Abstract

Abstract: Objective:to analyzevarious growth factor (GF) content in umbilical cord derived mesenchymal stem cell (UC-MSC) culture waste at harvest.Methods: We cultured UC- MSCs in complete medium, and collected the medium waste at harvests.  We noted the time of harvests, and measured the concentrations of EGF, VEGF, NGF and Pl GF in the various medium wastes using ELISA method. We grouped GF concentrations according to various culture lengthsat harvest, and calculated themean values and standard deviations of the various GF. We compareddescriptively the GF concentrations at various culture lengths at harvests to conclude which condition yielded the overall highest GF concentration.Results: EGF, VEGF and PlGF concentrations were highest at harvest day-3, whileNGF concentration was the highest at harvest-day-5, respectively. Therefore, overall highest concentration of most GF was at harvest day 3.Conclusion:UC-MSC culture waste at harvest day-3 containedEGF, VEGF, NGF and PlGF, which might be used in regenerative medicine.

Abstract

and its correlation to i mRNA expression in low-grade and high-grade EOC’s patients from Dr. Cipto Mangunkusumo General hospital, Indonesia. Methods: This study is cross-sectional with a total of 65 healthy subjects and 80 ovarian biopsies (15 ovarian cysts as expression calibrators, 36 low-grade EOC, and 29 high-grade EOC) were used in this study. The distribution of genotypes and alleles was analyzed using ARMS PCR. The mRNA expressions of RELA were determined by real-time polymerase chain reaction (qPCR) analysis.Results: There was no significant difference between genotype and allele distributions for RELA rs11820062 in normal and case group. RELA relative mRNA expression was significantly higher in low-grade and high-grade EOC compared to in ovarian cysts (p<0.01). RELA rs11820062 CC genotype correlated to higher RELA mRNA relative expression and the TT genotype of RELA rs11820062 correlated with lower RELA mRNA relative expression in low-grade and high-grade EOC.Conclusion: C allele in rs11820062 caused an increased expression of RELA mRNA, which individuals with CC genotype correlated with higher RELA expression in low-grade and high-grade EOC. In contrast, individuals with the T allele of RELA rs11820062 had a protective effect against EOC risk because the RELA TT genotype tended to have a lower RELA mRNA expression in EOC.Keywords: epithelial ovarian cancer, NF-kB, RELA, rs11820062.AbstrakTujuan: Mengetahui distribusi RELA rs11820062 dan korelasinya dengan ekspresi mRNA RELA pada pasien EOC low-grade dan high-grade di Rumah Sakit Dr. Cipto Mangunkusumo, Indonesia.Metode: Penelitian ini merupakan peneliatan potong lintang terhadap 65 sampel darah perempuan normal dan total 80 biopsi kanker ovarium dengan rincian: 15 kista ovarium sebagai kalibrator ekspresi, 36 EOC low-grade, dan 29 EOC high-grade. Distribusi genotipe dan alel dianalisis menggunakan ARMS PCR dan ekspresi mRNA RELA dikuantifikasi menggunakan teknik qPCR. Hasil: Tidak terdapat perbedaan distribusi genotipe dan alel antara kelompok normal dengan kasus EOC. Ekspresi relatif mRNA RELA meningkat secara signifikan pada kelompok EOC low-grade dan high-grade. Individu dengan genotipe RELA rs11820062 homozigot CC memiliki ekspresi mRNA yang lebih tinggi dibandingkan genotipe lain. Sebaliknya individu dengan genotipe TT memiliki korelasi dengan ekspresi mRNA RELA yang lebih rendah pada tipe low-grade dan high-grade EOC. Kesimpulan: Alel C pada RELA rs11820062 menyebabkan peningkatan ekspresi mRNA RELA pada pasien EO yang dilihat dari individu dengan genotipe CC cenderung memiliki ekspresi mRNA RELA yang lebih tinggi pada tipe EOC low-grade dan high-grade. Sebaliknya, individu dengan alel T RELA rs11820062 diduga memiliki efek protektif terhadap risiko EOC karena adanya korelasi antara genotipe TT dengan ekspresi mRNA RELA yang lebih rendah pada EOC.Kata kunci: kanker ovarium epitelial, NF-kB, RELA, rs11820062.

Abstract

BACKGROUND: Nephrotic syndrome (NS) is the most common glomerular disease in childhood. The proposed hypothesis for the pathogenesis of this disease has changed over time, from immune dysregulation theory and systemic circulating factors theory, to the growing recognition of podocytopathies’ role. The existance of podocytopathies is usually examined by using podocyte-derived microparticles (MPs), such as nephrin, podocin, and podocalyxin (PCX). Therefore in this study, the difference between nephrin, podocin, and PCX expressions in NS children and healthy children was investigated.METHODS: An observational cross-sectional study was conducted, involving 33 children with NS and 22 age-matched healthy children as controls. Urine samples were collected from each subject in the early morning, before being processed and incubated with antibodies to detect nephrin, podocin, and PCX. The processed samples were then analyzed with flow cytometer methods.RESULTS: NS subjects had significantly higher expression of all three urinary podocyte-derived MPs compared to the control subjetcs. Nephrin, podocin, and PCX showed good discrimination in NS subjects with the area under curve (AUC) of 0.895, 0.849, and 0.728, respectively.CONCLUSION: This study revealed the differential expression of podocyte proteins in NS subjects compared to healthy controls. This supports the role of podocytopathies in the pathogenesis of NS. Therefore, nephrin, podocin, and PCX might have potentials to be future non-invasive diagnostic tools in glomerular disease.KEYWORDS: nephrin, nephrotic syndrome, podocalyxin, podocin, podocyte, urinary microparticle

Abstract

BACKGROUND: Hypoxia inducible factor-1α (HIF-1α), nuclear factor erythroid 2-related factor 2 (Nrf-2), and vascular endothelial growth factor (VEGF), play a crucial role as neuroprotective factors. Currently, there is a lack of studies examining the biomolecular responses of the brain to intermittent hypoxia resulting from various pressures. This study was conducted to investigate the physiological responses, histopathological features, and cellular adaptive responses in the brains of rats that were intermittently exposed to hypobaric hypoxic conditions.METHODS: Thirty male Sprague Dawley rats were divided into six groups: a control group and five treatment groups exposed to hypobaric hypoxia. The treatment groups were placed in a hypobaric chamber simulating an altitude of 3,048 meters for 1 hour/day for 1, 7, 14, 21, and 28 days. After exposure, brain tissue was collected for histopathological analysis and protein quantification of HIF-1α, Nrf-2, cytoglobin (Cygb), neuroglobin (Ngb), VEGF, malondialdehyde (MDA), glutathione (GSH), superoxide dismutase (SOD), glutathione peroxidase (GPx), and catalase (CAT).RESULTS: In the brain, intermittent hypobaric hypoxia significantly increased HIF-1α expression (p=0.000) and its downstream proteins Cygb (p=0.000), and VEGF (p=0.001), with a peak at 14x IHH exposure compared to control. This was followed by a significant increase in Nrf-2 expression (p=0.000), SOD (p=0.000), Gpx (p=0.000), and CAT activity (p=0.000), indicating an adaptive antioxidant response. Conversely, MDA levels was decreased with prolonged exposure, suggesting reduced oxidative damage.CONCLUSION: IHH elevates HIF-1α, Nrf-2, and oxidative stress markers, triggering an adaptive antioxidant response in the rat’s brains.KEYWORDS: HIF-1α, intermittent hypobaric hypoxia, Nrf-2, oxidative stress

Abstract

Flight crews frequently experience intermittent hypobaric hypoxia (IHH), which may trigger oxidative stress and vascular remodeling in the pulmonary arteries. This study investigated the effects of IHH exposure on oxidative stress markers, tunica media thickness, and the expression of LR11 and HIF-1α proteins in rat pulmonary arteries using five groups of healthy male Sprague–Dawley rats (n = 5 per group). Twenty-five male Sprague–Dawley rats were divided into five groups: a control group and four treatment groups exposed to simulated altitudes of 25,000 ft (7,620 m) for 5 minutes per day over 1, 3, 7, or 14 days. Levels of HIF-1α and LR11 were measured using ELISA, oxidative stress markers such as γ-glutamyl-cysteinyl-glycine (GSH) and malondialdehyde (MDA) were analyzed spectrophotometrically, and tunica media thickness was assessed histologically. HIF-1α levels peaked after acute hypoxia exposure and declined with prolonged exposure, whereas LR11 levels progressively increased, peaking in the 14-day group. MDA levels, reflecting oxidative stress, were highest after 3 days of exposure, while GSH levels decreased in all hypoxia groups compared with the control. Histological analysis showed the greatest tunica media thickness in the 3-day exposure group, followed by gradual reductions. Although no statistically significant correlations were found between protein levels and tunica media thickness, similar directional trends suggest biologically relevant associations. Intermittent hypobaric hypoxia (IHH) induces dynamic changes in oxidative stress and vascular remodeling in rat pulmonary arteries. Acute hypoxia increases HIF-1α expression and oxidative stress, whereas repeated exposures appear to trigger compensatory adaptations, including LR11 upregulation and tunica media remodeling. These findings demonstrate measurable molecular and structural alterations underlying pulmonary vascular responses to repeated high-altitude exposure.

Abstract

Pendahuluan: Hipoksia hipobarik intermiten (IHH) dapat mengganggu homeostasis redoks dan menurunkan saturasi oksigen. Malondialdehid (MDA) mencerminkan peroksidasi lipid, sedangkan reduced glutathione (GSH) menggambarkan kapasitas antioksidan endogen. Penelitian ini mengevaluasi MDA dan GSH plasma serta saturasi oksigen perifer (SpO₂) setelah paparan IHH akut dan berulang pada tikus. Metode: Sebanyak 25 tikus jantan Sprague-Dawley dibagi acak ke dalam lima kelompok (n=5/kelompok): kontrol normobarik dan IHH 1×, 3×, 7×, atau 14×. IHH diberikan sekali sehari dalam ruang hipobarik setara 25.000 kaki (7.620 m) selama 5 menit. MDA plasma diukur dengan TBARS dan GSH dengan uji DTNB. SpO₂ dicatat segera setelah paparan menggunakan pulse oximetry untuk rodensia. Perbedaan antarkelompok dianalisis dengan ANOVA satu arah dan Tukey HSD. Hasil: GSH berbeda bermakna antarkelompok (F=30,022; p<0,001) dan lebih rendah pada seluruh kelompok IHH dibanding kontrol, dengan nilai terendah setelah 7 kali paparan. MDA juga berbeda bermakna (F=14,481; p<0,001), dengan nilai tertinggi setelah 3 kali paparan; IHH 3× lebih tinggi daripada kontrol (p<0,001) dan IHH 14× (p=0,007). SpO₂ lebih rendah pada seluruh kelompok IHH dibanding kontrol (p<0,001), dengan rerata terendah setelah 3 kali paparan (61,2±4,66%). Simpulan: IHH berkaitan dengan desaturasi oksigen dan perubahan biomarker stres oksidatif sirkulasi. Pola MDA dan GSH yang tidak linear konsisten dengan respons redoks yang bergantung waktu, tetapi belum membuktikan adaptasi fisiologis.
