Abstract

Doxorubicinisoneofchemotherapeuticagentwidelyusedinbreastcancertreatment,but in high dose doxorubicin gives negative side effect, including vomit, nausea, immune suppression, and cardiac toxicity. This toxicity hopefully could be reduced by combination chemotherapy using natural herbs such as ciplukan herb. This research was conducted to explorecytotoxicactivityofsingleciplukanherbsethanolicextractanditscombinationwith doxorubicinonT47Dbreastcancercells.Cytotoxicactivityofciplukanherbsethanolicextract only and its combination with doxorubicinwere tested on T47D cells using MTT assay toobtainIC50valueandcombinationindex(CI),respectively.Singleextractshowedcytotoxic activityonT47DcellswithIC50valueofwas160*g/ml.Thus,combinationtreatmentfrom ciplukanherbsethanolicextractanddoxorubicinshowedsynergisticeffect(CI<1,0).Thiseffect wasreachedatconcentrationofciplukanherbsethanolicextract-doxorubicin80μg/ml-2nM, 80 μg/ml-4 nM, and 80 μg/ml-8 nM. This research indicated that ciplukan herbs ethanolic extractispotentialtobeappliedasco-chemotherapeuticagentinbreastcancertherapy.

Abstract

Fungal endophytes have been known as sources for bioactives with high chemical structure variability. This study aimed to screen extracts of some endophytic fungi associated with plants from Enggano Island as antioxidant and antibacterial using thin layer chromatography (TLC) bioautography method. Antibacterial ativity was performed against Escherichia coli and Staphylococcus aureus. Seventeen out of 22 extracts showed growth inhibition activity against both bacteria, while 16 extracts were active as DPPH free radical scavenging agents. Further determination on IC50 value of four prominent extracts revealed that fungal endophytes AK3018-1, FC-1, KC-4, and SO-3 have IC50 value of 85, 84, 704, and 347 ?g/ml, respectively in DPPH radical scavenging method. Extracts of fungal endophytes CR-3, CS-2, and SM-2 showed prominent antibacterial activity among other extracts, indicated by wide clear white zone around the spot. Further evaluation on minimum inhibitory concentration (MIC) of those three extracts by microdilution method showed that CR-3, CS-2, and SM-2 have MICs value of 512, >512, and 64µg/ml, respectively against S. aureus.Fungal endophytes AK3018-1, FC-1, KC-4, SO-3, CR-3, CS-2, and SM-2 were respectively isolated from Dioscorea bulbifera tuber, Fibraurea chloroleuca twig, Knema cinerea twig, Smilax odoratissima stem, Cryptocarya sp. twig, Calophyllum soulattri twig, and Smilaxmacrophylla stem.

Abstract

Three species of Smilax spp. (Smilax macrophylla, S. odoratissima and S. zeylanica) collected from Enggano island were evaluated for their potential as an antibacterial and antioxidant. Stems and leaves of three species of Smilax spp. were extracted successively with nhexane,chloroform, ethyl acetate and methanol. Antibacterial activity was evaluated by TLC-bioautography against Eschericha. coli InaCC B5 and Staphylococcus. aureus Ina-CC B4. The antioxidant activity was analyzed by DPPH free radical activity by bioautography method. The value of minimum inhibitory concentration (MIC) and IC50 of active extracts were done by serial dilution in 96- well microplate. The results showed that 20 extracts have antioxidant activity, 13 extracts inhibited the growth of S. aureus Ina-CC B4, and 14 extracts inhibited the growth of E. coli Ina-CC B5. MIC values of active extracts against S. aureus Ina-CC B4 were in the range of 128 - > 512 µg / ml, while the values of MIC against E. coli B5 Ina-CC were > 512 µg / ml. IC50 values of extracts that has antioxidant activity were in the range of 184.11-4549.34 mg/L.

Abstract

Celery (Apium graveolens L.) is commonly used to lower blood pressure, antirheumatic, relaxant, mild diuretic, antiseptic for the urinary tract, antioxidant, and anti-inflammation. According to previous studies, a number of the phytochemicals found in the plant show cytotoxicity toward some types of cancer cells. However, studies on the cytotoxic effects of celery herb ethanolic extract (CEE) on breast cancer cell (T47D), colon cancer cell (WiDr), and cervix cancer cell (HeLa), however, has not been done yet. Our research aims at doing so. Cytotoxicity test was conducted using MTT assay and its absorbance was read using ELISA reader at λ = 595 nm. Results of the assay show that CEE reduces cell viability at concentrations of 100-750 µg/ml on HeLa cells, while reduction of T47D and WiDr cell viability was not achieved until concentrations of 500-750 µg/ml. Based on these results, we conclude that CEE hold many potentials for further developments as preventive and therapeutive agent in cancer treatment.

Abstract

Spirulina is a type of microalgae that contains many useful compounds having antioxidant properties. It has low biological activity and limited protein content when used in dry form. Proteins can be broken down through a hydrolysis reaction to increase their bioactivity, producing smaller peptides and free amino acids. This study aims to evaluate the effects of two cysteine-protease enzymes, bromelain and papain, on the hydrolysis of Spirulina protein. The research examined how these enzymes affect the degree of hydrolysis, protein content, molecular weight, and antioxidant activity of the resulting protein hydrolysate. A non-factorial, completely randomized design was used with three replicates per treatment. The results showed that the type of enzyme used significantly influenced all measured parameters. Bromelain was found to be more effective than papain. Spirulina protein hydrolyzed with bromelain had 32.15 ± 0.74% protein, 48.51 ± 0.94% hydrolysis, a density of 0.786 mg/mL, and 29.64 ± 0.82 ppm antioxidant activity. It also contained 18 types of amino acids, totaling 14.41 g/kg. The most efficient of physical extraction methods—particularly the combination of freeze-thaw and ultrasonication—for obtaining high-yield, high-quality protein from Spirulina. Further purification is needed to obtain the smallest peptide.

Abstract

Endophytic fungi living in medicinal plant tissues are potential bioactive metabolite sources for drug discovery and development. However, the research regarding endophytic fungi associated with Indonesian medicinal plants is still limited. This study aimed to explore the biodiversity of endophytic fungi associated with the medicinal plant Uncaria gambir (Hunter) Roxb. var. udang and their antibacterial activities. Twenty-seven endophytic filamentous fungi were isolated from the surface-sterilized leaves, stems, and roots of Uncaria gambir (Hunter) Roxb. var. udang collected in Jasinga, Bogor, West Java, Indonesia. The observation based on the morphological characters both macroscopically and microscopically, as well as chemical profiling using thin layer chromatography (TLC), revealed that some of the isolated endophytic fungi were identical. Overall, the isolated endophytic fungi consist of six Coelomycetes, three Dematiaceae, four Pestalotiopsis sp., two Penicillium sp., one Aspergillus terreus, and two fungi were unidentified. Antibacterial activities of the ethyl acetate extract from the isolated endophytic fungi associated with Uncaria gambir (Hunter) Roxb. var. udang were observed against E. coli, B. subtilis, S. aureus, and M. luteus. Twelve types of isolated endophytic fungi from Uncaria gambir (Hunter) Roxb. var. udang exhibited antibacterial activities.

Abstract

ABSTRACT. Endophytic fungi are recognized as a valuable source of bioactive compounds with significant therapeutic potential, with species from the genus Colletotrichum being particularly noted for their prolific production of structurally diverse secondary metabolites. This study aimed to evaluate the antiproliferative activity of the endophytic fungus Colletotrichum sp., isolated from the leaves of Myristica fragrans Houtt., commonly known as nutmeg against MCF-7 human breast cancer cells using an integrated in vitro and in silico approach. Bioassay-guided fractionation of the fungal extract followed by LC-HRMS analysis led to the identification of three major compounds: 2-amino-1,3,4-octadecanetriol, L-α-palmitin, and muscone-like compound. The antiproliferative activity of each subfraction was assessed using the MTT assay, while molecular docking studies were conducted to evaluate their binding affinities and interaction profiles with estrogen receptor α (ERα). Among the tested subfractions, F2.3 demonstrated the strongest antiproliferative activity, reducing MCF-7 cell viability to 22.08% at a concentration of 50 µg mL-1. The identified compounds demonstrated notable antiproliferative potential, with L-α-Palmitin showing the moderate binding affinity to ERα as indicated by a binding energy of -6.406 kcal mol-1. Molecular docking analyses revealed key interactions, including hydrogen bonding and hydrophobic contacts, contributing to receptor-ligand stability. These findings highlight Colletotrichum sp. as a promising source of antiproliferative agents, thus warranting further investigation into the isolation, structural characterization, and in vivo validation of its active constituents for potential pharmaceutical development.
