Abstract

ABSTRACT             This research has been conducted on June until December 2017 in Laboratory of Parasites and Fish Diseases of Fisheries and Marine Faculty, University of Riau. The aimed of this research was to know the structure of liver and kidney tissues of Pangasius hypophthalmus that were immersed in Lawsonia inermis L. solution for preventing the of Aeromonas hydrophila infection. This research was using experimental method and the data were analyzed by descriptively. This study was using five treatments, Kn(were not immersed and infected), Kp (were not immersed Lawsonia inermis L. solution but infected of Aeromonas hydrophila), P1(dose Lawsonia inermis L. solution 2000 ppm), P2(3000 ppm), and P3(4000 ppm). The result shown that abnormalities liver tissues of Pangasius hypophthalmussuch us hemorrhage, hypertrophy and necrosis and kidney abnormalities such us hemorrhage, necrosis, and congestion. The best result was P1 of Pangasius hypophthalmus were immersed lawsonia inermis L. solution in preventing the Aeromonas hydrophila infection. Keyword:Histophatology, Liver, Kidney, Lawsonia inermis L.,Pangasius hypopthalmus

Abstract

ABSTRACT    One of the plant (Artocarpusaltilis)is a plant that can be used as a drug and has many benefits, one of which is antibacterial to the disease Motile Aeromonad Septicemia (MAS). This research was conducted from February to August 2018 at the Laboratory for Fighting Pests and Diseases of Fish, Department of Aquaculture, Faculty of Fisheries and Marine, University of Riau. The purpose of the study was to determine the inhibitory potency of the breadfruit leaf solution in inhibiting the growth of Aeromonas hydrophila bacteria, the MIC (Minimum Inhibitory Concentration) dose and the toxicity of the solution of breadfruit leaves to immersion. The research method used is the experimental method using the KIRBY-BAUER disc method to reduce the level of error then repeated 3 times. Determination of treatment was PO control (Oxytetracyclin), P1 (100%), P2 (90%), P3 (80%), P4 (70%), P5 (60%), P6 (50%), P7 (40%) , P8 (30%), P9 (20%), P10 (10%), P11 (9%). The doses used in this study were 9% (900 ppm), 10% (1000 ppm), 20% (2000 ppm), 30% (3000 ppm) and controls . The results of this study indicate that the breadfruit leaf solution (Artocarpusaltilis) is able to inhibit the growth of A. hydrohila by presenting a 100% inhibition dose with an average of 10.56 mm and a minimum inhibition of 9%, giving an average inhibition of 6.46 mm. It can be said that the dose of MIC (minimum inhibitory concentration) of breadfruit leaf solution, at a dose of 10% with an average number of bacterial colonies of 268 Sel / mL, is the minimum dose capable of inhibiting the growth of A. hydrophila bacteria. The results of the LD50 toxicity tests of the breadfruit leaf solution on Siam ham (P.hypophthalmus) were immersed for 24 hours at a concentration of 1177 ppm. Keywords:  Artocarpus altilis, Aeromonas  hydrophila, antimicrobial, LD50

Abstract

The effect of containing curcuma (Curcuma xanthorrhiza Roxb) on diferentiation of leukocytes of Cat Fish (Mytus nemurus). The aim of the study is to find of diferentiation of leukocyte of Cat Fish (Mytus nemurus). The study was conducted from March to April 2014 in the Experimental Pond and Laboratory of Parasitic Diseases of Fish and Fisheries and Marine Sciences Faculty of the Riau University. The method used was experimental method using RAL with one factor with 4 level treatments and three repliations. the result indicate that the best differentiation of leukocytes in the P3 treatment is total leukocytes (96.538 cell/mm3), diferentiation of leukocytes (lymphocytes: 96,67%, monocytes: 1,67%, and neutrophils: 1,67%) and the best treament of survival rate is P2 and P3 with survival rate is 100%.Keywords: Differentiation, Leukocytes, Cat Fish, and Curcuma Solution

Abstract

This research was conducted on October 2015 - June 2016, at the Laboratory of Microbiology and the Laboratory of Parasites and Fish Diseases, Faculty of Fisheries and Marine Sciences, University of Riau. The purpose of this study is to comprehend the probiotic Bacillus sp. from the digestive tract of the giant prawns (Macrobranchium rosenbergii DE MAN) and the black tiger shrimp (Panaeus monodon) for the health status of tilapia (Oreochormis niloticus). This study uses a Completely Randomized Design (CRD), which consists of three treatments and each of them uses three replications for the probiotic Bacillus sp. treatment of the digestive tract either from the giant prawns or the tiger shrimp. The Bacillus sp. treatment from the giant prawns are P1G (UG4 isolates), P2G isolates combined (UG1, UG2, UG3, UG4, and UG5) and as a control (P0).While the tiger shrimp are using P1W (UWH9 isolates), P2W isolates that combined (UWH1, UWH2, UWH8, UWH9 and UWH 10) and as a control (P0). Probiotics from each treatment were put into the medium water about 3 mL / container with a density of 104 CFU/ml. The results showed that the P1W treatment is the best treatment after infected by Streptococcus iniae for the health statusof tilapia, it is showed by the number of Erythrocytes 246.67x104 sel / mm, Haemoglobin level of 6633 g/dl, Hematocrytes 30.33%, and total Leukocytes 79,33x103 sel/mm3. Given by probiotics of Bacillus sp that originatit from the digestive tract of giant prawns and tiger shrimp through the water were able to maintain the health of tilapia (Oreochormis niloticus).Key Words: Bacillus sp, giant prawns, black tiger shrimp, health status, tilapia

Abstract

ABSTRACT Lemon (Citrus limon) is a fruit that can be used as an antimicrobial/antibacterial because it contains secondary metabolites, such as flavonoids, tannins, terpenoid, and asam sitrat. Edwardsiella tardawas pathogenic bacteriathat can cause diseases in cultured fish.This research was conducted from Mei to Juli 2019, at Parasites and Fish Disease Laboratory, Faculty of Fisheries and Marine, University of Riau. The purpose of this study was to get the sensitivity of lemon(Citrus limon) in inhibit he growth ofEdwardsiella tarda, the range of Minimal Inhibitory Concentration (MIC) and the lethal dose (LD50)of lemon (Citrus limon) solution against to (Pangasius hypophthalmus) with length 7-9 cm by immersion for 24 hours. Sensitivity test is using the method of KirbyBauer disk blank 6 mm disc with dose used were 100-0,3% and control Oxytetracyclin. The results showed that the sensitivity of Lemon solution towardsE. tardaat dose 100% with clear zone 14,96 mm and dose 0,3% with clear zone 6,00 mm.  At a concentration of 0,5% was determined as the MIC value with a bacterial density 265,33 CFU / mL.Dose LD50 lemon solution against catfish by immersion for 24 hours in 0,49%Keyword :(Citrus limon), Edwardsiella tarda, Sensitivity, Minimum Inhibitory Concentration, LD50

Abstract

ABSTRACT This research was conducted in April until June 2018 at the Experimental Pond and Laboratory of Parasites and Fish Diseases, Faculty of Fisheries and Marine, University of Riau. The purpose of this study was to determine the effect of the addition of propolis on feed to improve the non-specific immune response of Pangasius hypophthalmus which is maintained in cages. The research method used in this study was an experimental method with a completely randomized design (CRD) consisting of 5 treatments and 3 replications, with the treatment of feeding containing propolis ie P0 (without the addition of propolis), P1 (addition of propolis600 ppm / kg of feed ), P2 (addition of propolis 700 ppm / kg of feed), P3 (addition of propolis 800 ppm / kg of feed) and P4 (addition of propolis 900 ppm / kg of feed). The test fish used was jambal siam fish with a size of 8-10 cm as many as600 tails. The parameters measured are; total leukocytes, leukocyte differentiation, phagocytic activity, survival, absolute weight growth and water quality measurements. The results obtained from the study, namely there is the effect of giving propolis in feed with the best dose is 800 ppm / kg of feed (addition of propolis 800 ppm / kg feed) is real P <0.05 and the best dose with a total leukocyte of 10.06 x104 cells / mm3, lymphocyte percentage 84.00%, neutrophils 6.33%, monocytes 9.67%, phagocytic activity 35.33%, survival 98% and absolute weight growth 39.18 g. Keywords: Leukocytes, Pangasius hypophthalmus, Propolis

Abstract

ABSTRAKThis research was conducted from October to December 2018 at the Laboratory of Parasites and Fish Diseases, Faculty of Fisheries and Marine, University of Riau. This study aims to obtain the best dose from the solution of harumanis mango seeds (Mangifera indica L.) to treat catfish (P. hypophthalmus) infected with A. hydrophila seen from the picture of leukocytes. While the expected benefits are a solution of harumanis mango seeds can be used as a medicine to treat Motile Aeromonas Septicemia (MAS) disease. The method used in the research is the experimental method using a completely randomized design (CRD) with five levels of treatment, namely (Kn): Infected fish A. hydrophila and untreated mango seed solution, (Kp): Fish infected A. hydrophila are not treated with mango seed solution, (P1): Fish infected A. hydrophila were soaked with mango seed solution at a dose of 1600 ppm, (P2): Fish infected A. hydrophila were immersed in a manga seed solution at a dose of 1800 ppm (P3): Infected fish A. hydrophila is soaked in a solution of mango seeds at a dose of 2000 ppm. The leukocyte parameters measured were total leukocytes, leukocrit percentage, leukocyte differentiation and phagocytic activity.The results showed that the solution of harumanis mango seeds was influential in treating catfish infected with A. hydrophila, the dose of 2000 ppm mango seed solution was the best dose in the treatment of catfish infected with A. hydrophila seen from a total leukocyte of 7.37 x 104 cells / mm3, leukocrit levels of 2%, phagocytic index of 35.67%, and survival rate reached 80%. While the percentage of 81% lymphocytes, 10.67% monocytes and 8.33% neutrophils. The results of measurements of water quality during the study, namely temperature 28.2-28.50C, pH 5-6.7 dissolved oxygen 5-6.7 mg / L, and ammonia 0.02 mg / L. Keywords: Description of Leukosit, Pangasius hypophtalmus, Mangifera indica L., Aeromonas hydrophila

Abstract

ABSTRACTThis study was conducted on June to October 2017 in the Laboratory of Parasites and Fish Diseases, Fisheries and Marine Science Faculty, Riau University. The objective of this research was to understand the best of Lawsonia inermis L. solution for immersion in preventing the  of Aeromonas hydrophila infection. There were five treatments applied namely negative  control (Kn) was the fishes were not infected with Aeromonas hydrophila and were not immersed with Lawsonia inermis L. solution, positive control (Kp)  was the fishes were not immersed Lawsonia inermis L.  solution but were infected with Aeromonas hydrophila. The dose of Lawsonia inermis L. solution were 2000 ppm (P1), 3000 ppm (P2), 4000 ppm (P3). The fish was immersed for five minute in once a week for five times immersion. The result shown that the immersing with Lawsonia inermis L. solution in 2000 ppm (P1) was the best dose with a total value of erythrocyte 1.92x106 cell/mm3, 37.33% of hematocrit and 7.80 g/dL of hemoglobin and  the survival rate  (P1) was 93,33 % .Keywords: Lawsonia inermis L., Pangasius hypophthalmus, Aeromonas hydrophila, erythrocyte, hematocrit

Abstract

 Abstract This research was conducted on May to June 2017in theLaboratory of Parasite and Fish Disease of Fisheries and Marine Faculty, Riau University.This research aimed to understand the health condition of Tambaqui reared in Floating Cageculture Placed in the Kampar River and in Pond in the Rumbio Village, Kampar Regency.The method used in this research was the survey method with two different locations namely up-down stream, and in pond. fish samples were of obtained in 3 times every 10 days.  The were 27 fishes, ranged from 13-16 TL. The blood was taken from the caudal vena using 1ml springe wet with 10% EDTA. Haematologycal parameters measured were blood glucose, erythrocyte, leucocyte, hematocryte and hemoglobin. Results shown that the blood glucose114mg/dL, total of erythrocytelevel 2,21x106cells/mm3, total of leucocyte 1,37x105cell/mm3, hematocryte 26,66% and hemoglobin 7,33 g/dl in floating cagesdownstream-upstreamhas a blood glucose 80,33 mg/dl, total of erythrocyte2,38x106cell/mm3, total of leucocyte1,29x105cell/mm3, hematocryte27,66% and hemoglobin 7,66  g/dl, while the fish that were rearead in the pond has a blood glucose 66mg/dl, total of erythrocyte2,37x106 cell/mm3, total of leucocyte1,27x105cell/mm3, hematocryte27,00% and hemoglobin7,66 g/dl. It can be concluded that the best result was in the fish result reared in Pond Rumbio village which was very suitable for Tambaqui fish. Key Words : Hematologycal,  Colossoma macropomum, Cageculture, Pond

Abstract

This study aimed to isolate and to find the bacteria that potential as probiotic candidate from Giant Gouramy (Osphronemus gouramy Lac.) intestine cultivated in Riau provincial to be used to control MAS (Motile Aeromonas Septicemia) desease. Survey method and experiment method were conducted in this study. Bacteria was isolated from the fish intestine, and was inoculated on TSA and MRS agar at different pH level (2,4, and 6). The isolateis were indentified by morphological observation, physiological and biochemical test, and by using API 50 CHL test kit. Antibacterial activity of the isolated against Aeromonas hydrophila was tested by the paper disk diffusion method. Eighteen isolates of probiotic candidates were found, and indicated optimal growth in all pH treatment. Based on morphological, physiological test, and biochemichals test, three genus of bacteria were identified as Bacillus sp., Lactobacillus sp., and Bifidobacterium sp. The highest antibacterial activity against A. hydrophila was indicated by Isolate G63 with inhibition zone of 13,2 mm. Based API 50 CHL test kit the isolate was categorized in to Lactobacillus planatarum at similarity level of 84%.Keywords: Giant Gouramy, intestine, isolate, probiotic candidate
