Abstract

Makanan tradisional merupakan salah satu jenis makanan yang mudah tercemar oleh bakteri karena diolah manggunakan cara yang sederhana serta adanya kandungan kandungan mineral, protein, karbohidrat dan air serta pH yang sesuai untuk pertumbuhan bakteri, sehingga dapat menyebabkan perubahan aroma maupun cita rasa akibat terkontaminasi oleh bakteri. Sotong pangkong merupakan salah satu jenis makanan tradisional yang ada di Kota Pontianak. Tujuan penelitian ini mendeteksi bakteri anggota Enterobactericeae pada makanan tradisional sotong pangkong. Pengambilan sampel dilakukan secara acak (random sampling) pada pedagang sotong pangkong di Kota Pontianak. Deteksi dan identifikasi bakteri dilakukan di Laboratorium Mikrobiologi, Jurusan Biologi, Fakultas Matematika dan Ilmu Pengetahuan Alam (MIPA), Universitas Tanjungpura, Pontianak, pada bulan November hingga  Berdasarkan hasil deteksi dan identifikasi ditemukan bakteri anggota Enterobactericeae pada makanan sotong pangkong, yang meliputi enam genus bakteri, yaitu anggota genus Escherichia, Enterobacter, Klebsiella, Salmonella, Proteus dan Shigella.

Abstract

Ayam broiler merupakan komoditas pangan yang memiliki nilai gizi yang tinggi dan mudah didapatkan, baik itu di pasar swalayan atau pasar tradisional. Penelitian ini bertujuan untuk mengetahui jumlah koloni bakteri pada daging ayam broiler yang dijual di pasar swalayan Kota Pontianak berdasarkan perhitungan Angka Lempeng Total (ALT). Sampel penelitian diambil dari tiga pasar swalayan di Kota Pontianak. Hasil pengamatan angka lempeng total bakteri dari 3 pasar swalayan yang terdapat di Pontianak melebihi ambang batas cemaran mikroba menurut SNI Tahun 2009 yaitu 1 x 106 cfu/ml, dengan hasil pengamatan sebagai berikut swalayan A berjumlah 3,2x106 cfu/ml, swalayan B berjumlah B 6,7 x 106 cfu/ml dan swalayan C berjumlah 3,8 x 106 cfu/ml.

Abstract

Jamur makroskopis dapat dimanfaatkan sebagai bahan makanan dan obat.Pada ekosistem hutan, jamur makroskopis berperan sebagai dekomposer. Penelitian ini bertujuan untuk mengetahui jenis-jenis jamur makroskopis anggota kelas Basidiomycetes yang terdapat di Hutan Bayur, Kabupaten Landak, Kalimantan Barat. Metode yang digunakan dalam penentuan stasiun sampling adalah purposive sampling dengan memilih 3 stasiun berdasarkan perbedaan rona lingkungan. Pengambilan sampel dilakukan dengan metode jelajah (Cruise Method). Identifikasi jamur makroskopis dengan mencocokkan karakteristik morfologis sampel dengan buku identifikasi. Hasil penelitian menunjukkan bahwa ditemukan 18 jenis jamur makroskopis yang termasuk ke dalam empat Ordo, yaitu Aphyllophorales, Agaricales, Auriculariales dan Polyporales. Jamur makroskopis yang ditemukan mendominasi berasal dari Ordo Aphyllophorales, seperti Pycnoporus sanguineus, Polyporus versicolor, Polyporus varius, Schizophyllum commune, Thelephora palmata, Earliella scabrosa, Lignosus rhinocerus, Trametes versicolor, Heterobasidion annosum dan Ganoderma applanatum. Beberapa jenis jamur makroskopis yang dimanfaatkan sebagai bahan makanan oleh masyarakat setempat adalah spesies Auricularia polytricha, Hygrocybe conica, Pleurotus ostreatus dan Schizophyllum commune.

Abstract

Jamur makro diketahui memiliki peran penting bagi kehidupan manusia dan kelestarian lingkungan. Penelitian ini bertujuan untuk mengetahui diversitas jenis-jenis jamur makro yang terdapat di Hutan Rubatn, Kecamatan Sompak, Kabupaten Landak, Provinsi Kalimantan Barat. Penelitian ini dilaksanakan selama 4 bulan dari bulan Mei sampai dengan Agustus 2018 menggunakan metode jelajah. Identifikasi jamur makro dilakukan berdasarkan ciri morfologi. Sebanyak 21 spesimen dari 15 marga jamur makro ditemukan di Hutan Rubatn, yaitu Earliella, Lignosus, Cerioporus, Pycnoporus, Trametes, Fomes, Lentinus, Ganoderma, Rigidoporus, Schizophyllum, Marasmius, Pleurotus, Mycena dan Stereum (Basidiomycetes), dan Daldinia (Ascomycetes). Jamur-jamur tersebut ditemukan di berbagai substrat, yaitu tanah, serasah dan pohon mati.

Abstract

Jeruk siam  (Citrus nobilis var.microcarpa) dapat terserang berbagai penyakit yang menunjukkan gejala pada daun. Penelitian ini bertujuan untuk mengetahui karakterstik morfologis jamur anggota genus Aspergillus dan Colletotrichum dari daun jeruk siam yang bergejala sakit di perkebunan kota Singkawang. Pengambilan sampel mengunakan metode purposive sampling di perkebunan Kelurahan Setapok Besar, Kecamatan Singkawang Utara. Penelitian dilakukan Laboratorium Mikrobiologi, Jurusan Biologi, Fakultas Matematika dan Ilmu Pengetahuan Alam, Universitas Tanjungpura, Pontianak, pada bulan Februari sampai November 2021. Penelitian ini menggunakan metode tanam langsung (direct plating). Identifikasi dilakukan berdasarkan karakteristik morfologis. Pengamatan karakteristik morfologis dilakukan dengan cara mengamati warna permukaan atas jamur, warna permukaan bawah jamur, karakter hifa, konidia dan konidiofor. Hasil penelitian diperoleh 3 isolat jamur Aspergillus dan 2 isolat jamur Colletotrichum yang memiliki karakter berbeda pada karakteristik makromorfologis dan mikromorfologis.

Abstract

Program Peningkatan Kapasitas Organisasi Kemahasiswaan (PPK Ormawa) Tahun 2022 dilakukan dalam bentuk penerapan sistem peringatan dini kebakaran hutan di Desa Jangkang Dua, Kabupaten Kubu Raya. Peralatan peringatan dini kebakaran hutan dibuat berbasis Internet of Things (IoT), sehingga dapat mengirimkan informasi kebakaran secara otomatis dan real time. Kegiatan ini bermitra dengan Pemerintah Desa Jangkang Dua, Kabupaten Kubu Raya. Permasalahan mitra adalah belum diterapkannya sistem peringatan dini kebakaran hutan di Desa Jangkang Dua, padahal desa ini termasuk dalam zona rawan terhadap kebakaran hutan dan lahan. Tujuan dari kegiatan ini adalah untuk menerapkan sistem peringatan dini kebakaran hutan dengan memasang peralatan peringatan dini dan mendorong masyarakat agar tanggap terhadap bencana kebakaran hutan. Kegiatan dilaksanakan selama 6 bulan, yaitu tanggal 27 Juni – 30 Desember 2022, oleh Himpunan Mahasiswa Geofisika (HMG) Fakultas Matematika dan Ilmu Pengetahuan Alam (FMIPA) Universitas Tanjungpura. Bentuk kegiatan pada program ini yaitu sosialisasi, edukasi, pemberdayaan masyarakat, pemasangan peralatan peringatan dini, dan pendampingan. Kegiatan ini telah berhasil menerapkan sistem peringatan dini kebakaran hutan di Desa Jangkang Dua. Hasil evaluasi menggunakan kuesioner dengan skala Likert menunjukkan bahwa penerapan sistem peringatan dini kebakaran hutan sudah terlaksana sangat baik dengan indeks persentase sebesar 91,44%. Hasil ini menunjukkan bahwa penerapan sistem peringatan dini kebakaran hutan dapat mencegah kebakaran hutan sehingga mengurangi dampak kerugian dari kebakaran hutan di Desa Jangkang Dua.The 2022 Student Organization Capacity Building Program (PPK Ormawa) was implemented by applying a forest fire early warning system in Jangkang Dua Village, Kubu Raya Regency. The early warning system devices are based on the Internet of Things (IoT), so they can send fire information automatically and in real time. This activity is in partnership with the Jangkang Dua Village Government, Kubu Raya Regency. The partner's problem is that a forest fire early warning system still needs to be implemented in Jangkang Dua Village, even though this village is in a zone prone to forest and land fires. This activity aims to implement a forest fire early warning system by installing early warning system devices and encouraging the community to be responsive to forest fire disasters. The action was carried out for about six months, from 27th July – 5th December 2022, by the Geophysics Student Association (HMG), Faculty of Mathematics and Natural Science (FMIPA), Universitas Tanjungpura. The activities in this program are socialization, education, community empowerment, installation of early warning system devices, and mentoring. This activity has successfully implemented a forest fire early warning system in Jangkang Dua Village. The evaluation result using a Likert scale questionnaire showed that the implementation of the forest fire early warning system was very well implemented, with a percentage index of 91.44%. This result indicates that implementing the forest fire early warning system can prevent forest fires and reduce the impact of losses from forest fires in Jangkang Dua Village.

Abstract

The objective of the present study was to detect presence of pathogenic fastidious bacterium, Candidatus Liberibacter asiaticus using PCR from leaves of Siam citrus showing Citrus Vein Phloem Degeneration (CVPD) symptoms in Singkawang City plantation, Pontianak, West Kalimantan, Indonesia. Citrus leaf samples were collected based on visual observation of symptoms showing CVPD infection. Typical symptoms of CVPD include leaf yellowing (chlorosis), vein banding, leaves become stiff, thicker and smaller in size. The pathogenic bacterium, Candidatus Liberibacter asiaticus was detected using two specific primers, namely, OI1/ OI2c amplified 16S rRNA gene and A2/J5, amplified ribosomal protein gene of the rplKAJL-rpoBC operon (β-operon). PCR amplification detected the presence of 1100 bp band using OI1/ OI2c primers, and 703 bp band using A2/J5 primers from symptomatic Siam citrus leaves. PCR products were not detected from healthy plants serve as control. By using two sets of specific primers to amplify 16S rRNA gene and ribosomal protein gene, Candidatus Liberibacter asiaticus was detected in symptomatic Siam citrus leaves in Singkawang City, Pontianak, Indonesia. Detection of the bacterial pathogen causing CVPD is important to prevent spreading of the disease which could affect production of citrus fruits.

Abstract

Lactic acid bacteria are included in the gram positive bacteria that are widely distributed in the digestive tract of living things, such as in the feces of the Pandan Luwak (P. hermaphroditus) and important role in improving the microbial balance in the body. The purpose of this study was to obtain phenotypic characters, types of bacteria and similarity relationship between lactic acid bacteria from feces of Pandan Civet (P. hermaphroditus) in West Kalimantan and lactic acid bacteria in the identification book Bergey's manual of determinative bacteriology. This research was conducted by the method of isolation and screening bacteria using MRSA media supplemented with CaCO3 1%. Characterization of lactic acid bacteria based on morphological, biochemical, and physiological characters in Bergey's Manual of Determination Bacteriology. The results of the analysis using the MVSP (Multivariant Statistic Package) program with SSM coefficients and the UPGMA method obtained 44 bacterial isolates  thought to belong to two genera, namely the genus Lactobacillus and Bifidobacterium with three species, namely L. casei, L. brevis and B. bifidum. The results of the analysis showed that 12 bacterial isolates had a similarity index of 87,6% to L. casei, 17 bacterial isolates had a similarity index of 92,2% to B. bifidum and 15 bacterial isolates had a similarity index of 84,5% to L. brevis.

Abstract

Latic acid bacteria play a significant role in the fermentation of food, such as kimchi, which is fermented vegetables. Sawi kampung ansabi is a vegetable cultivated by the Dayak people in Kalimantan during the rice farming season which can be fermented into kimchi. Based on phenotypic similaities, the aim of this study was to identify and acquire cultures of lactic acid bacteria on MRSA media (De Man Rogosa and Sharpe Agar) enriched 1% CaCO3 from kimchi sawi ansabi fermented food sawi ansabi. The results of the isolations were used to test 10 isolates of lactic acid bacteria using the MVSP (Multivariate Statistical Package) program, the UPGMA (Unweighted Pair Group Method with Arithmetic Average Mean) method based on the Simple Matching Coefficient and Jaccard Coefficient to form dendrograms. Simple Matching Coefficient analysis resulted 8 isolates suspected to be Lactobacillus plantarum with a similarity index of 94.6%, 1 suspected 91.9% to Lactobacillus dellbrueckii, and 1 suspected 91.9% to Pediococcus cerevisiae. The Jaccard Coefficient analysis resulted in 8 isolates suspected of Lactobacillus plantarum with a similarity index of 92.6%, 1 isolate suspected 87% to Lactobacillus dellbrueckii, and 1 isolate 88% to Pediococcus cerevisiae.

Abstract

A consortium of microorganisms is a mixture of microbial populations in the form of a community that has cooperative, commensal, and mutualistic relationships. Naturally, bacteria are able to communicate with each other. The relationship between bacteria can have a synergistic relationship if the substrate is sufficient and the bacteria do not inhibit each other's growth. In this study, Bacillus cereus isolates SP1, SP2, and SP3 isolated from Sungai Ambawang peatland, Kubu Raya Regency, West Kalimantan will be consorted. The purpose of this study was to determine the synergism of three strains of Bacillus cereus bacteria. All isolates tested for synergism against other isolates showed no inhibition zone at the intersection between isolates. This means that each isolate can synergize with one another. The synergism test results show that three strains of Bacillus cereus bacteria can synergize in the manufacture of biofertilizers.
