Abstract

The test of amylase inhibition activity of the leaves of Melicope lunu-ankenda and bark of Melicope glabra (BI.) T.G. Hartley extracts were done. The objective of this study was to determine in-vitro α-amylase inhibition activity against two samples by DNS method. The result showed that two samples of Melicope lunu-ankenda leaves and bark of Melicope glabra (BI.) TG Hartley displayed the enzyme inhibitory effect with percent inhibitions of 48.71% and 46.36%. Two sample extracts can inhibit amylase. It can be suggested that about sample were potential as antidiabetic agents.

Abstract

Endophytic bacteria found in plant tissues can produce enzymes, one of which is lipase. The purpose of this study was to screening endophytic bacteria from the leaves of Okra (Abelmoschus esculentus L. Moench) which produced lipases. The method used in this study is screening with media nutrient agar (NA) containing Rhodamine B and olive oil. The results showed that 4 of bacterial isolates (D1, D2, D3 and D4) from 14 endophytic bacterial isolates had lipase activity. The crude lipase extract was quantitatively tested for lipase activity by the titrimetry method. The highest lipase activity was shown by isolates with a D1 code of 1.50 U/mL.

Abstract

The research about in vitro trakeospasmolytic response of methanol extract of Macaranga lamellata Whitmore leaves was carried out. This research was conducted to determine the spasmolytic effect of the methanol extract of Macaranga lamellata Whitmore leaves on changes in separate organ of the guinea pig ring tracheal. The methods used include the bioassay isolated organ with two groups of treatment, control group and extract group. Methanol extract was given cumulatively on the organ bath. Positive control is used aminofilin and negative control is used DMSO solvent. Phytochemical test results showed that total methanol extract contained alkaloids, polyphenol, steroids, flavonoids dan saponins while phytochemical test results showed on ethyl acetate fraction contained alkaloids, polyphenol, steroids, quinons dan flavonoids. The Percentage change in tracheal smooth muscle as a spasmolytic response by using methanol extract with cumulative concentration of 0.3, 1 dan 3 mg/mL respectively -24.925%; -29.865% and -36.795% . This research is shown methanol extract of Macaranga lamellata Whitmore leaves have antiasthma effect based on the tracheospasmolytic activity. Keywords: Antiasthma, Bioassay isolated organ, Macaranga lamellata Whitmore, Marmot, Trakeopasmolytic.

Abstract

ABSTRACT Phytochemical test and antibacterial activity of methanol extract of Okra (Abelmoschus esculentus (L.) Moench) fruit have been carried out. Extraction using maceration method. The methanol extract of Okra fruit contains phenolic secondary metabolites, alkaloids and steroids. Antibacterial activity opposes the agar diffusion method against the bacteria Salmonella typhi and Steprococcus mutans. The Minimum Inhibitory Concentration Value of Okra fruit methanol extract against Salmonella typhi bacteria at a concentration of 0.625-1.25% and against Streptococcus mutans bacteria 1.25% -2.5%. Okra fruit methanol extract has antibacterial activity with wide variations. Keywords : Abelmoschus esculentus (L.) Moench, antibacterial activity, secondary metabolites, Minimum Inhibitory Concentration (MIC).

Abstract

Lipase is an enzyme that can hydrolyze triglycerol into glycerol and free fatty acids. The aim of this research is to obtain lipase-producing bacteria and determine the potential of lipase from water bacteria in the water of the Benanga Lempake Dam, North Samarinda for biodetergent applications through stability test of lipase activity against several commercial detergents. Screening of lipase-producing bacteria was carried out on nutrient agar media containing olive oil and Rhodamine B. The results showed that seven isolates were capable of producing lipase. Based on the stability test of lipase on detergent, it can be seen that the activity of lipase decreased by more than 70% at the addition of 1% detergent.

Abstract

Raw starch-degrading amylase (RSDA) is a hydrolytic enzyme that can degrade raw starch into glucose. This study was conducted to obtain raw starch-degrading amylase-producing bacteria from the soil of the Bukit Pinang Landfill, Samarinda City. Isolation of bacteria using the spread plate method. Screening of RSDA-producing bacteria using nutrient media to contain raw starch. Isolation of bacteria obtained 10 single colonies of bacteria. The isolated bacteria were selected for their ability to produce RSDA. All single colonies of bacteria could produce RSDA and their activity was tested quantitatively using the 3,5 Dinitrosalicylic Acid (DNS) method. Bacteria with the TPA 1 colony code had the highest activity of 3.837 U/mL.
