Abstract

The high cost of media for microbial culture encourages researchers to create alternative media using local raw materials at low prices. One of the most important media compositions for bacterial growth is protein. This content can be obtained from nuts, one of which is almonds (Prunus dulcis). The purpose of this study   is   to  find  natural   media  that    has   the   potential    for   the    growth of microorganisms at a low price. The  research method  used is an experimental method to determine the growth of Escherichia coli and Staphylococcus aureus bacteria on natural media of almonds. The results showed the growth of Escherichia coli and Staphylococcus aureus bacteria in this alternative media of almonds. Based on the results of the study, it can be concluded that  almond  nut  media  can  be  applied  as  an  alternative  medium in   laboratory research, especially in the field of_microbiology. It is suggested that further research should be carried out on natural media which are sources of carbohydrates and proteins as alternative growth media for bacteria.

Abstract

Background & Objective This study aims to determine the egg-laying behaviour of Aedes aegypti in water contaminated with animal waste and soil, count eggs caught in cages, observe and count the number of healthy and damaged eggs, and compare the number of eggs from each water medium. Method This study is descriptive, the data were analyzed using the SPSS application, presented in tabular form, and explained in a narrative manner. Result The results of observations of egg-laying behaviour indicate that water contaminated with animal waste and soil can be a breeding ground for the Aedes aegypti. Conclusion Based on the results of observations of the behaviour of Aedes aegypti in water media contaminated with animal faeces and soil, it shows a distinctive behaviour, that water contaminated with animal faeces and soil is favoured by Aedes aegypti as a place to lay eggs and breed. The total number of eggs caught was 1,716, with the number of healthy eggs ( 1,219) and the number of damaged eggs (497). Differences in the content contained in the water media affected the laying of Aedes aegypti eggs but did not affect the number of healthy or damaged eggs.

Abstract

Background & Objective Tuberculosis is an infectious disease caused by acid-fast bacteria, namely Mycobacterium tuberculosis, which mainly attacks the lungs. Pulmonary tuberculosis is a global health problem with the second deadliest case after HIV/AIDS infection, and ranks third with the most TB cases after India and China. The main source of transmission is patients with active TB pulmonary tuberculosis who can transmit to people around them. This study aims to determine whether pulmonary TB transmission occurs in people who are in household contact with patients with acid-fast bacteria pulmonary TB active. Method This study is descriptive, using the Ziehl neelsen examination method, with a total sampling technique, conducted on 34 people who are in household contact with pulmonary TB patients in Ciwangi Village, Balubur District, Limbangan. Result Based on the examination results in people who are in contact with people with TB, the results are negative, the presence of the bacteria that cause TB. Conclusion The results of Pulmonary Tuberculosis screening conducted on 34 people from 13 heads of families, it can be concluded that the results of TB microscopy with the Ziehl neelsen technique were negative.

Abstract

Background & Objective Thalassemia is a blood disorder caused by genetic factors that cause the globin chain in hemoglobin to not function normally. It is the most common hereditary disease in the world and in Ciamis Regency in 2021 there were 191 patients. Thalassemia prevention is carried out by screening to identify thalassemia carriers. Hematological examination in the form of erythrocyte index (MCV, MCH, MCHC) and mentzer index has been known to determine thalassemia screening.  The purpose of this study was to determine the description of erythrocyte index and mentzer index in thalassemia patient's father. Method The method used in this study is descriptive method. The population in this study were members of POPTI (Parents Association of Thalassemia Patients) Ciamis Regency. The sample in this study were 30 samples. Result Based on the results of the study obtained the results of erythrocyte index (MCV, MCH) low (MCHC) normal and Mentzer index obtained the results of 20 samples (66%) suspected as thalassemia carrier, 5 samples (17%) suspected iron deficiency anemia and according to the reference value there were 5 samples (17%). Conclusion The results of a study conducted on 30 fathers of thalassemia patients showed that 83% of fathers of thalassemia patients had MCV and MCH values below the normal threshold and 73% had normal MCHC values. Furthermore, from the results of the Mentzer index examination, it is known that 66% of the fathers of thalassemia patients are suspected of having minor thalassemia, 17% are suspected of having iron deficiency anemia disorders and 17% have a normal Mentzer index.

Abstract

Background & Objective: Total cholesterol is the total amount of HDL, LDL, and VLDL cholesterol in each deciliter of blood. Risk factors that can affect total cholesterol levels are age, gender, sleep patterns, genetics, physical activity, and body mass index. This study aims to determine the description of total cholesterol levels in productive age in the Imbanagara Health Center Working Area. Method: This research method is descriptive research with a purposive sampling technique and involves 31 respondents the instrument used is a photometer. This research was conducted in the Imbanagara Health Center working area from May to June 2023. Result: The results showed that the total cholesterol levels of 31 respondents, were 25 people (81%) with normal total cholesterol levels, 5 people (16%) with high threshold total cholesterol levels, and 1 other person (3%) with high total cholesterol levels. Conclusion: It can be concluded that of the 31 respondents in the Imbanagara Health Center work area, the average has normal Total Cholesterol results.

Abstract

Gram staining is a staining method used to distinguish bacterial species into two major groups, namely Gram-positive and Gram-negative, Crystal violet is the primary (main) dye that will colour the target microorganisms. Staining in bacteria can be replaced using alternative substances with low cost, readily available and harmless, one of which uses substances that have anthocyanin content, namely purple sweet potato. The purpose of this study was to determine the results of using purple sweet potato as an alternative substance to replace crystal violet in staining Streptococcus sp and Escherichia coli bacteria. The method used is experimental. Based on the results of research that has been done on the use of purple sweet potato juice as an alternative substance to crystal violet in gram staining of Streptococcus sp and Escherichia coli bacteria, it shows that purple sweet potato material can be used at pH 6.5 and 5.7 while at pH 5.5 and 5 the bacteria are not perfectly coloured. So it can be concluded that purple sweet potato juice can be used as an alternative substance to Crystal violet in gram staining.

Abstract

Pleural effusion is a clinical condition that may indicate serious underlying diseases, including infection, malignancy, and systemic disorders. Cytological examination of pleural fluid plays an important role in determining the possible etiology of pleural effusion and requires proper fixation to preserve cellular morphology and staining quality. This study aimed to evaluate the cytological characteristics and preparation quality of pleural fluid specimens fixed in 96% alcohol for 15 minutes and 30 minutes using the Papanicolaou staining method. This quasi-experimental study included 16 pleural fluid samples obtained from routine laboratory examinations. Each sample was processed into four cytological smears, consisting of two duplicate smears fixed in 96% alcohol for 15 minutes and two duplicate smears fixed in 96% alcohol for 30 minutes. A total of 64 smears were evaluated, with 32 smears in each fixation group. All smears were stained using the Papanicolaou method and assessed microscopically based on background appearance, cellular morphology, nuclear characteristics, and overall staining quality. Of the 32 smears fixed for 15 minutes, 20 smears (62,5%) were classified as poor quality and 12 smears (37,5%) as good quality. In contrast, among the 32 smears fixed for 30 minutes, 3 smears (9,4%) were classified as poor quality and 29 smears (90,6%) as good quality. These findings indicate that fixation using 96% alcohol for 30 minutes produces better cytological preparation quality than fixation for 15 minutes. Therefore, a 30-minute fixation period is recommended to support optimal cytological evaluation and diagnostic accuracy.

Abstract

Gastritis is an inflammation of the gastric mucosa that may be caused by various factors, including irregular dietary habits, nonsteroidal anti-inflammatory drug (NSAID) use, psychological stress, alcohol consumption, and Helicobacter pylori infection, which is recognized as a major cause of chronic gastritis. In primary healthcare settings, identification of the underlying etiology of gastritis is often limited by the availability of diagnostic laboratory facilities. Therefore, serological detection of anti-Helicobacter pylori immunoglobulin G (IgG) antibodies may serve as an alternative diagnostic approach. This study aimed to determine the serological profile of anti-Helicobacter pylori IgG antibodies among patients with chronic gastritis attending the Baregbeg Community Health Center. A descriptive study was conducted involving 24 outpatients diagnosed with chronic gastritis who met the inclusion criteria. Participants were recruited using a total sampling technique. Serum samples were collected and analyzed using an immunochromatographic assay at the Baregbeg Community Health Center Laboratory between April and June 2025. All 24 participants (100%) tested negative for anti-Helicobacter pylori IgG antibodies. No serological evidence of Helicobacter pylori infection was detected among the study participants. In conclusion, no serological evidence of Helicobacter pylori infection was detected among chronic gastritis patients included in this study, with an observed Helicobacter pylori seropositivity rate of 0%.

Abstract

Pediculosis capitis merupakan penyakit akibat infestasi Pediculus humanus capitis yang masih menjadi masalah kesehatan pada anak, terutama santri di pondok pesantren. Tingginya kepadatan hunian, interaksi antarsantri, perilaku personal hygiene yang kurang baik, serta penggunaan barang pribadi secara bersama meningkatkan risiko penularan. Selain menyebabkan pruritus, infestasi kutu kepala dapat menimbulkan dermatitis, infeksi sekunder, gangguan tidur, hingga anemia sehingga diperlukan upaya pengendalian yang tidak hanya berfokus pada pencegahan, tetapi juga pengobatan yang efektif dan aman. Penelitian ini bertujuan menganalisis hubungan faktor risiko dengan kejadian infestasi Pediculus humanus capitis pada santri di Kabupaten Ciamis serta mengevaluasi aktivitas pedikulosida kombinasi minyak lidah buaya (Aloe vera) dan minyak daun kemangi (Ocimum sanctum). Penelitian menggunakan desain analitik dengan pendekatan cross-sectional untuk menganalisis faktor risiko melalui kuesioner dan pemeriksaan langsung, serta penelitian eksperimental menggunakan lima kelompok perlakuan, yaitu T1 (1:3), T2 (1:1), T3 (3:1), T4 (kontrol positif), dan T5 (kontrol negatif) dengan lima kali pengulangan. Analisis hubungan faktor risiko menggunakan uji Chi-square, sedangkan data waktu mortalitas dianalisis menggunakan One-Way ANOVA. Hasil penelitian menunjukkan prevalensi infestasi sebesar 54,3%. Faktor yang berhubungan secara signifikan dengan infestasi meliputi jenis kelamin, panjang rambut, frekuensi mencuci rambut, penggunaan aksesoris kepala bersama, dan penggunaan alas tidur bersama. Kombinasi minyak lidah buaya dan minyak daun kemangi menunjukkan aktivitas pedikulosida, dengan perlakuan T1 menghasilkan waktu mortalitas tercepat, yaitu 115,0 ± 13,77 detik. Disimpulkan bahwa pengendalian Pediculosis capitis pada santri memerlukan pendekatan terpadu melalui pengendalian faktor risiko dan pemanfaatan kombinasi minyak lidah buaya serta minyak daun kemangi sebagai alternatif pedikulosida alami
