Abstract

Mutasi merupakan teknik untuk meningkatkan keragaman genetik sebagai modal dasar perakitan varietas unggul. Tujuan penelitian ini adalah untuk menganalisis keragaman genetik populasi M2 cabai hasil iradiasi sinar gamma berdasarkan karakter morfologi. Analisis keragaman genetik dilakukan melalui pendugaan ragam genetik, heritabilitas arti luas, serta sidik lintas karakter-karakter yang berpengaruh langsung maupun tidak langsung terhadap hasil. Sebanyak 350 tanaman M2 dievaluasi karakter morfologinya di Kebun Percobaan BB Biogen Bogor pada bulan Agustus 2020 hingga Januari 2021. Hasil analisis menunjukkan nilai keragaman genetik yang luas pada populasi M2 diperoleh pada karakter umur panen dan lebar tajuk. Nilai heritabilitas arti luas yang tinggi terdapat pada karakter panjang buah, diameter buah, bobot per buah, panjang daun, lebar daun, dan lebar tajuk. Sementara itu sidik lintas menunjukkan terdapat dua karakter yang memiliki nilai pengaruh langsung yang besar terhadap hasil yaitu diameter batang dan lebar tajuk. Abstract

Plant regeneration post-genetic transformation play an important an role in genome editing activities that should be optimized via several factors such as the composition of the medium and the concentration of plant growth regulators. This study aimed to optimize the kinetin concentrations and medium compositions for shoot multiplication originating from cotyledon node explants of several local citrus cultivars. The cotyledonary nodes from three citrus cultivars (Batu 55, Siam Madu, and Proksi-1 Agrihorti) were incubated in MS medium with Morel and Wetmore vitamins (VMW) supplemented with several kinetin concentrations (0; 0.2; 0.4; 0.6; 0.8; and 1 mg/L). The best kinetin concentrations for number of shoots variable were then combined with MT medium. The results showed that kinetin concentration at 0.8 mg/L gave the best number of shoot in Batu 55 cultivar as well as 1 mg/L concentration in Siam Madu and Proksi-1 Agrihorti cultivars. The combination 1 mg/L kinetin with Murashige and Tucker (MT) medium showed the best number of shoots, percentage of shoot formation, number of leaves, number of nodes, and shoot length in this study. This medium composition could be further used for shoot multiplication in genetic transformation in those three citrus cultivars, including genome editing activities in development of new improved citrus varieties.

Abstract

The evaluation of citrus resistance to Huanglongbing (HLB) disease is still challenging due to the incapability of the bacteria to be cultured purely in artificial medium, the complexity of inoculation methods, and the long duration required for phenotypic observation. Thus, the use of molecular markers is one of the alternatives to solve this problem. The focuses of this study were to perform in silico analysis of the nucleotide variations in NONEXPRESSOR OF PATHOGENESIS-RELATED GENES 1 (NPR1) gene sequence among several citrus genotypes whose resistance information to HLB have been known previously and to employ phylogenetic analysis among them. The NPR1 gene sequences from 20 genotypes which consisted of 14 citrus and six relative genotypes were collected in silico from Citrus Genome Database and analyzed using multiple sequence alignment program. A total of six interesting SNPs that could distinguish between susceptible and resistant citrus genotypes were detected in this study. As many as five SNPs were non-synonymous, while only one synonymous SNP that did not cause the amino acid change was identified in this study. The phylogenetic analysis also revealed the separation between susceptible and tolerant/resistant citrus genotypes in two main clusters. The SNPs found in this study are expected to be useful for designing new functional markers as a selection tool in future studies.
