Abstract

Totally 15 isolates of endophytic fungi were obtained from leaves and young stems of a medicinal plant Albertisia papuana Becc.(Menispermaceae). The antibacterial screening of the ethyl acetate extract derived from the fungi cultures in potato dextrose broth (PDB)were performed on a non eluted Thin Layer Chromatography Bioautography assay (TLC bioautography assay). From the screening test, itwas found that the ethyl acetate extract of the fungus Xylaria sp. DAP-KRI-5 culture showed strong antibacterial activity against Staphylococcus aureus. The isolation of active metabolite based on the bioautography guided assay from ethyl acetate extract of the Xylaria sp. DAP-KRI-5, led us to identify phloroglucinol as a main antibacterial compound. The chemical structure of phloroglucinol was deduced from it spectral data, including UV-Vis, 1H and 13C-NMR, GC-MS and published data elsewhere.

Abstract

ABSTRACTCitric acid demand increases steadily, and there is a need to increase productivity through selection of suitable carbon sources, and addition of substances that increase citric acids production rate. Methanol has been suggested to increase citric acid fermentation on high carbohydrate containing substances. The objective of the study was to evaluate the suitability of sweet sorghum juice for citric acids production and to verify the effect of methanol on citric acids production using Aspergillus niger InaCC F539  as inoculant. Sweet sorghum juice with the total initial reducing sugar of 11.5 % (w/v) was used as the sole carbon sources. To study the effect of total initial reducing sugar on citric acid production the initial reducing sugar was adjusted to the concentration of  30 to 75 g/L. Preliminary experiment was conducted to get the optimum methanol concentration that stimulate citric acid production. The optimum methanol concentration that stimulate citric acid production was 4% (v/v). Submerged fermentation was conducted as shake culture (125 rpm at 28 °C).  Citric acids production was affected by total initial reducing sugar. Higher total initial reducing sugar produced higher citric acids. Maximum citric acid production was 18.96g/L on sweet sorghum juice with 75 g/L total initial reducing sugar. Methanol 4 % (v/v) increase citric acid production by 41.35 to 65.89 %. Juice of sweet sorghum was a good medium for citric acids production, and methanol stimulate and increase citric acid production. It is a good basis for exploring efficient and cost effective industrial scale citric acid production. Keywords: Citric acid, Methanol, Sweet sorghum, Aspergillus niger

Abstract

Pathogenic bacterial resistance to antibacterial agents used in the treatment of recent formal systems have undergone significant increase. It is necessary to get a source of new antibacterial agents to overcome resistance. Endophytic fungi have been known as one of the organisms that have the ability to produce a variety of bioactive metabolites. In this study, 26 endophytic fungi have been isolated from various parts of Cinnamomum burmannii plant. The results of antibacterial activity screening of endophytic fungal extracts byTLC bioautography method without elution showed that as many as 18 extracts inhibited the growth of Staphylococcus aureus InaCC-B4 with the diameter of inhibition zone (DIZ) were in the range of 5.00 to 16.00 mm. On the other hand, the number of extracts that could inhibit the growth of Escherichia coli InaCC-B5 were 24 extracts with the DIZ were in the range of 4.00 to 28.00 mm. A detailed analysis of eluted TLC-bioautography method showed the chemical compounds of extracts that were responsible for antibacterial activity.

Abstract

Antifungal Activity of Tegeran Wood (Cudrania javanensis Trécul) Extracts against Fungus Peniophora sp.  ABSTRACTThe fungus Peniophora sp. can degrade natural dyes, causing discoloration on batik cloth. This study was aimed to evaluate the antifungal activity of tegeran wood (Cudrania javanensis) extracts against Peniophora sp. isolated from the aqueous extract of mahogany (Swietenia mahagoni) bark. Aqueous and methanol extraction procedures gave tegeran wood extracts whose concentrations were then varied into 100, 250 and 500 ppm. Phytochemical analysis of the extracts were carried out, and the results showed that the average values of the total polysaccharides and terpenoids of both aqueous and methanol extracts did not differ significantly (p<0.05), whereas those of polyphenols did (p<0.05). The total polyphenols of the methanol extract (484.723 mg GAE/g) was higher than that of the aqueous extract (389.903 mg GAE/g). Results showed that the methanol extract of tegeran wood showed higher antifungal activity against Peniophora sp. than the aqueous extract, and was also higher than ketoconazole control (100 ppm). The minimum inhibitory concentration of the methanol extract against Peniophora sp. growth was of 500 ppm with the antifungal activity value (AFA) of 76.30%.Keywords: antifungal activity, Cudrania javanensis, Peniophora sp., Swietenia mahagoni, wood extract ABSTRAKJamur Peniophora sp. dapat mendegradasi zat warna alam sehingga warna yang dihasilkan pada kain batik lebih pudar.
