Abstract

The study was carried out to isolate and detect the resistance of E. coli bacteria from four livestock in Surabaya, Indonesia. This research used 40 samples of chicken thigh meat. The first step is isolated Escherichia coli bacteria using BGBB media (Brillilliant Green Bile Broth), and then used EMBA (Eosin Methylen Blue Agar) media for confirmatory tests, IMViC tests (Indole, Methyl Red, Voges Praskauer, and Citrat) to identify the enterobacteriaceae bacteria, and performed the gram staining test. Determined the Escherichia coli sensitivity to antibiotic by using disk diffusion method. Resistance test has resulted for ampicillin against E.coli is 100% resistance in all farms. In this research 13 of 40 samples from chicken meat had positive E.coli. The result from electrophoresis product of PCR said that resistance Escherichia coli had a specific band. Total 13 samples of positive Escherichia coli resistance ampicillin has specific DNA band primary shv with 768 bp in length.

Amount of samples in this study were 40 samples of broiler meat from 10 markets, samples were taken purposively and each sample used 25 grams from Musculus pectoralis of broiler meat. Samples were examined using pour plate method of total plate count. The result showed that 80% (32 of 40 samples) were exceeded the TPC's limit. The highest value of TPC result was 3,7 í— 107 CFU/g from "H” market.

Abstract

Gastrointestinal helminth infection is the most common and the easiest infection to infect wild cats or pet cats but is often ignored by the cat owners because sometimes there are no symptoms or a lack of attention from the cat owner to their pet cat. The aim of this study was to estimate the prevalence and risk factors of gastrointestinal helminth infection in pet cats in Wonogiri District, Central Java. A total of 100 cat samples were evaluated using a questionnaire, then fecal microscopic examination using simple native method, sedimentation, and floatation methods. Data obtained from this study were presented descriptively and analyzed using the Chi-Square test. The result showed that 37 out of 100 fecal samples were found positive infected with gastrointestinal helminth including Toxocara cati and Dipylidium caninum or the prevalence of gastrointestinal helminth infection can be calculated at 37%. The result of the analysis of significant risk factors were breed, number of pet cat owners, grooming, deworming, and vaccination.
