Abstract

Background: Tooth extraction is an action taken to prevent or treat the onset of a disease. However, there are still many complications after tooth extraction, such as a wound healing process that runs slowly and even causes excessive bleeding which can cause death. Gallus domesticus egg-white gel is a medicament that has growth factors that can help the wound healing process run smoothly. Purpose: To determine the number of inflammatory cells involved in the wound healing process after tooth extraction by the application Gallus domesticus egg-white gel. Methods: This research is an experimental laboratory in vivo involving 32 male Sprague dawley rats divided into 4 groups: treatment group (egg-white gel), positive control group (iodine glycerin), negative control group (gel base and saline). Each group will have the left mandibular central incisor extracted under ketamine. All samples were sacrificed on day 3 and 7 for histopathological examination. Results: The Kruskal-Wallis test showed a significance value of 0.00 (p <0.05) meaning that there was a significant difference in the number of inflammatory cells on the 3rd and 7th day. The significance value between treatment groups was 0.320 (p>0.05), which means there was no significant difference between groups. Conclusion: There was a decrease in the number of inflammatory cells in the wound healing process after tooth extraction by application of Gallus domesticus egg-white gel histologically to iodine glycerin on the 3rd and 7th day, but statistically there was no significant difference (p>0.05).

Abstract

Background: Silk from Bombyx mori, a species of silkworm, contains fibroin, which has good biocompatibility and is potentially suitable for medical applications, especially in the treatment of tissue damage. Purpose: This study evaluated the potential interaction of B. mori fibroin peptides with bone morphogenetic protein 2 (BMP2) and transforming growth factor beta 1 (TGFB1), which are protein markers for dentine reparative activity. Methods: The research was carried out in silico. The three-dimensional structure of the proteins was obtained from the Research Collaboratory for Structural Bioinformatics Protein Data Bank (RCSB PDB), and the antimicrobial potential of fibroin was evaluated using Antimicrobial Peptide Scanner v.1, the Collection of Anti-Microbial Peptides (CAMPR3), PeptideRanker, ToxinPred, AlgPred, and AllergenFP. Molecular modeling and analysis were performed with trRosetta, PrankWeb, the HDOCK server, and Discovery Studio. Results: The light chain 1 peptide (LC1), light chain 2 peptide (LC2), heavy chain 2 (HC2), and heavy chain 7 (HC7) showed high binding affinity to BMP2, while LC2, HC1, HC3, and HC6 showed high binding affinity to TGFB1 compared to silicic acid as a standard anti-inflammatory drug. Conclusion: These seven peptides can potentially interact with BMP2 or TGFB1 and might have anti-inflammatory capability.

Abstract

Background: The trend of studies on dental medicaments is increasing rapidly. Antibacterial or anti-inflammatory activity is most frequently studied. Ovalbumin is one of the proteins whose benefits have been studied, but these benefits are still limited because of ovalbumin's potential for proliferative bioactivity. Purpose: The aim of this study is to examine ovalbumin's potential as a woundhealing medicament through molecular docking analysis on a protein related to the extracellular signal-regulated kinases/mitogenactivated protein kinase (ERK/MAPK) signaling pathway. Methods: Ovalbumin was hydrolyzed through BIOPEP-UWM (The BIOPEPUWMâ„¢ database of bioactive peptides). Protein target and interaction were predicted using Similarity Ensemble Approach target prediction webserver, SuperPred webserver, STRING webserver, and Cytoscape version 3.9.1. Selected fragments were docked using Autodock Vina in PyRx 0.8 with Tukey's multiple comparison test and Biovia Discovery Studio version 19.1.0.18287 for visualization. Results: This study found that ovalbumin has the potential to positively regulate cell proliferation, angiogenesis, and fibroblast growth factor production. Six of the 131 fragments of ovalbumin could interact with 73 proteins, and the 20 proteins with the highest probability and score of betweenness centrality showed potential for bioactivity. Five fragments and povidone-iodine interacted inside the Adenosine triphosphate (ATP) phosphorylation site of ERK2, whereas fragment 1 (F1) and glycerin interacted outside the site. F1 could decrease the binding energy required for adenosine 5"²-[,-methylene]triphosphate or an ATP-analogue chemical compound to interact with ERK2 compared to the control, with a score that was not significant. Conclusion: Ovalbumin has the potential to induce cell proliferation by affecting ERK2-ligand interactions.

Abstract

Background: Dental composite composed of natural filler is a trend nowadays. Silk fiber, Bombyx mori L., is one of the natural fibers that have good microhardness (66 VHN) and has a potency to become a renewable natural filler. This research aimed to determine microhardness differences of silk fiber composite B. mori L., and nanofiller composite resin as a gold standard.Method: A quasi-experimental laboratory using composite resin with silk fiber B. mori L., as a natural filler; nanofiller composite Z350 XT flowable (3M ESPE, USA) as a positive control and no-filler composite as a negative control. Four cylindrical specimens per group were made (d=3mm, h=6mm). Microhardness tests were done with Vickers hardness tester (Shimadzu, Japan). Indentation of 100 grams for 20 seconds.Result: There were significant differences in the natural filler, nanofiller group (positive) control and no-filler composite (negative control). Based on the Kruskal Wallis test result, there was a significant difference in microhardness (p=0.024). The highest microhardness was nanofiller (115,2 VHN), silk fiber filler (109,4 VHN), and no-filler composite (32,2 VHN).Conclusion: Silk fiber Bombyx mori L., has shown the good potential of being a renewable natural filler (109,4 VHN) however lower than nanofiller composites.

Abstract

Background: The human body can self-repair. However, if the damage occurs excessively or reaches a critical defect, it requires material substitution to restore its shape and function. Tissue engineering is developed based on the principle of reconstructing damaged tissues due to critical defects to restore, maintain, and repair damaged tissues or organs by applying three main components: scaffolds, signaling molecules, and cells. This study aims to investigate the ability of synthetic coral scaffold incorporated with Platelet Rich Plasma in bone remodeling after tooth extraction. Method: This study was conducted as in vivo experimental laboratory study with a post-test control group design. The 48 male Rattus norvegicus rats were divided into 4 treatment groups. Firstly, the rats’ teeth were extracted, then, on the tooth extraction area, it was treated with povidone-iodine (control group), curaspon, scaffold only, and scaffold-incorporated Platelet Rich Plasma. Result: The results of the Kruskall-Wallis test showed no significant difference (p > 0.05) between the four treatment groups. articleConclusion: The PRP-incorporated synthetic coral scaffold had the highest score of collagen formation density and significantly accelerated collagen formation in the bone remodeling process compared to other treatment materials.
