Abstract

The structure of follicular tissue affects the ability to maintain the structural integrity of follicles against cryoinjury post-vitrification. Histological analysis was conducted on the structural integrity of each stage of preantral follicles post-vitrification using 7.5% and 15.0% doses of ethylene glycol (EG), and ovarian sections with HE staining were observed using an Olympus CX21 microscope connected to Optilab 3.0 lens and Image Raster software. Analysis was conducted on the ovarian cortex in the tracing line area using polygon measure tools to obtain follicle density (follicles/mm2) and follicle index (%) data. The result showed that the EG group 7.5% (KP1) increased follicle density compared to the vitrified group (KKV) in primordial (15.83±1.77) and primary (22.94±8.51) stages. Meanwhile, KP2 (EG 15%) was in primordial (41.92±6.45), primary (11.69±1.95), secondary (33.48±3.63), and tertiary (5.93±0.69) stages. KP1 increased grade 3 follicle index compared to KKV in primary (27.66±2.34), secondary (32.41±6.99), and tertiary (25.00±5.00) stages. Meanwhile, KP2 was in primary (26.87±6.68) and tertiary (25.00±5.00) stages. Both doses of 7.5% and 15.0% EG were able to maintain structural integrity at certain stages of preantral follicles. Secondary and tertiary follicles are the best stages in maintaining grade 3 follicular integrity with the addition of 7.5% EG.

Abstract

Background: Gap Junction (GJ) plays a role in supporting the heart electricity. Connexin43 (Cx43) as the main protein constituent of GJ in left cardiac ventricle, will increase in number and slightly redistributed to the lateral sides of cardiomyocytes after aerobic exercise in adulthood. The effects of aerobic exercise that begin at childhood are not well known.Objective: This study aims to observe the effect of aerobic exercise which started from childhood on left ventricle Cx43 distribution.Methods: This study was conducted on 28 male Juvenile (4 weeks) and young adult (8 weeks) rats, divided into 7 groups: 1) Juvenile rats undergoing 4 weeks of exercise (E-J4); 2) Control E-J4 (C-J4); 3) Juvenile rats undergoing 8 weeks of exercise (E-J8); 4) Control EJ-8 (C-J8); 5) Juvenile rats undergoing 12 weeks of exercise (E-J12; 6); Young adult rats undergoing 8 weeks of exercise (E-Yo8); 7) Control E-J12 and E-Yo8 (C-JY128). Exercise group will undergo different length of duration, starting from week 4 until 12 weeks. Cx43 was identified by immunohistochemical staining and analyzed with ImageJ software. Comparison was analyzed using independent t-test.Results: Insignificant lower of total Cx43 expression in E-J4 (64200.45 + 4243.676 total area, p >0.05) compared to control. In contrast, a significant higher of total Cx43 expression was observed in EJ-8, EJ-12 and E-Yo8 (80152.95 + 3760.481, p = 0.001; 75596.775 + 3976.333, p = 0,002; 81216.85+ 2475.768, p = 0,000). Slightly higher of lateral Cx43 redistribution occurred in all aerobic exercise, with significant lateralization in E-J8 and E-Yo8.Conclusion: Aerobic exercise increases Cx43 and slightly redistributed to lateral myocytes under normal condition both in juvenile and young adult rats.

Abstract

Perlemakan hati merupakan penyakit degeneratif yang dapat berkembang pada masa pertumbuhan. Perlemakan hati dapat terjadi pada dua kondisi yang sangat berbeda jauh secara patofisiologi yaitu kondisi kekurangan kalori dan kelebihan kalori. Penelitian ini bertujuan untuk membandingkan ukuran berat hati pada pemberian diet tinggi kalori, cukup kalori, dan rendah kalori pada tikus selama masa pertumbuhan. Sampel penelitian menggunakan 15 organ hati Spraque Dawley usia 6 minggu yang diberi asupan kalori berbeda selama 8 minggu. Penelitian ini terdiri atas 3 (tiga) kelompok yaitu kelompok kurang kalori (KK), kelompok cukup kalori (CK), dan kelompok tinggi kalori (TK). Parameter penelitian adalah pemeriksaan berat hati utuk menilai hepatomegaly. Hasil penelitian menunjukkan kelompok TK mengalami kenaikan berat hati ±20% lebih besar dibandingkan dengan kelompok CK dan kelompok KK (p<0,05). Penelitian ini menyimpulkan bahwa diet tinggi kalori dapat menyebabkan terjadinya  hepatomegaly.

Abstract

BACKGROUND: Many hypotheses regarding the pathophysiology of enterocolitis in aganglionic megacolon or Hirschsprung disease (HSCR) has been proposed. Paneth cells are columnar intestinal epithelial cells that have an important role in maintaining of intestinal homeostasis as a bactericide. Since enterocolitis in HSCR may have association with Paneth cells metaplasia and hyperplasia, current study investigated Paneth cells metaplasia and hyperplasia in the sigmoid colon of HSCR rat model and its products, namely a-defensins and IL-1b, in the sigmoid colon tissues.METHODS: Aganglionosis-induced and control Sprague-Dawley rats were euthanized on Day (D)-7, -14, -17, -19, -21, -23, -25, and -28. Sigmoid colon tissue was isolated at each time point, and degree of enterocolitis as well as Paneth cells metaplasia and hyperplasia were analyzed by Hematoxylin-eosin staining, then protein levels of a-defensins and interleukin (IL)-1b were determined by enzyme-linked immunosorbent assay (ELISA).RESULTS: Enterocolitis scores increased with time. The Paneth cells metaplasia and hyperplasia were observed on D14 until D28 (p<0.01 vs. control group) followed by an increased in the levels of IL-1b. The levels of a-defensins protein expression were initially increased (D7-D14; p<0.01 vs. control group) but then undergo reciprocal changes on D19 until D28 (p<0.01 vs. D7 and D14). Positive correlations between the degree of enterocolitis and Paneth cells number were detected in the sigmoid colon (r=0.42).CONCLUSION: Paneth cells underwent metaplasia and hyperplasia in the sigmoid colon of HSCR rats corresponding to an increase in the degree of enterocolitis, but not followed by an increase in the level of a-defensins as well as IL-1b, suggesting that there is an involvement of Paneth cells in the pathophysiology of enterocolitis due to HSCR.KEYWORDS: Hirschsprung, enterocolitis, defensins; metaplasia, Paneth cell, animal model

Abstract

Topical capsaicin with high concentrations (8%) is effective in the treatment of pain in Post-Herpetic Neuralgia (PHN) and HIV-Associated Distal Sensory Polyneuropathy (HIV-DSP) patients. High concentrations of capsaicin encapsulated in transfersome nanovesicles aim to reduce the irritating ability of capsaicin when using as drugs. Gel preparations were chosen in the formulation because they are non-sticky, easy to wash, easy to apply, and more practical. The components contained in the gel affect the stability of the gel. Physical and chemical stability tests need to be carried out to ensure the quality, safety and efficacy of the gel so that it meets the expected specifications during its shelf life. This study aims to determine the stability of selected gel formulas containing high concentrations of transfersome capsaicin. The selected gel formula is a gel containing a mixture of phosphatidylcholine (Phospholiphon 90G®) and Tween 80 (80:20). Stability was carried out at high temperature (40±2oC), room temperature (30±2oC), low temperature (5±3oC), and cycling test. The stability parameters tested included organoleptic, pH, viscosity, and assay. The results of the transfersom capsaicin gel stability test showed no change in pH, viscosity, and assay in all storage conditions, but the gel showed a change in color, smell, and taste when stored at high temperatures (40 ± 2oC). It can be concluded that the resulting capsaicin transfersome gel formula with a phospholipid:tween ratio of 80 (80:20) was less stable during 12 weeks of storage at 40 oC

Abstract

Abstract:Human umbilical cord derived mesenchymal stromal cells (UC-MSCs) that are accessible from cell banks can be actuated to separate into different cell sorts, accordingly making them handy potential hotspots for cell-based treatments. In injured peripheral nerves, Schwann cells (SCs) add to useful recuperation by supporting axonal recovery and myelin remaking. Here, we first exhibit a framework to induce UC-MSCs to separate into cells with SC properties (UC-SCs) by treatment with β-mercaptoethanol took after by retinoic corrosive and an arrangement of particular cytokines. The UC-SCs are morphologically like SCs and express SC markers, including P0, as evaluated by immunocytochemistry and converse interpretation polymerase chain response. Transplantation of UC-SCs into transected sciatic nerves in grown-up rats improved nerve recovery. The viability of UC-SCs for axonal recovery was tantamount to that of legitimate human SCs taking into account histological criteria and useful recuperation. Immunohistochemistry and immunoelectron microscopy additionally showed myelination of recovered axons by UC-SCs. These discoveries demonstrate that cells with SC properties and with the capacity to bolster axonal recovery and reproduce myelin can be effectively actuated from UC-MSCs to advance practical recuperation after peripheral nerve injury. This framework might be pertinent for the advancement of cell-based treatments.

Abstract

Highlights: This study observed the histology of pancreatic β-cell damage without any intervention to the kidneys of the animal models. The histological analysis of the kidneys shows that STZ-induced animal models can be used for assessing kidney abnormalities due to hyperglycemia. A scoring system for the histological analysis was developed to evaluate the changes in the kidney cells. Abstract The kidneys are one of the organs affected by microvascular complications due to diabetes mellitus. Hyperglycemia plays an important role in glomerular, mesangial cell, and tubular damage in the kidneys. Metabolic dysregulation, including hyperglycemia, initiates cellular damage in the kidneys. Streptozotocin (STZ) is a chemical compound that is known to damage pancreatic cells and cause hyperglycemia. This study aimed to examine the effects of hyperglycemia on the morphology of the kidneys. Kidney tissues were observed histologically using a light microscope. Samples were taken from the kidneys of experimental animals administered with STZ to induce hyperglycemia. Observation was performed afterwards to investigate any damage to pancreatic cells. A total of 12 kidney samples were divided into two groups: the control group and the STZ-induced group. The samples were prepared before staining with hematoxylin-eosin and Masson's trichrome. The endothelium, podocytes, mesangial cells, and basement membrane of the glomerulus were examined. The tubules of the kidneys were also examined, and the presence or absence of connective tissue formation in both groups was statistically tested. The results suggested a significant difference in tubular damage (p<0.05) and an insignificant difference in an increase in the damage of other components of the kidneys (p>0.05) in the STZ-induced group. Significant morphological changes were observed in the hyperglycemic renal tubules due to the administration of STZ. In conclusion, STZ-induced hyperglycemia caused damage to the kidney components but overall had no significant impact on the kidney.

Abstract

Background: Aging occurs in all organs, including the skin. As the largest organ of the body, wrinkles on the skin show signs of aging that are visible and affect appearance. Thread embedding acupuncture (akupunktur tanam benang/ATB) is one of acupuncture methods used in various cases, including aesthetics. This case report describes the role of ATB in the management of nasolabial folds (NLF). Case Illustration: A 45-year-old woman came complaints of deep and long smile lines. On physical examination, the NLF length (measured with digital vernier caliper) right/left: 34.15/30.3 mm; Wrinkle severity rating scale (WSRS) right/left: 3/3. ATB was given once and the results were observed until week 12. The best results were obtained at week 12 of follow-up (NLF: 22.00/21.46 mm; WSRS: 2/2) Conclusion: NLF improvement in ATB is getting better over time. Further clinical trials are needed to determine the effectiveness of ATB in improving NLF. Keywords: Thread embedding acupuncture; Wrinkle; Aesthetic acupuncture

Abstract

ABSTRACT Environmental auditory exposure in a form of both enrichment and stressor environmental can differently modulate the development of auditory system Prenatal chronic noise exposure caused neurogenesis and neuroplasticity disorders in the brain's auditory pathway resulting in neurocognitive impairment. Meanwhile exposure to prenatal music positively modifies morphological and biochemical developments in the brain's auditory pathway that supports neurogenesis and neuroplasticity. This study aimed to determine the plasticity of brainstem nuclei after exposure to music, noise, and a combination of both. This research used an experimental method using 24 female chicks as subjects. Subjects were obtained from eggs after incubation, which were divided into 4 groups: control, music, noise, and combined noise and music. Brain stem nuclear plasticity was measured by total number of nuclei, neuronal nuclear area, and synaptophysin expression as parameters. Prenatal music exposure significantly increased the total number of neurons, neuronal nuclear area, and synaptophysin expression in brain stem nuclei (p < 0.001), whereas combined and noise exposure significantly decreased these three plasticity parameters (p < 0.001). In conclusion, prenatal music exposure potential to increase neuroplasticity of brainstem nuclei for better neurocognitive function ABSTRAK Paparan pendengaran lingkungan dalam bentuk enrichment dan stresor lingkungan dapat memodulasi perkembangan sistem pendengaran secara berbeda. Paparan kebisingan kronis prenatal menyebabkan gangguan neurogenesis dan neuroplastisitas pada jalur pendengaran otak sehingga mengakibatkan gangguan neurokognitif. Sementara itu, paparan musik prenatal secara positif mengubah perkembangan morfologi dan biokimia pada jalur pendengaran otak yang mendukung neurogenesis dan neuroplastisitas. Penelitian ini bertujuan mengetahui plastisitas nucleus batang otak setelah paparan musik, kebisingan, dan kombinasi keduanya. Penelitian ini menggunakan metode eksperimen dengan menggunakan 24 ekor anak ayam betina sebagai subjeknya. Subjek diperoleh dari telur setelah diinkubasi, yang terbagi dalam 4 kelompok: kontrol, musik, kebisingan, dan gabungan kebisingan dan musik. Plastisitas nukleus batang otak diukur dengan parameter jumah total nukleus, luas neuronal nuclear area, dan ekpresi synaptophysin. Paparan musik prenatal meningkatkan jumlah total neuron, luas neuronal nuclear area, dan ekspresi synaptophysin di inti batang otak secara signifikan (p< 0,001), sedangkan paparan bising dan kombinasi secara signifikan menurunkan ketiga parameter plastisitas tersebut (p < 0.001). Kesimpulannya, paparan musik prenatal berpotensi meningkatkan neuroplastisitas inti batang otak untuk fungsi neurokognitif yang lebih baik
