Abstract

       Ant nest plant (Myrmecodia tuberosa Jack.) is a medicinal plant which belongs to Rubiaceae family. Due to its potency as anticancer, ant nest plant tends to exploit. Direct exploitation of ant nest plant from nature might cause decreasing of its nature population. Therefore, the culture of ant nest plant should be done by using tissue culture. The purpose of this research was to determine the optimum effects of sucrose concentration in solid and liquid medium to increase wet weight callus of ant nest plant by using in vitro technique. A completely randomized design (CRD) with four different sucrose concentration, is 30g/L, 60g/L, 90g/L dan 120g/L were used in solid and liquid medium. The results showed that callus treated any sucrose concentrations had crumb textured form and green to yellowish green color. The highest wet weight callus were found on solid (2.12 g) and liquid (2.03 g) medium which were treated with 30 g/L of sucrose.       Key words: Myrmecodia tuberose Jack., Sucrose, solid and liquid medium, callus weight

Abstract

Propolis is one of natural products produced honey bees and has frequently used as a drug or supplements, antiinflammation, therapy disease, accelerated healing cuts and others. In addition, propolis has many benefits one having the nature as an antibacterial. This study aims to establish the nature of bees antibacterial propolis Trigona incisa against bacteria Klebsiella pneumonia and Staphylococcus aureus. The ektraksi done in a maceration and the antibacterial with the diffusion discs kirby-bauer. The antibacterial activity it uses random design complete (RAL) consisting of 12 treatment and test 3 for every type of bacteria. Variation concentration the treatment is DMSO (control negative), 10%, 20%, 30%, 40%, 50%, 60%, 70%, 80%, 90%, 100% and kloramfenikol (control positive). Antibacterial test extract propolis bees Trigona incisa against bacteria Klebsiella pneumonia and Staphylococcus aureus, showed potential as antibacterial. The best to extract propolis construction beehive Trigona incisa in pursuing the growth of bacteria Klebsiella pneumonia is 70 % with broad zone obstruent formed the 11,18 mm (category strong) while bacteria Staphylococcus aureus does not hinder (weak) category. On wrapping bee products Trigona incisa, a weakling now under inhibit bacterial growth Klebsiella pneumonia and Staphylococcus aureus.

Abstract

Aglaonema “Lipstik” merupakan Aglaonema hibrida yang memiliki corak warna merah atau merah muda pada tepi daun sehingga diberi nama lipstik. Keunikan dan variasi warna daunnya, Aglaonema “Lipstik” dimanfaatkan sebagai penghias ruangan seperti ruang tamu, kantor dan hotel. Selain karena indah, tanaman ini mampu menyerap polusi udara di dalam ruangan. Sehingga banyak pecinta tanaman hias membudidayakannya. Dalam prosesnya, diperlukan perawatan agar memperoleh daun yang indah dengan menggunakan pupuk. Eceng gondong (Eichornia crassieps) dapat diolah menjadi pupuk organik cair (POC) karena memiliki kandungan unsur organik yang dapat digunakan untuk pertumbuhan tanaman. Tujuan utama penelitian ini adalah untuk mengetahui respon pertumbuhan Aglaonema “Lipstik” yang diberi pupuk organik cair eceng gondok. Metode penelitian menggunakan rancangan acak lengkap yang terdiri dari empat konsentrasi perlakuan POC (0 ppm, 1,5 ppm, 3 ppm dan 4,5 ppm) dalam enam kali pengulangan. Parameter pengamatan meliputi pertambahan jumlah daun, pertambahan tinggi tanaman, luas daun dan warna daun. Hasil penelitian menunjukkan bahwa POC eceng gondok tidak berpengaruh terhadap beberapa parameter yaitu pertambahan tinggi tanaman, jumlah daun, luas daun dan warna hijau daun. Sedangkan pada parameter penambahan kecerahan warna merah daun berpengaruh signifikan pada konsentrasi 4,5 ppm sebesar 3,5%.

Abstract

Studi pengaruh Rasio Pupuk Organik Cair Limbah Ikan Nila (Oreochromis niloticus) dengan Pupuk Inorganik Komersial terhadap Pertumbuhan Sawi Pakcoy (Brassica rapa L.) secara Hidroponik Rakit Apung dilaksanakan pada bulan September 2018 sampai Januari 2019. Penelitian ini bertujuan untuk mengetahui pengaruh rasio pemberian pupuk organik cair limbah ikan nila terhadap pertumbuhan tanaman sawi pakcoy secara hidroponik rakit apung, untuk mengetahui konsentrasi pupuk organik cair limbah ikan nila terbaik untuk pertumbuhan tanaman sawi pakcoy yang optimal pada. Penelitian ini menggunakan rancangan acak kelompok (RAK) yang terdiri dari 5 perlakuan (100% AB mix, 25% POC dan 75% AB mix, 50% POC dan 50% AB mix, 75% POC dan 25% AB mix, 100% POC) dengan 4 ulangan. Pengamatan dilakukan selama 4 minggu. Parameter lingkungan yang diamati adalah suhu lingkungan (rumah hidroponik), suhu larutan, pH larutan dan TDS (Total Dissolved Solid). Parameter pertumbuhan yang diamati yaitu, tinggi tanaman, jumlah daun, berat segar, berat kering, luas daun, kandungan klorofil, laju pertumbuhan relatif tanaman, dan volume akar. Hasil penelitian ini menunjukkan perlakuan dengan menggunakan 100% AB mix dan perlakuan 25% POC dan 75% AB mix merupakan perlakuan terbaik pada parameter tinggi tanaman, jumlah daun, luas daun, berat segar, berat kering, volume akar, laju pertumbuhan relatif, dan kandungan klorofil. Pada penelitian ini dapat disimpulkan bahwa substitusi lebih dari 25% POC belum mampu memberikan pertumbuhan yang optimum seperti pupuk AB mix.

Abstract

Anthill plant (Myrmecodia tuberosa Jack.) or sarang semut is a native plant epiphytes from Indonesia. Anthill plants used as a medicinal because contains compounds active such as flavonoids, tannins and polyphenols that are beneficial to the human body. The aims of study to determine the influence of the media type of planting and the best of fertilization on the growth of anthill plants. This study consisted of two treatment are the first treatment plant seed germination anthill on different media of treatments that husk fuel, cocopeat, fern roots and moss. The second treatment is growth plant of anthill are using completely randomized design (CRD) with two factors different. The first factor is the growing media with 4 levels of fuel treatments, husk fuel, cocopeat, fern roots and moss. The second factor is a fertilizer with a second level of treatment that is with and without fertilizer (Gandasil D 2 g/L). Influence significantly of treatment we used Duncan test (DMRT) level of 95%. The results of study in the first treatment, anthill plant can germinate 100% in all treatment media. In the second treatment, the use of growing media and fertilizer significantly influenced the increase of plant height, leaf number and leaf area. The average of plant height in moss media with the addition of fertilizer of 35.75 mm, the highest number of leaves from all treatment media with the addition of fertilizer of 2,00 strands, the largest leaf area from cocopeat media with the addition of fertilizer of 77,02 mm2, and the increase of tubers diameter found in cocopeat media with the addition of fertilizer of 4.13 mm.

Abstract

This study aim was to determine the influence of types and different concentration of cytokinin on the callus formation. The different sources of explant (cotyledons, stems, tubers, roots) from sarangsemut plants was cultured in Murashige-Skoog medium, supplemented with various levels of 2.4 Dichlorophenoxy Acetic Acid (2.4 D)viz0.5; 1; 1.5; 2 mg/l and combined with 2mg/l kinetin were performed to investigate callus formation. The results showed that the best callus formation was found on medium-supplemented with 2 mg/l 2,4-D and 2 mg /1 Kinetin on four sources of explant used. In addition, phenolic, alkaloids, flavonoids, tannins and steroids were also present in the callus.

Abstract

Sebagai salah satu faktor penting dalam pendidikan, guru harus meningkatkan pengetahuan, baik teoritis maupun keterampilan, seperti praktik laboratorium, terutama untuk guru biologi. Berdasarkan hal ini, tujuan program pengabdian masyarakat adalah untuk meningkatkan keterampilan teoritis dan praktis guru biologi SMA. Program pengabdian masyarakat ini diikuti oleh 30 guru dari berbagai SMA di Samarinda. Teori tentang ekstraksi, penyaringan, dan teknik uji antimikrobia dari sumber daya alam dilakukan di ruang kelas, sedangkan penggunaan rotary evaporator, freeze drying, skrining uji fitokimia, dan uji antimikrobia dilakukan di laboratorium yang dibantu oleh teknisi laboratorium. Setelah menerima program pengabdian masyarakat ini, nilai rata-rata kepuasan yang diberikan oleh para guru melalui kuisioner yaitu: (skala 5) sebanyak 49%, baik (skala 4) sebanyak 47%, cukup (nilai skala 3) ) sebesar 5% dan lebih kecil (skala nilai 2) sebesar 1%. Kesimpulan, kegiatan pengabdian ini berjalan dengan baik dan mempunyai manfaat baik bagi guru biologi SMA di Samarinda.

Abstract

This feeding experiment was performed to determine the effects of Myrmecodia pendens bulb extract (MBE) supplementation in fish feed on the growth, survival, and hemato-biochemical profile of Clarias gariepinus. A group of fish was fed with 0.25; 0.50; 1.0; 2.0% MBE and compared to control group (without MBE) for 75 days observation. At the end of feeding trial, growth parameters, hematological profile such as red blood cells (RBC), white blood cells (WBC), Hemoglobin (Hb), Hematocrit (Htc), differential leukocyte, blood plasma biochemistry (glucose, total albumin, cholesterol, and triglyceride), the hepatosomatic (HSI) and intestinal somatic index (ISI) were measured. Survival of all fish was also counted every two weeks. Supplementation MBE above 0.25% resulted significantly higher final biomass weight (FBW), body weight gain (BWG), daily weight gain (DWG), and average weekly gain (AWG). Meanwhile, fish group fed dietary MBE above 1.0% had significantly higher specific growth rate (SGR) (3.32±0.15) than other groups. Fish fed 1.0% of MBE also showed better value of feed conversion ratio (FCR) (1.13±0.03), Hb, and HSI compared to others group. Survival, neutrophil, monocyte, and ISI of all groups was not affected by any concentration of MBE supplementation. Dietary MBE above 0.5% enhanced RBC, WBC, Hematocrit, platelet (PLT), lymphocyte, blood plasma biochemistry such as glucose, total albumin, and triglyceride. Cholesterol of fish fed MBE in the diet showed incrementally enhanced. Present finding suggested that 1.0% MBE in the diet of Clarias gariepinus is recommended to enhance growth, survival, and blood profiles

Abstract

Background: Aktivitas riset dan praktikum di bidang fisiologi hewan membutuhkan beberapa peralatan yang menunjang kelancaran dan keberhasilan kegiatan tersebut. Pengetahuan tentang alat-alat terkait bidang fisiologi hewan, termasuk teknik menggunakan mutlak diketahui dan dipahami. Tujuan pengabdian kepada masyarakat dalam bentuk workshop daring ini adalah untuk meningkatkan pengetahuan dan wawasan guru sains SMP/SMA setingkat dan muridnya dalam mengetahui dan memahami peralatan-peralatan yang terkait bidang fisiologi hewan baik untuk keperluan penelitian atau praktikum. Sasaran kegiatan ini adalah guru sains dan siswa dari beberapa daerah di Indonesia. Metode: Kegiatan diadakan melalui media zoom meeting, video demo penggunaan alat dan diskusi interaktif yang diikuti oleh 60 peserta, terdiri dari guru dan murid. Materi yang disampaikan adalah alat-alat penunjang penelitian dan praktikum bidang fisiologi, teknik penggunaan, serta video demo live serta diskusi interaktif. Hasil: Untuk mengukur keberhasilan program pengabdian ini, peserta workshop daring melakukan pre-test dan post-test. Hasil menunjukkan rata-rata 52,91 (pre-test) dan 72,86 (post-test). Terdapat peningkatan pengetahuan dan wawasan peserta workshop daring peralatan fisiologi hewan sebesar 19,95 poin. Kesimpulan: Kegiatan pengabdian ini berkontribusi meningkatkan pengetahuan dan wawasan peserta workshop daring tentang peralatan fisiologi hewan untuk menunjang penelitian dan praktikum.

Abstract

Bangle (Zingiber montanum) merupakan family zingiberaceae yang dimanfaatkan sebagai obat tradisional untuk mengobati beberapa penyakit. Tanaman bangle yang dikembangkan sebagai obat tradisonal tidak lepas dari kandungan senyawa aktif didalamnya. Kandungan senyawa aktif tanaman bangle memiliki efek aktivitas antioksidan dan antibakteri. Tujuan penelitian ini yaitu untuk mengetahui senyawa fitokimia, aktivitas antioksidan dan konsentrasi efektif ekstrak akar segar bangle (Z. montanum) dalam menghambat pertumbuhan bakteri Escerichia coli dan Streptococcus sobrinus. Penelitian ini menggunakan metode kolorimetri, DPPH dan kertas cakram (tes Kirby & Baurer). Sampel yang digunakan ekstrak etanol akar segar bangle dengan konsentrasi ekstrak 25%, 50%, 75%, 100%. Kontrol positif yaitu kloromfenicol dan kontrol negatif yaitu etanol 10%. Hasil penelitian menunjukkan bahwa ekstrak akar segar mengandung senyawa fitokimia berupa alkaloid, fenolik, flavonoid, saponin dan triterpenoid serta nilai TPC, TFC, dan antioksidan adalah 49.01 mg GAE/g, 394.07 mg CE/g dan 0.993 µg/mL. Aktivitas antibakteri memiliki daya hambat pada bakteri Escherichia coli dan Streptococcus sobrinus dengan konsentrasi 100% secara berturut-turut adalah 16.18 mm dan 9.86 mm.   Kata Kunci: akar segar bangle, aktivitas antioksidan, antibakteri, uji fitokimia ABSTRACT Bangle (Zingiber montanum) is the Zingiberaceae family that is used as a traditional medicine to treat some diseases. The Bangle plant developed as a traditional medicine cannot be separated from the active compounds contained in it. The active compound content of the bangle plant has the effect of antioxidant and antibacterial activity. The purpose of this study was to determine the phytochemical compounds, antioxidant activity and effective concentration of bangle fresh root extract (Z. montanum) in inhibiting the growth of Escerichia coli and Streptococcus sobrinus bacteria. This research used colorimetric method, DPPH and disc paper (Kirby & Baurer test). The sample used ethanol extract of bangle fresh root with extract concentrations of 25%, 50%, 75%, 100%. The positive control was chloramphenicol and the negative control was 10% ethanol. The results showed that the fresh root extract contained phytochemical compounds in the form of alkaloids, phenolics, flavonoids, saponins and triterpenoids and the TPC, TFC, and antioxidant values were 49.01 mg GAE/gr, 394.07 mg CE/gr, and 0.993 g/mL. Antibacterial activity had inhibitory activity in Escherichia coli and Streptococcus sobrinus bacteria with a concentration of 100%, respectively 16.18 mm and 9.86 mm. Keywords: bangle fresh root, antioxidant activity, antibacterial, phytochemical screening
