculture products. The research was conducted by extracting Amphora sp. using ethanol solvent with sonication method, the extraction results were analyzed by LC-HRMS, and the metabolite readings were then analyzed for drug potential and biological activity online using swissADME, Protox II, and Way2Drug applications. The results of the LC-HRMS analysis showed that the extracted Amphora sp. showed the highest abundance of valine compounds with 17.6% as well as other compounds L-Norleucin, stearamide, palmitoleic acid, isotretinoin, and arachidonic acid. The results of the ADME/T analysis showed that the six compounds of Amphora sp. extract had quite good potential according to the Lipinski rule of five. Biological activity also shows potential as an anti-inflammatory, antifungal, antibacterial, antiviral, and antineoplastic agent with Pa values ranging from 0.260 to 0.873. These results indicate that the production development of Amphora sp. and its use is very wide, including in the health sector.Keyword: ADME/T, PASSonline, Amphora sp., sonication, aquaculture

Abstract

Natural feed has a very important role in supporting the success of aquaculture. The availability of natural feed is a key factor in the seed / fry phase, where at that phase the fish are still unable to consume additional feed, namely pellet feed. The objectives of this activity are: (1) Introducing Artemia, Improving the entrepreneurial spirit and entrepreneurial motivation, (3) Increasing the knowledge and abilities of SMK Ibrahimy 1 Sukorejo students in terms of decapsulation and artemia culture. This activity targets the fishery students of SMK Ibrahimy 1 Sukorejo, Department of Agribusiness, Brackishwater and Marine Fisheries. Service activities are carried out using lecture and demonstration/practice methods. Evaluation was carried out on the ability of participants to practice artemia decapsulation and artemia culture, indicators of the success of this practice are theoretical and practical knowledge of Artemia sp culture. The success of this activity can be seen from the very high enthusiasm of students and the increasing knowledge of students about the decapsulation of Artemia sp. The obstacle that is used as an evaluation in this activity is that several additional tools are still needed to maximize the decapsulation practice of Artemia sp.

Abstract

The purpose of this study was to determine the intensive technique of rearing vannamei shrimp in shrimp hatchery installation Gelung, Brackish Water Aquaculture Center Situbondo, East Java. The data collection method used in this activity is a descriptive method that is the method carried out by looking at existing activites in the field with existing libraries. Activites undertaken include; pond preparation namely cleaning and washing of cultivation facilities, drying, HDPE plastic repair, map sterilization, installation of cultivation facilities and calcification of maps, preparation of aquaculture media including water filling and sterilization, fertilizer and POC application; selection and stocking of fry; feed management includes the type of feed, amount or dose of feed, frequency and technique of feeding, and check anco; water quality management includes observing water quality parameters, water change, water treatment and siphon; sampling; pest and disease control; harvest; and business analysis.

Highlight ResearchIndicated that white feces at Vanammei appeared along with the appearance of the disease besides its main trigger that is Vibrio sp.Increase the availability of carbon for photosynthesis process of phytoplankton.Clinical symptom of shrimp attacked by white feces disease is indicated by a change in intestine.AbstractShrimp disease that currently causes economic loss to shrimp farmers is White Feces Disease (WFD). This disease appeared due to several factors, such as poor pond management, unhealthy shrimp seed, and poor water quality which resulted in the appearance of Vibrio sp. bacteria and Dinoflagellate. This study aimed to analyze the cause of WFD outbreak in vannamei shrimp pond. The study method through direct experiment was applied in shrimp pond. Sampling was performed three times in each feeding tray to collect ten shrimps. Overall, sampling was performed twice a week. The Sample Survey Method was used to collect sample in this study. Result of study showed that clinical symptom was observed through changes in pattern and behavior of vannamei shrimp during culture. However, this observation resulted in insignificant data. Vannamei shrimp infected WFD tended to swim slower and often rose to the surface with body color turned red. Moreover, total organic matter (TOM) increased on week-7 along with the increasing growth of plankton, particularly from the Dinoflagellate group. However, bacterial growth of Vibrio sp. on week-7 was insignificant, yet many shrimps were found dead with white feces during that period. The result of analysis indicated that white feces disease was caused by Dinoflagellate besides the main trigger, namely, Vibrio sp.

Abstract

Water quality standard criteria for shrimp farming, as regulated by the CBIB, are a crucial aspect in intensive farming operations. This research aims to assess the suitability of water quality parameters in intensive cultivation of vaname shrimp (L. vannamei) with CBIB standards (Good Fish Cultivation Methods). The research method uses an ex post facto causal design during one shrimp cultivation cycle, with a focus on the condition of water quality parameters compared with CBIB standards in accordance with Minister of Maritime Affairs and Fisheries Regulation Number 75 of 2016. The research results show that most of the water quality parameters during the cultivation period in accordance with CBIB standards, except for alkalinity and organic matter exceeding the thresholds set by CBIB. Abnormal conditions in these two parameters are caused by unpredictable natural and seasonal factors, as seen in unstable fluctuations in temperature and salinity. However, overall, the cultivation system at the research location still complies with the cultivation quality standards set by CBIB. From this discussion, it can be concluded that the water quality parameters at the research pond location are mostly in accordance with CBIB quality standards, with the exception of alkalinity and organic matter which slightly exceed the limits set by CBIB..

Abstract

In order to strengthen the economy of the community in Sumberejo Village and overcome the limited stock to supply catfish, it is necessary to form a community business, namely a catfish cultivator group. There were twelve potential community members were gathered to be taught and assisted in the process of establishing and managing a catfish cultivation business. The members of the community were taught technically how to cultivate catfish right through to the marketing process, then facilitated the formation of farmer groups together with the Situbondo district fisheries instructor and the Sumberejo Village Head until the "Santri Aquaculture Ibrahimy" farmer group was formed. Assistance is provided starting from the process of building a pond, preparing the pond, keeping fish and harvesting, to the marketing process. The results of economic benefits began to be seen in the first harvest cycle, there was a profit of IDR 2,400,000, with capital issued of IDR 7,000,000,-. With the existence of the Santri Aquaculture Ibrahimy farmer group, it can be achieved at least to overcome the shortage of daily consumptive stock of catfish and to strengthen the village economy by opening up employment opportunities for the people of Sumberejo Village.

Abstract

Bugeman Village has great potential for vaname shrimp farming. However, vaname shrimp production in this village is still hindered by suboptimal pond water quality. This has resulted in high shrimp mortality rates and low yields. This community service aims to improve the water quality of round vaname shrimp ponds (KODAME) in Bugeman Village through mentoring undergraduate students studying water quality management. Water quality is very important for the aquaculture process as it can help increase aquaculture productivity and prevent environmental damage. Students will provide training to shrimp farmers on effective pond water management techniques, such as measuring water quality parameters, regulating aeration and filtration, and using probiotics. This program is expected to improve the water quality of shrimp farmers' ponds in Bugeman Village, thereby increasing shrimp production and increasing their income. Bugeman Village is expected to become a self-reliant vaname shrimp village that can improve the welfare of its people. In addition to training, students will also assist shrimp farmers in implementing the learned pond water management techniques. Students will also monitor and evaluate this program to ensure its effectiveness. It is hoped that this program can make a significant contribution to the development of vaname shrimp farming in Bugeman Village and improve the welfare of its people.

Abstract

Grouper production in Indonesia can be supported by hybridization technology which can produce grouper species with relatively faster growth and more resistance to disease. Cantang grouper is a type of grouper fish that was engineered by crossbreeding between female tiger grouper (Epinephelus fuscoguttatus) and male grouper Kertang (Epinephelus lanceolatus). The purpose of this study was to determine the stages and technology of nursery grouper Cantang which was carried out at BPBAP Situbondo. The method used in this research is the method of observation, participation, and interviews. The primary data needed in this study are data on infrastructure, data on the number and size of seeds, feed management, water quality management, and harvesting. The data is presented with a descriptive method by discussing the data that has been obtained compared to the theories that have developed at this time. The results of this study indicate that cantang grouper nursery activities start from tank preparation, water filling, seed selection and distribution, feed management, water quality, fish pest and disease control, grading, and harvesting of cantang grouper seeds. During the practice period, the seeds were spread from hatcheries with a stocking density of 3000 to 5000 individuals in one tank with a size of 4 x 2.5 x 1 m, or the equivalent of 300 to 500 individuals/m3. The distribution of seeds is carried out after the acclimatization process, namely the adaptation process before releasing the seeds. After the seeds are removed, the tub is given an antiseptic to prevent disease in the seeds. There are several factors that can affect the growth rate of seeds, namely water quality management and feeding. During the maintenance period, a flowing water system is used, so that the water used is continuously renewed. In the morning, the seeds are flushed and soaked using fresh water, with the aim of releasing mucus on the seeds so as to reduce the potential for disease outbreaks. Nursery grouper at BPBAP Situbondo can run optimally by paying attention to the stages of cultivation, especially in the feed management and water quality management.

Abstract

ABSTRACT  β-actin is a prominent protein in the immune system. An outcome from gene transcription, the protein protects against pathogen infection, such as receptor clustering, antigen internalization, and regulating vesicle for antigen processing. ß - actin expression will determine the success of immune response of the organism. This study aimed to understand the role of pigment-protein fraction (PPF) from Nannochloropsis oculata in the increase of β-actin expression in humpback grouper Cromileptes altivelis infected by viral nervous necrosis (VNN). The experiment was performed with one negative control (A: normal fish) and three PPF treatments (B: fish + PPF, C: fish + VNN, and D: fish + PPF + VNN). PPF was applied through the sonde method and analyzed with immunohistochemistry technique and Immunoratio software. The results showed that PPF was able to increase β-actin expression on all treatments: A (34.9%), B (38.1%), C (39.1%), and D (51.6%). It demonstrated PPF ability to induce the increase of β-actin expression indicating an improved defense of humpback grouper C. altivelis against VNN infection. Keywords: β-actin, pigment-protein fraction, Nannochloropsis oculata, VNN  ABSTRAK β-aktin adalah protein yang berfungsi penting sebagai salah satu sistem pertahanan tubuh. Protein ini merupakan hasil transkripsi dari gen yang memiliki fungsi penting dalam sistem pertahanan tubuh terhadap infeksi patogen, seperti kelompok reseptor, internalisasi antigen, dan mengatur keluar masuknya vesikel untuk pemrosesan antigen. Ekpresi β-aktin menjadi penentu berhasil atau tidaknya respons imun pada tubuh organisme. Penelitian ini bertujuan untuk mengetahui peran fraksi protein pigment (FPP) dari Nannochloropsis oculata dalam meningkatkan respons β-aktin pada ikan kerapu tikus Cromileptes altivelis yang terinfeksi viral nervous necrosis (VNN). Pengamatan dilakukan pada empat percobaan yaitu, satu kontrol (A : ikan normal) dan tiga perlakuan FPP (B : ikan + FPP, C: ikan + VNN, dan D: ikan + VNN + FPP). Pemberian FPP menggunakan metode sonde dan analisis hasil penelitian menggunakan teknik imunohistokimia dan Immunoratio. Hasil penelitian ini menunjukkan bahwa pengiduksian FPP mampu meningkatkan ekpresi β-aktin yaitu A (34,9%), B (38,1%), C (39,1%), dan D (51,6%). Hal tersebut menunjukan FPP mampu menjadi inducer yang baik untuk peningkatan ekpresi β-aktin. Meningkatnya ekspresi β-aktin bisa dijadikan sebagai indikator pertahanan tubuh ikan kerapu tikus C. altivelis terhadap infeksi VNN. Kata kunci: β-aktin, fraksi protein pigmen, Nannochloropsis oculata, VNN
