Abstract

Saluran pencernaan merupakan lingkungan kompleks yang terdiri atas berbagai spesies bakteri. Saluran pencernaan biota perairan terdiri atas bakteri aerob dan anaerob yang mampu memproduksi senyawa antibakteri dan enzim. Tujuan penelitian ini adalah untuk mengisolasi bakteri asam laktat yang menghasilkan protease dan senyawa bakteriosin-like dari saluran pencernaan kepiting bakau. Isolasi dan karakterisasi isolat dilakukan menggunakan media MRS. Supernatan netral bebas sel isolat telah diuji dengan menggunakan disc difusi agar terhadap bakteri patogen dan pembusuk. Uji produksi enzim protease telah diuji dengan metode disc diffusion agar menggunakan media kasein agar. Di antara seratus isolat, terdapat 96 isolat menunjukkan zona bening di MRS + CaCO3,, katalase negatif, dan bakteri Gram positif. Tiga puluh empat isolat bakteri asam laktat menghasilkkan protease dan hanya empat isolat (yaitu IKP29, IKP30, IKP52, dan IKP94) menunjukkan penghambatan yang kuat terhadap bakteri patogen dan pembusuk. Terdapat tiga pola inhibisi dari keempat isolate terhadap Bacillus subtilis, Staphylococcus aureus, Eschericia coli, dan Salmonella sp. Empat isolat tersebut berpotensi untuk dimanfaatkan sebagai starter pada produksi fermentasi produk hasil perikanan. Penelitian ini adalah penelitian pertama terkait isolasi bakteri asam laktat yang menghasilkan protease dan bakteriosin dari saluran pencernaan dari kepiting bakau.Kata kunci: Bakteri Asam Laktat, Bakteriosin-like substance, Protease, Scylla  sp. Digestive tract is complex environment consist of large amount of bacteria’s species. Fish intestine bacteria consist of aerobic or facultative anaerob bacteria which can produce antibacterial and enzym. The objectives of this research were to isolated lactic acid bacteria that produce bacteriocin-like and protease from mud crab digestive tract. Isolation and characterization of isolates were conducted employing media MRS.  Neutralized cell free supernatant of isolates were tested using disc diffusion agar of against pathogenic and spoilage bacteria to indicate bacteriocin-like-producing lactic acid bacteria. Protease-producing isolate was tested using disc diffusion method in casein agar. Among a hundred isolates, 96 isolates were showed clear zone in MRS+CaCO3,, catalase negative, and Gram positive bacteria. Thirty four isolates produced protease and only four isolates (i.e. IKP29, IKP30, IKP52, and IKP94) showed strong inhibition against pathogenic and spoilage bacteria. There were three patterns of inhibition among three isolates against Bacillus subtilis, Staphylococcus aureus, Eschericia coli, and Salmonella sp. All three isolates showed potential uses for produce starter culture for fishery product fermentation purpose. This is the first report of isolation lactic acid bacteria that produced protease and bacteriocin-like from digestive tract of mud crab. Abstract

The burrowing goby, Trypauchen vagina, is recorded for the first time from the Pangpang Bay of Banyuwangi, Indonesia. Types of fish T. vagina is a type that has not published in Indonesia. This fish was caught using trap net in waters Pangpang Bay, which is an essential area in Banyuwangi, East Java. The description of the morphology and fish habitat conditions shows that the region has a diversity of fish resources that need attention. This goby fish has a local name as fish bedhul picek due to the sense of sight on these fish as if it does not work. Eyes covered by skin and socket head section was not found their eyeballs. The body of the specimens is elongate and reddish-pink in fresh specimens in coloration. There is no barbel on the chin. Cycloid scales are present on the whole body except for the head. Muncar people commonly consume T.vagina. This fish has been food sources in several regions but uncommon at several local markets in Indonesia. The T.vagina inhabit at Pangpang Bay as the bay, which has the high primary productivity of these waters due to sufficient nutrient support. Enrichment of nutrient comes from the run-off land and also the input of the number of fishing activity (embankment). An earlier report said that this fish found in waters Banyu Asin, Palembang, Sumatra, Indonesia. It also confirms that the fish found in this study are also living in a mud substrate that is reasonably smooth with right mangrove conditions around the bay, which close to Alas Purwo National Park.

Abstract

A single specimen of saltwater crocodile Crocodylus porosus Schneider, 1801 (1.50 cm in length) was captured and photographed by a local fisherman by gill net at fishpond around mangrove ecosystem of east coast Surabaya on December 2020. The location where the estuarine crocodile caught is an important wetland area in Surabaya with an excellent mangrove cover. Wonorejo mangrove location is about 10 km from the centre of Surabaya, the second-largest city in Indonesia. This finding is considered as a first reported of C. porosus in Surabaya, and at the same time is proof of the importance of the mangrove area on the east coast of Surabaya as a habitat for wildlife that must be conserved appropriately. Routine monitoring of mangrove areas, including their wildlife, is essential so that they are not displaced by the development of the city which changes the land function to become a residential area.

Abstract

Aquaponics is a technology that combines the cultivation of vegetable production and fish farming. The aquaponics technology also to fertilization to give nutrients to the plant to support of survival. In facts, fertilization done to plants is located on land. Fertilizing the plants that live in the waters are still very rare, because the waters as a growing medium that is considered to always provide sufficient nutrients for plant growth. In this research, using biofertilizer Euchema cottonii of seaweed. The advantages of biofertilizer seaweed is to have the content of macro and micro nutrients are complete. It also contains substances hyper Grow (PGR) plant (auxin, cytokinin, gibberellin, abisat acid and ethylene).In this research to determine the effect of biofertilizer products of seaweed (E. cottonii) commercial on the growth of dumbo catfish (C. gariepinus) and pakcoy mustard (B. rapa L) on aquaponics system. The design of experiments in this research using a completely randomized design (CRD) with 5 treatments and 4 replications. The treatment used is fertilizer "AB mix®" 1% (A), without fertilizer (B) and the provision of products biofertilizer seaweed (E. cottonii) commercial 0,16% (C), 0,18% (D) and 0,20% (E). The main parameters measured were heavy growth of biomass, plant height pakcoy mustard (B. rapa L), growth SGR, length and survival rate of fish of dumbo catfish (C. gariepinus).The results showed that the dose of different commercial product biofertilizer seaweed (E. cottonii) gives a significantly different effect on the height growth of pakcoy mustard (B. rapa L) and the survival rate of dumbo catfish (C. gariepinus). The optimal dosage of commercial seaweed (E. cottonii) biofertilizer product in increasing the growth of pakcoy mustard (B. rapa L.) and African catfish (C. gariepinus) in the aquaponic system, namely treatment E at a dose of 0.20%.

Abstract

Increasingly rapid industrial development poses serious environmental problems such as wastewater.The higher the number of production, the more waste that wasted and can decrease the environment quality. Fieldwork practice activity is carried out starting from the date of January 20, 2014, until February 15, 2014. The purpose of this fieldwork practice is to acquire knowledge and skills on the field of wastewater treatment technique and to know the parameters of the wastewater inlet and outlet channels in comparison with wastewater quality standards. The method that used in Field Work Practice This is descriptive, the method to describe the circumstances or events in a particular region. The result obtained from fieldwork practice known that wastewater treatment techniques consist of preliminary treatment activities, primary treatment, secondary treatment, sedimentation, and disinfection. Preliminary treatment activities is a filtering process substantial in the wastewater that comes out of the inlet channel using four mesh size of the net. The primary treatment for separating solids that escaped from the screening process and disposed to wastewater reservoirs. Secondary treatment activity including Aerobacter sp., Nitrosomonas sp., Nitrobacter sp., Bacillus sp. of bacteria and oxygen addition inside of wastewater. Sedimentation phase separation process is carried out suspended solids in wastewater using clarifier pond. Disinfection stage which activities the addition of chlorine with 222 ppm of doses to kill harmful pathogens. The final stage test to determine the parameters of wastewater effluent quality by using living fishes and taking the sample from another institution.

Abstract

kan Cupang Betta splendens adalah salah satu jenis ikan hias peliharaan yang mempunyai daya tarik pada warna yang dimunculkan dari tubuhnya. Ikan cupang memiliki nilai ekspor US$ 4,911. Kualitas warna ikan hias menentukan nilai ekonomis, tampilan warna yang indah merupakan salah satu indikator yang menjadi daya tarik. Salah satu usaha yang dilakukan untuk mendapatkan warna cerah yang merata pada ikan adalah dengan teknik manipulasi pigmen ke dalam pakan. Mikroalga Porphyridium cruentum merupakan mikroalga merah yang memiliki manfaat yang digunakan sebagai sumber pigmen alami. Penelitian ini bertujuan untuk mengetahui pengaruh penambahan P. cruentum pada pakan terhadap kecerahan warna, dan kelulushidupan ikan cupang. Metode penelitian ini menggunakan rancangan Acak Lengkap (RAL) dan perlakuan yang dilakukan dalam penelitian ini menggunakan perbedaan dosis P. cruentum yang berbeda yaitu menggunakan P0 sebagai kontrol dengan pemberian cacing darah, P1 dengan dosis P. cruentum 0%,P2 dosis P. cruentum 1%, P3 dosis P. cruentum 3%, P4 dosis P. cruentum 5%. Kemudian dilakukan ulangan 4 kali. Hasil penelitian ini menunjukkan bahwa penambahan P. cruentum berpengaruh terhadap perubahan kecerahan warna ikan cupang. Perubahan kecerahan warna ikan cupang dilihat dari perubahan nilai hue (o). Penambahan P. cruentum tidak berpengaruh terhadap kelulushidupan. Penambahan dosis 1% P. cruentum pada pakan menghasilkan kecerahan warna terbaik selama empat minggu yaitu 8.50, 15.50, 21.00, dan 27.50 sedangkan dosis 0% menghasilkan kecerahan warna terendah yaitu 0.00. Nilai kelulushidupan selama penelitian pada tiap perlakukan tidak berbeda nyata yaitu 100%.

Abstract

The potential of freshwater fisheries in Bangka Regency, Bangka Belitung Province, is sufficient to support fisheries activities in this area. An activity that is quite prominent is catfish farming using swamps and tarpaulin ponds on limited land. In this study, the analysis of cultivation activities was carried out on the Bukit Dempo community group who became partners in the national community service activities, in cooperation with Universitas Airlangga and Bangka Belitung University. Characteristics of the pond using a combination of fixed nets with a net diameter of 0.5 cm. In the pond also found water plants in the form of water hyacinth which is expected to be able to make a symbiotic use of the organic material that has been left behind from feed and catfish waste. The cultivation system is classified as semi-intensive with a combination of commercial feed with high protein (31-33%).  The main problem in this cultivation system is low productivity and the lack of technology applied in an effort to increase production efficiency Furthermore, high production yields are marketed to Pangkal Pinang City and also to local areas around the Belinyu sub-district. Technical constraints include Motile Aeromonas Septicemia (MAS) due to the bacterium Aeromonas hydrophylla, which causes about 10% of mortality in commodities before harvest. There has been no treatment action in overcoming the disease, so it is necessary to be careful in avoiding death due to disease by observing catfish at the change of seasons.

Abstract

Crabs are a group of Decapoda (Portunidae) that act as keystone species from Pangpang Bay as the marine benthic organism. Besides having an ecological function, crab also provides essential components for human health. The crab identification technique is usually conducted based on morphology and anatomy characteristics, in which certain body parts as the key for identification. This study used two identification methods, i.e. morphological features and a molecular approach. Although morphological identification has been carried out, the molecular techniques provide better accuracy and, at the same time, provide additional information about the characteristics of mitochondrial DNA. The purpose of this study is to identify the blue swimming crab caught by a traditional fisherman at Pangpang Bay, Banyuwangi, based on mitochondrial DNA sequence on cytochrome c oxidase subunit I, and reconstructed the phylogenetic tree including genetic distance also was analysed. The nucleotide sequences of the COI gene were analysed by Chromas, Clustalω, Reverse-Complement, and the MegaX. The phylogenetic tree and genetic distance calculations were carried out using Mega X software through the Neighbor-Joining (NJ) Algorithm with the addition of several sequences from the NCBI online database. This study confirmed that the specimen of Pangpang Bay is Portunus pelagicus (BWIPP001 and BWIPP003) and Portunus sanguinolentus (BWIPP002). The species of P. pelagicus have 99.99% similarities with the same species (KJ168060) from China, while the P. sanguinolentus is close to the same species (EU284144) with a per cent identity is 99.97%. The genetic distance, for P. pelagicus and P. sanguinolentus, were in range of 0.00-0.066 and 0.00-0.005 respectively.

Abstract

The Pangpang Bay Banyuwangi has a high potential for fisheries, processing and capturing fisheries, especially the order Perciformes of the families Carangidae, Lutjanidae and Serranidae. Grouper (Serranidae) is one of the high-economic marine fish commodities. However, the grouper fishing data in this area is only known as grouper without knowing the type of species. It is challenging to identify grouper species because groupers have a high degree of morphological similarity and colour variation. Therefore, molecular identification is necessary to determine the type of grouper in this region. This study aims to identify the grouper species that landed from Pangpang Bay based on DNA barcoding. In addition, phylogenetic tree analysis between grouper species (Serranidae) was carried out, and genetic distance analysis of the identified species was compared to the NCBI database. Morphological observations on groupers were carried out first, and fish meat samples were taken for experiments on molecular identification. The results of molecular-based identification revealed two grouper species Epinephelus coioides and Cephalopholis miniata. The results of phylogenetic tree reconstruction of the two groupers form a separate clade. The genetic distance between Epinephelus coioides ranged from 0.00-0.122, while the species of Cephalopholis miniata had a genetic distance of 0.00-0.002.

Abstract

Utilization of natural materials sourced from the sea is very promising. These natural ingredients can function as antibacterials that are useful in a number of fields. This study aims to explore the extract of sea cucumber Phylloporus sp. as an antibacterial agent for Vibrio alginoliticus and Vibrio harveyi. The diffusion method was carried out with three treatments of sea cucumber extract starting at doses of 500, 1000 and 1500 mg/L with three replications for each treatment. Positive control was antibiotic Chloramphenicol 100 mg/ml (0.01%) and negative control with dimethylsulfoxide (DMSO) 10%. The test results showed that sea cucumber extract was able to inhibit both types of bacteria, visible from the inhibition zone formed. In Vibrio alginoliticus bacteria, the average inhibition zone formed was 0.433 ± 0.0288. In bacteria, Vibrio harveyii was inhibited by sea cucumber extract with an average inhibition zone of 0.516 ± 0.104. Inhibition zone of sea cucumber extract Phylloporus sp. lower than Chloramphenicol which is a commercial antibacterial.
