Abstract

The presence of heavy metals in the environment can be a serious problem because it is harmful to human health. One of the heavy metal ions that pollute the environment is the Cr6+ metal ion. The usual tool used to analyze metal ions is Cr6+ is the Atomic Absorption Spectrometry (AAS) and Inductively Couple Plasma (ICP) which are very expensive and require trained personnel. The purpose of this study was to design a simple detector for Cr6+ metal ions as an alternative to conventional methods using AAS and ICP. The detection tool was developed using the android-based digital image method with SLR data analysis techniques obtained from the reaction of the metal ion complex Cr6 + with the compound dhypenilcarbazida (DPC). The parameters of the tool measured were accuracy, selectivity and sensitivity with the reaction of DPC compounds as complexes. The comparison level of this method is the complex method with DPC using a UV-Vis spectrophotometer. This research was carried out in the Chemistry Laboratory FKIP UNIB in November 2018 until March 2019. The implementation of tool parameters was tested to samples of aquatic environments in Bengkulu City, namely PDAM Kota Bengkulu water, tap water of FKIP UNIB Dean, Lake UNIB water, Sungai Hitam water and water Pantai Panjang. The results showed that the detection of Cr6+ metal ions made by the android-based digital image method had better accuracy, selectivity and sensitivity than conventional methods. The DPC is selective for Cr6 + metal ions with a change in color from clear to violet, at a wavelength of 540 nm. Accuracy was checked using a UV-Vis spectrophotometer with 99% accurate results. After that, test sensitivity to Cr6 + metal ions. using the android application method generates a Limit of Detection (LOD) of 6.7 ppb. The detector that was developed was applied to water samples in the city of Bengkulu, it turned out that all of the water samples showed that the levels of Cr6+ metal ions were still below the threshold determined by the Minister of Health Regulation, so it was still safe to use in everyday life.

Abstract

Consuming food containing high level of purin leads to increase uric acid in the blood which, in turn, cause hyperuricemia or high uric acid level. The use of improper allopurinol for hyperuricemia medication that doesn’t follow medical doctor’s prescription can give harmful side effect. It is therefore required to use an alternative safe medication. Uncaria cordata (Lour.) Merr.) is a plant often used for medication of high levels of blood uric acid by communities in Pasar Ngalam village, Bengkulu Province. This research aims to evaluate the potency of extracted compounds from Uncaria cordata (Lour.) Merr.) in reducing uric acid of Mus musculus suffering from hyperuricemia. This research is a laboratory experiment using completed random design. As many as 25 Mus musculus were divided into 5 groups of treatment: PN = normal treatment, PA = treated with allopurinol, P1 – P3 = treated with extracted compound of Uncaria cordata (Lour.) Merr.) with a dose of 5.3 mg/30gBW (P1); 10.6 mg/30gBW (P2); and 21.2 mg/30gBW (P3). In order for Mus musculus to suffer from high uric acid level, potassium oxonate-induced hyperuricemia was injected. The result showed that extracted compounds of Uncaria cordata (Lour.) Merr.) using ethanol exhibited antihyperuricemia proven by decline of uric acid level of Mus musculus from hyperuricemia on each given dose. Decreasing uric acid level for each given dose of ethanol extract (5.3 mg/30gBW, 10.6mg/30gBW and 21.2mg/30gBW) was not significant different (0.541; with a ³0.05).

Abstract

The aim of this study was to determine the anticholesterol activity of the stem extract of Uncaria cordata (Lour.) Merr. on total cholesterol levels in male mice (Mus musculus). Before carrying out the anticholesterol activity test, phytochemical tests were carried out to determine the content of secondary metabolites in Uncaria cordata stems. Furthermore, the anticholesterol activity test was carried out in vivo using male mice, male mice were induced by high-fat diet (PTL) in the form of beef brain, quail egg yolk and propylthiouracil to increase cholesterol levels in mice until they experienced hypercholesterolemia, then mice were induced by plant extracts to see anticholesterol activity. Cholesterol levels were measured using a glucometer (Autocheck). Phytochemical test results showed the content of secondary metabolites in the form of flavonoids, saponins, alkaloids, tannins and terpenoids. The results of the anti-cholesterol activity test showed that there was anti-cholesterol activity as indicated by a decrease in cholesterol levels in mice with hypercholesterolemia. A decrease of 11.45% at a dose of 5.3 mg/30grW; 12.89% at a dose of 10.2mg/30grW and 21.05% at a dose of 21.2mg/30grW. Key words : Anticholesterol, Uncaria cordata (Lour.) Merr., Hypercholesterolemia
