Abstract

Abstract Isolation, identification and testing of antimicrobial activity of secondary metabolites of endophytic fungal culture TCBP4 isolated from bratawali (Tinospora crispa) has been performed. The fungus TCBP4 was cultivated in Potato Dextrose Broth (PDB) for 1 month, media and fungi extracted with ethyl acetate. The extract was separated and purified by several chromatographic techniques, from which 9 fractions were obtained. Fraction 3e was purified again and was obtained 6 fractions (3e1-3e6). Fractions 3e3-3e6 were tested against bacteria isolates Staphylococcus aureus Bacillus subtilis,Eschericia coli and yeast isolate Candida albicans by microdillution method. Antimicrobial activity test result showed that fractions 3e3-3e5 had better antibacterial activity compared to chloramphenicol as commercial antibiotic. It was indicated by MIC value of the fractions was lower (8 ug/ml) compared with the antibiotic chloramphenicol (16 ug/ml). Fraction 3e3 had better antifungal activity compared to commercial antifungal nystatin and cabisidin against C. albicans. GC-MS analysis showed that the chemical constituent of 3e5 fraction was identified as 3-acetyl -2,5,7-trihydroxy-1,4-naphtalenedione . Key words : Tinospora crispa, endophytic fungi, isolation, identification, antimicrobial Abstrak Isolasi, identifikasi dan uji aktivitas antimikrobia terhadap metabolit sekunder dari kultur jamur endofit TCBP4 yang diisolasi dari tumbuhan bratawali (Tinospora crispa). Jamur endofit TCBP4 dikultivasi pada media Potato Dextrose Broth (PDB) selama 1 bulan, selanjutnya media dan jamur diekstrak dengan pelarut etil asetat. Ekstrak dipisahkan dan dimurnikan dengan beberapa teknik kromatografi sehingga diperoleh 9 fraksi. Fraksi 3e dimurnikan kembali dan diperoleh 6 fraksi (3e1-3e6). Fraksi 3e3-3e6 diuji aktivitas antibakterinya terhadap beberapa isolate bakteri Staphylo-coccus aureus Bacillus subtilis, Eschericia coli dan isolate khamir Candida albicans dengan metode mikrodilusi. Hasil uji antimikrobia menunjukkan bahwa fraksi 3e3-3e5 mempunyai aktivitas antibakteri lebih baik bila dibandingkan dengan chloramfenikol, dimana nilai MIC dari fraksi (8 ug/ml) lebih rendah dari antibiotika chloramfenikol (16 ug/ml). Fraksi 3e3 mempunyai aktivitas antijamur lebih baik bila dibandingkan dengan antijamur komersial nistatin dan kabisidin terhadap  C. albicans. Analisis GC-MS menunjukkan bahwa komponen kimia dari fraksi 3e5 diidentifikasi sebagai 3-acetyl -2,5,7-trihydroxy-1,4-naphtalenedione . Abstract

Abstrak Daun cabe jawa (Piper retrofractum Vahl.) telah terbukti berkhasiat sebagai bahan antimikroba, akan tetapi belum ada informasi lebih lanjut tentang mekanisme aksi antibakteri dari minyak atsiri daun cabe jawa. Penelitian ini untuk mengetahui pengaruh minyak atsiri daun cabe jawa terhadap bakteri uji sensitif serta difokuskan untuk melihat kerusakan membran sel bakteri melalui analisis protein, asam nukleat dan ion-ion logam kalsium dan kalium. Distilasi uap minyak atsiri dari daun segar cabe jawa (Piper retrofractum Vahl.) menghasilkan 0.03% cairan kental kekuningan. Analisis dengan GC-MS mengidentifikasi 4 senyawa sebagai komponen utama, yaitu germakren D (24.20%), tetrametilsiklo[5.3.1.0(4.11)]-undek-8-ena (17.73%), ar-turmeron (11.55%) dan benzil benzoat (6.28%). Minyak atsiri cabe jawa terbukti aktif melawan beberapa bakteri patogen antara lain Staphylococcus aureus, Bacillus subtilis and Micrococcus luteus dengan diameter daya hambat berturut-turut 8.0; 9.7; 8.5 mm. Pengamatan lebih lanjut memperlihatkan nilai MIC dari minyak atsiri cabe jawa  terhadap B. subtilis adalah 2% (v/v), dan juga merubah morfologi sel dari bakteri yang diobservasi menggunakan SEM. Kata kunci: Piper retrofractum Vah., cabe jawa, minyak atsiri, aktivitas antibakteri Abstract The leaves of Javaneese pepper (Piper retrofractum Vahl.) has been proven efficacious as antimicrobial agent, however, there was no further information about action mechanism of antibacterial of Javaneese pepper leaves essential oil. This study was to determine the effect of essential oils of javaneese pepper leaves against bacterial sensitive test and is focused on investigating the damage of cell membranes of bacteria through the analysis of proteins, nucleic acids and metal ions, calcium and potassium. Steam distillation of essential oil from fresh leaves of Javaneese pepper (Piper retrofractum Vahl.) gave 0.03 % yellowish viscous liquid. Germacrene D (24.20 %), tetramethylcyclo[5.3.1.0(4.11)]-undec-8-ene (17.73 %), Ar-turmeron (11.55 %) and benzyl benzoate (6.28 %) were identified as major constituents analyzed by GC-MS. Javaneese pepper essential oil was active against pathogenic bacteria Staphylococcus aureus, Bacillus subtilis and Micrococcus luteus with diameter of clear zone of 8.0, 9.7 and 8.5 mm respectively. Further investigation showed MIC value of the oil against B. subtilis was 2 % (v/v), and was also alter the cell morphology of tested bacterium observed by SEM. Abstract

Academic supervision is series activity assist teachers to develop their abilities to organize learning process for reach the goal. Headmaster must arrange academic supervision at school as wellas implementation of Permendiknas No. 13 Tahun 2007. The problem ist facton the field which shows many headmasters feel reluctant to do it. So,it was held research at SMP Negeri 4 Pacitan with the goal to gain illustrations: (1) characteristic of academic supervision preparation at SMPN 4 P Pacitan, (2) Characteristic of academic supervision implementation at SMPN 4, (3) characteristic of the advantageous academic supervision implementationat SMPN 4 Pacitan.The research method was used of is qualitatif research with etnografi study approach by naturalistic paradigm. The data source an resource person were administration document, teacher, and headmaster. The techniques of collecting data were observation, interview, and documentation. The data analysis was descriptife qualitatif with the data validity, Examination used triangulation technique.Based on the research, was gained descriptif results: (1) characteristic of academic supervision preparation, (2) characteristic of academic supervision was done when teacher was doing learning proces in calss, (3) characteristic of advantageousacademic supervision implementation at SMPN 4 between: increasingteacher and headmaster competence, so the would more professional and increasing students achievement.

Abstract

Totally 15 isolates of endophytic fungi were obtained from leaves and young stems of a medicinal plant Albertisia papuana Becc.(Menispermaceae). The antibacterial screening of the ethyl acetate extract derived from the fungi cultures in potato dextrose broth (PDB)were performed on a non eluted Thin Layer Chromatography Bioautography assay (TLC bioautography assay). From the screening test, itwas found that the ethyl acetate extract of the fungus Xylaria sp. DAP-KRI-5 culture showed strong antibacterial activity against Staphylococcus aureus. The isolation of active metabolite based on the bioautography guided assay from ethyl acetate extract of the Xylaria sp. DAP-KRI-5, led us to identify phloroglucinol as a main antibacterial compound. The chemical structure of phloroglucinol was deduced from it spectral data, including UV-Vis, 1H and 13C-NMR, GC-MS and published data elsewhere.

Abstract

Fungal endophytes have been known as sources for bioactives with high chemical structure variability. This study aimed to screen extracts of some endophytic fungi associated with plants from Enggano Island as antioxidant and antibacterial using thin layer chromatography (TLC) bioautography method. Antibacterial ativity was performed against Escherichia coli and Staphylococcus aureus. Seventeen out of 22 extracts showed growth inhibition activity against both bacteria, while 16 extracts were active as DPPH free radical scavenging agents. Further determination on IC50 value of four prominent extracts revealed that fungal endophytes AK3018-1, FC-1, KC-4, and SO-3 have IC50 value of 85, 84, 704, and 347 ?g/ml, respectively in DPPH radical scavenging method. Extracts of fungal endophytes CR-3, CS-2, and SM-2 showed prominent antibacterial activity among other extracts, indicated by wide clear white zone around the spot. Further evaluation on minimum inhibitory concentration (MIC) of those three extracts by microdilution method showed that CR-3, CS-2, and SM-2 have MICs value of 512, >512, and 64µg/ml, respectively against S. aureus.Fungal endophytes AK3018-1, FC-1, KC-4, SO-3, CR-3, CS-2, and SM-2 were respectively isolated from Dioscorea bulbifera tuber, Fibraurea chloroleuca twig, Knema cinerea twig, Smilax odoratissima stem, Cryptocarya sp. twig, Calophyllum soulattri twig, and Smilaxmacrophylla stem.

Abstract

Three species of Smilax spp. (Smilax macrophylla, S. odoratissima and S. zeylanica) collected from Enggano island were evaluated for their potential as an antibacterial and antioxidant. Stems and leaves of three species of Smilax spp. were extracted successively with nhexane,chloroform, ethyl acetate and methanol. Antibacterial activity was evaluated by TLC-bioautography against Eschericha. coli InaCC B5 and Staphylococcus. aureus Ina-CC B4. The antioxidant activity was analyzed by DPPH free radical activity by bioautography method. The value of minimum inhibitory concentration (MIC) and IC50 of active extracts were done by serial dilution in 96- well microplate. The results showed that 20 extracts have antioxidant activity, 13 extracts inhibited the growth of S. aureus Ina-CC B4, and 14 extracts inhibited the growth of E. coli Ina-CC B5. MIC values of active extracts against S. aureus Ina-CC B4 were in the range of 128 - > 512 µg / ml, while the values of MIC against E. coli B5 Ina-CC were > 512 µg / ml. IC50 values of extracts that has antioxidant activity were in the range of 184.11-4549.34 mg/L.

Abstract

Terminalia is the second largest genus of Combretaceae family, consists of 200 species distributed in the tropics and subtropics. Many species of Terminalia are well kown in traditional medicine  in several countries of Africa and Asia. In this study, bark extracts of four species of Terminalia (T. catappa, T. citrina, T. bellirica and T. macadamii) were evaluated for their in-vitro anti-inflammatory and antioxidant activity. Determination of the anti-inflammatory activity was carried out using red blood cell membrane stabilization assay and the antioxidant activity was carried out using DPPH free radical scavenging and ferric reducing power assay. Estimation of total phenolic   content was carried out using Folin-Ciocalteu reagent, while  total flavonoid content using alumminium trichloride reagent. The results showed that extract of T. catappa has the most potent anti-inflammatory activity (IC50= 97,83±0,0100 ?g/mL) and the most powerful  antioxidant activity of DPPH free radical scavenging (IC50= 21,8900 ± 0,0264 ?g/mL) and ferric reducing power (IC50= 87,1533 ± 0,04163 ?g/ mL). The weakest anti-inflammatory activity was T. citrina extract (IC50= 159,35±0,0200 ?g/mL), whereas T. bellirica extract demonstrated the weakest antioxidant activity of  DPPH free radical scavenging (IC50= 44,4867 ± 0,0153 ?g/mL) and ferric reducing  power (IC50= 140,89±0,04 ?g/mL). The extract of T. citrina has the highest content of total phenolic (84,8167 ± 0,53407 mg GAE/g) and flavonoid (23,1200 ± 1,7149 mg RE/g) while the T. macadamii extract has the lowest content of total phenolic (24,3700 ± 0,0173 mg GAE/g) and flavonoid (13,2667 ± 0,3386 mg RE/g).

Abstract

Penelitian bertujuan untuk mendeskripsikan: (1) wawasan kevokasionalan (pemahaman tentang SMK sebagai pendidikan dunia kerja (X1), pemahaman jenis-jenis program keahlian (X2), pemahaman dunia kerja (X3), layanan bimbingan konseling(X6)), lingkungan belajar (X4), Potensi daerah (X5), dan latar belakang sosial ekonomi orang tua (X7) siswa SMP kelas IX terhadap minat melanjutkan di SMK; (2) pengaruh wawasan kevokasionalan siswa SMP kelas IX  (pemahaman tentang SMK sebagai pendidikan dunia kerja (X1), pemahaman jenis-jenis program keahlian (X2), pemahaman duni kerja (X3), layanan bimbingan konseling(X6)), lingkungan belajar (X4), Potensi daerah (X5), dan latar belakang sosial ekonomi orang tua (X7) siswa SMP kelas IX terhadap minat (Y) melanjutkan di SMK. Penelitian merupakan penelitian survei dengan pendekatan kuantitatif berjenis asosiasi. Hasil penelitian menunjukkan bahwa: (1) variabel X­1, X­2, X­3,X5,X­6cenderung mendekati kategori tinggi dengan nilai persentase secara berurutan 75%, 72,23%, 71,72%, 71,89%, 72,46%, dan variabel X4, X7 cenderung mendekati kategori rendah dengan nilai persentase secara berurutan 62,09%, 56,51%; (2) variabel X­1, X­2, X­3,X4, X­5,X­6, ­X7 ada pengaruh terhadap variabel Y dengan nilai koefisien korelasi secara berurutan 0.504, 0,590, 0,837, 0,178, 0,374, 0,706, 0,178 dan secara bersama-sama X1,X2,X3,X4,X5,X6,X7 terhadap Y ada pengaruh dengan koefisien korelasi sebesar 0,767.Kata Kunci: wawasan kevokasionalan, pemahaman, minat.

Abstract

The treatments of textile wastewater by chemical and physical methods have some drawbacks such as economically feasible and high cost, lack of effective color reduction and formation of by-product. Conversely, biological processes have been proposed as a less expensive and less environmentally intrusive alternative. In the present study, decolorization of various dyes was studied using microorganisms isolated from textile/dye wastewater inoculated into culture medium supplemented with 0.01% of anthraquinone and azo dyes (Disperse Red 73, Lanasol Blue 3G Disperse Orange 30, Kemachrome Black T, and Mixture of dyes). The effect of medium composition was investigated by applying four different media (BSM, ME, Basal, and GYP). From 42 isolates, 4 isolates showed the potential for decolorizing structurally different anthraquinone and azo dyes employed in the industry. Based on the microscopic observation, the 3 isolates designated as OW1, RW8, ATSN-3 were identified as bacteria and 1 isolate designated as BSS was observed as fungus. BSS fungus isolate showed the highest ability to decolorize various reactive dyes including anthraquinone and azo dyes. BSS grew well in the ME medium, resulting in approximately 96.4% decolorization efficiency after 72 hours. The result indicates the potential for this isolate to be used in the biological treatment of textile industry.

Abstract

This study aims to identify the chemical compounds of ethyl acetate extract of the bark of bayur (Pterospermum javanicum) and to evaluate its antibacterial activity and potential antioxidant. The estimation of total flavonoid was done by spectrophotometry method. The chemical compounds were analyzed by gas chromatography-mass spectrometry (GC-MS). The antibacterial assay against Staphylococcus aureus Ina-CC B5 and Escherichia coli Ina-CC B4 was done by TLC-bioautographic method, whereas the potential antioxidant was determined by DPPH radical scavenging assay. Meanwhile, its minimum inhibitory concentration (MIC) value and IC50 of the extract were done on microplate 96 well by serial microdilution method. GC/MS analysis revealed the presence of 31 identified chemical compounds in the extract with total flavonoid was 625 mg CE/gram extract. The MIC values against E.coli Ina-CC B5 and S. aureus Ina-CC B4 were >256 ug/ml. The IC50 from DPPH radical scavenging assay was 48.92 ppm indicating that ethyl acetate extract of the bark of bayur is potent antioxidant.
