Abstract

Abstrak This research was conducted  on June until October 2018 in Experimental Pond and Laboraty of Parasites and Fish Diseases, Faculty of Fish and Marine Science, University of Riau, Pekanbaru. The purpose of this study was to determine the effect of feeding containing curcumin extract with different doses on the image of red blood in siam jambal fish Pangasius hypophthalmus. The method used is the used in the experimental method with a Cumpletely Randomized Design (CDR) with one factor and 4 treatment . the namely: P0 (Feedingwithout addition of turmeric curcumin extract), P1 (Addition curcumin extract from turmeric in feed at a dose of 0,5 g/kg), P2 Addition curcumin extract from turmeric in feed ar a dose of 0,7 g/kg), P3 (Addition of curcumin extract extract from turmetric in feed at a dose of 0,9 g/kg). The test fish used was Pangasius hypophthalmus with a size was 7-8 cm as many as 600 fishs. The parameters measured are; total erythrocytes, haemaglobin, haematocrit, absolute weight growth, survival rate and water quality measurements. The resullts that the effect of giving curcumin extract from turmeric in feed with the best dose is 0,9 g/kg, which is indicated by a total erythrocyte of 2,91x106 cells/mm3, haemaglobin 10 g/dL , haematocrit 38,33%, absolute weight growth 32 %  and  survival rate 99,33%.                                                                                                                       Keywords:Pangasius hypophthalmus,Curcumin extract, Erythrocyte

Abstract

ABSTRACTResearch was conducted from September to October 2014, at the labory Examination of Fish Disease, Fisheries and Marine Sciences Faculty of Riau University. The Purpose of this study was to determine the sensitivity of mangosteen rind (G. mangostana L.) as antimicrobial to A. hydrophila, the range of Minimal Inhibitory Concentration (MIC) and the lethal dose (LD50) mangosteen rind (G. mangostana L.) solution against to catfish (Clarias gariepinus) (10-13 cm) by immersion. The research method used doses 10 % (19 g/L), 20 % (38 g/L), 30 % (57 g/L), 40 % (76 g/L), 50 % (95 g/L), 60 % (114 g/L), 70 % (133 g/L), 80 % (152 g/L), 90 % (171g/L), 100 % (190 g/L). The results showed that mangosteen rind was effectively to inhibit the growth of bacterial A. hydrophila at a dose of 50 % (95 g/L) inhibitory zone of 13,65 mm, with the number of A. hydrophila colonies in TSA medium dose 50 % (95 g/L) is 30,6 x 108 cfu/mL. Abstract

ABSTRACT Lemon (Citrus limon) is a fruit that can be used as an antimicrobial/antibacterial because it contains secondary metabolites, such as flavonoids, tannins, terpenoid, and asam sitrat. Edwardsiella tardawas pathogenic bacteriathat can cause diseases in cultured fish.This research was conducted from Mei to Juli 2019, at Parasites and Fish Disease Laboratory, Faculty of Fisheries and Marine, University of Riau. The purpose of this study was to get the sensitivity of lemon(Citrus limon) in inhibit he growth ofEdwardsiella tarda, the range of Minimal Inhibitory Concentration (MIC) and the lethal dose (LD50)of lemon (Citrus limon) solution against to (Pangasius hypophthalmus) with length 7-9 cm by immersion for 24 hours. Sensitivity test is using the method of KirbyBauer disk blank 6 mm disc with dose used were 100-0,3% and control Oxytetracyclin. The results showed that the sensitivity of Lemon solution towardsE. tardaat dose 100% with clear zone 14,96 mm and dose 0,3% with clear zone 6,00 mm.  At a concentration of 0,5% was determined as the MIC value with a bacterial density 265,33 CFU / mL.Dose LD50 lemon solution against catfish by immersion for 24 hours in 0,49%Keyword :(Citrus limon), Edwardsiella tarda, Sensitivity, Minimum Inhibitory Concentration, LD50

Abstract

ABSTRACTThis study was conducted from September-November 2014 in the Laboratory of marine Microbiology, Faculty of Fisheries and Marine Sciences, University of Riau. The purpose of this study was to determine the effect of probiotics strain Riau of giant prawn toward A. hydrophila. This research was continued of the Hasyimi’s research on 2014, the treatment in method cakram used is probiotics Bacillus sp. with six levels of treatment: P0 (disk blank+50 μL TSB), P1 (antibiotic novobiocin), P2 (50 μL isolate Bacillus F), P3 (50 μL isolate Bacillus G), P4 (50 μL isolate J) and P5 (50 μL isolate K), and the dilution method uses four methods (F, G, J and K) of four kind of dilution (10-2, 10-4, 10-6 and 10-8). The results showed that the genetic of probiotics bacterial strain Riau wich was isolated giant prawn could theat A. hydrophila, with the biggest blocked power is in K isolator (13,8 mm) and the smalest blocked power is in G isolator (7,6 mm), and MIC value is in 10-8 dilution and MBC value is in 10-4 retard.Key words: Bacillus sp., giant prawn, A. hydrophila

Abstract

This research was done in March - April 2014 in Fish Disease Parasite and Laboratories and an Experiment of the Faculty of Fisheries and Marine Science University of Riau. The purpose of this research is to know the influence of the addition of curcuma on feed to the total of erythrocytes , hematocrit , hemoglobin and the growth of catfish. A research method used is the design of random complete with one factor which feed containing curcuma with four standard treatment consists of , P0: control without given ) curcuma solution , the P1 ( curcuma solution with a dose of 2 g/kilogram feed ) , the P2 ( curcuma solution with a dose of 4 g/kilogram feed ) , the P3 ( curcuma solution with a dose of 6 g/kilogram feed ). The results of research shows that the addition of a solution on feed curcuma doesn’t impact the condition of fish blood because of poor environmental conditions. It can increase the growth of absolute weight catfish .The total erythrocytes is ranged between 1,25-1,91 mm3/ million cells , hematocrit 13,8-18,5 % and the percentage of hemoglobin 3,9-4,2 g/dL.Keywords: River catfish (Mystus nemurus), ginger, blood catfish

Abstract

ABSTRAKThis research was conducted from March to June at the Laboratory of Fish Parasites and Diseases, the Marine Microbiology Laboratory and the Marine Chemistry Laboratory of the Faculty of Fisheries and Maritime Affairs, Riau University, Pekanbaru. The purpose of this study was to analyze the effectiveness of heterotrophic bacteria to improve water quality and body resistance of salted tilapia (Oreochromis niloticus) from haematological (Erythrocytes, hematocrit, hemoglobin and leukocytes) against disease.The method used in this study is an experimental method, using a Completely Randomized Design (CRD), 1 factor, namely with five levels of treatment three replications. Kn: Without administration of heterotrophic bacteria, without being tested, Kp: Without administration of heterotrophic bacteria, tested by Aeromonas hydrophilla bacteria, P1G: Giving heterotrophic bacteria Vagococus fluvialis at a dose of 15ml /25L, P2G: Giving heterotrophic bacteria Bacillus cereus at a dose of 15ml/25L, P3G: Provision of combined heterotrophic bacteria Bacillus cereus and Vagococus fluvialis at a dose of 15ml/25L. The results showed that the best treatment was P3 treatment using combined heterotrophic bacteriaBacillus cereus and Vagococus fluvialis. This is based on the value of blood parameters after the challenge test with Aeromonas hydrophila bacteria showing total erythrocytes of 252.33x104 cells/mm3, total leukocytes 90.13x103 cells/mm3, hemoglobin levels 8.03 g/dL, hematocrit 32.23%, survival rate 96, 66% and absolute weight growth of 4.82 g/head. The results of measurements of water quality during the study temperature range between 27-27.40C, dissolved oxygen 1.8-2.3 ppm, pH of water ranged from 7.8 to 7.9. Keywords: Heterotrophic Bacteria (B. Abstract

ABSTRACT This research was conducted from April until May 2018. The purpose of this study was to calculate the density of Vibrio sp. bacteria insea water of Dumai,to determine the ratio of Vibrio sp.toheterotrophicbacteria to examine the resistence of Vibrio sp. to antibiotics. In this study the determination of sampling points using purposive sampling method, was set into three sampling points for each station. Density total of Vibrio sp. and heterotrofik bacteria by using spread plate count method. The result showed that density Vibrio sp.ranged from 1.3 x 103 - 1.6 x 103 CFU / ml. the haignest number of Vibrio sp. wasfound in stations 4, and the lowest was in station 5.  The total number of heterotrophic bacteria ranged from 7.37 x 106 - 10.38 x 106 CFU / ml. the highest number of heterotrophic was found in station 4 in the lowest was in station 5. The hignest percentange ratio of the density of Vibrio sp.to heterotrophic bacteria was in station 5 0.0178% and the lowest was in station 1 0.0144%. resistance ofVibrio sp. toward Chloramphenicol as antibiotic tested indicatedthat 31 isolates were categorized into intermediate with the inhibition zone ranged from 12-19.8 mm, and 1 isolate was sensitive with the inhibition zone was 21 mm. Toward antibiotics penicilin and isoniazid, all bacterial isolates were resistant with the inhibition zone ranged from 1.6 - 9 mm and 0.9 - 6.2 mm, respectively. Keyword: Density, Vibrio sp,Heterotrophic Bacteria, Antibiotics

Abstract

Heterotrophs is a bacteria that use organic carbon and inorganic nitrogen as a food source. The research was conducted from December 2016 to January 2017. The aim at this study was to isolate, determine the ability of bacteria isolates as an antibacterial to pathogenic bacteria and determine the characteristics of heterotrophic bacteria isolated from residential area and low salinity seawater based on PCR method technique 16S rDNA sequences. Isolation and identification morphology, biochemical test and antibacterial tests were analyzed in Marine Microbiology Laboratorium of Riau University. DNA and PCR analysis process was conducted in Biology Laboratorium of Riau University. The sequencing DNA process was conducted in PT. Genetika Science Indonesia, West Jakarta. The result showed that the twenty three pure isolates bacteria and only seven isolates could be sequenced. Bacterial isolates 20M2, 20M4, 20M5, 20M11, 30M6, 30M7 and 30M8 able to inhibit the growth of pathogenic bacteria. Based on the BLAST system accessed via World Wide Web, the isolated bacteria were: Bacillus toyonensis strain ACOPR1ISOXg, Bacillus subtilis strain Y37, Enterobacter cloacae strain ENC-3, Clostridium acetobulylicum strain S512, Bacillus cereus strain 4PLGES, Bacillus cereus strain ML267 and Bacillus thuringensis strain C17.Keywords: heterotrophic bacteria, isolation, characterization, 16S rDNA

Abstract

ABSTRACTCitrus hystrix is an essential oil producing plant. In addition to essential oils, there are several ingredients of Citrus hystrix as an antibacterial agent, namely: vitamin C, flavonoids, tannins and alkaloids. This research was conducted on June until August 2019. The aims of this research was to get obtain information about the effect of giving a solution of Citrus hystrix to the erythrocyte image of conjoined Pangasius hypophthalmus and find out the best dosage for the prevention of MAS (Motile Aeromonas Septicaemia) disease, seen from the total erythrocytes, hemoglobin, hematocrit, and morphology of erythrocyte patchouli fish (Motile Aeromonas Septicaemia). The method used is an experimental method with a completely randomized design (CRD), one factor with 5 treatment and three raplication of KN: (without immersed with Citrus hystrix solution and without infected by  Aeromonas hydrophila) KP: (without immersed with Citrus hystrix solution and infected by Aeromonas hydrophila.) P1: (1200 ppm), P2: (1500 ppm), P3: (1800 ppm). immersed of Citrus hystrix solution was carried out four times, namely on days 7,14,21 and 28, soaking was carried out for 2 minutes. Then on the 31st day a challenge test was carried out with Aeromonas hydrophila bacteria with a density of 108 CFU / ml of 0.1 ml /fish. The results showed that immersed of fish with a solution of Citrus hystrix gave effect to the erythrocyte image of conjoined Pangasius hypophthalmus. The best dosage is P2 1500 ppm treatment seen from total erythsoit 172.67 x 104 cells/mm3, hematocrit value 31.33% and hemoglobin value 6.93 g/dl. Siamese Pangasius hypophthalmus life reaches 100%. The quality of water obtained during the study were temperatures ranging from 27.3 - 28 oC, pH 6-7, DO 3.2 - 4.7 mg/L and Ammonia 0.02 mg/L.Key word :Citrus hystrix, Catfish, Aeromonas hydrophila, immertion,erythrocyte,

Abstract

ABSTRACT            Fungal infections is one of the secondary infectious that can reduce the aesthetics value of goldfish, and cause 50% death in goldfish. The objective of the study was to find out the species of pathogenic fungi in goldfish. the method used was survey at the location of the Delima village, Tuah Karya village, Simpang Baru village, Tampan Sub-District, Pekanbaru City. The fish used were 7 - 12 cm as many as 27 tails. Conventional identification includes macroscopic inspect of fungi colonies and inspect microscopic of fungi hyphae. Molecular identification was carried out by Polymerase Chain Reaction (PCR) technique and analyzed with Bioedit, BLAST and MEGA applications to find out the phylogenetic tree. Research parameters include water quality and fungi prevalence. The results of conventional identification techniques found the species of fungi, includes Aspergillus niger, Penicillium sp., Curvularia sp., Bipolaris sp., Fusarium sp. The result of molecular identification techniques found the species of fungi, includes Aspergillus tamarii with homolog value 99%,Fungal endophyte with homolog value 98%, Curvularia pseudorobusta with homolog value 93%. The conclusions obtained from this study are Fusarium sp. and Curvularia sp. The prevalence value in the second sampling with an average of 66.33% was higher than the value of the first sampling with an average 22,22% and third sampling with an average 11,11%. Keywords:Fungi, Carassius auratus, Polymerase Chain Reaction
