Abstract

A bipotentiometric FIA system has been developed for the determination of vitamin C. The system was a single stream manifold, which consisted of a peristaltic pump, a self made injector with internal loop and two Pt-electrodes connected to a PC by serial ADC or parallel ADC interface card. One of the Ptelectrodes was located in the injector before the internal loop, while the other electrodes at the end of the TEFLON reaction tube. The PC was supported with a self-made software for controlling the pump, signal acquisition, processing, and visualisation, and further peak determination, followed by calibration of standard solutions and calculation of sample concentrations. The above FIA-system configuration was applied to determine vitamin C by the injection of sample solution into a stream of potassium hexacyanoferrate (III) solution. The optimum result was achieved with knitted tube reaction of 0.8 mm i.d., 50 cm length, at flow rate 0,64 ml/minute, and consumed 3.6 ml of carrier solution per five injection, and gave linearity range at 0.001 – 0.100 N vitamin C, with the coefficient of correlation 0.9999, RSD peak to peak better than 3.0% and the error of artificial samples were less than 3.4 %.Key Words : Bipotentiometric-FIA, injector with internal loop, module-program, vitamin C

Abstract

Resistance to the traditional antifolates is now widespread in Plasmodium falciparum and Plasmodium vivax. To find the interaction model of some compounds derived from 1,6-dihydro-1,3,5-triazine-2,4-diamine, molecular docking technique was carried out using these compounds ligand and pDHFR as the receptor. Complex ligand and the receptor from Protein Data Bank (PDB ID 1J3K) were chosen as an interaction model between a ligand and its receptor. Each compound was optimized using ab initio methods with 6-311G basis set. Partial charges of ligand were added using AM1-BCC methods. Each ligand was docked to the receptor utilizing Dock6 software. Theoretical prediction based on the binding energy between these compounds as the ligand with pDHFR as receptor resulted in 1 compound, namely 6,6-dimethyl-1-[3-(2-chloro-4,5-dibromophenoxy)propoxy]-1,6-dihydro-1,3,5-triazine-2,4-diamine possessing binding affinity better than that of WR99210 which was known to have good antimalarial activity.

Abstract

Guru mempunyai kewajiban besar memastikan anak didik memahami materi pelajaran. Teachers are responsible for ensuring students catch on to the subject matter. However, the limited study time in class and students' varying abilities cause face-to-face learning methods to be less effective in increasing student understanding. The global challenge of education demands teachers become competent in digital mastering technology. Learning videos that are presented virtually are an effective learning method for students. Consequently, teachers should have the ability to make digital videos independently. This study introduced digital technology types, installation, and video-making applications for teachers in Muhammadiyah's junior and senior high schools in Gresik (SMP-SMA Muhammadiyah Gresik) on June 21, 2022, increasing teacher competency in using digital technology in learning. The implementation method includes providing material by experts, having participants practice accompanied by mentors, and making videos independently. Based on the analysis of the pre-test and post-test scores during the activity, there was an increase in the participant's score from before the training (pre-test), which was 76.8. After participating in the training (post-test), the participant scored 97.2. Participants gain an increased understanding of making learning videos, as shown by the results of learning videos made individually by participants. The learning videos produced by the participants also met the criteria for good digital videos. Activity evaluation data show that this workshop is necessary and right on target for professional development, especially for SMP-SMA Muhammadiyah Gresik Teachers. The activity concept is also suitable for solving problems in junior high school level education. Thus, this training succeeded in increasing the competence of participants regarding the use of video and digital technology as one of the learning innovations.

Abstract

Structure-based was performed to understand the mechanism inhibition of pyrimethamine (CP6) against Plasmodium Falciparum mutants at the molecular level. A molecular docking process was carried out to obtain the initial conformation of each system. The data showed in the form of RMSD values, hydrogen bonds, and grid scores. The results show that the RMSD value in the redocking process meets the criteria of the CP6-1J3J and WRA-1J3K complexes with a value of ≤ 2 í…. Several hydrogen bonds bind to receptor-active sites, including Ile14, Asp50, Ile164, and Asn108. Additionally, the grid score (kcal/mol) binds well on the active site: CP6-1J3J (-46.86), WRA-1J3K (-65.40), and CP6-1J3k (-44.71). Furthermore, molecular dynamics simulation was carried out to determine the free energy (∆Gbind) of each complex using the MM/GBSA approach. The results show ∆Gbind (kcal/mol) in each complex, namely CP6-1J3J (-28.24), WRA-1J3K (-36.62), and CP6-1J3k (-24.23). Information on this research was expected to provide molecular insight into pyrimethamine as an antimalaria inhibitor.
