Abstract

Bacteria are one of the microorganisms that have the potential to produce hydrolytic enzymes. This research was carried out to isolate bacteria from the soil of the Bukit Pinang Samarinda Final Disposal Site and select bacteria that produce cellulase, protease and lipase enzymes. Bacteria from landfill soil were isolated using the spread plate method and 10 single colony bacterial isolates were selected which were then coded TPA 1 – TPA 10. The results of the selection of bacteria producing hydrolase enzymes showed that 9 isolates were capable of producing protease enzymes, 6 isolates were capable of producing cellulase enzymes and 4 isolates were capable of producing cellulase enzymes. The isolate was able to produce the lipase enzyme.

Abstract

Pacing plants (Costus sp.) Potentially as medicinal plants, but there is no research on the endophytic bacteria they contain. This research was conducted to filter endophytic bacteria from Pacing stems (Costus sp.) And to find out the secondary metabolites contained by each endophytic bacteria. Endophytic bacteria obtained from Pacing Stems (Costus sp.) Totaled 17 colonies. Crude extracts from all bacterial colonies containing alkaloids, 15 crude extracts containing saponins. Steroids are only contained by 3 types of bacterial extracts and triterpenoids are only contained by 2 types of bacterial extract.

Abstract

Uji Fitokimia dan aktivitas antibakteri ekstrak metanol daun salam (Syzygium polyanthum) telah dilakukan. Penelitian ini bertujuan untuk mengetahui metabolit sekunder yang terkandung pada ekstrak metanol daun salam dan konsentrasi hambat minimum (KHM) ekstrak tersebut. Uji aktivitas antibakteri dilakukan dengan metode difusi menggunakan kertas cakram. Ekstrak metanol daun salam (Syzygium polyanthum) metanol daun salam mengandung steroid, flavonoid dan fenolik. Konsentrasi hambat minimum ekstrak metanol daun salam (Syzygium polyanthum) terhadap Staphylococcus aureus sebesar 0-1% dan terhadap Salmonella typhi sebasar 1-2%.

Abstract

Isolasi bakteri penghasil lipase dari sumber air panas Long Kali dilakukan dengan menyeleksi koloni bakteri pada media Nutrient Agar (NA) yang mengandung Rhodamin B dan minyak zaitun. Hasil isolasi menunjukkan 5 dari 14 isolat bakteri mampu menghasilkan lipase. lima isolat tersebut diuji aktivitas lipase dengan metode titrimetri, isolat dengan kode 14 memiliki aktivitas tertinggi dengan aktivitas 2,20 Unit/mL.

Abstract

Endophyte bacteria isolate from the leaves of Macaranga hullettii King ex Hook.f. has potential to producer lipase. In the study was conducted to filter lipase producing endophyte bacteria from Macaranga hullettii King ex Hook.f. and determine the optimum production time. The results of screening endophyte bacteria showed 6 isolates of endophyte bacteria that had the potential to produce lipase and 1 isolate was chosen which is number 5 to determine the optimum production time. The optimum production time of lipase from Macaranga hullettii King ex Hook.f. leaves for 72 hours with activity that is 2.92 U/mL.

Abstract

This research aims to isolate the crude extract amylase of sprouted seeds jackfruit (Artocarpus heterophyllus Lam) and determine the specific activity. Determination of total protein concentration and amylase activity of enzyme crude extract on seed germination jackfruit (Artocarpus heterophyllus Lam). Determination of total protein concentration using the method of Bradford, whereas the determination of amylase activity was performed using dinitrosalisilat acid method (DNS). The results showed a total protein concentration of the enzyme crude extract of sprout seeds jackfruit (Artocarpus heterophyllus Lam) is 319,47 ug / mL, amylase activity of 13,96 U / mL and specific activity is 1748,05 U / mg.

Abstract

Endophytic bacteria are bacteria that live in plant tissues. These bacteria produce extracellular enzymes, one of which is the lipase. This research was conducted to isolate the endophytic bacteria of Ginseng Jawa (Talinum triangulare (Jacq.) Willd) leaves which produced lipase and lipase screening from these bacteria. Isolation was carried out on nutrient agar (NA) media and screening of endophytic bacteria were carried out on nutrient agar (NA) media containing olive oil and Rhodamin B. Isolation results obtained by 5 single colonies and one colony of positive endophytic bacteria that produces lipase which characterized by the presence of orange-colored luminescence on the surface of the bacterial colony

Abstract

Endophytic bacteria is a bacteria that live in plant tissues without disrupt the plant's life cycle. This research aims to isolate endophytic bacteria from okra fruit (Abelmoschus esculentus L. Moench). Before isolation, okra fruit was sterilized by the outer surface with aquadest, 5.25% NaClO and 95% ethanol. The okra fruit sample is then cut and the inside of the okra fruit is cultured in a nutrient agar (NA). The results of this test indicate that there are endophytic bacteria that grow characterized by the presence of turbid areas around the sample. The results of isolation of endophytic bacteria using the method of stocking cup were obtained by 28 isolates of endophytic bacteria.

Abstract

Raw starch degrading amylase (RSDA) is an enzyme that can degrade raw starch into glucose without going through the gelatinization process. This research was conducted to isolate RSDA producing bacteria originating from the Karang Mumus River in Samarinda City. Bacterial isolation was carried out using the spread plate method. Screening for RSDA producing bacteria using nutrient media containing raw starch. The results of bacterial isolation obtained 14 single colonies of bacteria. A total of 5 bacterial colonies out of 14 bacteria were able to produce RSDA. Then the RSDA enzyme from the 5 bacteria was tested for amylase activity quantitatively using the dinitrosalicylic acid (DNS) method. Then, of the 5 bacteria, 1 bacterium with the colony code KM 5 had the highest activity, namely 0,332 U/mL.

Abstract

Penelitian ini dilakukan untuk menseleksi bakteri halofilik penghasil lipase dan pengaruh ion logam terhadap ekstrak kasar lipase. Seleksi bakteri penghasil lipase ditunjukkan dengan adanya pendar jingga di sekitar koloni di bawah sinar UV pada media NA yang mengandung minyak zaitun dan Rhodamine-B. Isolat yang memiliki aktivitas lipase tertinggi kemudian diproduksi lipasenya dan di uji aktivitasnya. Hasil penelitian diperoleh waktu produksi lipase pada 72 jam. Penambahan CaCl2 meningkatkan aktivitas lipase dengan aktivitas relatif sebesar 115% sedangkan, KCl, BaCl2 dan MgCl2 menurunkan aktivitas lipase dengan aktivitas relatif berturut-turut sebesar 78,82, 96,5 dan 80% terhadap aktivitas lipase tanpa penambahan ion logam. Penambahan FeCl3 menyebabkan ekstrak kasar lipase terdenaturasi.
