Abstract

Background: One of the conditions for releasing standardized herbal medicines is the presence of pre-clinical testing that can be conducted with mice. Emulsion products with the main composition of pumpkin, red palm oil, and dragon fruit have high levels of β-carotene. The purpose of this study was to observe changes in weight, cholesterol, and sub-chronic toxicity from mice treated with emulsion products.Materials and Methods: Mice observed consisted of 6 groups: (1) untreated control (given standard ration); (2) negative control (given standard ration and 2.8% (v/b) egg yolk); (3) positive control (given standard ration and 0.4% (v/b) β-carotene); (4) standard ration, 0.4% (v/b) β-carotene and 2.8% (v/b) egg yolk; (5) given a standard ration and 2% (v/b) emulsion products; and (6) given a standard ration, 2% (v/b) emulsion products and 2.8% (v/b) egg yolk. Observations of sub-chronic toxicity were done by measuring creatinine levels, kidney weight, and visual observation of kidney swelling. Results: The use of β-carotene-rich emulsions does not cause a decrease in the average weight of mice. In general, β-carotene and emulsion can overcome cholesterol increases on the 7th day, but not for the 15th day as a result of standard food intake. The increase in creatinine levels only occurs in mice treated with egg yolk control. The additional treatment of β-carotene and emulsion successfully prevents the increase in creatinine levels. Conclusion: Emulsion products did not cause changes in weight, were able to reduce cholesterol levels in a limited manner and were not toxic to mice. Keywords: cholesterol changes, sub-chronic toxicity, b-carotene, emulsion

Abstract

Application of natural deep eutectic solvent (NADES) citric acid-glucose-based microwave-assisted extraction (MAE) method for total polyphenol content (TPC) enriches the use of Eleutherine bulbosa (Mill.) bulb more widely and commercially. Therefore, this study aims to optimize NADES citric acid-glucose-based MAE on the TPC from E. bulbosa bulbs using response surface methodology (RSM). In the present study, the dried sample of E. bulbosa bulb was extracted using the NADES based MAE. Determination of TPC was conducted using Folin-Ciocalteau reagent and standard gallic acid, then measured using a UV-Vis spectrophotometer at 761 nm. Extraction condition optimization of the NADES based MAE method was performed using RSM with Box Behnken Design (four-factors-three-levels and 29 runs) on Design Expert v12. Based on the results study, the optimum condition extraction on the TPC enrichment was obtained at the NADES ratio (citric acid: glucose) 1:1 g/g; solid-liquid ratio 1:8 g/mL; extraction time of 15 min; and 270 Watt microwave power. The confirmation test and scale-up (50 g samples) were obtained with a 61.63 ± 2.23 mg GA/g sample.

Abstract

Silver nanoparticles themselves have been very much developed both through the method of synthesizing silver nanoparticles both physically, chemically and biologically. In the chemical method we can do by reducing Ag+ ions into silver nanoparticles which have an attachment to the reductant and also have a very strong ability to reduce Ag+ ions to become silver nanoparticles have a small size. Based on the literature review used, the reductor is Sodium Borohydrate (NaBH4). Chitosan acts as a capping agent which functions as a stabilizer in the silver nanoparticles. Silver nanoparticles modified using chitosan from previous research, namely for activity tests as antioxidants, as metal sensors and as antibacterials Where there is a change in color on silver nanoparticles from clear to yellow. Abstract

Silver nanoparticles with a size of 1 to 100 nm with unique properties have many advantages that they are widely applied in the medical and industrial fields. Various methods can be carried out in synthesizing silver nanoparticles both physically, chemically and biologically. In the chemical method, it is done by reducing Ag+ ions into silver nanoparticles so that this method depends on reducing agents and stabilizers used. The use of different reducing agents greatly affects the size of the nanoparticles produced. Based on the literature review, sodium borohydride (NaBH4) has a strong ability to reduce Ag+ ions into silver nanoparticles become a small size. In addition, referring to previous studies if modified with L-cysteine, the silver nanoparticles formed can be stable for more than 2 months at room temperature storage. L-cysteine-modified silver nanoparticles from previous studies were used for colorimetric analysis of vitamin B1 detection in food and water samples, where the color change in silver nanoparticles from yellow to colorless showed remarkable chemical sensor selectivity for detecting target analytes.
