Abstract

Diabetes is a disease characterized by increased levels of glucose in the blood that occur due to a lack of insulin production or the cessation of insulin production in the body. Sidaguri herb (Sida Rhombifolia) is known to have antidiabetic activity. This study was conducted to evaluate the antidiabetic activity of the ethanolic extract of the leaves and stems of sidaguri (Sida rhombifolia) in animal of type 2 diabetes model and to find out which part is the most effective as antidiabetic. This research method is an in vivo experimental study. Mice were divided into 9 groups, that is negative control group, positive control group, Glibenclamide 0.65 mg/KgBW, sidaguri leaf extract at a dose of 3.5; 7; and 14 mg/KgBW, as well as sidaguri stem extract at a dose of 3.5; 7; and 14 mg/KgBW. The test was carried out curatively, the test animals except negative controls were given induction of alloxan monohydrate 80 mg/KgBW via the intravenous route, then after 3 days post-induction, fasting blood glucose levels were checked. The treatment was continued by giving therapy for 14 days via the oral route. The test parameters in this study were the measurement of blood glucose levels on days 0, 7, and 14, and pancreatic histopathology was also performed. The test results show that was found that the extract that gave the best antidiabetic effect was the ethanolic extract of sidaguri leaves at a dose of 14 mg/KgBW but had not been able to fully improve the condition of the pancreatic cells

Abstract

Obesity is currently a disease with a prevalence that continues to increase every year and is a significant risk factor for cardiovascular disease. The relationship between obesity and inflammation is characterized by increased levels of TNF- which triggers inflammation. This study was conducted to determine the activity of katuk (Sauropus androgynus (L.) Merr) leaves as anti-obesity and anti-inflammatory properties. The study was conducted with a preventive method on white male wistar rats aged 2-3 months and body weight of 180-200 g. The test animals were divided into 6 groups that were induced by high-fat and carbohydrate diets except the normal group and were measured for anti-obesity and anti-inflammatory parameters. The results showed that katuk (Sauropus androgynus (L.) Merr) leaves can maintain activity as an anti-obesity and anti-inflammatory in test rats. Katuk leaf ethanol extract at a dose of 300 mg/kgBW has an effective activity as an anti-obesity, while at a dose of 150 mg/kgBW it has an anti-inflammatory activity.

Abstract

Kardiovaskular adalah Penyakit tidak menular yang menyerang masyarakat di negara maju hingga negara berkembang seperti Indonesia, salah satu dari penyakit kardiovaskular adalah hipertensi yang disebut sebagai penyumbang kematian terbanyak di dunia setiap tahunnya, seorang dengan tekanan darah abnormal dapat mengalami beberapa penyakit seperti gagal jantung, gagal ginjal, diabetes dan stroke. Tujuan dari penelitian ini untuk mengetahui potensi Carica Papaya dalam penanganan hipertensi dan perkembangan penelitian terhadap potensi Carica Papaya. Subjek review ini adalah artikel nasional dan internasional yang terindeks secara nasional dan international 10 tahun terakhir yang membahas kemampuan antihipertensi pada Carica Papaya dengan melakukan klasifikasi berdasarkan metode pengujian yang digunakan, mekanisme kerja senyawa kimia yang berperan aktif dalam aktivitas hipotensi dan bagian tumbuhan yang digunakan dalam pengujian. Pada studi literatur didapatkan bahwa Carica Papaya memiliki berbagai mineral dan metabolit sekunder yang mampu menurunkan tekanan darah melalu mekanisme yang mirip dengan obat konvensional, Carica Papaya memiliki senyawa saponin, alkaloid, flavonoid dan kalium yang mampu menurunkan tekanan darah dengan mekanisme vasodilatasi, ACE Inhibitor dan diuretik. Carica Papaya memiliki kemampuan antihipertensi yang baik pada seluruh bagian tumbuhan namun perlunya studi lebih lanjut secara spesifik pada senyawa aktif dan mekanisme kerja yang belum diteliti.

Abstract

The Costus speciosus plant, commonly known as pacing, is widely used both as an ornamental plant and in traditional medicine. All parts of the plant, particularly the rhizomes, are known to possess various pharmacological activities, including anti-diabetic, anti-cancer, anti-bacterial, and anti-inflammatory properties. However, with the extensive use of the rhizomes in traditional medicine, it is essential to assess their safety. This study aims to determine the LC50 value of ethanol extract from Costus speciosus rhizomes (EERP) and evaluate its acute toxicity level. The Fish Embryo Acute Toxicity (FET) Test was used to assess toxicity, employing zebrafish (Danio rerio) embryos as the model organism. Zebrafish are chosen because their genetic makeup shares about 60-70% similarity with humans, making them a suitable model for mapping toxic effects. The study involved seven experimental groups: two control groups (negative and positive controls) and five groups treated with varying concentrations of EERP (62.5 ppm, 31.25 ppm, 15.63 ppm, 7.81 ppm, and 3.91 ppm). The results revealed an LC50 value of 11.707 ppm for EERP, indicating that the extract falls under the "slightly toxic" category. These findings provide valuable insights into the potential toxicity of Costus speciosus, which is crucial for determining safe dosage levels in its use as a traditional medicine. Further research is needed to explore the underlying toxic mechanisms of this extract.

Abstract

Masyarakat secara tradisional menggunakan daun Kate Mas (Euphorbia heterophylla Desf.) untuk mengatasi kesulitan buang air besar. Penelitian sebelumnya melaporkan daun E. heterophylla memiliki aktivitas antioksidan DPPH dan anti-inflamasi pada tikus diinduksi karagenan. Penelitian ini bertujuan untuk menganalisis pengaruh pemberian ekstrak E. heterophylla (EEH) secara in vitro pada suspensi sel darah merah yang diinduksi larutan hipotonik, serta untuk mengevaluasi efek EEH dalam uji in vivo pada hewan yang diinduksi dengan larutan putih telur 1%. Studi in vitro menggunakan metode HRBC (Human Red Blood Cell) dengan darah yang dirusak larutan hipotonik sebagai media uji yang diukur menggunakan spektrofotometer UV-vis pada panjang gelombang 560 nm. Metode in vivo dilakukan pada tikus yang dibagi secara acak menjadi lima kelompok (n=5): kelompok kontrol positif (Na-CMC), kelompok pembanding (diklofenak natrium 4,5 mg/kg BB), dan tiga kelompok uji (EEH 50, 100, 200 mg/kg BB). Semua kelompok diinduksi dengan putih telur 1% pada telapak kaki tikus. Parameter yang diukur adalah nilai IC50 dan persentase inhibisi edema pada kaki tikus setiap 30 menit selama 180 menit. Hasil: Skrining metabolit sekunder pada EEH menunjukkan adanya flavonoid, alkaloid, saponin, tanin, kuinon, dan steroid/triterpenoid. EEH menunjukkan nilai IC50 yang tergolong aktif sebagai anti-inflamasi dan mampu mengurangi volume edema. Kesimpulan: EEH berpotensi untuk dikembangkan sebagai pengobatan anti-inflamasi. Kata Kunci: Anti-Inflamasi, Euphorbia heterophylla, Putih Telur, Larutan Hipotonik, Human Red Blood Cell The community traditionally uses Kate Mas (Euphorbia heterophylla Desf.) leaves to alleviate constipation. Previous studies reported that E. heterophylla leaves possess DPPH antioxidant and anti-inflammatory activities in carrageenan-induced rats. This study aims to analyze the effect of E. heterophylla extract (EEH) administration in vitro on red blood cell suspensions induced by a hypotonic solution, and to evaluate the effects of EEH in vivo on animals induced with 1% egg white solution. The in vitro study was conducted using the Human Red Blood Cell (HRBC) method, where red blood cells were subjected to damage by a hypotonic solution as the test medium. The measurement was performed using a UV-Vis spectrophotometer at a wavelength of 560 nm. The in vivo method was conducted on rats randomly divided into five groups (n=5): positive control group (Na-CMC), comparison group (diclofenac sodium 4.5 mg/kg BW), and three test groups (EEH 50, 100, 200 mg/kg BW). All groups were induced with 1% egg white on the rat's paw. The parameters measured were the IC50 value and the percentage of edema inhibition in the rat’s paw every 30 minutes for 180 minutes. Results: Secondary metabolite screening of EEH showed the presence of flavonoids, alkaloids, saponins, tannins, quinones, and steroids/triterpenoids. EEH demonstrated an IC50 value classified as active anti-inflammatory and was able to reduce edema volume. Conclusion: EEH has the potential to be developed as an anti-inflammatory treatment.
