Abstract

The aim of this research was to formulate of mucoadhesive ketoprofen granules using chitosan polymer. Ketoprofen granules were prepared by wet granulation method with several concentrations of chitosan polymer 0%, 10%, 20%, and 30% (w/v). Granules were evaluated physically and chemically. In vitro dissolution test was carried out for 6 hours using a basket method in buffer phosphate medium pH 7.4. General evaluation of granules showed the yield was relatively good. In vitro mucoadhesive test exhibited that mucoadhesive ketoprofen granules using chitosan polymer could provide mucoadhesive properties to stomach and intestinal of rabbits. The statistics tests for dissolution efficiency with one ways ANOVA (SPSS 17) showed a significant different among F0, F1, F2 and F3 (P0.05). All the formulas followed Langenbucher kinetic release model.

Abstract

Ketoprofen yang beredar di Indonesia sebagian besar dalam bentuk sediaan tablet salut enterik, tetapi metoda analisis untuk penetapan kadar dan uji disolusinya belum tersedia dalam farmakope.  Tujuan dari penelitian ini adalah  mengembangkan metode kromatografi cair kinerja tinggi (KCKT) dan spektrofotometri ultraviolet visibel (UV) untuk  melakukan analisis tablet salut enterik ketoprofen. Penetapan kadar  dan keseragaman kandungan ketoprofen tablet salut enterik ditentukan secara KCKT isokratik fase terbalik yang telah divalidasi menggunakan kolom reverse phase (RP-18 ) (250 x 4,6 mm) diameter partikel 5 µm, fase gerak metanol-dapar fosfat 13 mM pH 6,5 perbandingan 60:40 v/v, laju alir 1,0 mL/menit dan detektor UV 258 nm. Spesifisitas, linieritas, akurasi, dan presisi  memenuhi persyaratan International Conference on Harmonization (ICH). Metode KCKT memberikan lineritas yang sangat baik (r > 0,999) pada rentang konsentrasi 15 – 35 µg/mL, presisi dinyatakan dalam persen deviasi standar relative (% RSD <0,87) dan perolehan kembali yang baik (R> 99,97%). Metode KCKT lebih sensitif dibandingkan metode spektrofotometri UV, dengan nilai LOD masing-masing adalah 0,18 dan 0,67 µg/mL serta LOQ 1,20 dan 2,49 µg/mL.  Hasil validasi dan uji penetapan kadar ketoprofen pada tablet salut enterik dengan metode KCKT tidak berbeda nyata dibandingkan dengan metode spektrofotometri UV (P>0,05)

Abstract

Paracetamol was an active ingredient, which is poorly soluble in water. Absorption of poorly soluble or slightly soluble in water is influenced by the rate of dissolution. Solubilization is an alternative to improve drug solubility in water with the addition of surfactant. The addition of Ryoto® sugar ester surfactant and propylene glycol cosolvent conducted to determine the concentration of Ryoto® sugar ester and prophylene glycol that increase the solubility and stability of the dosage form. Solubilization of Paracetamol with the addition of Ryoto® sugar ester without propylene glycol and combinations Ryoto® sugar ester and propylene glycol at a concentration below the CMC point (Critical Micell Concentration), at the point of CMC, and above the CMC points in a row is 0.005 mg/ml, 0.006 mg/ml, and 0.007 mg/ml with 10% propylene glycol. The highest solubility of Paracetamol achieved in the formula 7 with a combination of the addition Ryoto® sugar ester (0.007 mg/ml) and propylene glycol (10%) by the recovery percentage of 99.6%, and the combination is also obtained the best preparation stability which is not occur the color change in the preparations during storage at room temperature and place protected from direct sunlight for a month. The addition of Ryoto® sugar ester surfactant and propylene glycol increase the solubility and stability of the solubilization of Paracetamol preparations.

Abstract

A research on formulation of edible film from basil leaves extract as mouth freshener has been done. The extract of basil leaves were used in various concentrations which are 2.5%, 5% and 7.5%. The products were evaluated for some parameters such as organoleptic, friability, drying shrinkage, pH, thickness, flavonoid contents and respondents preference. The results of evaluation showed that edible filmsfrom basil leaves extract meet requirements as required by Standard Nasional Indonesia (SNI) and have such quality as product in the market. Statistical analysis using Kruskal Wallis test showed that respondents preferred for the F0 formulation in term of their appearance and taste while as mouth freshener, respondents preferred the F3 formulation.

Abstract

Pada penetitian ini pembentukan kokristal asam usnat-N-methyl-D-glucamine bertujuan untuk meningkatkan kelarutan dan laju disolusi asam usnat serta uji efektivitasnya dalam kadar IL-8 pada tikus inflamasi. Kokristal asam usnat–N-methyl-D-glucamine dibuat dengan perbandingan 1:1 ekuimola dengan metode penguapan pelarut menggunakan etanol. Karakterisasi sampel dilakukan dengan difraksi sinar-X, Differential Scanning Calorimetry (DSC), Fourier Transform Infrared (FTIR), dan Scanning Electron Microscopy (SEM). Evaluasi kokristal yang dilakukan mencakup uji kelarutan, uji laju disolusi, dan penentuan kadar interleukin-8 (IL-8) pada tikus inflamasi. Uji kelarutan dilakukan menggunakan pelarut air suling bebas CO2 sedangkan uji laju disolusi menggunakan media air suling bebas CO2 0.25% (b/v) sodium lauril sulfat (SLS), kedua larutan uji dianalisis menggunakan spektrofotometer UV-Vis. Pemeriksaan IL-8 tikus inflamasi menggunakan metode ELISA. Hasil karakterisasi difraksi sinar-X menunjukkan profil difraktogram yang berbeda dari komponen penyusun yang mengindikasikan terbentuknya fase kokristal. Hasil termogram DSC menunjukkan adanya puncak endotermik baru yang berbeda dengan kedua komponen penyusun. Spektrum FT-IR menunjukkan terjadinya pergeseran bilangan gelombang pada spektrum kokristal dibandingkan spektrum asam usnat dan N-methyl-D-glucamine. Analisis SEM menunjukkan perubahan morfologi partikel. Kelarutan dan laju disolusi kokristal meningkat secara signifikan (P<0,05) sebanyak 24 kali dan 23 kali dibandingkan dengan senyawa asam usnat murni. Kokristal dapat menurunkan kadar IL-8 pada tikus inflamasi yang setara dengan aktivitas antiinflamasi asetosal (P<0,05). Disimpulkan bahwa koristal asam usnat - N-methyl-D-glucamine dapat meningkatkan kelarutan, laju disolusi, dan menurunkan kadar IL-8 tikus inflamasi.

Abstract

Bromelin merupakan unsur pokok dari nanas yang penting dan berguna dalam bidang farmasi dan makanan, untuk mendapatkan enzim bromelin dari buah nanas diperlukan proses isolasi enzim bromelin. Tujuan dari penelitan ini adalah untuk mengisolasi dan mengkarakterisasi serbuk enzim bromelin dari batang nanas (Ananas Comosus (L.) Merr). Isolasi enzim bromelin dari batang nanas telah dilakukan dengan menggunakan pelarut dapar fosfat pH 7.0 disentrifugasi pada 3.500 rpm selama 15 menit. Serbuk enzim bromelin diperoleh dengan menggunakan metoda freeze drying dan didapat persentase randemen sebesar 2,032 %. Karakterisasi dari serbuk enzim bromelin terdiri dari pengukuran kosentrasi protein, penentuan aktivitas enzim dan aktivitas spesifik enzim. Pengukuran konsentrasi protein menggunakan metoda Biuret memberikan hasil konsentrasi sebesar 1,66±0,003 mg/ml. Penetuan aktivitas enzim menggunakan metoda Murachi memberikan hasil aktivitas enzim sebesar 43,09±0,01 U/ml dan aktivitas spesifik enzim sebesar 26,02±0,003 U/mg.

Abstract

Tablet salut enterik Ketoprofen adalah obat yang beredar luas di  Indonesia. Digunakan  untuk mengatasi nyeri arthiritis tulang, rematik dan demam. Namun, sejauh ini uji disolusinya secara spesifik belum tersedia dalam farmakope manapun.  Uji disolusi termasuk parameter yang harus diperhatikan untuk mengetahui kualitas produk obat sediaan padat. Tujuan dari penelitian ini adalah  mengembangkan dan memvalidasi metode pegujian disolusi dari tablet salut enterik ketoprofen. Profil disolusi diamati terhadap 3 produk tablet salut enterik ketoprofen yang beredar di kota Padang. Uji disolusi dilakukan dua tahap menggunakan 750 mL larutan HCl 0,1 N ( tahap asam) dan 1000 mL larutan dapar fosfat pH 6,8 dan 7,4 (tahap basa), alat tipe 1 (keranjang) dan tipe 2 (dayung) kecepatan 50 dan 75 rpm. Hasil uji disolusi selanjutnya ditentukan secara spektrofotometri UV. Metoda uji disolusi  hyperdiscriminating  diperoleh pada uji disolusi menggunakan alat tipe 1,  kecepatan rotasi 75 rpm dan  media disolusi 1000 mL dapar fosfat pH 6,8 dengan nilai Q45 ≥ 75%. Spesifiitas, linieritas  (r = 0,9988), presisi (RSD = 1,12%) dan akurasi (recoveri = 95,7 - 97,6%)  memenuhi syarat keberterimaan sesuai pedoman ICH dan USP. Uji disolusi yang dikembangkan dapat digunakan untuk tujuan pengawasan mutu tablet salut enterik ketoprofen .

Abstract

Eco-Enzyme, a multifunctional liquid produced from a mixture of organic waste such as vegetables, fruits, brown sugar, coconut sugar, corn sugar, sugarcane, and water that has undergone fermentation, has various benefits for the environment and human health. Eco-Enzyme contains beneficial enzymes that accelerate biochemical reactions in the environment. The enzymes found in Eco-Enzyme can be used for various applications, such as organic liquid fertilizer for plants, an additive in detergents, floor cleaners, pesticide residue cleaners, and rust removers. Using Eco-Enzyme as a liquid soap can help reduce soil pollution. Meanwhile, using Eco-Enzyme as a cleaning agent can help reduce the use of hazardous chemicals that damage the environment. This integrated program is carried out in three stages: the first is to socialize the benefits of using Eco-Enzyme, the second is to make Eco-Enzyme, and the third is to make soap from Eco-Enzyme. The production of Eco-Enzyme begins with the collection of organic waste consisting of fruit and vegetable waste, which is then mixed with brown sugar and water and left to ferment for three months. After the Eco-Enzyme product is ready, soap-making uses ingredients such as potassium hydroxide (KOH), coconut oil, cooking oil, distilled water, orange-scented perfume, and Eco-Enzyme. Training on Eco-Enzyme and soap making is conducted in the Andalas Subdistrict, which is welcomed enthusiastically by the local community. This program is expected to serve as a reference for transforming organic waste into more practical and environmentally friendly sustainable products.

Abstract

Ketoprofen is a non-steroidal anti-inflammatory compound (NSAID) which has low solubility in water. This study aimed to increase the dissolution rate of ketoprofen by forming multicomponent ketoprofen-glutamine in ratios of 1:1, 1:2, and 2:1 using the solid-state grinding method. The samples were characterized using Powder X-Ray Diffractometry (PXRD), Differential Scanning Calorimetry (DSC), Fourier Transform Infrared (FTIR) Spectroscopy, Scanning Electron Microscope (SEM), solubility, dissolution rate, and stability tests. These results indicated the formation of a eutectic mixture of ketoprofen-glutamine. From the study of the ketoprofen–glutamine eutectic mixture, the eutectic point at a ratio of 1:1 was obtained at 95.51°C. The PXRD spectra did not show new crystal habits; the solubility test of the eutectic mixture of ketoprofen-glutamine showed increases of 2.450, 2.518, and 2.374 times, respectively. The eutectic mixture of ketoprofen–glutamine had rod-like crystal particles that formed and aggregated. The samples showed an increased dissolution rate of ketoprofen. The stability test showed that ketoprofen-glutamine was stable during storage at 75% relative humidity (RH) and 40 °C for 6 months; there was only a decrease in intensity, as seen from the PXRD spectrum. Abstract

Capsule is capsule preparations which consisting of a drug in a soft capsule and hard capsule sell. Capsule is the preparations that most people used to discover the unpleasant taste of the drug. Most of capsule sell are made of gelatin, but it can also made with starch or other ingredients. The other ingredients which can substitute gelatin is pectin. Pectin is an ingredient belong to the class of carbohydrates. This study aim to find others alternative that can be used as new ingredients of capsule sell, which is jack fruit peels. Dissolved 20 mg jack fruit peels in 1000 ml of distilled water and, added 5% of citric acid. Then extracted on a hot plate with a stirring speed 600 rpm, temperature of 85ͦC, for 120 minutes. The yield obtained was 0.319%. The best formulations results were 2 (0.7 gram pectin, 5.00 gram carrageenan, 100ml disttiled water). The result of the evaluation of capsule sell standards with a body diameter of 6,2 mm, 6,3 mm, of a lid diameter, a body length of 15,2 mm, a lid length of 07.8 mm, and those that are unqualified with 0.09 gram of capsule weights. The organleptic results are light brown color, texture a little loud and there is no fragrance. The capsule sell disintegration should meet the requirements set by the Indonesian Pharmacopoeia V edition, is 13 minutes 47 seconds.Keywords: jackfruit skin (Artocarpus heterophyllus Lam), pectin, capsule shell
