Abstract

Al-toxicity stressed soybean will show its tolerance response by changing pod number per plant. Hence, Al-toxicity tolerance of soybean can be predicted using that character. The objective of the research was to study inheritance of pod number trait of soybean on acid soil. Development of base population was carried out by crossing the genotypes of W3898- 14-3 with MLGG 0583 and MLGG 0709 from March 2002 to January 2003. Then, from March to June 2003, 13 populations resulting from soybean base population were grown on Pulung Kencana, Tulang Bawang, Lampung, where the soil is Ultisol with Al saturity of 32.84%. The design was randomized completely block design with three replications. The space was 40 x 15 cm, one plant per hill. Results showed that based on pod number per plant, soybean tolerance was polygenically controlled, where the gene action was additive and there was no allelic and non-allelic interaction genes. Abstract

Studies in transgenic sugarcane (Saccharum officinarum L.) demonstrated that sucrose transporter (SUT) genes were essential in sucrose translocations. Sucrose transporter gene isolated from sugarcane were designated as SoSUT1 and SoSOSUT2 respectively as encode protein of 518 and 747 amino acids. The genes were constructed into plasmid pYES2 for SoSUT1, and pYX112 for SoSOSUT2. cDNA SoSOSUT2 had also constructed into plasmid pBIN-At-GFP which was possible to trace the gene inserted. The constructed plasmid was transformed into yeast (Saccharomyces cerevisiae) and grown in minimal medium (SD–urasil) as selection medium. The transformed colony was confirmed using PCR. The functional expression was studied by growing yeast in YPD medium with 2% Sucrose, then the sucrose uptake was measured in number interval time using resorcinol method. The result showed that yeast INVSc1-pYES2-SoSUT1 and BF264- pYX112-SoSOSUT2 had higher ability in sucrose transport compared to the control-INVSc1 for SoSUT1 and control-BF264 for SoUT2. Moreover, the result showed that SoSUT1 had higher ability to transport sucrose than SoSOSUT2. Confocal microscope observation showed that transformation gene SoSOSUT2 was successful, which was indicated by green exposure of GFP protein.Keyword: sugarcane, sucrose transporter, yeast

Abstract

This research was aimed at investigating both direct and indirect impacts, and heritability values of characters regarding the potential of corn production and resistance to downy mildew. The result of this investigation is required to determine some criteria taken into account for selection process of downy mildew-resistant corn breeding with high yield. The field experiment was conducted at Research Centre of Agriculture Faculty, Brawijaya University from January to April 2012. Five varieties of hybrid crown and five inbreeding lines were employed, and Randomised Block Design was applied with two replications. As observed, the characters held heritability ranging from average to high, except for heritability of length and width of stomata on the lower surface of the leaves which was categorised as low. Moreover, the stomata density found on lower surface of the leaves was directly and positively correlated to the intensity of attack by downy mildew, which, then, was used as criteria selection in downy mildew-resistance. The intensity of disease and the density of the stomata on lower surface of the leaves accounted for direct and negative correlation to corn production, while the length and diameter of corncob was responsible for direct and positive correlation to corn production. The betterment of corn production can be coped by improving the plant resistance to downy mildew and characters of corncob diameter.Keywords: path-analysis, corn, downy mildew

Abstract

ABSTRAK: Persilangan antar spesies kacang hijau dan kacang uci telah berhasil dilakukan. Kacang uci memiliki keunggulan yaitu tahan terhadap hama kumbang bubuk dan jumlah polongnya banyak. Dengan persilangan antar spesies ini dapat memperbaiki daya hasil dan ketahanan kacang hijau terhadap hama kumbang bubuk. Penelitian ini terdiri atas 4 tahap kegiatan yaitu: 1) persilangan antar spesies kacang hijau dengan kacang uci, 2). evaluasi dan seleksi beberapa populasi keturunan persilangan (F1) berdasarkan daya hasil dan ketahanan terhadap hama kumbang bubuk. Evaluasi dilakukan terhadap komponen hasil dan hasil, dan ketahanan terhadap kumbang bubuk menggunakan metode Chiang dan Talekar. Hasil dari penelitian ini adalah bahwa : 1). Terjadi perbaikan daya hasil dan ketahanan terhadap hama kumbang bubuk pada populasi hasil persilangan antara kacang hijau dan kacang uci. 2). Tingkat daya silang antara varietas yang satu dengan yang lainnya berbeda. Varietas Manyar dan Sampeong memiliki daya silang terhadap kacang uci dibandingkan dengan varietas Vima dan Merak, 3). Karakteristik populasi keturunan hasil persilangan berada diantara kedua tetuanya.

Abstract

This study aims to determine the relationship between lower limb muscle endurance and functional mobility among elderly individuals at Puskesmas II Tabanan. This observational analytic study employed a cross-sectional design and included 63 elderly individuals aged 60–69 years with normal BMI based on Asian adult standards. Participants were selected through simple random sampling. Lower limb muscle endurance was assessed using the 30-Second Chair Stand Test (30s-CST), while functional mobility was measured using the Timed Up and Go Test (TUGT). Data were analyzed using the Spearman’s Rho correlation test due to non-normal distribution. The Spearman’s correlation test showed a strong and statistically significant negative correlation between lower limb muscle endurance and functional mobility (r = -0.535, p = 0.000). Participants with higher repetition counts in the 30s-CST completed the TUGT in shorter times, indicating better mobility performance. There is a significant inverse relationship between lower limb muscle endurance and functional mobility in older adults. Routine assessment and targeted interventions focusing on muscle endurance should be integrated into geriatric healthcare programs to enhance mobility and reduce fall risk.

Abstract

Dalam pembelajaran Konsentrasi Keahlian Otomotif (KKO), siswa kerap mengalami kesulitan dalam memahami konsep sistem suspensi kendaraan, yang berdampak pada rendahnya hasil belajar dan motivasi belajar yang kurang optimal. Hal ini diperparah oleh pendekatan pembelajaran yang masih bersifat satu arah serta keterbatasan sarana ajar yang digunakan di kelas. Menanggapi permasalahan ini, penelitian ini bertujuan mengembangkan media pembelajaran interaktif berbasis Canva AI yang dirancang dengan pendekatan Problem Based Learning (PBL), sehingga siswa dapat belajar melalui pemecahan masalah nyata yang relevan dengan dunia otomotif. Penelitian ini dilakukan di SMKS Musda Perbaungan dengan melibatkan siswa kelas XI TKRO 1 sebagai subjek. Metode yang digunakan adalah Research and Development (R&D), mengacu pada model ADDIE yang mencakup lima tahap: analisis, desain, pengembangan, implementasi, dan evaluasi. Hasil pengembangan media dinilai oleh para ahli dan menunjukkan tingkat kelayakan yang sangat baik, dengan rata-rata skor dari ahli media sebesar 0,95, ahli desain pembelajaran sebesar 0,95, dan ahli materi sebesar 0,96 semuanya termasuk dalam kategori "valid". Pengujian dilakukan melalui tiga tahap: uji coba individu dengan skor rata-rata 68,33, uji coba kelompok kecil 67,78, dan uji coba kelompok besar 67,63 seluruhnya menunjukkan bahwa media termasuk dalam kategori layak. Secara keseluruhan, skor kelayakan rata-rata adalah 34,43. Dari segi efektivitas, terdapat peningkatan signifikan dalam hasil belajar siswa, dari rata-rata Pretest 26,96 menjadi 92,27 pada Posttest. Nilai N-Gain sebesar 0,89 tergolong dalam kategori peningkatan tinggi, dan efektivitas pembelajaran mencapai 89,21%, yang menunjukkan bahwa media ini sangat efektif. Berdasarkan hasil penelitian, media pembelajaran interaktif berbasis Canva AI dengan pendekatan PBL terbukti valid dan efektif dalam meningkatkan pemahaman dan hasil belajar siswa pada materi sistem suspensi.

Abstract

Pada mata pelajaran Konsentrasi Keahlian Otomotif (KKO), peserta didik menghadapi berbagai kendala, seperti rendahnya motivasi belajar, kurangnya pemahaman terhadap materi, serta hasil belajar yang belum memuaskan. Kondisi ini disebabkan oleh penerapan metode pembelajaran konvensional yang berpusat pada pendidik, serta keterbatasan media pembelajaran yang digunakan, seperti papan tulis dan proyektor LCD. Untuk mengatasi permasalahan tersebut, penelitian ini bertujuan mengembangkan media pembelajaran interaktif berbasis laman web dengan pendekatan Project Based Learning (PjBL) yang dinilai layak dan efektif. Penelitian ini dilaksanakan di SMKS Musda Perbaungan dengan melibatkan peserta didik kelas XI TKRO 1 sebagai subjek penelitian. Metode yang digunakan adalah Research and Development (R&D) dengan mengacu pada model pengembangan ADDIE, yang meliputi tahap Analisis, Perancangan, Pengembangan, Implementasi, dan Evaluasi. Hasil penelitian menunjukkan bahwa media pembelajaran yang dikembangkan memenuhi kriteria kelayakan. Penilaian oleh ahli media, ahli desain pembelajaran, dan ahli materi memperoleh skor rata-rata berturut-turut sebesar 0,87; 0,92; dan 0,98, yang termasuk dalam kategori “valid”. Uji coba perseorangan memperoleh nilai rata-rata 59,33; uji coba kelompok kecil sebesar 59,55; dan uji coba kelompok besar sebesar 59,7 — seluruhnya tergolong dalam kategori “valid”. Rata-rata keseluruhan nilai kelayakan adalah 30,23. Dari aspek efektivitas, terdapat peningkatan signifikan pada hasil belajar peserta didik, dari rata-rata nilai prates sebesar 49,44 menjadi 91,66 pada pascates, dengan selisih peningkatan sebesar 42,22. Nilai N-Gain sebesar 0,83 menunjukkan kategori peningkatan tinggi, dan tingkat efektivitas mencapai 83,49% (efektif). Berdasarkan temuan tersebut, media pembelajaran interaktif berbasis laman web dengan pendekatan PjBL dinyatakan sangat layak dan efektif dalam meningkatkan hasil belajar peserta didik pada mata pelajaran KKO di kelas XI TKRO SMKS Musda Perbaungan.
