Abstract

Background: Some studies have reported a relationship between the sperm DNA fragmentation index (DFI) and the rate of fertilization and pregnancy. This study was designed to assess the mean sperm DNA fragmentation in pregnancies that occur in infertile couples, whether in natural pregnancy, intrauterine insemination (IUI), and in vitro fertilization (IVF) – intra cytoplasmic injection (ICSI). Methods: This research is an observational analytic study with a cross-sectional design. The sample taken in this study were infertile patients that underwent natural pregnancy or IUI or IVF-ICSI at Yasmin Infertility Clinic of Dr. Ciptomangunkusumo Hospital period 2018-2020 with a consecutive sampling technique. The research data was processed and analyzed by the Mann-Whitney test using the SPSS application. Results: The mean DFI of sperm in men with infertile couples who successfully conceived naturally was 10.7% (mild), while IUI was 20.4% (moderate), and IVF-ICSI was 30.5% (poor). The mean DFI in semen samples of men from infertile couples who underwent a natural program was significantly lower in those who successfully conceived compared to those who did not. Similar results were also shown in the IUI and IVF-ICSI programs, which showed a significantly lower DFI compared to non-pregnant women. Conclusion: DFI can be applied as a marker for selecting the type of pregnancy program in infertility management.

Abstract

In the absence of sperm retrieved from azoospermic male, thus considered in utilizing developed spermatogenic cells including spermatogonia to spermatid in restoring male fertility. Spermatogenic cells obtained from testicular tissue culture need to be expanded prior its utilization, yet the excessive apoptosis of resulting cell culture is a challenge. This review was aimed to discuss article dealing with the factors and the underlying mechanism inducing apoptosis of spermatogenic cells culture. A computerized search of PubMed and google scholar database was conducted for general terms such as “male infertility”, “azoospermia”, “human spermatogenesis, “spermatogenic cells” and “apoptosis” and followed by screening and evaluation of collected paper accordingly. Various agents induced the apoptosis in spermatogenic cells such as high temperature, hypothermic storage, metabolite of cell culture derived and gonadotropin suppression driving to apoptosis. Thus all of the agents were paving the specific pathways of inducing apoptosis, through extrinsic and intrinsic, leading to the activation of caspase initiators followed by the activation of caspase executor in various types of spermatogenic cells. Keywords: Human spermatogenic cells, Apoptosis, Culture.

Abstract

The cryopreserve technique using liquid nitrogen is used to preserve human sperm. Nevertheless, this technique has several disadvantages, such as bacterial/viral contamination and its expensive cost. Freeze-drying process offers more simple and cheaper of sperm storage technique. This study aimed to investigate the effect of storage temperature and period of freeze-dried human sperm on DNA integrity. Human sperm samples of 15 donors were prepared with simple washing techniques. Post-washing samples (T0) were divided into 4 groups with the same volume. The four group of samples carried on the freeze-drying process and stored based on different temperature and period: 4°C for 1 week (T1), room temperature (24-25ºC) for 1 week (T2), 4°C for 3 months (T3), and room temperature for 3 months (T4). Sperm DNA integrity was analyzed before and after the storage period of each group. There was a statistically significant difference of sperm DNA integrity of freezes - dried at 4°C in 1 week versus 3 months (T1 vs T3), at room temperature in 1 week versus 3 months (T2 vs T4) (p

Abstract

Abstract:Human umbilical cord derived mesenchymal stromal cells (UC-MSCs) that are accessible from cell banks can be actuated to separate into different cell sorts, accordingly making them handy potential hotspots for cell-based treatments. In injured peripheral nerves, Schwann cells (SCs) add to useful recuperation by supporting axonal recovery and myelin remaking. Here, we first exhibit a framework to induce UC-MSCs to separate into cells with SC properties (UC-SCs) by treatment with β-mercaptoethanol took after by retinoic corrosive and an arrangement of particular cytokines. The UC-SCs are morphologically like SCs and express SC markers, including P0, as evaluated by immunocytochemistry and converse interpretation polymerase chain response. Transplantation of UC-SCs into transected sciatic nerves in grown-up rats improved nerve recovery. The viability of UC-SCs for axonal recovery was tantamount to that of legitimate human SCs taking into account histological criteria and useful recuperation. Immunohistochemistry and immunoelectron microscopy additionally showed myelination of recovered axons by UC-SCs. These discoveries demonstrate that cells with SC properties and with the capacity to bolster axonal recovery and reproduce myelin can be effectively actuated from UC-MSCs to advance practical recuperation after peripheral nerve injury. This framework might be pertinent for the advancement of cell-based treatments.

Abstract

DAZ-like (DAZL) on chromosome 3 and BOULE on chromosome 2 are genes included in the DAZ gene family. These genes have a role as regulator of specific cell cycle in germ cells. Mutations on DAZ gene family cause meiotic arrest and infertility. DAZL and BOULE are known to have interaction with CDC25 in regulating meiosis in the cell cycle. The examination of infertility on azoospermia cases, which are caused by spermatogenesis arrest, has so far been limited by histological examination from testicular biopsy. Therefore, molecular research is needed to determine the candidate genes that could be used as a marker in improving the quality of testicular biopsy examination. This research is a cross-sectional study which used 40 biopsy testes samples based on Johnsen scores 2 to 8. The mRNA expression analysis of DAZL and BOULE used qRT-PCR. The DAZL gene began to be expressed in scoring 5-8, where there was an increase from scoring 5-7 then a decrease in scoring 8. The BOULE gene began to be expressed in scoring 2 – 8, but there was an increase from scoring 4 – 7 and then a decrease in scoring 6 and 8. The correlation between the mRNA expressions of DAZL and BOULE showed a significant positive correlation (r = 0.415, p<0.05). These results indicated that both DAZL and Boule contribute in the failure of spermatogenesis. Keywords: Male infertility, Spermatogenic failure, Johnson score, DAZL, BOULE

Abstract

Abstract: This study was aimed to determine the differences in embryo quality between spermatozoa with good or poor of DNA fragmentation index and chromatin maturation, through spermatozoa preparation with the addition of ALA. This experimental study compared the quality of embryos that fertilized by spermatozoa with DNA fragmentation and chromatin maturation, with and without ALA supplementation. In this study semen samples from 20 patients were collected, which were then prepared and examined for DNA fragmentation index (DF) and chromatin maturation (CM). Then the sample was divided into 4 groups, namely the good DF - good CM, good DF - poor CM, poor DF - good CM and poor CM - poor CM, further divided again with and without treatment (ALA supplementation). After injection of spermatozoa into the oocyte, the development of embryo was observed in each group. In the group without treatment, significant differences in embryo quality were obtained between groups of good DF - good CM with poor DF - good CM, good DF - good CM with poor DF - poor CM, good DF - poor CM with poor DF - good CM and good DF - poor CM with poor DF - poor CM (p <0.05). In the treatment group, there were not significant difference in the quality of the embryo between treatment groups, namely with or without ALA, in the group with good or poor DF, and with good or poor CM (p> 0.05). Embryo fertilized by spermatozoa with good or poor DF and with good or poor CM, with the supplementation of ALA, the quality was not significantly better than without the supplementation of ALA.    Keywords: spermatozoa DNA fragmentation, spermatozoa chromatin maturation, ALA, in vitro fertilization, embryo quality

Abstract

BackgroundThe endometrial preparation phase preceding embryo transfer assumes pivotal significance in achieving optimal endometrial receptivity and ensuring the success of pregnancy. The natural cycle (NC) and artificial cycle (AC) are the preferred methods for many in vitro fertilization (IVF) specialists. The objective of this study was to compare the difference between NC and AC in women undergoing frozen-thawed embryo transfer (FET) after IVF. MethodsA cross-sectional study was conducted involving 150 adult women who underwent FET with a single autologous blastocyst stage embryo in a 5-year period (2014-2019). Bivariate analysis was conducted to discern implantation and pregnancy rates associated with NC and AC for endometrial preparation. Multiple logistic regression was used to assess the association between endometrial preparation and clinical outcomes while adjusting for potential confounders. ResultsOf the 150 subjects meeting study criteria, 19 underwent NC, while 131 underwent AC for endometrial preparation. Natural cycle exhibited a higher biochemical pregnancy rates compared to AC (89.5% vs. 53.4%; p=0.003). Artificial cycle is a significant determinant for biochemical pregnancy rates compared to NC (aOR: 0.132;95% CI: 0.028 – 0.623; p=0.010) ConclusionIn women undergoing FET, NC resulted in higher biochemical pregnancy rates compared to AC. However, clinical pregnancy rates and ongoing pregnancies rates did not exhibit significant disparities between NC and AC. Future studies will hopefully further illuminate which protocol is most suitable for the individual patient. Further multi-center randomized clinical trials are needed to confirm the relationship between biochemical pregnancy rates and NC.
