Abstract

Ikan komet (Carassius auratus auratus) merupakan ikan hias air tawar yang banyak dibudidayakan dan memiliki warna yang menarik. Produksi benih ikan komet belum bisa memenuhi tingkat permintaan pasar yang terus meningkat setiap tahunnya, sehingga perlu dilakukan peningkatan pada kegiatan pembenihan. Salah satu faktor yang memberikan pengaruh sangat besar terhadap tingginya kematian ikan pada kegiatan pembenihan adalah suhu. Suhu merupakan faktor lingkungan yang dapat mempengaruhi pertumbuhan rata-rata serta menentukan waktu penetasan serta berpengaruh langsung pada proses perkembangan embrio. Secara umum fase awal kehidupan ikan merupakan fase yang paling sensitif dan mudah menjadi stres dalam menerima pengaruh lingkungan. Tujuan penelitian ini yaitu untuk mengetahui pengaruh suhu terhadap embriogenesis telur ikan pelangi, waktu inkubasi telur ikan pelangi, dan suhu optimal untuk menghasilkan daya tetas maksimal telur ikan pelangi. Penelitian ini dilaksanakan pada bulan Juni-Juli 2015 di Unit Pengembangan Budidaya Air Tawar (UPTBAT) Punten, Batu, Jawa Timur. Metode yang digunakan yaitu metode eksperimental. Variabel yang diamati adalah perkembangan embrio dan daya tetas telur ikan komet. Hasil penelitian menunjukkan bahwa suhu berpengaruh terhadap waktu perubahan fase embriogenesis dan waktu inkubasi telur ikan komet. Semua perlakuan suhu (22,5-29,5°C) merupakan suhu optimal pemeliharaan telur ikan komet.

Abstract

Collagen is a connective tissue protein that is mostly produced from cattle and pigs. The use of cows and pigs as the manufacture of collagen causes a disease case, among others are bovine spongiform encephalopathy and transmissible spongiform disease. One of the alternative materials in the isolation of collagen is the use of fish skin. The purpose of this study was to determine the effect of the concentration of acetic acid solution on the characteristics of red snapper collagen produced by the hydroextraction method and to determine the optimum concentration of acetic acid solution in the red snapper skin collagen isolation process using the hydroextraction method. This study was experimental with Completely Randomized Design (CRD) consisting of 3 acetic acid treatment: (P1) acetic acid with concentration 0,1 M; (P2) 0,15 M and (P3) 0,2 M. This study showed that the use of different acetic acid had a significant effect (p<0.05) on the yield parameters that is 1.72 – 2.46%, water content 11.12 – 12.8%, and protein content 83.66 – 84.81% and had no significant effect on the pH. The best treatment was P3 which use the higher concentration of acetic acid that produced higher yield 2.46% and protein content of 84.81%. P3 with concentration of acetic acid 0,2 M can be used for found the best result of characterization of collagen.

Abstract

Fish Cantang grouper is a cross between a female tiger grouper (Ephinepelus fuscoguttatus) and a male kertang grouper (Ephinepelus lanceolatus), has a high economic value. Salinity is a water quality factor that affects cultivation because salinity has osmotic pressure that can cause changes in physiological activity and stress in fish if it is not in accordance with the habitat. Stress in fish causes the release of cortisol and catecholamine hormones. The catecholamine hormone then increases the glycogenolysis process, regulates cardiovascular and respiratory function, in addition to increasing cortisol and blood glucose. Stress in fish is also indicated by a hematological response in the form of changes in the number of erythrocytes. This study aims to determine the effect of different salinity treatments on physiological and hematological responses. The results of the analysis of variance (ANOVA) carried out from the initial observation to 24 hours showed that each salinity treatment had a significantly different effect on blood cortisol, blood glucose, and erythrocyte levels, with a value (p <0.05). The highest average blood cortisol was in the 36 ppt treatment at 24-hour observation time. The average blood glucose increased from before being treated, until receiving treatment for up to 24 hours. The highest average was obtained at a salinity of 36 ppt, with observation for 24 hours. The results obtained from the study showed that the highest number of erythrocyte cells at 24 hours was in the P2 treatment (36 ppt), namely 1.15 x 106 cells/mm³.
