Abstract

Penelitian pengaruh pemberian ekstrak daun honje hutan Etlingera hemisphaerica (Blume) R.M.Sm terhadap gejala Parkinsonisme pada mencit Mus musculusL. (1758) Swiss Webster yang telah disuntik paraquat telah dilakukan pada bulan November sampai Desember 2014 di Kebun Biologi, FKIP Universitas Bengkulu. Perlakuan terdiri 3 kelompok yaitu: perlakuan kontrol (R0), Perlakuan paraquat dengan dosis 10 mg/kg bb secara intraperitoneal (R1) dan perlakuan paraqaut 10 mg/kg bb + ekstrak E. hemisphaerica dengan dosis 0,39 mg/g bb (R2). Hasil penelitian menunjukkan bahwa pemberian paraquat menyebabkan mencit mengalami gejala parkinsonisme dengan memperlihatkan perilaku postural instability dan rigiditas. Secara umum gejala parkinsonisme ditunjukkan pada penurunan kemampuan refleks menghindari jurang, refleks geotaksis negatif, kemampuan berenang. Tikus yang menunjukkan gejala parkinsonisme diberi ekstrak daun honje memulihkan kemampuan penciuman dan kemampuan penglihatan, memulihkan kemampuan pendengaran dan kemampuan mengangkat badan tetapi tidak 100%. Abstract

The research aimed to know the effect of total protein extracted from black sea cucumbers (Holothuria leucospilota) on agglutination of mouse sperms (Mus musculus). The flesh of black sea cucumber  was homogenized in Tris Buffer Saline (TBS) at pH 7.4. The homogenate was added with ammonium sulphate 50%(w/v) for presipitating protein. The presipated protein was purified with 12.000 MWCO dialysis bag to remove ammonium sulphate. The purified protein was quantitated with Biuret Method. The concentration of purified protein was 4,97g/dL. There were five concentration proteins used mouse sperm agglutination test, i.e. 4.97, 3.52, 2.48, 1.17 g/dL and 0% (control). Mouse sperm suspension was prepared by soaking an epypydimal duct in 0,9% (w/v) NaCl solution. The purified protein in varied concentration was then used for agglutination of suspension of mouse sperm. This research used Completely Randomized Design (CRD) with 5 concentrations of protein  and 5 replications. The data of this research were analyzed by using ANOVA. The result showed that total protein extract of black sea cucumberwas significantly different on mouse sperm agglutination. Abstract

Penelitian  ini  bertujuan menggumpalkan sperma mencit dengan ekstrak  total protein dari endosperm biji ketapang. Biji ketapang  mengandung  protein yang berperilaku seperti lektin. Lektin adalah  glikoprotein yang dapat berikatan dengan residu karbohidrat spesifik. Buah ketapang masak dipecahkan, diambil bagian endospermnya dan dicuci bersih dengan akuades. Endosperm biji dihomogenasikan dalam larutan buffer pengekstrak. Homogenat diaduk semalam dengan batang magnet berputar pada suhu 4oC. Selanjutnya homogenat disentrifus 2500 rpm 4oC selama 15 menit. Supernatan ditambahkan ammonium sulfat sampai konsentrasi 50%  (w/v), diaduk selama 3 jam, selanjutnya disentrifus 10.000 rpm at  4oC  selama 15 menit. Protein terpresipitasi dimurnikan dengan kantong dialisis ukuran 12.000 MWCO (molecular weight cut off).  Protein terpresipitasi dilarutkan dalam buffer dan diukur kadar proteinnya dengan metode Biuret.  Mencit dimatikan secara  cervical dislocation, dibedah, dan diambil saluran epididymis. Suspensi sperma dibuat dengan merendam  epididymis dalam larutan NaCl 0,9% (w/v), selanjutnya bagian proksimal epidydimis dipotong lalu ditekan perlahan hingga sekresi cairan epidydimis keluar dan tersuspensi dengan NaCl 0,9%. Uji agglutinasi dilakukan dengan mencampur suspensi 5 ul sperma mencit dengan 5 ul ekstrak potein total fraksi dialyisis dari konsentrasi 4,23;  8,97;  13,2  dan 17,4 µg/dl, fraksi ammonium sulfat, dan fraksi ekstrak kotor. Laju penggumpalan sperma mencit yang paling cepat yaitu fraksi amonium sulfat  yang mengandung  17,4 µg/dl protein.  Endosperm biji ketapang mengandung lektin yang dapat menggumpalkan sperma mencit.

Abstract

Indonesia is one of the world's mega biodiversity needs to be preserved, due to estimated the extinction rate of one species is estimated at 10,000 times per year caused by human activity. Various efforts have been made by various sectors to preserve the environment, including in education through Environmental Education (EE). This research aim to implement Environmental Literacy (EL) domain in Biodiversity learning assisted validated e-booklet (CVR0,67) to enhance student knowledge, cognitive skills (identification of issues, analysis of issues, and action plans), attitude (verbal commitment, environmental sensitivity, and feelings towards the environment), and responsible behavior towards the environment. The EL domains related to real context of biodiversity in Gegas Dam is implemented in order to avoid the availability of disconnecting youth generation with their native biodiversity. Data analysis and identification of diversity- diversity index, homogeneity, dominance and correlation among chemical-physical environmental factors- are presented in an e-booklet to strengthen students' knowledge and cognitive skills, while attitude and responsibility behavior are strengthened by presenting local, national and global environmental issues. Through one group pre-posttest design, 30 high school students in South Sumatra as youth generation were involved in this research. Students’ EL is measured by 66 questions EL domain. The results showed that students' verbal commitment got the highest n-gain score (0.62 medium), while environmental sensitivity was the smallest (0.2 low). These results indicate that verbally students or youth generation already know how to conserve the environment. Cultivation of environmental conservation habits could be continuously improved by realizing their verbal ideas.

Abstract

Untuk membandingkan jenis dan jumlah kejadian cacat bawaan pada bayi lahir, kami telah melakukan dua kali survey rekam medis, dan observasi langsung di RumahSakit M.Yunus dalam jarak lebih dari sepuluh tahun. Pada tahun 2000 kami mengolah data rekam medis periode Agustus 1998 s/d Juli 1999, dan mengamati setiap kelahiran bayi yang lahir di rumah sakit tersebut secara langsung selama bulan Juli 1999. Pada tahun 2013 kami sekali lagi mengolah yang sama pada periode Januari 2010 s/d Juli 2011, dan mengamati setiap kelahiran bayi selama bulan Juli 2011. Dari survey dan pengamatan langsung tersebut kami mencatat jenis-jenis cacat bawaan bayi lahir di Rumah Sakit M.Yunus Bengkulu, dan membandingkan jumlah dan jenisnya. Hasil penelitian menunjukkan ada 6 jenis cacat bawaan bayi lahir yang cenderung meningkat dari tahun ke tahun, ialah Atresia Ani, Hirschprung, Gastrochisis, Spina bifida, Hernia, dan Hydrocephalus. Peningkatan resiko cacat bawaan bayi lahir tersebut diduga berkaitan dengan semakin banyaknya penggunaan bahan kimia beresiko teratogenik, dan mutagenik di dalam asupan bahan makanan danobat-obatan di luar resep dokter. Kata kunci:   neonatus congenital, rekam medis, Atresia Ani, Hirschprung, Gastrochisis, Spina bifida, Hernia, Hydrocephalus, teratogenik

Abstract

We have reported here a specific human short stature population in Kedurang District, South Bengkulu. AH those are men, described as a mild dwarf (130-150 cm), detected after 5 to 10 years old, caused by the failure on lumbar backbone growth and development, and only found among the men. In this investigation, we examine the inheritance pattern of the dwarfism by using pedigree analysis, study the morphological comparison between the disorder and normal person, and finally we ascertain the variation among the disorder. We accomplished a survey on pedigree family to at least 3-4 generations related to the disorder. The data are then tested with Chi-square test, and transformed to tree families and pedigree analysis. To investigate morphological performance and their variation, we collected morphometric measurements to both disorder and normal group of sample; they are the height, the vertebral bones length, head measurement, the leg and arm length (upper and fore side), and their indexes. The data were analyzed descriptively. To find out the variation, the quantitative expression and penetrance, we evaluated the data according to normal distribution with Z test. The result showed all the dwarfisms in Kedurang are men, who inherited his gene from his carrier mother. As a result, the gene has tightly linked to X chromosome and supposedly belongs to Spondylo Epiphypeseal Dysplasia Tarda (SEDT). These were concluded from 32 related family of dwarfism. We found 19 dwarf persons, grouped into young, adult, and older persons. The their height were shorter than normal (136,5 cm), and the length of vertebrae was under normal value (51,5 cm). Their frontal extremity were shorter than the normal one, but they are in proportion with any part comprised them. Their feet were shorter, but the proportion of leg is longer than calf. The rest is as normally. Among the dwarf person, there are variations of the length. The phenotypic expressivity is varied in the vertebrae, leg, and calf; but the rest is relatively invariable. Their penetrance is strong, mostly in the height, length of vertebrae, and legs. Based on morphological data we concluded that the disorder in the Kedurang, South Bengkulu belongs to SEDT (Spondyloepiphyseal dysplasia tardd).

Abstract

Introduction : Jatropha multifida Linne is known as Betadin plant by the Bengkulunese. The stem extract of J. multifida has antibacterial activity. This study examined the potential of stem extract of J. multifida to be developed as an anti-malarial drugs through trials in M. musculus whose erythrocytes infected by Plasmodium berghei.Methods : Phytochemical test of the stem of J. multifida to test the flavonoids, alkaloids, tannins, saponins, terpenoids and steroids. The stem extract of J. multifida obtained by maceration with 96% ethanol. 30 M. musculus divided 6 groups, each consist of 5 animals. P0 is the negative control groups that were not infected by P berghei. P1 is the positive control groups that were only infected by P berghei, P2 is a comparison groups that were infected with P berghei and chloroquine, groups P3, P4, and P5 were infected by P berghei and treated with stem extract of J multifida with consecutive doses 0.028 g / kgbw, 0.056 g / kgbw and 0.084 g / kgbw. After 24 hours, the number of erythrocytes was observed with hemocytometer then was counted the number of uninfected erythrocytes by P berghei using a microscope at 1000x magnification.Results : The stem extract of J. multifida at doses of 0.028 g/kgbw, 0.056 g/kgbw and a dose of 0.084 g/kgbw were able to increase the number of uninfected erythrocytes by P berghei in M. musculus, respectively an average of 9.135million cell eritrosit/mm3, 7.618 millioncell eritrosit/mm3, and 9.856 millioncell eritrosit/mm3. The ability of stem extract of J. multifida in increasing the number of erythrocytes uninfected with P berghei was much higher than the malaria drug chloroquine diposphat. On one way ANOVA analysis of Fcount ( 13,2 ) > Ftable (2.76 ) , with (α = 0.05), there are noticeable differences in the provision of treatment. The increasing number of uninfected erythrocytes by P berghei was due to the content of flavanol glycosides in J multifida stem. Flavanol glycosides was expected to form Flvanolglycosides-heme complex that could inhibit the formation of P berghei parasites HemazoinConclusion : The stem extract of J. multifida could be developed as a potential anti-malarial drugs since it could increase the number of uninfected erythrocytes by P berghei in M. musculus. Abstract

Telah dilakukan penelitian tentang pengaruh kelebihan seng sulfat terhadap perkembangan tulang femur embrio ayam (Gallus gallus) galur Tegel TM 70. ZnSO4 dengan dosis 0,2 mg, O,4 mg dan 0,9 mg per telur, diinjeksikan secara tunggal melalui kantung yolk pada hari ke-2, ke-4 dan ke-6 inkubasi. Keadaan embrio diamati pada hari ke-13 inkubasi. Perlakuan dengan O,2 mg ZnSO4 pada hari ke-2 inkubasi cenderung meningkatkan panjang dan luas tulang femur, namun dosis 0,9 mg nyata menurunkannya (p<O,05 dan p<O,01). ZnSO4 lebih berpengaruh terhadap luas penampang bagian yang menulang daripada terhadap panjang bagian yang menulang. Disimpulkan bahwa seng dalam konsentrasi tertentu dibutuhkan untuk perkembangan optimal tulang femur, sedangkan kelebihan seng lebih berpengaruh dalam menghambat aktivitas osteogenik periosteum daripada aktivitas lempeng epifisis. The effect of excess zinc sulphate (ZnSO4) on the development of the femur in the Tegel TM 70 chick embryo (Gallus gallus) has been investigated by injecting the substance into the yolk sac of 2, 4 and 6 days incubated eggs. Three kinds of a single dose of ZnSO4 0.2 mg, 0.5 mg and 0.9 mg per egg were used. The embryos were examined on day 13 of incubation. Treatment with 0.2 mg ZnSO4 on day 2 incubation inclined to increase the width and length of the femur. Conversely, the dose of 0.9 mg significantly decreased the width and length of the femur (p<0.05 and p<0.01). ZnSO4 exerts its influence on the width of ossified parts of the femur, rather than on the length. It can be concluded that a certain concentration of zinc was necessary for optimal development of the femur, while zinc in excess influenced more the delay of periosteal osteogenic activity than that of epiphyseal plate osteogenic activity.

Abstract

AbstraCT[Adaptation Study of Male and Female Notochelys Platynota in Ex-situ Conservation Area University of Bengkulu]. The purposes of this research were to compare the growth of male N. platynota and female N. platynota, and to know the condition of abiotic factors in ex-situ conservation area at the University of Bengkulu. Nine N. platynotas consisting of four males and five females were kept individually in three cages, each of which consisting of three spaces. Cages were placed at three different spots in the ex-situ conservation area of Bengkulu University. N. platynota was fed with water spinach (Ipomea aquatica) as much as 10% of their body weight, every two days. Data collection was done once a week for five weeks. The data were analyzed to determine: a) weight growth, b) body-thick growth, c) growth of carapace length, d) growth of carapace width, e) growth of plastron length, f) growth of plastron width, and g) environmental factors (air temperature, soil temperature, water pH, soil pH, and humidity. The results showed that: a) N. platynota males experienced a growth of 5.04% higher than the female N. platynota, i.e.only 2.26%, b) the abiotic factor conditions of habitat of N. platynota were the followings: air temperature 30.8ºC, ground temperature 31.8ºC, soil pH 5.1, water pH 7.5 and relative humidity 71.2%. Based on the data of N. platynota growth and abiotic factor condition, it can be concluded that the ex-situ conservation area of Bengkulu University is suitable to be the new habitat for N. platynota. Keywords: Adaptation; ex-situ conservation; growth; notochelys platynota.(Received August 13, 2018; Accepted January 8, 2019; Published February 25, 2019) ABSTRAKPenelitian ini bertujuan untuk: a) membandingkan pertumbuhan Notochelys platynota jantan dan N. platynota betina, (b) mengetahui kondisi faktor abiotik di area konservasi ex-situ Universitas Bengkulu. Sembilan ekor N. platynota yang terdiri dari empat ekor jantan dan lima ekor betina dipelihara secara individu dalam tiga keramba yang terdiri atas tiga plot pada masing-masing keramba.  Keramba di letakkan pada tiga titik berbeda di area konservasi ex-situ Universitas Bengkulu. N. platynota diberi pakan berupa  Kangkung (Ipomea aquatiqa) sebanyak 10% dari berat badan, pemberian pakan dilakukan setiap dua hari sekali. Pengambilan data dilakukan setiap satu kali seminggu selama lima pekan. Data dianalisis untuk mengetahui: (a) pertumbuhan berat badan, (b) pertumbuhan tebal badan, (c) pertumbuhan panjang karapaks, (d) pertumbuhan lebar karapaks, (e) pertumbuhan panjang plastron, (f) pertumbuhan lebar plastron, (g) faktor lingkungan abiotik (suhu air, suhu udara, pH air, pH tanah dan kelembaban udara). Hasil penelitian menunjukkan bahwa: (a) N. platynota jantan mengalami pertumbuhan sebesar 5.04%. lebih tinggi dari pada pertumbuhan N. platynota betina yaitu 2.26% (b) Kondisi faktor abiotik yang menjadi habitat N. platynota antara lain dengan rata-rata: suhu udara 30.8ºC, suhu tanah 31.8ºC, pH tanah 5.1, pH air 7.5 dan kelembaban udara 71.2%. Berdasarkan data hasil pengamatan pertumbuhan N. platynota dan kondisi faktor abiotiknya, dapat di katakan bahwa area konservasi ex-situ Universitas Bengkulu sudah cocok untuk dijadikan habitat baru bagi N. platynota. Kata Kunci: Adaptasi; konservasi ex situ; pertumbuhan; Notochelys platynota.
