Abstract

Cancer prevalence is steadily increasing each year and becomes the second leading cause of death worldwide. In Indonesia, breast cancer had a prevalence of 16.7% in 2018. Free radicals contribute to the occurrence of breast cancer, while antioxidants play a vital role in protecting cells and repairing the damage caused by free radicals. Ihau, an endemic fruit in Kalimantan Island, contains phytochemical compounds with potential antioxidant and anticancer properties. Utilizing local food as natural antioxidants could serve as an alternative for breast cancer prevention and treatment. This study aims to assess the anticancer potential of Ihau fruit extract on the MCF-7 breast cancer cell line. A post-test-only control group design method using the MTT assay was used, with two treatment groups: water and 96% ethanol extract, and four different concentrations (125, 250, 500, and 1000 ppm), and each was replicated three times. Statistical analysis using the ANOVA test found no significant difference among all concentrations. The IC50 values of the cytotoxic activity of water and ethanol extracts were 1,197.7 ppm and 1,148 ppm, respectively. It can be concluded that both water and ethanol extract of Ihau fruit exhibited very weak cytotoxic activity.

Abstract

Composition adulteration in a product is still a serious problem. For that we need a DNA-based test method that is fast, specific and simple. In this study, a rapid test technique with the loop-mediated isothermal amplification method was applied to identify the presence of pork DNA in meatballs. Six primers were used for the identification of mitochondrial DNA cytochrome oxidase subunit II. Samples were prepared in the form of mixed beef meatballs containing pork with a concentration of 50; 5; 0.5; 0.05%, respectively. The results showed that the optimal concentration for primers was 4 pmol inner primers, 2 pmol loop primers, and 1 pmol outer primers. The LAMP reaction showed a change in color in the meatball sample with the pork mixture to the lowest concentration. The LAMP technique provides a faster test with the final result of on-the-spot color visualization so that it is potential to be developed as a quick detection kit for identification of halal food processed products

Abstract

Diabetes Melitus Tipe 2 (DMT2) merupakan penyakit dengan karakteristik hiperglikemia yang disebabkan oleh resistensi insulin, gangguan sekresi insulin, maupun keduannya. DMT2 dapat diperbaiki melalui penatalaksanaan secara non farmakologi salah satunya melalui terapi nutrisi medis. Beras analog menjadi solusi alternatif bahan pangan sumber karbohidrat yang lebih sehat sebagai pengganti beras putih bagi pasien DMT2. Penelitian ini bertujuan untuk membuat produk beras analog kombinasi tepung kedelai edamame dan rumput laut untuk mengetahui hasil organoleptik dan kandungan gizinya. Uji organoleptik dilakukan kepada 25 panelis terlatih untuk mengetahui tingkat kesukaan berdasarkan rasa, aroma, warna, dan tekstur. Untuk mengetahui kandungan gizi, dilakukan uji proksimat dan uji anthrone. Penelitian ini merupakan penelitian eksperimental dengan 5 perlakuan dan 3 ulangan. Data hasil analisis diuji secara statistik menggunakan uji ANOVA dan dilanjutkan dengan uji Post Hoc Tukey. Persentase tingkat kesukaan tertinggi secara keseluruhan berada pada beras analog F3 (50%TE+50%TRL) yang mengandung karbohidrat sebesar 83,71 g, protein 2,76 g, lemak 0,23 g, kadar air 12,89 ml, abu 0,41 g, gula 0,18 g, serat larut 3,89 g, serat tidak larut 3,44 g total serat 7,29 g dan energi sebesar 346,92 kkal dalam 100 g. Uji statistika menunjukkan terdapat perbedaan yang signifikan antara kandungan gizi beras analog dengan beras putih (p<0,05).
