Abstract

Aedes aegypti is a primer vector of dengue virus (DENV) and chikungunya virus (CHIKV). The susceptibility of mosquitoes to DENV and CHIKV depends on their recognition receptor of pathogens. C-type lectins (CTLs) are an important mediator of virus infection in A. aegypti. This study aims to identify potential receptors and determine the binding affinity between ligand–receptor interaction, CTLs and virus envelopes (DENV-1, 2, 3, and 4 and CHIKV) interaction based on in silico analysis. Sample sequences were obtained from GenBank (NCBI), and 10 CTLs were acquired from VectorBase. Homology modeling based on a minimum standard of 20% was processed using the SWISS-MODEL server. Molecule interaction was visualized as tertiary protein structure in PyMol V1.7.4. Homology modeling revealed all 3D protein model scores to be at 20%. The highest homology modeling score of 98.72% is obtained for CHIKV envelope protein (KF925315.1). The CTL GenBank ID: AAEL014382-PA has the lowest score at −1407.20 kcal/mol. The predicted mechanism of DENV-1, 2, 3, and 4 co-infections with CHIKV-1 is through the binding of the viral envelope to the CTL. Interaction analysis showed that the viruses enter through an endocytic mechanism. The results of homology modeling and lowest interaction energy could serve as the basis for in vivo studies on virus co-infection in A. aegypti. This information illustrates that CTL receptors facilitate DENV and CHIKV co-infection in A. aegypti.

Abstract

Non-small cell lung cancer (NSCLC) presents a significant global health challenge, with its prevalence and mortality rates rising steadily. In Indonesia, Syzygium cumini (L.) Skeels, known for its flavonoid richness, has a long history in traditional medicine. However, its specific mechanisms of action against cancer, particularly in inducing apoptosis in NSCLC, have not been fully elucidated. This study utilized an in silico approach to evaluate the pro-apoptotic potential of S. cumini flavonoids against NSCLC by targeting key proteins: Bcl-2, Bax, and Caspase-3. We retrieved flavonoid structures from PubChem and protein data from the Research Collaboratory for Structural Bioinformatics Protein Data Bank (RCSB PDB). The drug-likeness of these compounds was assessed using Swiss ADME, adhering to Lipinski's rule of five, while their anti-NSCLC probability was predicted using PASS Online. Molecular docking and screening were performed with PyRx, and the results were visualized using Discovery Studio. Our findings identified epigallocatechin 3-O-gallate and ellagic acid as the most promising anti-NSCLC candidates. Ellagic acid demonstrated the strongest binding affinity to Caspase-3, suggesting a potent pro-apoptotic effect. Epigallocatechin 3-O-gallate, on the other hand, exhibited the lowest binding energy across multiple target proteins, particularly Bcl-2 and Bax, indicating its broad pro-apoptotic potential. These results collectively suggest that flavonoids from S. cumini may hold significant promise as a source of novel anti-NSCLC agents, warranting further in vitro and in vivo investigations.

Abstract

Mangrove ecosystems play an important role in maintaining the balance of the coastal environment and become a habitat for various types of fauna, including arthropods. This study aims to identify the types of arthropods and determine the level of diversity in the mangrove ecosystem of Cemara Beach, West Lombok Regency. The method used is an exploratory survey method with sampling techniques using Hand Collecting and Sweep Netting in several mangrove vegetation zones.  The presence of these arthropods suggests that the mangrove ecosystem in Cemara Beach still supports the life of various small organisms, although some environmental changes, such as deforestation and increased garbage are beginning to affect the presence of some species. Therefore, the results of this study can be the initial basis for sustainable mangrove conservation and management efforts, while affirming the importance of the role of arthropods in maintaining the balance of coastal ecosystems.

Abstract

This study aims to identify the diversity of surface-dwelling insects in Jeruk Manis, located within Gunung Rinjani National Park, and to analyze species diversity, dominance, and evenness indices. A survey method employing stratified random sampling based on habitat type was applied. Insect sampling was conducted at eight observation points on April 26, 2025. The study identified seven species of surface-dwelling insects: Largidae, Pachycondyla, Tapinoma sessile, Calliptamus italicus, Gray bird grasshopper, Pyrrhocoridae, and Thanasimus formicarius. Data analysis using the Shannon-Wiener Diversity Index yielded a diversity value (H') of 2.70, indicating a moderate level of species diversity. The Dominance Index (C) of 0.126 suggests that no single species dominates the community. However, the Evenness Index (J'), which varies between 0.126 and 7.397, reveals an uneven distribution of individuals among species, with certain species exhibiting significantly higher abundance than others. This research contributes to a deeper understanding of the insect ecosystem in the region and highlights the crucial role of surface-dwelling insects as bioindicators of environmental health.
