Abstract

Infection caused by hepatitis B virus (HBV) is still a major global health problem and can cause liver cirrhosis and hepatocellularcarcinoma as well. Telbivudine is one among the drugs used to treat the disease routinely. However, using this drug in a long term therapymight cause mutations in HBV polymerase gene that decreases the effectiveness of the therapy. Here with the researchers report the geneticvariations of the gene isolated from telbivudine-which is used treated chronic hepatitis B patients in Surabaya, Indonesia. The blood serawere collected at Dr. Soetomo hospital from 10 telbivudine-treated and 10 untreated chronic hepatitis B patients. The DNA viral wasisolated and purified from each serum. Sequence polymerase gene at nucleotides 455 to 796 was amplified by PCR, and then analyzedbio informatically to determine their mutation profile. This study revealed a point mutation in HBV25 sample at nucleotide A1525G thatgives rise to I509V modification. Such mutation is also observed in a sequence that is available in Gen Bank with an accession numberAY641562. Additionally, the researchers found point mutations A1554G, T1593C, and C1629T in HBV25 sample and a point mutationA1554G in HBV20 sample. However, these mutations are silent. To conclude, the mutation in HBV polymerase gene among telbivudinetreatedchronic hepatitis B patients in Surabaya is known as A1525G.

Abstract

The aims of this research were to know the ability of recombinant xylanolytic enzyme from recombinant E. coli DH5a (pTP510) to hydrolyze several commercial xylan and analysis the reduction sugar product. Recombinant xylanolytic enzyme (exo-xylanase, b-xylosidase and a-L-arabinofuranosidase) could hydrolyzed several commercial xylan (oat-spelt xylan, birchwood, wheat, rye, and arabinan) with xylanolytic activities are: oat-spelt xylan (1.73 U/mL), birchwood (0.92 U/mL), wheat (6.52 U/mL), rye (4.94 U/mL), and arabinan (3.40 U/mL). Xylanolytic enzyme assay use specific substrate p-nitrophenyl-b-D-xylopyranoside (pNP-X) shown xylosidase activity 15.869 U/mL. Hydrolysis product was analyzed by HPLC. The results showed that xylose, arabinose, and xylo-oligosaccharide were produced from birchwood, wheat, rye, and arabinan hydrolysis, although xylose and arabinose were produced from hydrolysis of oat-spelt xylan.

Abstract

Chronic activivity of Hepatitis B Virus (HBV) infection can lead to liver cirrhosis or hepatocellular carcinoma. The objective of thisstudy was to know by analyzing the distribution of HBV genotypes and subtypes from hepatitis B patients suffering from chronic activehepatitis B infection in Surabaya. The HBV genotypes were determined by comparing the S gene sequences to those kept in the GeneBank. The phylogenetic tree was constructed by means of the unweighted-pair group method using arithmetic averages. Furthermore,the subtypes were deduced based on the prediction of amino acid residues 116 to 183 of HBsAg on multiple sequences alignment withClustalW2. This study involved 20 sera obtained from patients suffering chronic active hepatitis B infection. After PCR and sequencing,it was found that 13 samples could be used for sequence analysis. The results showed that all sequences were clustered into HBV genotypeB. The subtype adw2 was identified from 12 of 13 sequences, whereas one (1) belonged to ayw1. The subtype adw2 is most prevalent inIndonesia, namely in the islands of Sumatra, Java, South Kalimantan, Bali, Lombok, Ternate, and Morotai, while ayw1 is found in theislands of Nusa Tenggara and Moluccas. Based on this study, it was found that the patients with HBV subtype adw2 were from Surabaya, whereas with ayw1 was from Nusa Tenggara. It can be concluded that the HBV infected patients with chronic active hepatitis B inSurabaya have the genotype B with subtype adw2 which was originally from Surabaya, whereas, ayw1 was a patient originally fromNusa Tenggara.

Abstract

Glucoamylase, the enzyme which converts starch to dextrins and glucose, is used in the starch-processing industry, especially for the production of glucose crystal and high fructose syrup. The aim of this work was to produce glucoamylase by Aspergillus niger BCS using rice husk as solid substrate support. For this purpose, rice husk added to the mixture was optimized on a fixed weight ratio of cassava starch and rice bran. After that, using optimum rice husk concentration, optimization of the weight ratio of cassava starch and rice bran was carried out. Glucoamylase A. niger produced in this work was characterized by determining its optimum pH and temperature, its stability under various pH and temperature, its kinetic parameters, its hydrolysis products using soluble starch as substrate, and its molecular weights. The optimum conditions for glucoamylase production by A. niger obtained when the weight ratio of cassava starch: rice bran was 1:1 (dry basis, db) and the rice husk concentration in the fermentation medium was 20% by weight (db). Under these optimized conditions, enzyme units as high as 1774 U/g dry fermented substrate was obtained when fermentation was carried out for 5 days at 30°C. Optimum pH of glucoamylase A. niger was 4.5 and its optimum temperature activity was 65°C. This enzyme was stable at pH 3.0 - 7.0 and at a temperature up to 50°C.

Abstract

A thermophilic aerobe microorganism designated NP4, was isolated from the heated sugar cane. It grew on dextran, and produced a thermoactive extracellular dextranase. Screening and isolation was done by assay of dextranase activity semi quantitatively on solid medium containing blue dextran. It provided several colonies with different morphology exhibited decolourized zones around, on culture plates containing blue dextran 2000R. The screening resulted in isolation of one microbe which efficiently assimilate dextran as carbon source. Dextranase production from the choised strain in liquid medium was conducted at room temperature for 8 hours with shaking speed of 125 rpm. The dextranase enzyme showed optimum pH of 8 and optimum temperature of 60 oC.

Abstract

Guru mempunyai kewajiban besar memastikan anak didik memahami materi pelajaran. Namun, waktu belajar di kelas yang terbatas dan beragamnya kemampuan siswa menyebabkan metode pembelajaran tatap muka di kelas menjadi kurang efektif dalam meningkatkan pemahaman siswa. Tantangan dunia pendidikan menuntut guru harus memiliki kompetensi penguasaan teknologi digital. Video pembelajaran yang disajikan secara virtual adalah salah satu metode pembelajaran efektif untuk siswa. Oleh karena itu, guru seyogyanya memiliki kemampuan membuat video digital secara mandiri. Pengenalan jenis teknologi digital, instalasi dan penggunaan aplikasi pembuatan video telah dilakukan ke 45 orang Guru SMP-SMA Muhammadiyah Gresik pada tanggal 21 Juni 2022 untuk meningkatkan kompetensi guru terhadap pemanfaatan teknologi digital dalam pembelajaran. Metode pelaksanaan kegiatan meliputi pemberian materi oleh tenaga ahli, praktik peserta didampingi oleh mentor, dan pembuatan video secara mandiri. Berdasar hasil analisis nilai pre-test dan post-test saat kegiatan, terdapat peningkatan nilai peserta dari sebelum pelatihan (pre-test) adalah 76,8 dan setelah mengikuti pelatihan (post-test), nilai peserta menjadi 97,2. Peserta mendapatkan peningkatan pemahaman dalam membuat video pembelajaran yang ditunjukkan dengan hasil karya video pembelajaran yang dibuat secara individu oleh peserta. Video pembelajaran yang dihasilkan peserta juga telah memenuhi kriteria video digital yang baik. Data evaluasi kegiatan menunjukkan workshop ini diperlukan dan tepat sasaran untuk pengembangan profesional khususnya Guru SMP-SMA Muhammadiyah Gresik. Konsep kegiatan juga sesuai untuk menyelesaikan permasalahan yang ada di pendidikan tingkat SMP-SMA. Dengan demikian, pelatihan ini berhasil meningkatkan kompetensi peserta tentang penggunaan video dan teknologi digital sebagai salah satu inovasi pembelajaran.Teachers are responsible for ensuring students catch on to the subject matter. However, the limited study time in class and students' varying abilities cause face-to-face learning methods to be less effective in increasing student understanding. The global challenge of education demands teachers become competent in digital mastering technology. Learning videos that are presented virtually are an effective learning method for students. Consequently, teachers should have the ability to make digital videos independently. This study introduced digital technology types, installation, and video-making applications for teachers in Muhammadiyah's junior and senior high schools in Gresik (SMP-SMA Muhammadiyah Gresik) on June 21, 2022, increasing teacher competency in using digital technology in learning. The implementation method includes providing material by experts, having participants practice accompanied by mentors, and making videos independently. Based on the analysis of the pre-test and post-test scores during the activity, there was an increase in the participant's score from before the training (pre-test), which was 76.8. After participating in the training (post-test), the participant scored 97.2. Participants gain an increased understanding of making learning videos, as shown by the results of learning videos made individually by participants. The learning videos produced by the participants also met the criteria for good digital videos. Activity evaluation data show that this workshop is necessary and right on target for professional development, especially for SMP-SMA Muhammadiyah Gresik Teachers. The activity concept is also suitable for solving problems in junior high school level education. Thus, this training succeeded in increasing the competence of participants regarding the use of video and digital technology as one of the learning innovations.

It is still very rare for offices to process incoming and outgoing letters using virtual storage media, even though in the case of making the letter they already use a computer, but in terms of storing it is still done manually, it has not been stored computerized so that it will be very difficult, especially when searching for mail data that has been processed. pass. And not only in terms of storage that is carried out by the system with a computerized system, there are many other things that are very easy to do, including the approval process or letter signing which will be carried out in a structured manner, both incoming and outgoing letters. Abstract

Posyandu (Integrated Health Service Post) is a vital community health facility that provides health services for toddlers and pregnant women. However, the data management and reporting processes often face challenges such as limited access to information and data recording errors. Therefore, this research aims to develop a Web-based Information System for Melati Posyandu Service using the CodeIgniter Framework to enhance the efficiency and accuracy of data management and reporting. During the development phase, system requirements analysis and application architecture design were carried out for the web-based application. The implementation of the system utilized the CodeIgniter Framework as the foundation to produce a faster, more efficient, and reliable application. Proposed features include recording health data for toddlers and pregnant women, immunization schedules, weight measurements, and health reports. The results of this research demonstrate that the Web-based Information System for Melati Posyandu Service can improve efficiency in health data recording and reporting. Users, including Posyandu staff, midwives, and administrators, can easily access and manipulate data in real-time. Additionally, the system enhances the quality of service by providing more accurate and comprehensive health information for toddlers. In conclusion, the implementation of the Web-based Information System for Melati Posyandu Service using the CodeIgniter Framework offers significant benefits in data management and health service delivery at Melati Posyandu in Melandang Village. Suggestions for further development encompass maximizing system utilization, adding additional features, conducting regular maintenance, and ongoing evaluation based on user feedback. With these steps, it is expected that the system will more effectively contribute to improving the quality of health services at Posyandu and support comprehensive community health efforts

Abstract

This research aims to develop and implement a file encryption method based on the cryptography library in Windows 10 to enhance data security. The encryption uses the Advanced Encryption Standard (AES) algorithm with a 256-bit key in Galois/Counter Mode (GCM), known for its high security. The encryption key is generated using the Scrypt algorithm, ensuring resistance to brute-force attacks. The implementation includes file encryption and decryption with secure handling of salt, initialization vector (IV), and authentication tag. The research results show that this method is effective and efficient in protecting data from unauthorized access in the Windows 10 environment. In conclusion, the use of AES-256-GCM with the cryptography library provides a robust and reliable solution for data security, which is expected to serve as a reference in the development of information security technology

Abstract

The purpose of this study was to analyze the PAI learning model in improving the character of students by using a social interaction model. The social interaction learning model focuses on the actual environmental setting so that students understand the essence of togetherness and diversity in living together in society later. In carrying out life in society, the ability to socialize is needed because we are social beings and cannot be separated from what is called social interaction with the community by being wrapped in religious norms. The research method used is descriptive qualitative. With a qualitative approach, researchers can observe, understand, and analyze accurately and comprehensively related to PAI learning with social interaction learning models to improve the character of students. The findings are as follows: a) The implementation of the social interaction learning model in PAI lessons in the classroom emphasizes problem-solving in small groups which will later illustrate the group results in front of the class, b) The PAI learning model using social interaction is very influential in improving the character of participants education that is realized by better social interaction and prioritizing Islamic norms.
