Abstract

Costus speciosus (commonly known as pacing rhizome) has been traditionally used for various therapeutic purposes, yet data on its long-term safety, particularly regarding renal function, remain limited. This study aimed to evaluate the subchronic toxicity of C. speciosus rhizome extract through renal function assessment in white Wistar rats, addressing a critical gap in preclinical safety evidence and supporting the development of standardized herbal preparations. Forty healthy Wistar rats, equally divided by sex, were acclimatized for seven days and randomly assigned to control and treatment groups. The treatment group received C. speciosus rhizome extract orally at a dose of 300 mg/kg body weight daily for 90 days, while the control group received a vehicle solution (Na-CMC). Body weight was monitored regularly, and at baseline (T0) and at the end of the study (T90), blood samples were collected for biochemical analysis of serum creatinine and urea levels. Additionally, kidney organ indices were determined, and histopathological examinations were performed to detect potential structural changes. The results indicated that body weight gain in both male and female rats was comparable between control and treatment groups throughout the study, with no statistically significant differences. Serum creatinine and urea levels remained within normal physiological ranges at both T0 and T90, and no significant differences were observed between groups. Kidney organ indices showed minor variations that were not statistically significant, and histopathological evaluation revealed no treatment-related abnormalities. In conclusion, subchronic oral administration of C. speciosus rhizome extract at 300 mg/kg body weight for 90 days did not produce nephrotoxic effects in Wistar rats. The extract was well tolerated, and renal function remained intact, supporting its safety profile. These findings provide foundational evidence for the safe development of C. speciosus as a standardized herbal medicinal product, addressing the urgent need for scientifically validated data on its long-term safety.

Abstract

Obesitas adalah kondisi patologis akibat akumulasi lemak berlebih yang meningkatkan risiko gangguan metabolik, termasuk disfungsi endotel yang ditandai oleh penurunan produksi Nitric Oxide (NO) sintesis oleh enzim Endothelial Nitric Oxide Synthase (eNOS). Penelitian ini bertujuan mengevaluasi efek kombinasi ekstrak etanol rimpang kunyit (Curcuma longa L.) dan daun pegagan (Centella asiatica (L.) Urb) terhadap kadar NO serum dan ekspresi gen eNOS pada jaringan hati tikus obesitas. Penelitian eksperimental ini menggunakan tikus jantan yang diinduksi obesitas dengan pakan tinggi lemak dan karbohidrat selama 60 hari. Tikus dibagi enam kelompok: normal (kontrol negative), obesitas (kontrol positif), pembanding orlistat 10,8 mg/kgBB, ekstrak kunyit 200 mg/kgBB, ekstrak pegagan 20 mg/kgBB, dan kombinasi kunyit-pegagan 200:100 mg/kgBB. Parameter yang diukur meliputi berat badan, kadar NO serum (metode Griess), ekspresi gen eNOS (qPCR), kadar SGOT dan  SGPT. Hasil menunjukkan kombinasi ekstrak secara signifikan menurunkan berat badan dan meningkatkan kadar NO dibandingkan kelompok obesitas (p < 0,05), meningkatkan ekspresi gen eNOS serta secara (p< 0,05) signifikan menurunkan kadar SGOT dan SGPT . Penelitian ini mengindikasikan potensi kombinasi ekstrak kunyit dan pegagan sebagai agen preventif disfungsi endotel pada obesitas melalui peningkatan produksi NO dan regulasi ekspresi gen eNOS serta berperan sebagai hepatoprotektor. Kata Kunci: Ekstrak Kunyit, Ekstrak Pegagan, Ekspresi Gen eNOS, Jaringan Hati, Obesitas  Obesity is a pathological condition caused by excessive fat accumulation that increases the risk of metabolic disorders, including endothelial dysfunction characterized by decreased Nitric Oxide (NO) synthesis by the enzyme Endothelial Nitric Oxide Synthase (eNOS). This study aims to evaluate the effects of a combination of ethanol extracts of turmeric rhizome (Curcuma longa L.) and gotu kola leaves (Centella asiatica (L.) Urb) on serum NO levels and eNOS gene expression in the liver tissue of obese rats. This experimental study used male rats induced to become obese with a high-fat and high-carbohydrate diet for 60 days. The rats were divided into six groups: normal (negative control), obese (positive control), orlistat 10.8 mg/kgBW, turmeric extract 200 mg/kgBW, gotu kola extract 20 mg/kgBW, and turmeric-gotu kola combination 200:100 mg/kgBW. The measured parameters included body weight, serum NO levels (Griess method), eNOS gene expression (qPCR), SGOT and SGPT levels. The results showed that the combination of extracts significantly reduced body weight and increased NO levels compared to the obesity group (p < 0.05), increased eNOS gene expression, and significantly (p < 0.05) decreased SGOT and SGPT levels. This study indicates the potential of the combination of turmeric and pegagan extracts as a preventive agent for endothelial dysfunction in obesity through the enhancement of NO production and regulation of eNOS gene expression as well as acting as a hepatoprotector.

Abstract

The Boarding School partner has many students but the habit of washing their hands before eating is not accompanied by washing with soap. This is a concern because washing with soap ensures the disappearance of pathogenic microbes. Teaching the importance of washing hands with soap before meals aims to raise awareness about the benefits of maintaining hygiene and supplementary nutrition could improve health. We trained the pupils in the Boarding School to make liquid hand-washing soap and yoghurt to stimulate a hygienic lifestyle, provide in-house supplementary nutrition, and gain economic benefit from the products. The hygienic lifestyle knowledge was delivered through a presentation in an open-class setting, while soap and yoghurt production skills were taught by an in-hand workshop, followed by post-monitoring for both activities. The yoghurt was produced by fermentation, and the soap was made by formulating a macerate of fennel leaf extracted into hand-washing soap. The pupils’ knowledge of hygienic lifestyles improved with an average score of 68 to 90.

Abstract

The application of information technology in the health sector, especially the e-Puskesmas system, is intended to improve the quality of services at the Grobogan Regency Community Health Center. The main objective of the study was to analyze the comparison between two models: the Technology Acceptance Model (TAM) and the Unified Theory of Acceptance and Use of Technology 2 (UTAUT 2). The study was conducted at the Grobogan Regency Community Health Center from January to March 2025. The population in this study was pharmaceutical personnel at the Grobogan Regency Community Health Center, and the sample in the study consisted of 90 people. The method in this study was comparative quantitative, with data quality testing using validity tests, reliability tests, and one-way ANOVA tests. The results of the one-way ANOVA test showed no significant difference between the TAM method and the UTAUT 2 method, with a sig value of 0.773 > α (0.05), which means there was no significant difference between the TAM method and the UTAUT 2 method in measuring the success of the implementation of the Puskesmas at the Grobogan Regency Community Health Center. This study canit can be concluded that in this study there is no significant difference between the TAM Method and the UTAUT 2 Method in measuring the success of the implementatione-Puskesmasat the Community Health Centers in Grobogan Regency.

Abstract

Fructose has long been known as a sugar additive or sweetener, commonly found in soft drinks, pastries, and other processed foods. High levels of added sugar in the daily diet increase the risk of various metabolic disorders, including insulin resistance. Insulin resistance can lead to impaired endothelial function (endothelial dysfunction), which can lead to impaired vasodilation responses. Endothelial dysfunction can lead to an increased risk of cardiovascular diseases such as atherosclerosis, coronary heart disease (CHD), hypertension, and other vascular diseases. The aim of this study was to identify the potential of a combination of dadap serep (Erythrina subumbrans) and pegagan (Centella asiatica) to reduce the risk of endothelial dysfunction (as measured by nitric oxide (NO) levels) triggered by high fructose consumption. This research method is an in vivo experimental preventive using 36 rats divided into normal group, induction group, comparison group of curcumin 9 mg/kgBW, test group 1 EEPDS 50:100 mg/kgBW, test group 2 EEPDS 100:100 mg/kgBW and test group 3 EEPDS 100:50 mg/kgBW. Measurement of NO levels using the Griess method, while insulin resistance examination was carried out using the insulin tolerance test method. The results showed that the combination group of EEPDS 100:50 mg/kgBW was the group that showed the most optimal effect in maintaining endothelial function and had the best insulin sensitivity. The conclusion of this study shows that the combination of EEPDS 100:50 mg/kgBW is effective in maintaining insulin sensitivity and maintaining endothelial function in rats.
